Objective:To investigate relationships between the single nucleotide polymorphisms(SNPs) of interleukin-1B(IL-1B) and variable number of tandem repeat (VNTR)of interleukin-1RN(IL-1RN) promoter genes and susceptibility of the patients with gastric adenocarcinoma or the patients with gastric adenocarcinoma to helicobacter pylori (Hp) infection.Methods:The SNPs of the IL-1B(-31C/T and -511C/T) were determined by gene chip and The VNTR of IL-1RN were determined by agrose gel electrophoresis with a total of 130 cases of gastric adenocarcinoma and 142 healthy controls.The sera concentrations of IgG,IgM and IgA of Hp antibodies were measured by ELISA in all cases and controls.Results:H.pylori infection was detected in 69.2% of 130 patients and 46.5% of 142 controls (P=0.007,odds ratio [OR]=2.53).Frequencies of IL-1B-31TT genotype in patients with gastric adenocarcinoma or in patients with pooly-differentiated gastric adenocarcinoma were significantly higher than those in healthy controls or in well-differentiated groups,(P

Objective:To explore side population(SP) cells in human epithelial cells and to observe the expression of the universal stem cell marker ABCG2,epidermal stem cell markers ?6 integrin and ?1 integrin on SP cells.Methods:Epithelial cells were obtained by digesting human skins with Dispase II and Trypsin and stained with Hoechst33342 and PI.The SP cells were analyzed and sorted by the fluorescence-activated cell sorter.Then the expression of ABCG2,?6 integrin and ?1 integrin was analyzed by flow cytometry.Results:SP cells accounted for 0.2%-0.3% of total human epithelial cells.Only a small part of cells expressed ABCG2,integrin ?6 and integrin ?1 of both SP cells and total human epithelial cells detected by flow cytometry.The difference of positive rates between SP cells and total human epithelial cells was not significant.Conclusion:SP cells accounted for 0.2%-0.3% of total human epithelial cells.The difference of positive rates of stem cells marker ABCG2,integrin ?6 and integrin ?1 between SP cells and total human epithelial cells was not significant.

Objective To measure HPSE and VEGF-C expression in pancreatic cancer tissues and to observe the effect of small interfering RNA targeting heparanase gene on VEGF-C expression and malignant biological behavior of pancreatic cancer cell lines BxPC-3.Methods We measured the relative mRNA levels of HPSE and VEGF-C in 34 pancreatic ductal cell adenocarcinoma(PDAC) specimens by RT-qPCR.The recombinant plasmid GV230/HPSE was constructed.BxPC-3 was transiently transfected with GV230/HPSE and HPSE-siRNA.The expression levels of HPSE and VEGF-C were measured by performing florescent real-time quantitative PCR (RT-qPCR) and immunoblotting.The invasion and migration potential of treated BxPC-3 was evaluated by transwell invasion assay and Scratch-wound assay.Results Spearman rank correlation analysis indicated a positive correlation between HPSE and VEGF-C (r =0.812,P <0.01)in pancreatic cancer tissues.HPSE and VEGF-C overexpression was correlated with tumor differentiation,lymph node metastasis and TNM stage(all P <0.01).RT-qPCR and Western blot showed high expression of HPSE mRNA and VEGF-C mRNA in GV230/HPSE group than that in control group,compared to low expression of HPSE mRNA and VEGF-C mRNA in siRNA group (all P < 0.01).Invasion chamber assay and Scratch-wound assay showed higher invasion in GV230/HPSE group than that in control group,while lower invasion in siRNA group (all P < 0.01).Conclusions Joint detection of HPSE and VEGF-C may be of significance in predicting prognosis of patients with pancreatic cancer.HPSE regulates the expression of VEGF-C and facilitates invasion and migration of BxPC-3 in vitro.
10.Total mesorectal excision with double mastomosis instruments in lower rectal carcinoma

ObjectiveTo determine the effect of total mesorectal excision with double anastomosis instruments(DAIs) in lower rectal carcinoma. MethodsFrom 1995 to 1998,57 cases of middle and lower segement rectal carcinoma were treated by total mesorectal excision with DAIs, and were followed for 3 years. Results No operative death and no anastomotic leakage were noted. Two patients suffered from anastomotic stenosis,but no dysuria and sexual dysfunction occurred. Local recurrence rate was 5.3%(3/57)in the follow-up period. ConclusionsTotal mesorectal excision with DAIs is not only an effective method to maintain pelvic organ function in lower rectal cancer, but also able to improve the living quality of patients and reduce local reccurence.
