1.Microsurgical anatomy of optic canal in Chinese adults.

Objective To investigate the anatomic characteristics of optic canal in chinese adults. Methods Twenty chinese cadaveric speciments fixed in formalin of chinese patients without intracranial and intraorbital disease. We measured the width and height of the intracranial opening and the intraorbital opening of the optic canal of all cases. The length of roof, floor, medial and lateral wall of the optic canal, and the width, the height of the middle protion were also measured after unroofing the canal with high-speed drilling. The degree of pneumatization of ethmoidal and sphenoidal sinuses was recorded. Results The intracranial opening of the optic canal was a horizontal oval shape. The width of it was (5.05±0.42) mm and the height was (4.61±0.27) mm. The length of optic roof, optic floor, medial wall and lateral wall of the optic canal was (9.67±1.51) mm, (5.41±0.84) mm, (9.58±0.72) mm and (9.26±0.66) mm, respectively. The intraorbital opening of the optic canal was a vertical oval shape, the height and width of it was (5.30±0.64) mm and (5.62±0.83) mm respectively. Conclusions The wall of the optic canal is thinner and there is a narrow portion in the middle of it. Depending on the degree of the pneumatization of sphenoidal and ethmoidal sinus, three types were divided into: 1. reversal U shape, 2. C shape, 3. D shape. The dura fold of the optic canal is long and its edge is sharp.
2.STUDY ON THE NEURONAL APOPTOSIS IN THE HIPPOCAMPAL CA1 REGION AFTER EXPERIMENTAL SUBARACHNOID HEMORRHAGE

Objective To investigate the mechanism of neuronal apoptosis in hippocampal CA1 region and the death signaling transduction pathway initiated by CD95(Fas) molecules during cerebral vasospasm(CVS) after subarachnoid hemorrhage(SAH). Methods SAH model was developed by injecting arterial blood twice the citernamagna.The morphological changes of the neurons were studied by light microscope and laser scanning confocal microscope.RT-PCR was performed for detecting the expression changes of Fas,FasLmRNA. Results Neurons in the hippocampal CA1 region after SAH showed typical apoptotic features under light microscope and confocal microscope.The expressions of Fas,FasLmRNA in hippocampus increased in SAH group,reached the highest level 3 days after SAH and still were significantly higher 7 days after SAH compared with normal group and then decreased to normal 14 days after SAH.Conclusion The death signaling transduction pathway initiated by CD95 molecules may play important roles in hippocampus neuronal apoptosis following SAH.
4.Effect of early life stress on locomotion and corticosterone in peripheral blood in adult rats

Objective To investigate the effect of early life stress on the locomotion and concentration of corticosterone in peripheral blood in adult rats.Methods Maternal deprivation and isolation were used in the model of early life stress. Chronic stress was used in adult rats.Open field test was used for the measurement of locomotion activity. ELISA was used to measure the concentration of corticosterone in peripheral blood.Results The total activity time of locomotion[(296.96±6.63)s, (291.06±8.40)s;P<0.01] and the concentration of corticosterone [(15.51±2.41)nmol/L,(11.27±2.49)nmol/L;P<0.05]were increased in adult rats by maternal deprivation. The total activity distance [(23231.29±2030.35)mm,(14042.81.06±2875.34)mm;P<0.05] and time[(296.39±3.48)s,(259.18±4.71)s;P<0.01]were increased after chronic stress which was used in adult rats in which maternal deprivation were used before. The concentration of corticosterone[(20.60±1.83)nmol/L,(14.49±1.37)nmol/L;P<0.01]was also increased in the same condition.Conclusions The concentration of corticosterone and locomotion activity of adult rats can be increased by early life stress. The reaction of HPA axis for chronic stress was improved by early life stress.
5.Progress in the study of biological characteristics of mesenchymal stem cells derived from human umbilical cord

Recent research found that human umbilical cord is rich in mesenchymal stem cells. These cells have the potencies of expressing many surface markers of mesenchymal stem cell and of differentiating into a variety of cells of three germ layers, synthesizing and secreting a set of trophic factors and other cytokines, and supporting the expansion and function of other cells hematopoietic stem cells and other cells. In addition, immunogenicity of these cells is realatively low.
6.Development of a test phantom used for MRI’s quality control

Objective: Design a test phantom with a full function, simple structure and high accuracy which is used for MRI’s Quality Control. Methods: Through a intensive study in imaging principle of MRI and parameters which effect the image quality ,first come up with the phantom’s performance data, and then determine the technical solutions including the choosing material, processing technology, the machining precision, at last develop a trial set of MRI test phantom using the method of numerical control machining. Make a comparative experiment between this phantom and the similar imported one to analyze the consistency of the results by the two phantoms. Results: The result of all the technical parameters of this phantom is consistent with the result of the imported one. There are some extra test components which the imported phantom doesn’t contain. Some performance data like the low contrast resolution are superior to the imported phantom. Conclusion:This MRI test phantom can satisfy the requirement of the daily quality control work in hospital.
7.Construction of Nedd4 knockout BMDM cell line by CRISPR/Cas9 technology

Objective To construct Nedd4 knockout bone marrow-derived macrophages(BMDM) cell line by CRISPR/Cas9 technology and to provide an effective tool for studying the function and mechanism of Nedd4 in macrophage.Methods First,three high-grade sgRNAs targeting Nedd4 gene exons were screened using the online tool before synthesized sgRNAs were inserted into the PX330 plasmid respectively.Secondly,the recombinant plasmids were transferred into BMDM cells and monoclonal cells were obtained by limiting dilution method.The protein levels of NEDD4 in monoclonal cells were detected by Western blotting.Finally,the DNA sequence of the monoclonal cells was confirmed by sequence analysis.Results One Nedd4 knockout BMDM cell line was obtained.The sequencing result showed that the Nedd4 gene had 16bp deletion mutation in this cell line.Conclusion The Nedd4 knockout BMDM macrophage cell line constructed by CRISPR/Cas9 technology will be a useful tool for studying the function and mechanism of Nedd4 in BMDM cells.
8.The perioperative use of steroids including dexamethasone in hyperthyroidism

ObjectiveTo explore the effects of steroids for hyperthyroidism management during the perioperative period. Methods198 cases were analyzed in terms of the perioperative use of steroids such as dexamethasone for hyperthyroidism. ResultsFT3 and FT4 decreased gradually to the nomal level after use of dexamethasone. 15 cases had hypokalemic limb paralysis, 2 cases had hyperthyroidism crises, 9 cases had transient numbness of mouth, lip and limb, and the rest resumed normal. ConclusionsThe use of steroids such as dexamethasone for hyperthyroidism can shorten the preoperative preparation time, improve the success rate, and reduce postoperative complications.
9.Expression of aquaporin 4 during development of experimential presyrinx state in rabbits

Objective:To investigate the expression of AQP4 during the development of presyrinx state of experimental syringomyelia in rabbits.Methods:The experimental syringomyelia models of rabbits were established by intra-cisternal injection of Kaolin.The expression of AQP4,AQP4mRNA and the water content of upper cervical spinal cord were measured with immunohistochemistry,Western blot,RT-PCR and dry-wet measurement on days 1,3,7,14,and 21 after operation,respectively.Results:Compared with animals of control group,the water content increased in those of Kaolin group from the 1st day(68.35%?0.70%),reached its peak on the 7th day(72.92%?0.86%),lasted to the 14th day(72.58%?0.55%),and then began to drop on the 21st day(70.03%?0.77%),while AQP-4 immunoreactive expression decreased on the 3st day[integral optical density(IOD)320.5?44.2],reached its minimum on the 7th day(IOD 258.7?26.5),lasted to the 14th day,and recovered partially on the 21th day approximately(IOD 321.5?46.1).RT-PCR found the decreasing of AQP4 mRNA coincided well with that of AQP4 immunoreactive expression in presyrinx state.The linear regression analysis indicated that expression of AQP4 and its mRNA in cervical cord had a negtive correlation with the change of spinal water content(r=-0.769,P
