1.Experience of vagina reconstruction with axial pattern skin flap

Objective To summarize the experience of vagina reconstruction with six kinds of axial pattern skin flaps Methods Six kinds of axial pattern skin flaps have been used for vagina reconstruction in 126 cases,including of hypogastric skin flap,thoraco umblical skin flap,labia minora skin flap,scrotal skin flap,penis skin flap and pudendal femoral skin flap Results All flaps were survived,but one case of necrosis occurred in hypogastric skin flap Conclusion The pudendal femoral skin flap is the first choice of flap for vagina reconstruction The advantages of this method are (1) the reconstructed vagina has good sensation and flexibility;(2)the linear scars of donor site are hidden and inconspicuous;(3)the flap is close to recipient site the procedure is simple
2.Effects of vitamines on human preadipocyte proliferation and differentiation

ObjectiveTo study the effects of vitamins on human pr eadipocyte proliferation and differentiation. MethodsVit amins (A, B, C, D, E and K families) were added to the culture media of human p readipocytes, then the proliferation of cells, the expression of GPDH and lipid droplet accumulation in the cytoplasm were recorded. ResultsVitamin A inhibited human preadipocyte proliferation and differentiation. Vi tamin C enhanced differentiation and proliferation. Vitamin B 6 stimulated diff erentiation. vitamin D 3 inhibited differentiation. Vitamin E had a strong inhi bitory effect on human preadipocyte differentiation. Vitamin K 1 had an unexpec tedly great stimulatory effect on human preadipocyte differentiation. Vitamin K 3 inhibited both human preadipocyte proliferation and differentiation. Other v itamins tested showed no noticeable effects. ConclusionT hese data are advisory to our balanced daily intake of vitamins.
3.THE ANTI-OTOTOXIC EFFECT OF IMINOETHYL-LYSINE ON GUINEA PIG'S COCHLEA DAMAGED BY GENTAMI- CIN

The object of this study was to investigate the anti-ototoxic effect of iminoethyl-lysine on cochlea of guinea pig damaged by gen-tamicin. The experiment consisted of three groups: normal control group, experimental treatment control group and experimental treatment group. Guinea pigs of the latter two groups were subjected to hypodermic injection of gentamicin, at the same time guinea pigs of the experimental treatment group were treated with intraperitoneal injection of iminoethyl-lysine. while guinea pigs of the experimental control group were treated with the same volume of saline. Guinea pigs of the normal control group were treated with hypodermic and intraperitoneal injection of the same volume of saline as that of the other two groups. The expression of iNOS was examined in the cochleas of all groups by immunohisto-chemistry. ABR hearing threshold shift were examined, and the damage of hair cells of cochleas of all groups was investigated by scanning electronic microscopy. The results showed that iNOS expression was negative in cochleas of the normal control group, and positive in that of other two groups. The expression of iNOS in the experimental control group was much more stronger than that of the experimental treatment group. The ABR threshold shift of the experimental treatment group was much more lower than that of the experimental control group. The damage of cochlea of the experimental control group was much more severe than that of the experimental treatment group. These results suggest that the expression of iNOS is positive in gentamicin-damaged cochleas. Iminoethyl-lysine has significant anti-ototoxic effect on the cochlea damaged by gentamicin, suggesting nitric oxide plays a very important part in the process of the cochlea injury by gentamicin.
4.Anti-toxical effect of iminoethyl-lysine on guinea pig renal damaged by gentamicin

AIM: To investigate the anti toxic effect of iminoethyl lysine (Imi), an inhibitor of inducible nitric oxide synthase (iNOS), on the guinea pig renal injury with gentamicin (Gen). METHODS: The experiment was consisted of four groups: normal animal group, gentamicin model group (Gen group), Gen+Imi group and Imi group. Guinea pigs of both Gen group and Gen+Imi group were given (sc) Gen 100 mg?kg -1 ?d -1 for 10 d, and the guinea pigs of Gen+Imi group were treated with Imi intraperitoneally. Gen group were treated with the same volume of saline. Guinea pigs of normal animal group were treated with subcutaneous and venous injection of the same volume of saline. Guinea pigs of Imi group were treated with the same dose of Imi. The renal function of guinea pigs of all groups was examined before and after experiment. Renals of all groups were embedded with paraffin, sectioned, stained with HE, and examined with the light microscope. iNOS was examined in the kedney of all groups with immunohistochemistry. RESULTS: The expression of iNOS was negative in kidney of normal animal group and Imi group, and positive in both Gen group and Gen+Imi group. The activity of iNOS of Gen group was significantly stronger than that of Gen+Imi group. The damage of renal of Gen group was much more severely than that of Gen+Imi group. CONCLUSION: The expression of iNOS is positive in the renal damaged with Gen. Imi can significantly inhibit the activity of iNOS, and has also the significant anti toxical effect on the renals damaged with Gen. It suggests that nitric oxide takes part in the process of pathology of renals damaged by gentamicin.
5.Effects of Lidocaine on Respiratory Failure and the Airway Peak Pressure in Patients with Severe Asthma: A Clinical Research

OBJECTIVE: To observe the effect of lidocaine on respiratory failure and the airway peak pressure in patients with severe asthma. METHODS: The severe bronchial asthma patients treated with mechanical ventilation were randomly divided into treatment group and control group. The change in airway peak pressure, man-machine counteraction, and the correcting time of respiratory failure of the two groups were recorded. RESULTS: The average airway peak pressure was(41.18?10.66) cmH2O in the control group vs.(29.23?9.07) cmH2O in the treatment group; the incidence of man-machine counteraction was 100% for the control group vs. only 40% for the treatment group; the correcting time of respiratory failure was(6.42?1.73) h for the control group vs.(3.31?1.08) h for the treatment group. The femora! arteries in 38 rabbits were transected and repaired with autogenous venous grafts with different diameters. The measurements of blood flow, patency rate and thrombosis rate, and scanning electron microscopic examination were made over a period of 14 days. The result showed that a venous graftl with a diameter 20 per cent less than that of the recipient artery was most satisfactory.
7.Simple Method for Preparing Tissue Chip and Its Application in Pathological Study

【Objective】To establish a simple method for preparing tissue chip.【Methods】One hundred and thirty mammary cancer and intestinal cancer specimens were fixed by formaldehyde(40?g/L) and embedded by wax,and then were made into routine tissue slices and stained by haematoxylin and eosin(HE).Wax slices with tissue chip were prepared by fixing a tissue slice(0.1 cm in diameter)into the hole(0.2 cm in diameter and 2~4mm in depth) of a blank wax slice and were sliced into 4?m-thick slices.The tissue chips were mounted on the slide and stained by immunohistochemistry after treatment.【Results】A slide held 45 tissue chips and 130 specimens could be all detected only on 3 slides,which saved time and the amount of reagent.The immunohistochemical results showed that the positive location was accurate and the background was clear.【Conclusion】This method for preparing tissue chip is simple and effective,and can save the amount of reagent.It is easy for the comparative and separated detection of specimens.
8.Function of the regenerative axon after peripheral nerve end-to-side neurorrhaphy in rats

Objective: To compare regeneration of motor fibers with that of sensory fibers by end to side neurorrhaphy and compare the effect of end to side neurorrhaphy with that of end to end neurorrhaphy. Methods: 20 SD rats were randomly divided into two groups: group A and group B. In group A, the right peroneal nerve was sectioned and the distal end was sutured laterally to the tibial nerve; in group B, the right peroneal nerve was sectioned and sutured with end to end neurorrhaphy. All left sides of two groups were used as control. Retrograde transportation of HRP was observed after 28 weeks. Results: The labelled neurons were also found in the anterior horn of the spinal cord and the spinal ganglia. Conclusion: The regenerative nerve fibers had motor fibers as well as sensory fibers by end to side neurorrhaphy. The effect of end to side neurorrhaphy is not as good as that of end to end neurorrhaphy. [
9.Expression and significance of the cell cycle regulators in laryngeal carcinogenesis detected by flow cytometry.

OBJECTIVE:
To investigate the expression and clinicopathological significance of the cell cycle regulators cyclin E, cyclin D1, p21, p16 in laryngeal carcinogenesis tissus.
METHOD:
The expression of cell cycle regulators were detected by flow cytometry method in 23 cases of polyps of vocal cord, 69 cases of laryngeal precancerous change and 33 cases of laryngeal squamous cell carcinoma (LSCC), which tissue was paraffin embedded, sliced, dewaxed, and prepared into the cell suspension, then fluorescently labeled by cyclin E, cyclin D1, p21 and p16.
RESULT:
In polyps of vocal cord, laryngeal precancerous change and LSCC, The positive expression rate of cyclin E and cyclin D1 were respectively 13.04%, 20.29D, 42.420 and 26.09%, 43.48% and 93.94%. The positive expression rate of p16 and p21 were respectively 61.90%, 40.98%, 14.28% and 47.62%, 23.81%, 26.23%. Those showed the positive expression rate of cyclin D1, cyclin E gradually decreased from vocal cord polyps, laryngeal precancerous change to LSCC, (P < 0.05, P < 0.01), while the positive expression rate of p21 and p16 gradually decreased (P < 0.01).
CONCLUSION
The abnormal expression of cell cycle regulatory factors is the molecular events of laryngeal carcinoma. High expression of positive regulatory factors cyclin D1 and cyclin E, and low expression of negative regulatory factors p16 and p21, which showed the imbalance of multiple positive and negative regulatory factors related with cell cycle play an important role in the occurrence of laryngeal cancer.

10.Effects of microfilaments on the expression of collagenases and tissue inhibitor of metalloproteinases-1 in fibroblasts.

OBJECTIVETo investigate the roles of microfilaments in the expression of collagenase and TIMP-1 in normal and hypertrophic scar.
METHODSCell culture and Northern blot hybridization methods were used in this study.
RESULTSAfter the disruption of microfilaments with cytochalasin B, mRNA contents of collagenase and TIMP-1 both increased significantly in normal and hypertrophic scar fibroblasts.
CONCLUSIONThe result suggest that the microfilament cytoskeleton may regulate the synthesis and degradation of ECM, which may be a mechanism of gene regulation during wound healing.
