1.Enterovirus 71 infection induced HFMD related to IL-6 and IL-13 level

Objective To study the clinical significance of interleukin (IL)-6 and IL-13 levels in serum of hand,foot,and mouth disease (HFMD)caused by enterovirus 71,and carries on the preliminary discussion on serum IL-6 and IL-13 cell origin. Methods Serum levels of IL-6 and IL-13 in HFMD and healthy control were detected by enzyme linked immunosorbent assay (ELISA),serum IL-6 and IL-13 levels among different severity of HFMD were compared.Serum IL-6 and IL-13 levels in HFMD after treatment were also detected.The secretion of IL-6 by monocyte and the secretion of IL-13 by T cells in peripheral blood mononuclear cells (PBMC)were detected by Flow Cytometry (FCM).Monocyte and T cells from health individual were treated with EV71,and the secretion of IL-6 by monocyte and the secretion of IL-13 by T cells were detected by FCM too.Results Serum levels of IL-6 and IL-13 were (79.63±29.45)pg/mL and (36.45±1 5.13)pg/mL respectively in HFMD,and serum levels of IL-6 and IL-13 were (27.26±7.82)pg/mL and (13.46±3.14)pg/mL respectively in health control,serum IL-6 and IL-13 levels in HFMD were significantly higher than that of healthy subjects (P < 0.01 ),and the mean serum IL-6 and IL-13 levels of HFMD were increased with the severity of the disease (P <0.01).After treatment,serum levels of IL-6 and IL-13 were (48.23 ±23.14) pg/mL and (23.25±9.63)pg/mL respectively,when compared with before treatment,serum levels of IL-6 and IL-13 were signifi-cantly decreased (P <0.01).The secretion ability of IL-6 by monocyte and the secretion of IL-13 by T cells in HFMD were signifi-cantly higher than that of healthy subjects.After infection by EV71,the secretion ability of IL-6 by monocyte and the secretion of IL-13 by T cells were significantly increased.Conclusion Infection of EV71 probably by increase the secretion ability of IL-6 by monocyte and the secretion of IL-13 by T cells,resulted in elevated of serum IL-6 and IL-13 levels,which involved in the occurrence and development of HFMD.
2.Safety risks assessment of the multi-parameter physiological monitor

Objective:To strengthen the safety management of monitor according to the analysis of risks in the process of using multi-parameter monitor.Methods:1200 multi-parameter monitors were investigated in the 25 medical units for monitor safety risks using statistical methods, to investigate and analyze the risk within three years and classified the related factors. Results: Within three years, there were 420 cases with security risks in 1200 sets of multi-parameter monitor, including 220 cases with clinical application, 120 cases with equipment management problems, and 80 cases with maintenance problems. Clinical operation of multi-parameter monitors problems led to safety risks with high proportion.Conclusion:Multi-parameter monitor safety risk is mainly caused by improper use in clinical. It can ensure the safety of multi-parameter monitor in clinical on strengthening ideological understanding, improving the clinical training, and paying attention to the maintenance.
3.Effects of recombinant human interferon α2a suppository on levels of inflammatory factors in cervical mucus of HPV-infected patients

Objective To investigate the effects of recombinant human interferon α2a(rhIFNα2a) suppository on the levels of inflammatory factors in the cervical mucus of patients infected with human papillomavirus(HPV).Methods A total of60 HPV-positive patients admitted to the Second Affiliated Hospital of Xi'an Jiaotong University from March to August in 2022 were selected as study objects,and then divided into observation and control groups,30 cases for each group,according to the random number table method.The observation group was given rhIFNα2a suppository therapy by vaginal medication,once every other day,continuous 10 times a month as a course of treatment,and 3 consecutive courses of treatment.The control group did not use drugs.The cervical secretions were collected and the levels of IL-1β,IL-2R,IL-6,IL-8,IL-10 and tumor necrosis factor-α(TNF-α) were measured by chemiluminescence assay.Results After 3 months of treatment,the levels inflammatory factors IL-1β,IL-6,IL-8 and TNF-α in cervical mucus of patients in the observation group were significantly lower than those in the control group(t=-2.717,-2.686,-3.178 and-3.25,respectively,each P <0.05).Compared with before treatment,the levels of IL-1β,IL-6,IL-8 and TNF-α in cervical mucus of patients in the observation group also decreased significantly(t=5.934,4.092,6.495 and 3.287,respectively,each P <0.01),while in the control group,only the level of IL-8 in cervical mucus was significantly different(t=2.345,P=0.024).Conclusion rhIFNα2a suppository can reduce the level of inflammatory factors in cervical mucus,attenuate the inflammatory response and accelerate the clearance of HPV.
4.Effect of tooth preparation and bonding agents on cementation of metal full crown

Objective To find out the effect of tooth preparation and bonding agents on cementation of metal full crown,to provide research basis for clinical practice of full crown restoration .Methods 60 dissociation teeth were randomly divided into six groups ,10 teeth in each group .Different adhesive materials or different polymerization angle were used in different groups ,the six groups were 3 polymerization angle zinc group ,3 polymerization angle glass ionomer group,3 polymerization angle resin cement group ,5 polymerization angle zinc group ,5 polymerization angle glass ionomer group ,and 5 polymerization angle resin cement group .In the first three groups ,polymerization angle was 3,and in the latter three groups ,polymerization angle was 5.The retentive force values of all groups were tested and compared.Results The metal crown retentive force of 3 polymerization angle zinc group[(198.5 ±11.6)N]was sig-nificantly better than that of 5 polymerization angle zinc group[(138.2 ±14.1)N](t=7.46,P<0.05).Polymeriza-tion angle showed no statistically significant effect on metal crown retentive force values of glass ionomer groups and resin cement groups (t=1.52,2.03,all P>0.05).Bonding zinc retention of 3 degree of polymerization group was (198.5 ±11.6)N,glass ionomer and resin cements were (302.7 ±19.2) N and (352.7 ±21.8) N.Bonding zinc retention of 5 degree of polymerization group was (138.2 ±14.1)N,glass ionomer and resin cements were (317.4 ± 23.8)N and (362.9 ±22.6) N.Different adhesives showed statistically significant effects on metal crown retentive force values of 3 polymerization angle groups and 5 polymerization angle groups (F=7.88,8.42,all P<0.05). Conclusion A suitable adhesive,standard operation can get the best cementation effect ,and improve clinical repair effect.
5. Selection of peptides bind to PreS1 antigen from a phage-displayed peptide library

Objective: To screen for peptides that specifically bind to PreS1 antigen from a phage-displayed peptide library. Methods: The PreS1 antigen was used as the target molecule to screen the binding peptide from the Ph. D. -7 peptide library with phage display technique, and the positive clones were identified by ELISA. Results: After three rounds of biopanning, the binding peptides were screened from the peptide library by ELISA and competitive inhibiting ELISA. Sequencing result showed that the binding peptides had high affinity and specificity. Conclusion: A peptide binding PreS1 antigen has been successfully obtained by screening the phage display library, which paves a way for the diagnosis and treatment of hepatitis B infection.
6.Key advances in podocyte studies using zebrafish model

Zebrafish have been emerging as a valuable model for kidney research , including podocyte biology , nephrogenesis and kidney disease .The zebrafish kidney has similar structure , common function and the same cell types with human kidney .Rapid development and feasible genetic editing approaches in zebrafish provide an ideal model for high -throughput gene screening involved in kidney disease and developmental process .Loss or injury of podocytes is the key mechanism , which can lead to proteinuria , glomerulo-sclerosis and renal failure .Here, we introduce structure and function of zebrafish kidney , filtration estimating assays , podocytes injury model and the highlights of recent zebrafish podocytes studies .
7.Relative factors analysis of type 2 diabetes mellitus complicated with osteoporosis in elderly men

Objective To investigate and analyze the relative factors in elderly men with type 2 diabetes mellitus(T2DM)complicated with osteoporosis(OP),thus to provide guidance for prevention of OP complicated with T2DM in elderly men.Methods Dual X -ray absorptiometry determination of bone mineral density (BMD)of 67 cases of elderly men with diabetes,men were divided into OP group (n =32),NOP group (n =35).Then,we compared differences in body mass index(BMI),diabetes duration,fasting plasma glucose (FPG),glycosylated hemo-globin(HbA1c),patients'age,fasting insulin(FINS),insulin sensitivity index(ISI),quantitative of urinary albumin (ALb),quantitative of urinary protein in 24 hours and alkaliphosphatase level(ALP),low density lipoprotein choles-terin(LDL -C),triglyceride(TG),and made correlation analysis.Results Compared with those in NOP group,pa-tients in OP group had older age,longer disease course,smaller baric index,lower BMD,higher ALP,lower FINS,low-er ISI,higher HbA1c,higher quantitative of urinary protein in 24 hours and urinary albumin,higher LDL -C,which were significantly different (P <0.05);BMD of elderly men with T2DM was negatively correlated to patients'age, disease course,HbA1c,quantitative of urinary protein in 24 hours,ALb,ALP and LDL -C,but positively correlated to BMI,FINS and ISI.Conclusion Many factors,such as elderly,low body weight,long duration of the disease,high level of blood sugar,insulin dysfunction,low insulin sensitivity,high level of blood suger,insulin dysfunction,low insulin sensitivity,high serum alkalinephosphatase,high LDL -C and diabetic nephropathy,contribute to osteoporosis in elderly men with T2DM.

Epigenetic modifications such as DNA methylation,histone post-translational modifications,non-coding RNA are reversible,heritable alterations which are induced by environmental stimuli.Major risk factors of diabetes and diabetic complications including hyperglycemia,oxidative stress and advanced glycation end products,can lead to abnormal epigenetic modifications in retinal vascular endothelial cells and retinal pigment epithelium cells.Epigenetic mechanisms are involved in the pathogenesis of macular edema and neovascularization of diabetic retinopathy (DR),as well as diabetic metabolic memory.The heritable nature of epigenetic marks also plays a key role in familial diabetes mellitus.Further elucidation of epigenetic mechanisms in DR can open the way for the discovery of novel therapeutic targets to prevent DR progression.
