AIM: To investigate the expression of Fas/FasL system in human lung carcinoma cell lines (A549, EBC-1, LCSC) and T-lymphocytes (Jurkat), and to search the possibilities of immune escape and counterattack mediated by Fas/FasL pathway in human lung carcinoma cells. METHODS: The protein and mRNA expression of Fas and FasL were detected by FACScan, RT-PCR, respectively. Cell apoptosis was assessed by fluorescent staining. 3.Caveolin-1 expression is downregulated by shear stress and TNF-? in human aortic endothelial cells

AIM: To observe effects of shear stress and TNF-? on caveolin-1 expression. METHODS: Cultured human aortic endothelial cells (HAECs) of passage 3-5 were used in the experiment. Cells were exposed to a laminar flow (shear stress 1.0 Pa) by using a parallel rectangular flow chamber for different time. Caveolin-1 mRNA and protein expression were measured by RT-PCR and Western blot, respectively. Caveolin-1 expression of the cells stimulated by TNF-? were also studied to elucidate the influence of this inflammatory factor. Objective:To observe the the effects of sustained-release coatings of HU-308 on the biological characteristics of rat oste-oblasts(ROBs)on Ti surface.Methods:The heparin/chitosan sustained release coatings containing HU-308 at different concentra-tions were prepared by layer-by-layer assembly technology on the alkali-and heat-treated Ti surface.The alkali-and heat-treated Ti samples were randomly divided into six groups,the samples in C group were used as control,in P group were treated by heparin/chi-tosan coatings and in T1,T2,T3 and T4 groups were treated by heparin/chitosan sustained-release coating containing HU-308 at dif-ferent concentrations.The adhesion,proliferation and ALP activity of ROBs cultured on the coatings were assessed at different times. Results:The adhesion,proliferation and ALP activity of ROBs of T4 group (the impregnating concentration of HU-308 was 0.025 mg/L)were the highest.Conclusion:The sustained-release coating with low concentration of HU-308 may improve the adhesion , proliferation and ALP activity of ROBs.
5.Pathogenic Bacteria in Hospital:Distribution and Drug Resistance of 1 213 Strains

OBJECTIVE To determine the distribution of bacterial flora in hospital infection and to provide laboratory(evidence) for controlling hospital infection and selecting rationally antibiotics in clinic practice.METHODS All(isolates) were identified by routine procedure.MRSA and ESBLs-producing rate of Escherichia coli and Klebsiella pneumoniae were(examined.) RESULTS Among all these clinical infectious specimens,there were 202 strains of Gram negative bacilli,(accounting) for 40.9%(202/495);166 strains of fungi,accounting for 33.5%;621 strains of Gram positive cocci,for 20.6%(102/495).Candida albicans,E.coli,Pseudomonas aerugionosa,C.tropicalis and C.glabrata took the first five bacteria in infection.Analysis of drug resistant bacteria suggested that the isolated rate of ESBLs-producing strains in Staphylococcus aureus be 47.6%,be CNS in MRCNS 78.1% and MRSA in SA be 42.3%.CONCLUSIONS Multidrug resistance and fungus infection are the main risk factors in our hospital.We must improve means of treatment on clinical work and use antibiotic rationally to reduce the infection rate.

Objective:
To analyze the epidemic trend of viral hepatitis in Nanjing from 1989 to 2019 and predict the incidence in 2020, so as to provide reference for the prevention and control of viral hepatitis.
Methods:
The incidence data of viral hepatitis in Nanjing from 1989 to 2019 was retrieved from Nanjng Center for Disease Control and Prevention and National Infectious Disease Reporting System. The epidemic trend was analyzed by estimating the annual percent change ( APC ) and the average annual percent change ( AAPC ). The seasonal incidence of different types of viral hepatitis was analyzed by seasonal index. The autoregressive integrated moving average model ( ARIMA ) was built to predict monthly incidence rate of viral hepatitis in 2020.
Results:
The annual incidence rate of viral hepatitis was 62.00/100 000 in Nanjing from 1989 to 2019, showing a downward trend ( AAPC=8.4%, P<0.05 ). From 1998 to 2019, the annual incidence rates of hepatitis A, B, C and E were 1.98/100 000, 14.31/100 000, 2.30/100 000 and 2.60/100 000. The incidence of hepatitis A and B showed downward trends ( AAPC=-11.81%, -6.02%, both P<0.05 ); the incidence trend of hepatitis C was not obvious ( P>0.05 ); the incidence of hepatitis E showed an increasing trend ( AAPC=4.82%, P<0.05 ). From 2015 to 2019, the third and fourth quarters were the epidemic seasons of hepatitis A, B and C, while the first and second quarters were the epidemic seasons of hepatitis E. The ARIMA model predicted that the monthly incidence rates of viral hepatitis in 2020 would range from 1.26/100 000 to 3.69/100 000, among which hepatitis B ranged from 1.21/100 000 to 2.58/100 000, hepatitis C from 0.20/100 000 to 0.48/100 000, hepatitis E from 0.09/100 000 to 0.25/100 000.
Conclusions
The incidence of viral hepatitis in Nanjing shows a downward trend. Among different types of hepatitis, hepatitis B has a higher incidence. All types of hepatitis have epidemic seasons. It is predicted that the monthly incidence rates of viral hepatitis will be 1.26/100 000 to 3.69/100 000 in 2020.
9.Preliminary study of tumor cells and bio-engineering cells carried by returned satellite

ObjectiveTo study the effects on growth characteristics of cells mutated by outer space. Methods5 tumor cell lines and 1 bio-pharmaceutical cell line were carried in the recoverable satellite No.18 and returned after 18 days flying in outer space.The cell living systems were utilized in passive carrying. Some survival cells were monocoloned, and the experimental methods such as cell morphological observation,MTT assay and FACS were used for analysis of cell growth characteristics. ResultsThe survival cell rates of tumor cell lines and bio-pharmaceutical cells were markedly different. Compared with the control group, mutated cells appeared to have multiple cell morphological changes and the cell number of G1 phase was increased markedly (P<0.05). Growth rate in mutated cells were varied without specific discipline. ConclusionEffects of outer space on growth characteristics were complicated and multiplex in mutated cells,which were not all heredity in further passage. The differences of the monoclonal cell lines provide the potential to obtain optimizing cell lines by screen.
10.Development of long-term cell culture facility in hypothermia and its application in space biology research

ObjectiveTo develop the facility of mammalian cell culture in hypothermia for long-term, and to lay the base for carrying mammalian cells to outer space and establishing outer space biomedical research stage. MethodsMultiple cell lines were maintained in the spacial designed hypothermia cell culture facility and carried to space by Shenzhou 4 and No.18 recoverable satellite. Cell morphology, cell multiplication and the ability to be monocloned were observed.ResultsCells were successfully maintained in the hypothermia cell culture facility for up to 26 days without obvious changes of cell morphology. The cells could normally grow, multiple and be monocloned after inoculated in 37 ℃ for 8 h.ConclusionA hypothermia cell culture facility was successfully established,which ensure technically mammalian cells to be carried by space craft and further research on space radiation.
