1.Screening affinity peptides of transforming growth factor beta type Ⅱreceptor from the phage display peptide library

Objective: To screen high-affinity short peptides of the transforming growth factor beta typeⅡ(TGF-? Ⅱ) receptor from the phage display 12-mer peptide library.Methods: The recombinant solube human TGF-? Ⅱ receptor was used as the target to screen its affinity short peptides from the phage display 12-mer peptide library.The positive phage clones were obtained by 3 rounds of biopanning,followed by single stranded DNA extraction and sequence analysis.Results: Four nucleotide sequences were obtained from 10 positive phage clones with high-affinity TGF-? Ⅱ receptor.Neither conservative nor homologous sequence was found by sequence analysis compared with the transforming growth factor beta 1.Conclusion: Phage short peptides have been screened from the phage display library,which helps pave the ground for further researches on affinity short peptides and their role in inhibiting hepatic fibrosis.
2.Comparative analysis of surgical treatment of secondary hyperparathyroidism with uremia

Objective:Comparative analysis of total parathyroidectomy (total parathyroidectomy, TPTX) and total parathyroidectomy with autoransplantation (total parathyroidectomy with autoransplantation, TPTX+ AT) for the treatment of uremia secondary hyperparathyroidism Clinical efficacy.Methods:A total of 82 cases of SHPT treated with TPTX or TPTX + AT in First Affiliated Hospital of Chengdu Medical College from June 2015 to July 2019 were collected According to the operation methods, 82 cases were devided into TPTX group and TPTX+ AT group.There were 34 patients in the TPTX group, 17 males and 17 females, with an average age of (45.97±8.96) years, and 48 patients in the TPTX+ AT group, 21 males and 27 females, with an average age of (49.12±7.30) years. SPSS 20.0 software was used for statistical analysis. The measurement data conforming to normal distribution were represented by ( Mean± SD), and the comparison between groups was performed by t test. The data not conforming to normal distribution were represented by quaternary M( P25, P75), and the comparison between groups was performed by non-parametric test. Enumeration data were expressed as cases (%), and comparison was performed by χ2 test. The postoperative complications, postoperative efficacy and recurrence rate were compared between the two procedures. Results:Both TPTX and TPTX+ AT had a low incidence of complications after SHPT, and both of them could effectively relieve symptoms such as bone pain, skin itching and subcutaneous soft tissue calcification.After 1 year of follow-up, both of the two surgical methods can effectively reduce blood calcium (TPTX group: t= 14.522, P<0.05; TPTX+ AT group: t=13.704, P<0.05), blood phosphorus (TPTX group: t=35.022, P<0.05; TPTX+ AT group: t=42.303, P<0.05) and iPTH(TPTX group: t= 20.166, P<0.05; TPTX+ AT group: t= 23.976, P<0.05), but the reduction of iPTH in TPTX group was more effective than that in TPTX+ AT group ( t=-3.416, P<0.05). Conclusion:Both TPTX and TPTX+ AT can effectively treat SHPT, and have high safety and can be used as an alternative for SHPT treatment.

Objective To investigate the effects of sea voyage on cellular immune function and nutrition status of naval servicemen. Methods A total of 173 male naval servicemen were included in the present study. The percentage of T, B and NK cell subpopulations from peripheral blood were assessed by flow cytometry. CD3+, CD3+CD4+, CD3+CD8+ and CD4+/CD8+ were employed as the markers of T cell subpopulation, CD3-CD19+ served as the marker of B cell subpopulation, and CD3-CD56+ served as the marker of NK cell subpopulation. The serum total protein (TP), albumin (ALB), glucose (Glu), total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HLD-C), low-density lipoprotein cholesterol (LDL-C), apolipoprotein A1 (ApoA1), apolipoprotein A2 (ApoA2), apolipoprotein B (ApoB), apolipoprotein C2 (ApoC2), apolipoprotein C3 (ApoC3), apolipoprotein E (ApoE), lipoprotein a (LPa), Fe2+, total iron-binding capacity (TIBC), Hb, K+, Na+, Cl-, Ca2+ and Mg2+ levels were determined by 7600DDP. Objective To evaluate HRCT features of otosclerosis.Methods HRCT findings of 61 ears with the diagnosis of otosclerosis based on clinical diagnostic criteria in 34 patients were evaluated retrospectively.Results Hypodense regions in the bony otic capsule were found on HRCT in 55 ears and no abnormality was identified on HRCT in 6 ears.In 55 ears with abnormal HRCT findings, HRCT demonstrated the hypodense region of bony otic capsule anterior to oval window alone in 6 ears, the hypodense region anterior to oval window associated with thicked stapedial footplate and pericochlear hypodensity in 6 ears, the hypodense region anterior to oval window associated with thicked stapedial footplate and hypodensity posterior to oval window in 11 ears, the hypodense region anterior to oval window associated with thicked stapedial footplate in 20 ears, the hypodense region anterior to oval window associated with pericochlear hypodensity in 10 ears, and pericochlear hypodensity in the bony otic capsule alone in 2 ears.Conclusion HRCT can detect abnormalities in the bony otic capsule and the stapedial footplate,contributing to confirming diagnosis of otosclerosis.
5.A meta-analysis on the effects of dietary approaches to stop hypertension (DASH) diet on blood pressure of children and adolescents

Objective To estimate the effects of the dietary approaches to stop hypertension (DASH) diet on blood pressure (BP) of children and adolescents by using meta-analysis. Methods The articles of the randomized controlled clinical trials (RCT) on the DASH diet were collected and selected according to inclusion and exclusion criteria. Meta-analysis was performed to calculate the weighted mean difference (WMD) and 95% confidence interval (CI) to assess the blood pressure change associated with DASH diet in children and adolescents. Results Eight RCT papers involving experimental group for 838 and the control group for 1 222 were included. All the papers had high methodological quality. According to comparative analysis between the lowest tertiles of DASH Score and highest tertiles of DASH Score, the DASH-style pattern led to lower systolic BP(SBP) (WMD=-2.29 mmHg, 95%CI:-3.20--1.38) (1 mmHg=0.133 kPa)and diastolic BP(DBP) (WMD=-2.18 mmHg, 95%CI:-3.00--1.35) levels. Comparing with pre-DASH diet, the DASH diet was more effective in lowing SBP (WMD=-5.83 mmHg, 95%CI:-9.55--2.11)and DBP(WMD=-2.78 mmHg, 95%CI:-4.74--0.83). Comparing with the routine care (RC), the DASH diet was proved more effective than RC in lowing SBP(WMD=-4.98 mmHg, 95%CI:-7.37--2.59) and DBP(WMD=-1.47 mmHg, 95%CI:-2.53--0.41) in children and adolescents. The DASH diet was more effective in lowing SBP(WMD=-3.25 mmHg, 95%CI:-5.46--1.05)than DBP in children and adolescents. Conclusion This research suggested that DASH diet had a positive clinical efficacy on blood pressure in children and adolescents, and caused a greater decrease in SBP than in DBP.
6.The role of interleukin 6 and interleukin 6 receptor in gliomas

Purpose:To discuss the role of interleukin 6 (IL 6) and IL 6 receptor (IL 6R) in gliomas. Methods:We detected IL 6 and IL 6R gene in 30 human glioma tissue specimens and adult normal astrocytes. A complementary DNA copy of total RNA was synthesized and amplified with specific primers using the reverse transcription polymerase chain reaction (RT PCR) method. Results:IL 6 gene was positive of in 24 (80.0%) samples of gliomas, while IL 6R gene was positive of in 26 (86.7%) cases. Co expression of IL 6 and IL 6R was identified in 22 (73.3%) specimens of gliomas. On the other hand,the expression of IL 6 was weakly positive in adult normal astrocyte, but IL 6R was negative.Conclusions:Our results indicate that IL 6/IL 6R autocrine or paracrine loop may exist in glioma, which could enhance the glioma cell proliferation.
7.Arsenic Trioxide Induces Apoptotic Cell Death through Mitochondrial Pathway in Human Leukemia HL-60 Cells

Objective:To investigate the potential pro-apoptotic activity of arsenic trioxide (ATO) in human leukemia HL-60 cells,as well as the potential mechanism with focus on mitochondrial pathway.Methods:After treatment with different concentrations of ATO (1 μg/mL,5 μg/mL or 10 μg/mL) for 24 h,apoptotic cell death was detected by flow cytometry,oxidative stress was determined by measuring MDA and GSH levels,the expression of apoptotic factors was detected by western blot,and mitochondrial membrane potential (MMP) was determined by immunofluorescence staining.Results:ATO at the concentrations of 5 μg/mL or 10 μg/mL induces apoptotic cell death and increases oxidative stress in human leukemia HL-60 cells.ATO significantly increases the expression of pro-apoptotic factors (Bax and Caspase-3),whereas decreases the expression of anti-apoptotic factor Bcl-2.Compared with the control group,ATO treatment significantly decreases the MMP level in HL-60 ceils.Conclusions:Arsenic trioxide induces apoptotic cell death through mitochondrial pathway in human leukemia HL-60 cells.

ObjectiveTo observe the effect of immunoloregulation induced by tripterygium glycosides on experimental IgA nephropathy in rats.MethodsRat model of IgA nephropathy was established with bovine serum albumin and staphylococcal enterotoxins compound immunization.The urine erythrocyte,urine protein in 24 h,renal function,erythrocyte C3b receptor rosette rate and kidney constitution pathology alteration among the normal control group,model group and tripterygium glycosides group were compared.ResultsCompared with the model group,urine erythrocyte,urine protein,serum Cr and BUN reduced( P<0.05),erythrocyte C3b receptor rosette rate was higher(P<0.05),IgA depositing on glomerular mesangial region was less and renal function improved in rats of the tripterygium glycosides group.ConclusionTripterygium glycosides have better therapeutic effect and adjusting action for immune disturbance.
9.Inhibitory effect of dexamethasone on TGF-beta1 expression of rabbit ciliary pigment epithelia cultured in vitro.

In order to elucidate the effect of dexamethasone on the expression of transforming growth factor-beta1 (TGF-beta1) in ciliary pigment epithelial (CPE) cells cultured in vitro, rabbit CPE cells were cultured in vitro, treated with DMEM medium containing 0, 1 x 10(-8), 5 x 10(-8), 10 x 10(-8) and 50 x 10(-8) mol/L dexamethasone respectively for 5 days. The TGF-beta1 expression was detected by immunohistochemistry Supervision methods and analyzed semi-quantitatively by HMIAS-2000 image system. As opposed to in vivo, rabbit CPE cells expressed TGF-beta1 under cultured circumstance in vitro. The gray scales of the positive yellow staining in the groups of 1 x 10(-8), 5 x 10(-8), 10 x 10(-8) and 50 x 10(-8) mol/L dexamethasone were 136.57 +/- 4.43, 140.20 +/- 6.10, 142.98 +/- 2.99, 146.80 +/- 1.68 and 150.05 +/- 1.94 respectively. When the concentrations of dexamethasone were equal to or higher than 5 x 10(-8) mol/L and, the expression of TGF-beta1 was inhibited. 10(-7) mol/L dexamethasone showed a significant inhibition. It was suggested that CPE cells possess the potential ability of synthesizing and expressing TGF-beta1. The inhibition of TGF-beta1 expression by dexamethasone may be beneficial to the treatment of proliferative vitroretinopathy, also exert some influence on the secretion of aqueous humor and ciliary inflammation.
10.CT and MRI study of ophthalmoplegia

Objective To analyze the diseases responsible for ophthalmoplegia and determine the optimal technique identifying the lesions. Methods CT and MR imaging findings of 1376 patients with ophthalmoplegia were analyzed. The total positive rate and ratio of the diseases causing ophthalmoplegia were calculated. The efficiency of various methods and sequences was compared in the evaluation of cavernous sinus inflammation and other lesions. Multi-paired samples Friedman test was used to compare five kinds of images from different methods and sequences, and Wilcoxon test was used to compare between every two kinds of images. Results The total positive rate was 91.9% (1264/1376). In 50 patients who underwent both CT and MRI, the positive rate of MRI (92. 0% ,46/50) was higher than that of CT (48.0% ,24/50)(Z = -4. 8, P < 0. 01). There were 552 cases (43.7%) of cavernous sinus lesions, 518 cases (41.0%)of extraocular muscle diseases, 108 cases (8. 5%) of cranio-orbital communicating lesions and 86 patients (6. 8%) of other lesions. The five kinds of images from various methods and sequences had significant difference in the detection of 283 cavernous sinus inflammation (χ2 = 1047. 1, P < 0. 01) cases. Transverse T1WI with thin slice thickness[(2. 71 ± 0. 69)scores]was better than that with thick slice thickness [(1.67 ± 0. 64) scores], contrast transverse T1 WI with thin slice thickness[(3.92 ± 0. 27) scores]was better than transverse T2WI with thick slice thickness, transverse T1WI and coronal T1 WI with thin slice thickness[(3. 10 ± 0. 39) scores]. Coronal T1 WI with thin slice thickness was better than transverse T1 WI with thin slice thickness and transverse T2WI, and the contrast coronal T1WI with thin slice thickness [(3.95 ± 0. 22) scores]was better than transverse T, WI with thin slice thickness, transverse T2 WI and coronal T1WI (P <0. 01 separately). The positive rate of enhanced MRI (100% ,39/39) was higher than that of nonenhanced MRI (82. 1% ,32/39) (Z = - 2. 1, P < 0. 05). Conclusion CT and MRI can show the lesions responsible for ophthalmoplegia. MRI is the best examination method in displaying these lesions.
