PKS gene was screened by PCR from thirty strains of spone-associated bacteria including twenty-one actinomycetes isolated from Craniella anstrialiensis and nine bacillus isolated from Dysidea avara in the South China Sea.As a result,a 669 bp KS domain gene was successfully amplified from Bacillus C89.BLAST analysis showed that the KS domains were most closely related to the KS sequences of Bacillus subtilis subsp.subtilis str.168 with 96% similarity.Phylogenetic analysis demonstrated the KS domain belong to trans-AT KS domains. This study demonstrated the existence of PKS gene in bacteria associated with sponge Dysidea avara for the first time, and provided proof for the hypothesis that sponge-associated bacteria are perhaps the true producers of many novel bioactive compounds in sponge.Meanwhile, this study lays a basis for the microbial screening for polyketide compounds production.

Objective To investigate the different killing effect to human pulmonary adenocarcinoma cell line cells GLC-82 with coexpressed double suicide genes compared with single gene. Methods Recombinant expression vectors containing CD (cytosine deaminase) and/or TK(thymidine kinase) gene under CMV promoter were constructed successfully. The vectors were transfected to GLC-82 tumor cell lines by use of lipofectamine. The clones were picked out after G418 selection. Extraneous gene integration and expression were confirmed by PCR and semi-quantitative RT-PCR. The cytotoxicity to these transgenic cells under treatment with 5-Fc and GCV were measured by MT1 assays. Results Double and single suicide gene transfer were both stably expressed in GLC-82 cells . The cytotoxic effects of co-expressed TK-CD genes were superior than that of the single gene. Conclusion The CD + TK/5-Fc + GCV coexpression system is more effective for killing effect of tumor cells than CD/5-Fc or TK/GCV system alone.

Objective To approach the effect of Hemorthoidectomy using LigaSure vessel sealant system with different bowel preparation means. Methods Seventy-eight patients with hemorrhoids were divided into three groups which are the Epsom salt group(21 cases),the Kaiselu group(32 eases),and the no bowel preparation group(25 cases)according to their operation's day.The observation items are the satisfaction rate of the operative doctors, the degree of comfort to patient, the severe pain duration time and scores, the first defecation time and the pain scores after operation, and the intention time. Results 61.90%patients in Epsom salt group felt uncomfortable but did not influence the life.6.25%patients in Kaiselu group felt uncomfortable but did not influence the life and 6.25%patients felt uncomfortable and influence the life. The degree of comfort of patient Was significant difference in three groups(H=13.39,P=0.001 2).There were no significant differences in other observation items among three groups(P>0.05).Condusions There are similar effect in three groups with Hemorrhoidectomy patient using LigaSure vessel sealant system, but in the degree of comfort to patients, the no bowel preparation means is the hest choice for patient. If the doctor order to do bowel preparation for patient, the bowel preparation using Kaiselu is better than that using Epsom salt.
7.Effects of Sargassum confusum polysaccharide on the expression of p53 and Rb genes in mouse sarcoma S180 cells.
Journal of Southern Medical University 2008;28(8):1378-1381
OBJECTIVETo investigate effects of Sargassum confusum polysaccharide (SP) on the expressions of p53 and Rb genes.
METHODSThe mice bearing S180 sarcoma were treated with abdominal SP injection at the daily dose of 50, 100 or 200 mg/kg for 10 consecutive days, The mice injected with 0.85% saline served as the negative control group and those with 5-fluorouracil (5-FU) injection as the positive control group. The tumor weight was measured to calculate the tumor inhibition rate. RT-PCR and Western blotting were used to detect the expression levels of p53 and Rb genes at the mRNA and protein levels, respectively.
RESULTSThe 10-day SP treatment at 50, 100 and 200 mg/kg resulted in tumor growth inhibition rates of 30.5%, 47.6% and 63.5%, respectively. SP treatment upregulated the mRNA expressions and increased the protein levels of p53 and Rb in the tumors.
CONCLUSIONSP has inhibitory effect against S180 sarcoma in vivo and can increase the expression levels of p53 and Rb genes at both the mRNA and protein levels.

WRKY transcription factors are novel transcriptional regulatory factors, which play an important role in regulating plant development, metabolism and other physiological processes. In this study, a new Dendrobium officinale WRKY transcription factor, designated as DoWRKY1 was cloned by using RT-PCR and RACE (GenBank Accession No. KF953910). Bioinformatic analysis demonstrated that, the full-length cDNA of DoWRKY1 was 1,704 bp. And DoWRKY1 contained a 1,629 bp open reading frame (ORF) that encoding a peptide of 542 amino acid residues. The putative DoWRKY1 protein contained two conserved WRKY domains and it belonged to the group I WRKY family protein. Yeast one-hybrid experiment showed that DoWRKY1 had transcriptional activation ability in yeast, and it could activate the expression of downstream report genes (His3 and Ade2). Semi-quantitative RT-PCR experiment showed that DoWRKY1 expressed in roots, stems, leaves and protocorm-like bodies. Real-time qRT-PCR proved that DoWRKY1 could be induced by methyl jasmonate (MeJA) and chitosan (Chitosan), and the expression level of this gene can reach the expression peak at 2 h and 1 h, respectively. These results are useful for further determination of the regulation function of this gene in secondary metabolism of D. officinale.

OBJECTIVETo determine the serotype and genotype of a sample with ABO blood group discrepancies.
METHODSSerotype was determined with serological method. Sequence specific primer polymerase chain reaction (SSP-PCR) was carried out based on the serotype. Sequences of exons 6 and 7 of ABO gene was analyzed by sequence-based testing (SBT).
RESULTSCompletely agglutinated A antigen, half agglutinated B antigen and weak agglutinated anti-B antibody were detected in both erythrocytes and serum, which suggested presence of a ABw serotype. An A/Bw12 genotype was revealed by B subgroup detection. Sequences of exons 6 and 7 were 278CT, 297GA and 467CT, 526CG, 657CT, 703GA, 796CA, 803GC, 930GA, respectively. The genotype fit with A102/B101 except for a nt278 C>T mutation. Blood group antigen gene mutation database (BGMUT) search has confirmed the mutant allele to be Bw12.
CONCLUSIONAn A102/Bw12 genotype has been found in the Chinese population.
