1.Determination of the Content of Dehydroepiandrosterone in Dehydroepiandrosterone Tablets by HPLC

OBJECTIVE:HPLC method was established for the determination of dehydroepiandrosterone(DHEA)in tablets.METHODS:The chromatographic system consisted of ?-Bondapak C18 column and mobile phase of methanol-water(75∶25)with a detection wavelength at 292nm.RESULTS:The linear range of DHEA was 0.2~1.0mg/ml,r=0.9 999.The recovery rate was 99.13%(RSD=0.42%).CONCLUSION:This method is simple and accurate for determination of DHEA in tablets.
2.Effect of Danshen Tablet combined with Simvastatin on hepatic function in patients with hypercholesterolemia

AIM:The present study is to observe the effect of Danshen Tablet combined with Simvastatin on hepatic function in patients with hypercholesterolemia.METHODS:Patients with hypercholesterolemia were randomly divided into Simvastatin treatment group(n=20)and Danshen Tablet combined with Simvastatin treatment group(n=20).Before study and after 12 weeks treatment,serum lipid(TC,TG,LDL-C,HDL-C,AST,ALT and CPK)were measured.RESULTS:After treatment,serum lipid in both groups were improved significantly,and there was no difference between them.In Simvastatin treatment group,there was a tendency to increase in ALT and AST,but we could reject at the significant level.In the same way,the serum lipid in combination treatment group decreased,but no statistic difference.Compared with Simvastatin treatment group,ALT and AST in combination treatment group reduced significantly.CPK in both groups did not change.CONCLUSION:Danshen Tablet can reduce liver injury by statin and could serve as an adjuvant in clinical hypolipidemic treatment.
3.Experimental study on rabbit endothelial progenitor cells-seeded stents in vitro

Objective To fabricate rabbit endothelial progenitor cells(EPCs)-seeded stents in vitro and evaluate its feasibility.Methods The mononuclear cells of rabbit were separated by density-gradient centrifugation using lymphocyte isolation.Cells were seeded on fibronectin-coated 6-well plates and maintained on M199 with endothelial cell growth supplements(ECGS,30 ?g/mL).After 2 weeks,cultured cells were identified by immunofluorescence and immunocytochemistry.EPCs proliferation and migration were measured by drawing a growth curve and modified Boyden chamber assay.EPCs adhesion assay was performed by replating EPCs on fibronectin-coated dishes.VEGF and G-CSF were assayed by ELISA while NO was tested by nitrate reductase method.EPCs were poured on fibronectin-coated or uncoated stents.After 6 days,scanning electron microscopy and fluorescence microscopy observation were performed.Results About 14 days after seeding,a majority of spindle-like cells appeared which were characterized as cells,double positive for Dil-acLDL and FITC-UEA-I.EPCs were further confirmed by its expression of the endothelial cell marks of vWF.After 2 weeks culture,EPCs proliferated in exponential growth.They showed high ability of adhesion(96.7?3.2%),migration(15?4) and could secret VEGF,G-CSF and NO.Compared with uncoated stents group,more EPCs migrated and adhered on fibronectin-coated stents group(27.80?4.26 vs 6.10?3.07cells/?400 field,P
5.Analysis of Chinese medicine on differentiation of symptoms and treatment of 36 cases of Sjāgre's syndrome

BACKGROUND: Sjogre's syndrome is a common disease of aged people,in which, dry mouth and throat have disturbed chawing, swallowing and speech and have seriously affected living quality of patients. OBJECTIVE: To probe into the improvement of oral function treated with Chinese medicine based on differentiation of symptoms. DESIGN: Retrospective analysis on cases was designed. SETTING: Department of Periodontal Disease, College of Stomatology,Fourth Military Medical University of Chinese PLA. PARTICIPANTS: Thirty-six cases of Sjogre's syndrome were selected from clinic of Department of Periodontal Disease of Stomatology College of Fourth Military Medical University of Chinese PLA from May 2000 to February 2004, with agreement and in the know of patients. Of those, 5cases were male, 31 cases were female, aged varied from 27 to 70 years,with duration of sickness in range from 0.5 to 10 years. METHODS: ① On basis of moistening, drying up heat, producing fluid and stopping thirst, the drugs for activating blood circulation, resolving stasis, clearing away heat and detoxification were supplemented. Formula:shashen (Radix Glehniae), maidong (Radix Ophiopogonis), shihu (Herba Dendrobii), yuzhu (Rhizoma Polygonati Odorati), huafen (Radix Trichosanthis), xuanshen (Radix Scrophulariae), shengdi (Radix Rehmanniae), chenpi (Pericarpium Cirri Reticulatae), danshen (Radix Salviae Miltiorrhizae), banzhilian (herba Scutellariae Barbatae), bahuasheshecao (Herba Hedyotis Dissusae), etc. ②Treatment of vitamins and transfer factor were associated.③ Artificial saliva in local was applied to prevent from secondary infection. ④ It was to test 99Tc m parotid function (After 99Tc m was injected intravenously on elbow, the parotid uptaking and concentrated 99Tc m curve were traced immediately. When radioactivity intensity was increased to maximum, the patients were required to chaw Vit. C tablet, 200 mg. In following, it was to trace the minimum of 99Tc m radioactivity on decreased curve. Normal imaging was illustrated clearly, the development after Vit. C stimulationwas declined swiftly. Time-radioactivity curve illustrated obvious uptaking and excreting segments. Positive results were unclear parotid development or the development after Vit. C stimulation was not declined obviously. Time-radioactivity curve was of horizontal type, low-horizontal type, parabola type or persistent rising type), erythrocyte sedimentation (ERS) (Normal value: 0-15 mm/h for male adult, 0-20 mm/h for female adult) and a part of immunology, such as rheumatoid factor, immunoglobulin (I gG normal value: 7.6-16.6 g/L), immune complex, etc. The effect was determined as increased saliva secretion and improved oral function. MAIN OUTCOME MEASURES: 99Tc m parotid function test, rheumatoid factor, ERS, IgG and immune complex were applied to determine the therapeutic effects of Chinese herbs. RESULTS:Thirty-six cases all entered result analysis. ① After treatment, the positive rate of rheumatoid factor was lower significantly compared with that before treatment [36.7%, 73.3%,(x2=6.796, P < 0.01)]. The levels of ERS, IgG and immune complex were all decreased significantly compared with those before treatment [(35.50±5.65) mm/h,(13.47±1.58) g/L,(8.30±1.24) mg/L;(55.20±8.85)mm/h,(23.57±2.37) g/L,(14.96±2.21) mg/L; (t=3.549-4.962,P < 0.01)]. The result of 99Tc m parotid function test and main determined indexes had been improved for all. ② After treatment, dry mouth, dry eye and general symptoms were all improved of various degrees and saliva secretion was increased. Total effective rate reached 80.7%. CONCLUSION: Treatment based on differentiation of symptoms is applied in Chinese medicine. On basis of moistening, drying up heat, producing fluid and stopping thirst, the drugs for activating blood circulation, resolving stasis, clearing away heat and detoxification are supplemented and effectively promote rehabilitation of oral function in Sjogre's syndrome,such as chawing, swallowing, speech, etc.

Objective:To prepare anti-AchR monoclonal antibodies from rat bone marrow.Methods:The extracellular region of human AchR ?1 subunit was expressed in E.coli and purified using a Ni-NTA column.Then the protein was refolded by dialyzing.Lewis rats were immunized with purified AchR ?1 subunit and B cells from bone marrow and spleen were fused with myeloma cells to generate monoclonal antibodies.Results:Human AchR ?1 subunit was successfully expressed in E.coli.Rats immunized with the refolded protein were shown symptoms of Myasthenia gravis.Hybridoma cells were produced using B cells from rat bone marrow and spleen.Conclusion:Bone marrow cells can be immortalized using hybridoma method,which can be used for further study of Myasthenia gravis.

9.Clinical study of renal function of newborns with sclerema neonatorum

Objective To investigate the changes of urinary brush border membrane enzymes and serum cystatin C (Cys C) in newborns in order to develop early diagnostic indicators for sclerema neonatorum (SN) renal dysfunction.Methods Sixty-night cases with sclerema neonatorum and 30 cases of normal newborns were enrolled in this study.Of all sclerema neonatorum cases,39 cases were mild and 30 cases were moderate to severe sclerema neonatorum.Immunoca talytic assay method was adapted to detect the level of urinary brush border membrane.The levels of serum Cys C,blood urea nitrogen (BUN),creatinine (Cr) and urinary β32 microglobulin (β2-MG) in two groups were also measured.Results The levels of urinary brush border membrane enzyme,β2-MG and serum Cys C in sclerema neonatorum group were (40.09 ± 7.29) U/L,(4.65 ± 1.33) mg/L and (1.84 ± 0.32) mg/L,higher than those in control group ((23.19 ± 5.62) U/L,(2.49 ± 0.77) mg/L and (1.07 ± 0.25) mg/L; t =10.34,7.47 and 10.55,P < 0.01).The levels of urinary brush border membrane enzyme and serum Cys C in moderate to severe sclerema neonatorum group were (42.06 ± 7.59) U/L and (1.93 ± 0.34) mg/L,higher than those in the mild group ((38.57 ± 6.70) U/L,(1.77 s0.29) mg/L;t =2.24,2.11,P <0.05).The abnormality rates of urinary brush border membrane enzyme and serum Cys C in sclerema neonatorum group were 79.7% (55/69) and 72.5% (50/69),higher than β2-MG abnormal rate (52.2% (36/69),x2 =12.95,12.11,P < 0.01).In sclerema neonatorum group,urinary brush border membrane enzyme was positively correlated with β2-MG (r =0.560,P < 0.01),and serum Cys C was positively correlated with BUN,Cr (r =0.314,0.287,P < 0.05).Conclusion Renal injury is common in SN.Urinary brush border membrane enzyme and serum Cys C are served as the diagnostic parameters for early detection of renal injury in SN.
10.Clinical research on the effect of abdominal Tuina and acupuncture in treating Swiss generalized anxiety disorder

Objective To observe the clinical effect of abdominal Tuina and acupuncture in treating Swiss generalized anxiety disorder (GAD). Methods 80 patients of GAD caused by liver depression-qi stagnation were randomly recruited into a treatment group and a control group, 40 patients in each. The treatment group was treated with abdominal tuina & acupuncture; while the control group was treated with acupuncture exclusively. Clinical effects and hamilton anxiety scale (HAMA) score were observed in both groups before and after the treatment. Results ①There was significant difference in total efficacy between the two groups CP<0.05), and the treatment group was better than the control group. ②The HAMA score of the two groups after the treatment was lower than that before the treatment, and the difference was significant (P< 0.05). There was also significant difference of HAMA score between the two groups after the treatment (/><0.05), with the treatment group showing better results than the control group. Conclusion Abdominal tuina combined with acupuncture has sound effects in treating Swiss GAD due to liver depression-qi stagnation.
