1.Relationship between IL-1 gene polymorphisns and gastric adenocarcinoma

Objective:To investigate relationships between the single nucleotide polymorphisms(SNPs) of interleukin-1B(IL-1B) and variable number of tandem repeat (VNTR)of interleukin-1RN(IL-1RN) promoter genes and susceptibility of the patients with gastric adenocarcinoma or the patients with gastric adenocarcinoma to helicobacter pylori (Hp) infection.Methods:The SNPs of the IL-1B(-31C/T and -511C/T) were determined by gene chip and The VNTR of IL-1RN were determined by agrose gel electrophoresis with a total of 130 cases of gastric adenocarcinoma and 142 healthy controls.The sera concentrations of IgG,IgM and IgA of Hp antibodies were measured by ELISA in all cases and controls.Results:H.pylori infection was detected in 69.2% of 130 patients and 46.5% of 142 controls (P=0.007,odds ratio [OR]=2.53).Frequencies of IL-1B-31TT genotype in patients with gastric adenocarcinoma or in patients with pooly-differentiated gastric adenocarcinoma were significantly higher than those in healthy controls or in well-differentiated groups,(P
2.Strategies to choose scaffold materials for tissue engineering.

Current therapies of organ failure or a wide range of tissue defect are often not ideal. Transplantation is the only effective way for long time survival. But it is hard to meet huge patients demands because of donor shortage, immune rejection and other problems. Tissue engineering could be a potential option. Choosing a suitable scaffold material is an essential part of it. According to different sources, tissue engineering scaffold materials could be divided into three types which are natural and its modified materials, artificial and composite ones. The purpose of tissue engineering scaffold is to repair the tissues or organs damage, so could reach the ideal recovery in its function and structure aspect. Therefore, tissue engineering scaffold should even be as close as much to the original tissue or organs in function and structure. We call it "organic scaffold" and this strategy might be the drastic perfect substitute for the tissues or organs in concern. Optimized organization with each kind scaffold materials could make up for biomimetic structure and function of the tissue or organs. Scaffold material surface modification, optimized preparation procedure and cytosine sustained-release microsphere addition should be considered together. This strategy is expected to open new perspectives for tissue engineering. Multidisciplinary approach including material science, molecular biology, and engineering might find the most ideal tissue engineering scaffold. Using the strategy of drawing on each other strength and optimized organization with each kind scaffold material to prepare a multifunctional biomimetic tissue engineering scaffold might be a good method for choosing tissue engineering scaffold materials. Our research group had differentiated bone marrow mesenchymal stem cells into bile canaliculi like cells. We prepared poly(L-lactic acid)/poly(ε-caprolactone) biliary stent. The scaffold's internal played a part in the long-term release of cytokines which mixed with sustained-release nano-microsphere containing growth factors. What's more, the stent internal surface coated with glue/collagen matrix mixing layer containing bFGF and EGF so could supplying the early release of the two cytokines. Finally, combining the poly(L-lactic acid)/poly(ε-caprolactone) biliary stent with the induced cells was the last step for preparing tissue-engineered bile duct. This literature reviewed a variety of the existing tissue engineering scaffold materials and briefly introduced the impact factors on the characteristics of tissue engineering scaffold materials such as preparation procedure, surface modification of scaffold, and so on. We explored the choosing strategy of desired tissue engineering scaffold materials.

3.Clinical experience of single port VATS procedure in treatment of thoracic disease

Objective Summarize the experience of single port video-assisted thoracoscopic surgery (SPVATS).Evaluate the value of SPVATS for thoracic diseases.Methods 32 patients underwent SPVATS from March 2011 to May 2011,including 28 males and4 females,with an average age of (26.5 ±9.2) years (ranged 18-72 years).There were twenty-seven cases of spontaneous pneumothorax ( including 2 cases of single stage bilateral surgery),three cases of unknown pleural effusion,one case of bilateral pulmonary nodules,and one case of pulmonary metastasis tumor.Results All procedures were successfully accomplished without conversion to conventional multi-port VATS or open approach.There was no major complication or operative death.All procedures were using self-developed specialized surgical instruments and two-incision joint protection.The procedures inluded resection of lung bullae in 27 cases,purulent drainage fiber stripping pleural lavage in 3,parietal pleural biopsy in 1,and pulmonary wedge resection in 1.The operating time ranged from (22.0 ± 9.5 ) min.The average intraoperative blood loss was ( 14.5 ± 8.8 )ml.The mean chest tube duration was (3.1 ± 0.6 ) days.The average postoperative hospital stay was (6.8 ± 1.6 ) days.Conclusion SPVATS is a new approach for treatment of thoracic diseases.This technique is preferred as a minimally invasive and cosmetic procedure with decreased operative time,quickened postoperative recovery,few complications,and satisfactory short-term outcome.
4.Serum level and genotype of interleukin-1β or interleukin-1 receptor antagonist in patients with gastric cancer

Objective To investigate the relationships of the serum level of interleukin-1β(IL-1β), the single nucleotide polymorphism(SNP) of IL-1B and interleukin-1 receptor antagonist (IL-1RN) genes with the gastric cancer or the gastric cancer infected by Helicobacter pylori(Hp). Methods The SNP of the IL-1B(-31C/T and -511C/T) was determined by gene chip and the variable number of tandem repeat(VNTR) of IL-1RN were detected by agarose gel electrophoresis. The sera level of IL-1β and the concentrations of IgG, IgM and IgA of Hp antibodies were measured by ELISA. Results The serum level of IL-1β increased significantly in patients with gastric cancer than that in control group(P<0.001). Hp infection was detected in 69.2% of 260 patients and 46.5% of 284 controls[P<0.001, odds ratio (OR)=2.59]. Frequency of genotype IL-1B-31TT or IL-1B-511TT in patients with gastric cancer were significantly higher than that in healthy controls (P<0.01, OR=1.95; P<0.05, OR=1.62), respectively. Frequency in Hp+ gastric cancer group was higher than that in Hp- group (P<0.05, OR= 2.00), and frequency of haplotype T-T in patients group was significantly higher than that in healthy control(χ2=4.45, P<0.05). The serum level [(802±148) ng/L] of IL-1β of the gastric cancer group was significantly higher than that of the control group [ (501±125) ng/L, P<0.01]. The serum level of IL-1β in patients with -31T or -511T allele was (845±156) ng/L or (871±148) ng/L, significantly higher than that without -31T [(555±116) ng/L] and -511T allele [(581±128) ng/L]. Furthermore, The serum level of IL-1β in Hp+ group with T allele were significantly higher than that in Hp- group (P<0.001). There was no association of IL-1RN gene and other IL-1B gene with gastric cancer or Hp+ gastric cancer. Conclusion IL-1B-31TT genotype was related to gastric cancer. IL-1B-511TT genotype was related to gastric cancer or with Hp+ gastric cancer. Both IL-1B-31T and -511T are associated with IL-1B gene. The haplotype T-T may be the genetic susceptible factor to gastric cancer.
5.Optimization of the method for isolating and culturing rat mesenchymal stem cells.

OBJECTIVETo optimize the protocols for isolation and culture of mesenchymal stem cells from rat bone marrow (BMSCs).
METHODSBMSCs were isolated by adherence to plastic with frequent medium change and reduced trypsinization time. The cell growth curves were drawn and the surface markers of BMSCs were detected by flow cytometry. The cells were induced to differentiate into osteogenic, adipogenic, hepatic and cholic lineages.
RESULTSThe cells isolated using this method were positive for CD29, CD44, and CD90 and negative for the hematopoietic surface markers CD45. The osteogenic and adipogenic differentiation of the BMSCs was verified by alkaline phosphatase staining, Alizarin red staining and Oil red staining. The cell subcultures up to passage 10 maintained capacities of differentiation into osteogenic and adipogenic lineages. The BMSCs induced with sequential addition of growth factors, cytokines and hormones differentiated into cells expressing hepatocyte- and cholangiocyte-specific markers.
CONCLUSIONThe optimized method allows efficient isolation of homogenous populations of MSCs from rat bone marrow, which can be induced into multiple cell lineages.

6.Clinical study of single kidney transplantation from young pediatric donors after death: 86 cases report

Objective By analyzing the perioperative management in our hospital to explore the clinical effect and safety of single kidney transplantation from deceased juveniles' donors.Methods We retrospectively analyze 86 cases of kidney transplantations from deceased juveniles' donors in our hospital from 2007 December to 2015 August.Results The success rate of the operations was 100%.The postoperative complications occurred as fellows:7 cases of acute rejection (8.14%);10 cases of drug intoxication (11.62%);21 cases of DGF (24.44%),4 cases of leakage of urine (4.65%),7 cases of lung infection (8.14%).Two cases (2.32%) died after the operation because of serious lung infection,and by corresponding treatment 47 cases recovered after 2-4 weeks.The creatinine level in 37 cases without any complications was 131.88 ± 44.20 μmol/L during discharge.Conclusion With strict selection,the organ from a deceased juvenile donor is safe and practicable.
7.Research on the role of resveratrol against breast cancer

Breast cancer (BC) ranks first in the incidence rate of female malignant tumor, the notable features of which include high invasive behavior, high malignant degree and poor prognosis. Resveratrol, a plant antioxidant, has been identified as a potential therapeutic agent for the occurrence and progress of BC. This article explores the mechanism of resveratrol intervention in BC by evaluating several in vitro and in vivo studies. It was found that resveratrol can weaken the proliferation and survival ability of BC cells, suppress their growth, metastasis, and invasion, and reverse their resistance to adriamycin by promoting cell apoptosis, regulating autophagy, inhibiting glycolysis and regulating the tumor microenvironment, expressions of matrix metalloproteinases, epithelial-mesenchymal transition and drug-resistant proteins, etc. The limited number of clinical trial studies on resveratrol, mainly focusing on prevention effect of it on breast cancer, may be one of the reasons that affect the comprehensive evaluation of the anti-cancer efficacy of resveratrol.

9.Correlation analysis of eye rubbing and sleeping posture with keratoconus

Objective To investigate the habits of eye rubbing and sleeping posture of patients with keratoconus(KC)and analyze the correlation of the onset and severity of KC with eye rubbing and sleeping posture.Methods In this retrospective case-control study,KC patients who received diagnosis and treatment in the Eye Hospital of Shandong First Medical University from January 2018 to June 2022 were collected as the KC group,and myopia patients with matching ages and sexes who visited our hospital from February to June 2022 were recruited as the control group.All patients were inves-tigated by questionnaire and the details of eye rubbing,sleeping posture and related habits were recorded.According to the severity of the disease(corneal curvature),KC patients were divided into different stages,and the eye rubbing and sleep-ing posture of KC patients in different stages were compared.Results In this study,406 KC patients and 394 myopia pa-tients were included.It was found that 67.98%of KC patients and 24.87%of myopia patients rubbed their eyes,with a sig-nificant difference(P<0.001);45.32%of KC patients and 24.87%of myopia patients were accustomed to sleeping on their side or stomach,with a significant difference(P<0.001).The multivariate binary logistic regression analysis indica-ted that the following variables showed significant differences:patients with eyes at the later stages(steeper eyes)were more likely to rub eyes[odds ratio(OR)=3.547,95%CI:2.02-6.24,P<0.001],sleep on side or stomach(OR=10.725,95%CI:5.77-19.94,P<0.001),and turn head to the steeper eye when sleeping(OR=12.171,95%CI:7.23-20.49,P<0.001).The statistical analysis demonstrated that the proportion of KC patients at the IV stage who were habitu-ated to rub their steeper eyes and turn their heads to the steeper eyes when sleeping was significantly higher than that of pa-tients at stages Ⅰ to Ⅲ(all P<0.001).Conclusion Eye rubbing and sleeping on the side or stomach are risk factors for the onset of KC and are related to the severity of the disease.
10.Fostering organ donation culture for facilitating high-quality development of hospital-level organ donation management center in China

"Chinese model" of organ donation and transplantation in China has won acclaims from all over the world. Current contradictions between unbalanced and inadequate development of organ donation and transplantation and surging public demands for transplant services remain serious. And an acute shortage of donated organs is still the greatest difficulty. Improving organ donation rate per million population (PMP) and organ utilization rate has been a great challenge for organ donation teams in China. This review summarized the relevant experiences of Second Affiliated Hospital of Guangxi Medical University in fostering organ donation culture atmosphere and connotation to accelerate the high-quality development of organ donation. It was intended to provide references for disciplined construction of other organ donation management teams and promote the development of organ donation and transplantation in China.
