1.Effect of miR-218 on proliferation and cell cycle of gastric cancer cells

Objective To investigate the effect of miR-218 on cell proliferation and cell cycle in the gastric cancers (GC).Methods SGC-7901 and MGC-803 cells were transfected with miR-218 mimics.Meanwhile,SGC-7901 and MGC-803 cells were transfected with control mimics (Scramble mimics) as negative control.Quantitative reverse transcription polymerase chain reaction (qRT-PCR) was used to detect the miR-218 expression in these transfected cells.The effect of miR-218 overexpression on cell proliferation was evaluated with direct cell counting and MTS [3-(4,5-dimethylthiazol-2-yl)-5 (3-carboxymethonyphenol)-2-(4-sulfophenyl)-2H-tetrazolium inner salt] assay.Moreover,the effect of miR-218 overexpression on cell cycle was examined by fluorescence activated cell sorting (FACS).Results miR-218 expression was remarkably induced in miR-218-transfected cells,miR-218 overexpression led to a significant decrease in cell proliferation rate compared to control cells (P < 0.05).miR-218 overexpression induced GC G1 arrest.Conclusions miR-218 can suppress GC cell proliferation and block cell cycle progression.It maybe play an important role in tumorigenesis and development of GC.
2.Expression level and clinical features of STAT3 in breast cancer

Objective To detect the expression of STAT3 in breast cancer and adjacent paracancerous tissues ,then analyze the correlationship between its expression level and some other clinical features of breast cancer,and to evaluate its significance in prognosis of patients with breast cancer. Methods 130 breast cancer patients were enrolled in the Jiangmen Central Hospital ,and 61 cases of adjacent normal tissues. The expression of STAT3 was detected by IHC. We compared the expression of STAT3 between the two groups and analyzed the relationship between STAT3 and other clinical features. Results The expression of STAT3 in breast cancer group was much higher than the adjacent paracancerous tissues. STAT3 expression showed obvious correlationship with clinical stage ,tumor size ,lymph node metastasis ,the expression of ER and PR ,but no statistical correla-tion with age ,histological grade and Her-2 expression. Survival analysis showed that patients with high STAT3 expression level had a significantly poor prognosis. The Cox regression analysis showed that PR and STAT3 expres-sion were independent prognostic factors for patients with breast cancer. Conclusion These results may implicate that STAT3 will promote the development of breast cancer and higher expression of STAT3 indicates the poor survival rate.
3.Measuring the displacement and deformation under the internal fixation with compressing steel plate for femoral fractures by digital speckle method

Objective Measuring the displacement and deformation of the fifth screw under different tensile force before or after adultal internal fixation with compressing steel plate for femoral fractuer by digital speckle correlation method.And analysis of the displacement and deformation after reducing the number of screws.Aimed at providing a theoretical basis for clinical femoral internal fixation with compressing steel plate about the number of screws. Methods Taking the antisepsis femoral specimen of grown-ups, embedding and fixing with tooth powder after removing the soft tissue, producing a fixed model in the 1/2 femur.Fixing steel plate below the lateral femur subperiosteal with five screws through the fourth floor of cortical bone on both sides of the fractures, samples were divided into 10 states, group A-J were measured by orders, under the rally of 400 N,500N , measuring the displayment of the fifth screw with CSS-44100-e-universal test machine for loading rally experiment ,and automatic data processing. Results For analysis of variance, the mean displacement measurements of the fifth screw under rally of 400N,500N, B-J group and A group have significant differences(P<0.05). The mean deformation measurements of the fifth screw under rally of 400N,500N, group A and group F-J group have significant differences(P<0.05);group A and B-E group have no significant differences.Conclusion In a certain range, the fracture displacement and the numuber of the screws show negative correlation. The screws near the fracture line for femoral compression plate internal fixation of femoral fractuer is easily broken because of the too large deformation.
4.Impact of anthracene addition on microbial community structure in soil microcosms from contaminated and uncontaminated sites.

OBJECTIVEThis paper aims to investigate the impact of anthracene addition on microbial community in agricultural soil irrigated with tap water or reclaimed wastewater.
METHODSThe changes of microbial community were characterized by terminal restriction fragment length polymorphism in combination with 16S rRNA gene clone library analysis.
RESULTSA significant change in microbial community composition was observed during the biodegradation of anthracene, with dominantly enriched members from the genus Methylophilus.
CONCLUSIONThis work might be useful for developing techniques for the isolation of novel putative PAH degrader.

5.EGFR gene mutation analysis among 185 cases of NSCLC patients in south of China

ObjectiveTo investigate the characteristics of EGFR gene mutations among NSCLC patients in south of China and analyze the correlation between mutations and clinical features.Methods Specimens of lung cancer tissues were collected from 185 NSCIC patients in our hospital.DNA was extracted from specimens.Exon 18,19,20 and 21 of EGFR gene were amplified by FQ-PCR to be bi-directional sequenced.ResultsEGFR gene mutations in 62 (33.5%) of 185 NSCLC patients was identified in carcinoma tissues,of which,2cases,41cases,5 cases and 14 cases respectively located at exon 18,exon 19,exon 20 and exon 21.The mutation of Del L747 → P752 (P753S) ( proportion 8.1% ),Del E746 → A750 ( proportion 45.1% ) at exon 19 and L858R ( proportion 22.6% ) at exon 21 were the predominant mutation in 16 kinds of mutations.Four cases of mutation at exon 19 got the different results in bi-directional sequencing.The silent mutation 2361G→A at exon 20 was observed (28.1% ).The mutation rate in women was significantly higher than men (46.2% vs 24.3%,x2 =9.670,P =0.002).Non-smokers had significantly higher mutation rate than smokers (41.4% vs 17.1%,x2 =7.380,P =0.007) ; Adenocarcinoma patients had significantly higher mutation rate than squamous cell carcinoma (38.3% vs 6.3%,x2 =6.426,P =0.011).Clinical stage Ⅲ patients had significantly lower mutation ratethan patients with stage Ⅱ orⅣ ( 10.8% vs 53.8%,x2 =8.026,P =0.003 ;10.8% vs 41.3%,x2 =9.518,P =0.002).No statistically significance correlation was found between the mutation ratio and age.ConclusionsEGFR gene mutation has a close relationship with females,non-smokers and adenocarcinoma.Most mutations occur in exon 19 and 20 among patients in south of China.
6.Application of the Simodont dental trainer in preclinical manual dexterity training

Objective:To explore the application of the Simodont dental trainer in preclinical manual dexterity training for dental students of different grades, and to discuss its effect by quantitatively evaluating their practice results.Methods:The evaluation was conducted among 118 students in the Department of Stomatology, Shantou University Medical College from Batch 2015 to Batch 2018. Each student had five different manual dexterity modules for training. Each module had three levels of difficulty and was required to be completed in 30 minutes. The assessment index included Target, Leeway Bottom and Sides, Container Bottom and Sides, which was provided by the Simodont dental trainer. The operating time (in seconds), the displacement of the dental hand pieces and the dental mirrors (in meters) were simultaneously recorded. SPSS 25.0 was used for statistical analysis.Results:For the index Target, Leeway Bottom and Sides, Container Bottom and Sides, the results showed that there was a statistical difference between students who passed pre-clinical training and who did not take the professional course ( P < 0.001). For the index Target, differences were showed among students from different grades in all modules ( P < 0.001). Conclusion:The circle module, channel module, hollow-circle module and cross-module in the Simodont dental trainer have sensitivity to discriminate the manual dexterity of different levels of dental students. The further assessment of the discrimination of the manual dexterity is required for assume-block module. The Simodont dental trainer can quantitatively measure the manual dexterity of dental students, which is important for the quantitative evaluation of dental preclinical education.

8.Ethyl Lithospermate Reduces Lipopolysaccharide-Induced Inflammation through Inhibiting NF-κB and STAT3 Pathways in RAW 264.7 Cells and Zebrafish.

OBJECTIVE:
To explore the anti-inflammatory effects of ethyl lithospermate in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine-derived macrophages and zebrafish, and its underlying mechanisms.
METHODS:
3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazoliumbromide (MTT) assays were performed to investigate the toxicity of ethyl lithospermate at different concentrations (12.5-100 µ mol/L) in RAW 264.7 cells. The cells were stimulated with LPS (100 ng/mL) for 12 h to establish an inflammation model in vitro, the production of pro-inflammatory cytokines interleukin (IL)-6 and tumor necrosis factor α (TNF-α) were assessed by enzyme linked immunosorbent assay (ELISA). Western blot was used to ascertain the protein expressions of signal transducer and activator of transcription 3 (STAT3), nuclear factor kappa B (NF-κB) p65, phospho-STAT3 (p-STAT3, Tyr705), inhibitor of NF-κB (IκB) α, and phospho-I κB α (p-IκB α, Ser32), and confocal imaging was used to identify the nuclear translocation of NF-κB p65 and p-STAT3 (Tyr705). Additionally, the yolk sacs of zebrafish (3 days post fertilization) were injected with 2 nL LPS (0.5 mg/mL) to induce an inflammation model in vivo. Survival analysis, hematoxylin-eosin (HE) staining, observation of neutrophil migration, and quantitative real-time polymerase chain reaction (qRT-PCR) were used to further study the anti-inflammatory effects of ethyl lithospermate and its probable mechanisms in vivo.
RESULTS:
The non-toxic concentrations of ethyl lithospermate have been found to range from 12.5 to 100 µ mol/L. Ethyl lithospermate inhibited the release of IL-6 and TNF-α(P<0.05 or P<0.01), decreased IκBα degradation and phosphorylation (P<0.05) as well as the nuclear translocation of NF-κB p65 and p-STAT3 (Tyr705) in LPS-induced RAW 264.7 cells (P<0.01). Ethyl lithospermate also decreased inflammatory cells infiltration and neutrophil migration while increasing the survival rate of LPS-stimulated zebrafish (P<0.05 or P<0.01). In addition, ethyl lithospermate also inhibited the mRNA expression levels of of IL-6, TNF-α, IκBα, STAT3, and NF-κB in LPS-stimulated zebrafish (P<0.01).
CONCLUSION
Ethyl lithospermate exerts anti-Inflammatory effected by inhibiting the NF-κB and STAT3 signal pathways in RAW 264.7 macrophages and zebrafish.
