Objective To investigate the role and the mechanism of Apr-1 gene on cholangiocarcinoma QBC939 cell lines proliferation and cell cycle regulation. Methods Apr-1 gene was transfected into QBC939 cells by using liposomes to establish a QBC939 cell model ( QBC939-Apr-1 ) stably expressing Apr-1 gene. Apr-1 mRNA expression and the changes in cell cycle and cell growth of QBC939 cells were analyzed by RT-PCR, flow cytometry ( FCM ) and growth curve before and after transfection. The regulatory effect of Apr-1 gene on the expression of cell cycle-related genes was investigated in QBC939 cells before and after Apr-1 transfection using cell cycle gene microarrays. Results Significant suppression of cell growth was observed with the cell model stably expressing Apr-1 gene. Apr-1 over-expression caused cell arrest from 9% to 13% (P ＜0. 01 ) increase in G2 population. Cell cycle gene microarrays demonstrated that the expression of Skp2 、UBE1 was up-regulated, while the expression of MRE11A 、CKS2 、CDK8 、CDC45 was down-regulated by more than 3 folds. Conclusions Apr-1 gene suppresses QBC939 cell proliferation in vitro, QBC939 cells presented with differences in the expression of cell cycle-related genes after Apr-1 gene transfection.

Objective To investigate the predictive value of urinary exosomal microRNA(miR)-29 c in the clinical outcome of organ-and non-organ-confined bladder urothelial carcinoma(BUC).Methods From January 2017 to March 2022,152 patients with BUC were recruited from the Department of Urology in our hospital as a validation set.In addition,126 non-cancer controls were selected from the physical examination center of our hospital.The expression level of urinary exosomal miR-29c was detected by real-time quantitative PCR.Results In the validation set,urinary exosomal miR-29c level in BUC patients was significantly lower than that in non-cancer control group(P＜0.05),while urinary exosomal miR-17-5p level and miR-590-5p level were not significantly different(P＞0.05).The area under ROC curve of urinary exosomal miR-29c for the diagnosis of BUC was 0.969(95％CI:0.953～0.986),and the corresponding sensitivity and specificity were 92.1％and 90.2％,respectively.In subtype analysis,urinary exosomal miR-29c levels were further reduced in patients with non-organ-confined BUC compared with patients with organ-confined BUC(P=0.009).Overall survival(OS),disease-free survival(DFS)and disease-specific survival(DSS)were longer in the urinary exosomal miR-29c high expression group(P＜0.05).Conclusion Low levels of urinary exosomal miR-29c are an adverse prognostic factor for survival in patients with BUC,and are promising as a predictor of adverse clinical outcomes of organ-and non-organ-confined BUC.

Objective To study nursing of ureteral calculi patients treated by holmium laser lithotripsy by ureteral soft mirror under general anesthesia.Methods A total of 118 patients with ureteral calculi in our hospital were randomly divided into control group and observation group according to the double blind number method,all patients carried out general anesthesia by soft ureteral holmium laser lithotripsy,the control group was given routine nursing care and the observation group was given intensive care.And the nursing results of patients were statistically analyzed.Results The observation group had higher one-time success rate of clear stone than the control group (91.53％ vs.81.36％,P ＜ 0.05);The incidence rates of complications in the observation group was 3.39％,which was significantly lower than 8.47％ in the control group(P ＜0.05).The hospitalization time,SDS score,VAS score and SAS score of the observation group were significantly better than that of the control group (P ＜ 0.05).The observation group had significantly higher related knowledge and treatment compliance than that in the control group(88.14％,91.53％ vs.76.27％,74.58％,P ＜0.05).Conclusion Holmium laser lithotripsy by ureteral soft mirror under general anesthesia can strengthen treatment cooperation,the awareness of related disease knowledge,improve the rehabilitation effect of patients,and it is worth of popularizing.
