ObjectiveTo study the effects on growth characteristics of cells mutated by outer space. Methods5 tumor cell lines and 1 bio-pharmaceutical cell line were carried in the recoverable satellite No.18 and returned after 18 days flying in outer space. The cell living systems were utilized in passive carrying. Some survival cells were monocoloned, and the experimental methods such as cell morphological observation,MTT assay and FACS were used for analysis of cell growth characteristics. ResultsThe survival cell rates of tumor cell lines and bio-pharmaceutical cells were markedly different. Compared with the control group, mutated cells appeared to have multiple cell morphological changes and the cell number of G1 phase was increased markedly (P<0.05). Growth rate in mutated cells were varied without specific discipline. ConclusionEffects of outer space on growth characteristics were complicated and multiplex in mutated cells, which were not all heredity in further passage. The differences of the monoclonal cell lines provide the potential to obtain optimizing cell lines by screen.

Objective To investigate the protective effect of methylene blue (MB) on blood-brain barrier (BBB) injury after focal cere-bral ischemia-reperfusion in rats. Methods 18 male Sprague-Dawley rats were randomly divided into sham-operated group (n=6), model group (n=6) and MB treatment group (n=6). The left middle cerebral arteries were occluded for 1 hour and reperfused. MB was infused intra-venously immediately after reperfusion (3 mg/kg) and again 2 hours post-reperfusion (1.5 mg/kg), while normal saline was administered in the model group. The sham-operated group was treated as same as the model group without occlusion and infusion. HE staining was used to observe the histological injury in the cortex around the infarcted region 47 hours after reperfusion, while albumin immunohistochemistry was used to evaluate the permeability of the BBB, and immunohistochemistry and double immunofluorescence staining were used to exam-ine the expressions of glial fibrillary acidic protein (GFAP) and aquaporin-4 (AQP-4). Results HE staining showed that cells and blood ves-sels were not intact in the cortex around the infarcted region in the model group and they were better in the MB treatment group. The expres-sions of the albumin, GFAP and AQP-4 were higher in the model group than in the sham-operated group (P<0.01), and were lower in MB treatment group than in the model group (P<0.05). The double immunofluorescence staining showed the colocalization of GFAP and AQP-4 in the astrocytes. Conclusion MB may ameliorate the BBB disruption induced by focal cerebral ischemia-reperfusion through reducing glio-cyte proliferation and down-regulation of AQP-4 expression in rats.

OBJECTIVETo investigate the cytotoxicity of bromoxynil on SH-SY5Y cells and its effect on the expression of nuclear factor-kappa B (NF-κB) and I kappa B alpha (IκBα) in SH-SY5Y cells.
METHODSSH-SY5Y cells were exposed to bromoxynil (10, 50, or 100 µmol/L) for 24 and 48 h, and other SH-SY5Y cells, which were used as a control, were exposed only to dimethyl sulfoxide. After 24 and 48 h of exposure, the morphological changes of these cells were observed under an inverted microscope, and the cytotoxicity of bromoxynil was measured by MTT assay. The cellular proliferation was examined by cell counting after 12, 24, 48, 72, and 96 h of exposure. After 24 h of exposure, the expression of NF-κB was evaluated by Western blot and immunocytochemistry, and the expression of IκBα was evaluated by Western blot.
RESULTSThe cellular proliferation inhibition rates (CPIRs) of 50 and 100 µmol/L groups were significantly higher than that of the control group after 24 and 48 h of exposure (P < 0.05); the CPIR was significantly higher after 48 h than after 24 h in the two groups (P < 0.05). The growth curve revealed that these groups began to show differences in cell count at the 24th of exposure and that the differences were even more marked as the exposure went on (F = 17.15, P < 0.05). The control group had a significantly increased cell count at the 48th, 72nd, and 96th h of exposure (P < 0.05); the 10 and 50 µmol/L groups had a significantly increased cell count at the 72nd and 96th h of exposure (P < 0.05); the 100 µmol/L group showed no significant change in cell count during 96h of exposure. The 50 and 100 µmol/L groups hada significantly longer cell doubling time than the control group (P < 0.05). The immunocytochemistry showed that as the dose of bromoxynil increased, the brownish yellow particles in the cytoplasm and nuclei became darker, the expression of NF-κB was upregulated, and the nuclear translocation of NF-κB was increased. The Western blot showed that the 100 µmol/L group had significantly higher expression of NF-κB in the nuclei than the control group (P < 0.05) and that the 50 and 100 µmol/L groups had significantly lower expression of IκBα in total proteins than the control group (P < 0.05).
CONCLUSIONBromoxynil can inhibit the proliferation of SH-SY5Y cells under this experimental condition, which may be related to activation of NF-κB.

Objective To investigate the respective effects of highly concentrated amino acids, highly concentrated glucose and mixture of the above two on the proliferation and extracellular matrix synthesis of glomerular mesangial cells. Methods Mesangial cells were cultured with mannitol (13.3 mmol/L ), highly concentrated amino acids (13.3 mmol/L), highly concentrated glucose (30.5 retool/L) and the mixture of highly concentrated amino acids and highly concentrated glucose repectively. Mesangial cell proliferation was examined by MTT and flow cytometry. The mRNA expression of TGF-β1,ColⅣ and fibronectin (FN) was detected by semi- quantitative RT-PCR, and the protein expression was detected by ELISA, Western blot and radioimmunoassay. Results (1) Cellular proliferation was increased in response to amino acids and was further enhanced by increased levels of both amino acids and glucose in a time-depentent manner. And the most prominent action occured at 48 hours. (2) Among the three group, mesangial cell cycle was affected, and the number of G0/G1 mesangial cells was reduced, but the number of cycle S cells was improved. (3) In the three group, the mRNA and protein expression of TGF-β1 was improved [protein unit: ng·ml-1·(105 cells)-1, 3023±85, 3163±80, 3321+85 vs 1506±63, P< 0.05]. (4)In the three group, the mRNA and protein expression of Col Ⅳ and FN was enhanced [protein unit: ng'ml-1(105 cells)-1, Col Ⅳ:16.78±2.56, 18.65±2.56, 22.83±2.45 vs 10.22±2.54, P<0.01; FN:18.47±2.34, 13.58±3.13, 17.98±2.56 vs 8.22±2.54, P<0.05]. The expression of Col Ⅳ mRNA and protein was stronger when the cells were cultured with the mixture of highly concentrated amino acids and highly concentrated glucose. Conclusions Highly concentrated amino acid can promote cell proliferation similarly to highly concentrated glucose, which may be through increasing the activity of mesangial cells and the synthesis of extracellular matrix via TGF- β1 signaling way. Highly concentrated amino acid has the synergism with highly concentrated glucose.

Objective To estimate the impact of various extent of coronary stenosis at rest on myocardial perfusion and functional indices from myocardial contrast echocardiography(MCE) and two-dimensional strain echocardiography(2D-Strain) and characterize the relationship between them. Methods Three levels of coronary stenosis:mild (50%~75%),moderate (76%~89%),severe (≥90%) in 25 patients with coronary artery stenosis and 15 healthy subjects in the control group were examined. Plateau videointensity (value A) and time to plateau (value β) of the replenishment curve from MCE were calculated for perfusion analysis, and longitudinal peak systolic strain(SL) from 2D-Strain were measured for functional analysis. Results Value A tended to decrease with increasing severity of stenosis, ahhough these differences did not reach the level of statistical significance, β and SL were significantly increased in the context of moderate and severe coronary stenosis when compared with control group (β, moderate 0.75±0.67 and severe 0.67±0.53 vs normal 0.97±0.65;SL,moderate 11.60±5.89 and severe 9.58±6.51 vs normal 19.46±6.17, P <0.05, respectively). β was positively correlated with SL (r = 0.65, P <0.05).Conclusions β of the replenishment curve from MCE and SL from 2D-Strain both can detect greater than 75% coronary stenosis inducing myocardial ischemia, and they had good correlation.

Aim To study anti-inflammatory and antibacterial effects of total flavones of oldenlandia diffusa Willd(FOD).Methods The models of dimethylbenzene-induced ear swollen in mice,turpentine-induced granuloma pouch in rats, paw edema caused by egg white in rats, increased vascular permeability by acetic acid in mice and the antibacterial activity in vitro were used to study FOD. Results FOD(15、30 and 60 mg?kg~-1 )show a marked suppressive effect on dimethylbenzene-induced ear swollen and increased vascular permeability in mice. In addition,FOD(12、24 and 48 mg?kg~-1 ) could reduce the paws edema and the granuloma pouch in rats.On the other hand,FOD had significally bacteriostatic effects against eight kinds of bacterial in vitro, in which effect on coccus is superior to that on bacillus. Conclusion FOD has anti-inflammatory and antibacterial effects.
