1.Observation of the effect of combination of TCM and Western treatment of allergic purpura Medicine

Objective To explore clinical treatment efficacy of allergic purpura by combination of TCM and Western medicine.Methods 138 patients with allergic purpura were chosen in this study.Divided 138 cases into 3 groups,which were group A,B and C.Patients in group A received Western medicine treatment of vitamin C,cimetidine,anti-infection treatment.Patients in group B received the same treatment as group A with combination of Xijiaodihuang Decoction.Patients in group C received the same treatment as group B with combination of acupuncture treatment.Total efficacy,clinical symptoms disappear interval and recurrence rate in following 6 months were observed.Results Compared with group A,total efficacy in group B increased significantly(P<0.05),which was more significant in group C(x2 =33.662,P<0.05);Erythra disappear interval in group B and C decreased significantly (P<0.05);Recurrence rates in group B and C were significantly lower than in group A in the following 6 months (x2 =15.240,P<0.05).Conclusion Combination treatment of TCM and Western medicine showes a positive effect in treatment of allergic purpura.Besides,course of disease and recurrence rate could be decreased by this treatment.

Objective To express recombinant human cardiotrophin 1 (huCT 1) protein with biological activity in E coli Methods huCT 1 open reading frame gene from plasmid pBSSK huCT1 was cloned into plasmid pMD18 T by PCR and T A cloning, and then cloned into prokaryotic GST fusion expression vector pGEX 2T to give pGEX2T huCT1 After pGEX2T huCT1 expression was induced by IPTG in E coli at different temperature and different time, the expression level and the proportion of the soluble protein were analyzed by SDA PAGE Then the soluble GST/huCT1 was purified by immobilized glutathione columns The GST fusion protein was cleaved by thrombin and purified again The recombinant huCT 1 with biological activity was identified according to the survival of axotomized sciatic motoneurons Results After the induction of pGEX2T huCT1 DH5? cells by IPTG at 29 ℃ for 4 h, the highest expression level of the recombinant protein was about 1/5 of total cell proteins, and the soluble portion was about 2/5 of fusion protein Purification of the soluble portion and thrombin cleaved fusion protein resulted in 85% and 80% purified recombinant GST fusion protein and huCT 1 protein, respectively Recombinant huCT 1 protected 55% motoneurons in spinal cord against sciatic axotomy in vivo in adult rats Conclusion Recombinant huCT 1 has biological activity in rat neurons
3.Pathological study of rabbit femoral after high-intensity focused ultrasound irradiation in vivo

Objective To inquire into the hurt-efforts to the rabbit femoral in vivo irradiated by high intensity focused ultrasound(HIFU). Methods Using a HIFU with the operating frequency of 0.8 MHz; the focal distance of 135 mm and the sound-intensity of 8000 W/cm 2. The average interal diameter of the rabbit femoral artery was about 1.1 mm.Thirty rabbits were divided into 3 groups randomly and unilateral-femoral was chosen at random. To observe by using the method of pathology and victorian blue of elastic fibre. Results After irradiation all the endothetial cells of blood vessels of the rabbit peeled off,disappeared and tiny thrombuses were formed in the vessels the core of smooth muscles withered, cell disorder appeared. Conclusions Irradiation by HIFU could cause injury to the rabbit femoral artery, change blood dynamics and produced contribution to the further research on the energy accumulation of the targets in HIFU system.
4.Study on Operation Status and Development Suggestions of Traditional Chinese Medicine Clinic in China

Objective To know the current status and problems of Traditional Chinese Medicine Clinic and to provide countermeasures.Methods Using literature research and statistical analysis.Results From 2008 to 2014,the number of institutions,health personnel and services of Traditional Chinese Medicine Clinic rose year by year.The Traditional Chinese Medicine Clinic was ahead of TCM-WM Clinic and Minority Clinic.The increase number and increase rate of the number of Traditional Chinese Medicine institutions,health personnel and services are more than TCM-WM Clinic and Minority Clinic.The age constitute is unbalance and the quality of personnel is different in Traditional Chinese Medicine Clinic.Conclusion The government should conduct multi-site practice.Traditional Chinese medicine clinics and other private medical institutions should have the equitable health care treatment.The development in preventive treatment of disease by TCM should be focused on.
5.Genotyping and Polymorphism Analysis of Plasmodium falciparum Isolates by Nest-PCR and PCR-RFLP

Objective To analyze the efficiency and specificity of MSP2 alleles genotyping for Plasmodium falci-parum isolates by Nest-PCR and PCR-RFLP. Methods MSP2 alleles from Plasmodium falciparum isolates of Yunnan and Hainan were genotyped by Nest-PCR and PCR-RFLP, respectively, and the efficiency and specificity of the two me-thods were analyzed. Results The conventional Nest-PCR method could detect 79.8% (166/208) alleles of MSP2,and 65.7% (65/99) for 3D7 family, but could not identify the type of any allele. While PCR-RFLP showed 25.3% higher genotyping efficiency than Nest-PCR. Moreover, this method could identify the allele types. Conclusion PCR-RFLP genotyping technique is more efficient and specific than conventional Nest-PCR, and it is a convenient tool in the study on molecular epidemiology of malaria.
6.Construction and purification of human cardiotrophin-1 adenovirus vector and its expression in vitro and in vivo

Objective To construct human cardiotrophin 1(huCT 1) adenovirus vector for central nervous system(CNS) gene therapy in vivo . Methods The huCT 1 and eGFP genes were cloned into shuttle plasmid pDC316 to construct pDC316 huCT 1 and pDC316 eGFP. Virus genome plasmids pBHGloxdeltaE1,3Cre and pHG140 were purified by CsCl banding certification. Recombinant replication defective adenovirus vectors AdCMV huCT1 and AdCMV eGFP were rescued in 293 packaging cells by co transfection and Cre mediated recombination of both plasmids pDC316 huCT1 and pBHGloxdelta1,3Cre. The insert gene and its expression were identified by PCR, RT PCR and immunohistochemistry after recombinant adenovirus transfected 293 cells and NIH 3T3 cells. Recombinant adenovirus vectors were purified by CsCl banding and titrated by plaque forming test. AdCMV huCT1 expression in vivo was analyzed by RT PCR and immunohistochemistry after transfection of the cervical spinal cord in adult rats. Results We have constructed two recombinant adenoviral vectors: AdCMV huCT1 and AdCMV eGFP, containing MCMV promoter, foreign DNA and SV40 PolyA with deletions of E1 and E3 regions. The positive huCT 1 mRNA and protein were identified in AdCMV huCT1 transfected NIH 3T3 cells and rat cervical spinal cord. The titer of virus stocks was generally up to 3.0?10 10 plaque forming units(pfu) per milliliter. Conclusion Recombinant purified AdCMV huCT1 vectors can be highly expressed in vitro and in vivo and is suitable for CNS gene therapy in vivo .
7.Accelerating effects of high glucose on NE-induced cardiac hypertrophy of the cultured myocardial cells

AIM To study the effects of high glucose on the cardiac hypertrophy,cultured cardiomyocytes were used to study the accelerating effects of high glucose on norepinephrine(NE) induced cardiac hypertrophy. METHODS Using cultured myocardial cells as a model, the cellular hypertrophy was observed. The protein content was assayed with Lowrys method. The cardiomyocytes' volumes was measured by computer photograph analysis system. The protein synthesis was assayed with leucine intake method. RESULTS The total cellular protein content?cellular volumes?cellular protein synthesis showed an increase in high glucose group, NE group, high glucose and NE group compared with control group, respectively. High glucose and NE group showed a further increase compared with NE group. CONCLUSIONS The high glucose itself induced hypertrophy of the cultured myocardial cells slightly. Meanwhile high glucose can further accelerate hypertrophy of the cultured myocardial cells induced with NE. The effective mechanism between high glucose and NE on cardiac hypertrophy need further study.

9.Clinical features and gene mutations in a Chinese family with choroideremia

Objective To observe the clinical features, phenotypes and genotypes in a Chinese family with choroideremia (CHM). Methods A Chinese four-generation family (15 members) with CHM, including 5 patients (4 males/1 female), 2 female carriers and 8 healthy members, was enrolled in this study. Initially all family members underwent best corrected visual acuity (BCVA), indirect ophthalmoscopy, fundus fluorescein angiography, optical coherence tomography (OCT), visual field and full view electroretinogram (ERG). BCVA was followed up for 3 years. Venous blood samples were collected, and all of the 15 coding exons and flanking intron regions were amplified in the proband by polymerase chain reaction followed by direct sequencing. Protein structure was modeled based on the protein data bank and mutations in DeepView v4.0.1 to predict the effect of the mutations. A total of 180 healthy volunteers were enrolled as control group to matching CHM gene sequences. Results The visual acuity (VA) of 3/4 adult male patients began to decrease at less than 10, 10 and 30 years old, the average BCVA was 0.43. There were characteristic signs and symptoms of CHM including narrow visual field, extinguished rod and cone response in ERG, disappeared junction line and intermediate line of photoreceptor inner segment/outer segment on OCT. After 3 years, the mean BCVA decreased to 0.11. The BCVA of one young male patient was 1.0 in both eyes with minor changes fundus and visual field. The VA of the female patient began to decrease at 50 years old, her BCVA of two eyes were 0.5 and 0.25, respectively. The fundus changes were typical of CHM, with relative scotomas in the peripheral visual field of OD, and big scotomas in the OS. After 3 years, her mean BCVA decreased to 0.2. Of 2 female carriers, one had minor fundus changes (patches of pigmentary deposits, atrophy spots of retinal pigment epithelium cells), and the other was normal. A novel heterozygous c.1837G>A mutation in exon 15 of CHM was detected in the proband, which resulted in the substitution of serine by proline at codon 613 (p.D613N). Based on molecular modeling, the misfolded protein caused by the mutation might destabilize the structure of the helix that potentially could affect the global stability of the Rep-1 protein. Conclusions A novel c.1837G>A (p.D613N) mutation may be the causative mutation for CHM in this family. Female CHM carriers may have some signs and symptoms.
10.Pathogenesis and treatment of overwhelming inflammation induced by Pseudomonas aeruginosa keratitis in rabbit

Background Infectious keratitis is common blinding eye disease in China.Inflammatory runaway reaction often occurs in infectious keratitis, and its mechanism and treating approach are worthy of research.Objective This study was to investigate the clinical manifestation,mechanism and treatment outcome of doxycycline in overwhelming inflammation induced by Pseudomonas aeruginosa keratitis.Methods Pseudomonas aeruginosa suspension was intrastromal injected in 60 right eyes of 60 New Zealand white rabbits,and gatifloxacin eye drops was frequent instilled for consecutive 3 days to establish corneal inflammatory runaway reaction models,and 42 eyes with worse keratitis were defined as overwhelming inflammation.Then the models were randomly divided into gatifloxacin treatment group (15 rabbits), combined treatment group (gatifloxacin with doxycycline treatment, 15 rabbits) and balance salt solution (BSS) control group (12 rabbits), and corresponding eye drops was topically administered in the rabbits 8 times per day for 14 days.The symptoms of the models were observed under the slit lamp microscope on day 1,3,7 and 14 after treatment,and the corneal infiltration area was calculated.The corneal morphology was examined by using hematoxylin-eosin staining.The expressions of matrix metalloproteinase-2 (MMP-2) and MMP-9 in the corneal tissue were examined by immunohistochemistry.The contents of tumor necrosis factor α (TNF-α) and interleukin-1β (IL-1β) in corneal homogenate were detected by ELISA.The use and care of the animals complied with the ARVO Statement.Results The inflammatory scores were lower and infiltration area of cornea was smaller 7 days after treatment than those before treatment in the combined treatment group and gatifloxacin treatment group (both at P<0.05).Hematoxylin-eosin staining result showed that the inflammation response of cornea subsided 14 days in comparison with 5 days after treatment in the combined treatment group, with the proliferation and rearrangement of fiberal tissue.The MMP-2 and MMP-9 were strongly expressed in corneal tissue with overwhelming inflammation in day 5 after treatment, however, the expression intensity weakened in 14 days after treatment in the combined treatment,showing significant differences in the absorbance between them (all at P<0.05).ELISA assay showed that the contents of TNF-α and IL-1β in corneal tissue were significantly lower in the combined treatment group than those in the BSS group (P =0.00,0.03).In addition, contents of TNF-α and IL-1β in corneal tissue were significantly declined at day 14 in comparison with day 5 and day 7 after treatment in the combined treatment group (all at P<0.05).Conclusions Overwhelming inflammatory keratitis models can be successfully established by corneal intrastromal injection of Pseudomonas aeruginosa suspension and immediately frequent 3-day instillation of gatifloxacin eye drops in rabbits.The topical application of doxycycline plays treating effect on overwhelming inflammatory keratitis by inhibiting the release of pro-inflammatory cytokines and MMPs.
