1.THE MORPHOLOGICAL AND CELLULAR DYNAMIC CHANGES IN ARSENIC TRIOXIDE-TREATED RAT LIVER CANCER

Objective To study the efficiency of the treatment of experimental hepatocellular carcinoma in rats with arsenic trioxide and to elucidate the possible mechanism.Methods Wistar rats were fed with diethylnitrosamine (DEN) to induce HCC, then treated with As2O3. The histological changes in liver tissue were observed under microscope, and the cellular dynamic parameters were studied by flow cytometry. Results Treatment with As2O3 caused HCC cells death via both apoptotic and non-apoptotic mechanisms when the dose was high (5 mg/kg), the necrosis was seldom and apoptosis was common when the dose was appropriate (1 mg/kg). Proliferation index (PI) decreased sharply in high-dose (5 mg/kg) group (P<0.01), but not in other two (1 mg/kg, 0.2 mg/jg) groups (P>0.05). However, S phase fraction (SPF) decreased dramatically in all three groups (P<0.01). Although apoptosis of HCC cells was common in all three groups, it reached the top only when the dose (1 mg/kg) was appropriate (P<0.001), and it was obviously accompanied with accumulation of cells in G2/M (G2/M restriction). Conclusion These date demonstrate that arsenic trioxide induces apoptosis of rat HCC cells, and it is closely associated with G2/M restriction when apoptosis reaches the top. The data also suggest that arsenic trioxide can inhibit cell proliferation, which is dose-dependent and time-dependent. The fact that continuous intermittent i.p. injection of arsenic trioxide can also be effective may afford a novel way to use the drug more safely.

Objective To study the influence of RNA interference Caspase-3 gene on apoptosis of transformed human embryonic kidney 293 cells induced by cadmium in vitro. Methods Transformed human embryonic kidney 293 cell were treated with siRNA for 36 h. The treated cells were incubated with 40 ?mol/L CdCl2 for 12-24 h and the cells viability were measured with methyl thiazolyl tetrazolium(MTT) assay. In addition,the expression of caspase-3 gene in 293 cells was detected by the methods of reverse transcription polymerase chain reaction (RT-PCR) and Western-blot analysis,and the occurrence of apoptosis was determined by flowcytometry with Annexin-V and propidium iodide (PI) double labeling method. 3.The experimental study of the effects of icariin on increasing Smad4 mRNA level in MC3T3-E1 cell in vitro

Objective To explore the effect of icariin on stimulating Smad4 mRNA level in MC3T3-E1 cells. Methods MC3T3-E1 cells were treated by 0 ng/ml, 10 ng/ml, 20 ng/ml, 40 ng/ml icariin respectively. MTT method and population diploid time were applied to observe the cell proliferation. The cell ALP level was measured by atomic absorptiometry and the type Ⅰ collagen was observed by immunohisto-chemistry. RT-PCR was used to find the Smad4 mRNA level in the MC3T3-E1 cells. Results Under the inversion phase contrast microscopy, MC3T3-E1 cells were oligodendrocytes-shaped rich with cell organs. The cells density in 10 ng/ml, 20 ng/ml were higher than those in 0 ng/ml,40 ng/ml. 4.Complications of interventional therapy in maxillofacial region

砄bjective: To study the complications of interventional therapy in maxillofacial region. Methods : Retrospective analysis was conducted in 73 cases(96 times) treated with interventional therapy in maxillofacial region. Intraarterial angiography, drug infusion and embolization were conducted in 71 cases of maxillofacial tumor and 2 cases of injury by Seldinger method. Results: Complications with clinical significance occurred were observed in 6 cases (6.2%): allergic reaction to angiographic agent in 2 cases, necrosis of skin in 1, local pain in 1, facial paralysis in 1 and death because of brain damage in 1. 5 cases were cured. Conclusion: The most serious complication in maxillofacial interventional therapy is nerve injury, which may result in death . Certain measures should taken to prevent or reduce the reaction.
5.Effects of 4-aminopyridine on the gastrointestinal function of murine

Aim To investigate the effects of 4-aminopyridine (4-AP),K+ channel blocker, on the gastrointestinal function of murine. Methods Charcoal suspension was used to detect the effects of 4-AP on gastrointestinal motility of mice in vivo, contraction of isolated fundus longitudinal strips and duodenum of rats were studied in vitro,gastric acid secretion was measured by Gastric Secretion Test. Results 4-AP(5 mg?kg -1,ig)inhibited the gastrointestinal motility of mice. 4-AP(5 mmol?L -1) increased the maximum contractive force and minimum relaxation force, decreased the amplitude and frequency of the isolated duodenum peristaltic contraction. 4-AP(2.5 mg?kg -1,ip) significantly enhanced gastric acid secretion of rats. Conclusion 4-AP inhibited gastrointestinal motility and enhanced gastric acid secretion in murine.
6.Value of video electroencephalogram in the identification of neonatal epileptic and non-epileptic seizures

Objective To evaluate the clinical effectiveness of video electroencephalogram (VEEG) in neonatal seizure. Methods Seizures attacks of 64 newborns with gestational ages 28-44 weeks were monitored by VEEG. Results Incidence of abnormal VEEG was 32. 8%. Among 64 cases found in the VEEG, 13 cases had epileptic discharge in the ictal VEEGs. Three cases showed epileptic discharge in the interictal VEEGs with normal background rhythm. Two cases presented epileptic discharge in the interictal VEEGs with abnormal background rhythm. Three cases had abnormal background rhythm only. Discharge frequency during sleeping was more quick than that during awake-ness, mostly in the quiet sleep phase. Most frequently ictal epileptic discharges were a focal onset (16/ 18) , and 13 cases happened in the temple lobe. No epileptic discharge in the ictal and interictal VEEGs were tested in 46 cases attacked with seizures, which were diagnosed as non-epileptic seizures. Conclusions VEEG plays an important role in the diagnosis and differential diagnosis in epilepsy and non-epileptic seizure in newborns, and is valuable to perform early to improve prognosis.
8.Clinical study on immune neutropenia

Objective To explore the diagnosis and of immune neutopenia.Methods Clinical data of 150 patients with immune neutropenia were collected and analyzed.Results Of 150 patients with immune neutropenia,35 cases went male(23.3%),115 cases were female(76.7%),and male to female ratio was 1 ∶ 3.3.Based on the difference produced way of granulocytes associated antibody,all patients could be divided into alloimmune neonatal neutropenia 9 cases (6.0%),drug-induced immune neutropenia 28 cases (18.7 %),primary autoimmune neutropenia 22 cases(14.6%),and secondary autoimmune neutropenia 91 cases (60.7%).Secondary autoimmune neutropenia included 39 cases of systemic lupus erythematous,21 cases of rheumatoid arthritis,2 cases of immune hemolytic anemia accompany with neutropenia,6 cases of immune thrombocytopenia accompany with neutropenia,3 cases of autoimmune hepatitis,16 cases of autoimmune thyroiditis,and 4 cases of Sjogren's syndrome.Conclusion The majority of primary immune neutropenia are female,which is usually secondary to autoimmune diseases.The diagnosis of primary immune neutropenia is very complicated,which requires a long period of clinical observation and follow-up.The majority of granulocytes associated antibody is IgG in the autoimmune neutropenia patients.Yet sometimes two or three kinds of antibodies were increased as well.
9.Literature Analysis of Moxifloxacin-induced Adverse Drugs Reactions in 3 445 Cases

OBJECTIVE:To investigate the characteristics and regularity of moxifloxacin-induced adverse drug reaction(ADR) and provide reference for the rational clinical use of drugs. METHODS:Retrieved from CNKI (2005-2014) about the moxifloxa-cin-induced ADR,3 445 cases and the related ADR information were statistically analyzed. RESULTS:Moxifloxacin-induced ADR had a certain relationship with gender,and the male had a high incidence,especially the elderly patients. ADR could appear within 10 min;the clinic features were allergic reaction,the nervous system and digestive system at most. CONCLUSIONS:Great impor-tance should be attached to ADR monitoring and rational use of drug to reduce or avoid the occurrences of moxifloxacin-induced ADR.
10.Effect of polysaccharide from Porphyra on spleen cell function in immunosuppressive mice

Objective To investigate the effects of polysaccharide from Porphyra on spleen cell function in immunosuppressive mice. Methods The polysaccharide at dosages 0.025, 0.050 and 0.150g?kg~ -1 was injected intraperitoneally for 7 days in the immunosuppressive mice induced by cyclophosphamide. On day 8, the splenic lymphocytic proliferation and activity of TNF were determined. Results The splenic lymphocytic proliferation and cytotoxicity of tumor necrosis factor (TNF) of the group treated with 0.150g?kg~ -1 of porphyra polysaccharide were higher than those of model group. Conclusion Polysaccharide from Porphyra could promote splenic lymphocytic proliferation and raise the level of TNF.
