Objective To investigate the role of Treg/CD4~+ T in the pathogenesis and prevention of bowel dysfunction after acute enteritis in rats. Methods Studies were conducted in 40 female Wistar rats with weights of 170-180 g.The rats were divided into normal control, experimental control,antibi- otics-treated,probiotics-treated and combined treated groups. The latter 4 groups were first infected with shigella flexneri by garage to establish the model of acute infectious enteritis, then normal control and experimental control group were administered 1 ml saline once everyday for 10 days.Antibiotics- treated group was given gentamycin 1 ml (8.32 mg?kg~(-)?d~(-1)) for 3 days, then was given saline 1 ml once everyday for 7 days.Probiotics-treated group was given medilac-S 1 ml (52 mg?kg~(-1)?d~(-1)) by gavage for the first 7 days, and 1 ml saline for the latter 3 days. The combined treatment group was given gentamycin for the first 3 days and medilac-S for the latter 7 days using the same dose. All rats were given colon-rectal balloon distention 16 to 22 days after the occurrence of bowel inflammation. The contraction of enteric smooth muscle was measured in vitro, the proportion of Treg cells in blood and mesenteric lymph node were analyzed by flow cytometry, and the histological changes were also observed at the same time.Statistical analyses were performed by using t test or one way ANOVA. Results No obvious changes of distal colon were histological observed in different groups. The perceptual threshold of rat gut decreased and the tension integral of enteric smooth muscle increased, and the ratio of Treg/CD4~+ T in blood and mesenteric lymph node was significantly lower in experimental control group [(4.40?0.43)%;(2.47?1.36)%] than normal controls [(5.15?0.47)%;(4.01?1.12)%,P<0.05],while that of combined treatment group had no difference from normal controls [(4.95+0.41)%;(3.22?1.86)%].Conclusions The ratio of Treg/CD4~+ T may play an impor- tant role in the pathogenesis of post-infectious bowel dysfunction in animals.Treating acute enteritis with both antibiotics and probiotics can normalize the abnormalities, and thus prevent the occurrence of bowel dysfunction.

Background Establising the culture model of Müller cells for obtaining the highly putified target cells is essential for the study about the physiology and pathology of retinal Müller cells. The exsiting purifing method for culturing Müller cells is dissatisfactory. Objective This study was to establish a method to obtain high purifing Müller cells. Methods The retina from 5 clean newborn SD rats were isolated and digested by 0. 01% trypsin and cultured in DMEM containing 10% fetal bovine serum. The cellular suspension was then prepared, and the target cells were screened using flow cytometry based on the size and the quantity of cells. Cultured and passaged cells were identified by transmission electron microscope and light microscope. Immunocytochemistry was used to detecte the expression of GFAP in cultured cells for the determination of type and purity of the cells. Results The cells showed the similar shape to retinal Müller cells after primarily culture with the large volume, and some small other types of cells could been seen. The growth of cells was quickly 3 weeks later. The fibroblasts were removed using sticking-wall by steps, and neurons were eliminated following passage. Aboundent of cellular organs were seen under the transmission electron microscope. The positive response rate of the cells for CFAP was 100%. Conclution Flow cytometry offer a rapid and feasible approach for purifying Muller cell and it builds the foundation for further study about Müller cells.

OBJECTIVE:To investigate clinical efficacy and safety of goserelin in the treatment of adenomyosis complicated with severe dysmenorrheal before placing levonorgestrel intrauterine system (LIS). METHODS:A total of 140 adenomyosis pa-tients with severe dysmenorrheal were selected from Sanya Family Planning Technology Service Center during Jan. 2014-Jan. 2016, and then divided into control group and observation group according to lottery method, with 70 cases in each group. Both groups re-ceived LIS within first 7 d of menstruation, and observation group was given Goserelin acetate sustained-release implant 10.8 mg subcutaneously via anterior abdominal wall one week before placing. Therapeutic efficacy was evaluated 6 months later. The im-provement of dysmenorrhea was compared between 2 groups. The uterine volume,COX dysmenorrhea score (CMSS),menstrual volume, hemoglobin (Hb) and CA125 content were compared between 2 groups before and after treatment. The occurrence of ADR was also compared. RESULTS:The rate of dysmenorrheal improvement in observation group was 95.71%,which was signifi-cantly higher than 81.43%of control group, with statistical significance(P<0.05). Before treatment, there was no statistical signif-icance in above indexes between 2 groups (P>0.05). Compare to before treatment,CMSS score, menstrual volume and CA125 content of 2 groups were decreased significantly after treatment, while Hb content was increased significantly;the observation group was significantly better than the control group, with statistical significance(P<0.05). There was no statistical significance in uterine volume between 2 groups before and after treatment (P>0.05). There was no statistical significance in the incidence of ADR(10.00% vs. 14.29%)between 2 groups(P>0.05). CONCLUSIONS:Goserelin in the treatment of adenomyosis complicated with severe dysmenorrheal before placing LIS can effectively relieve dysmenorrheal, reduce menstrual volume and improve Hb and CA125 levels, but do not increase the risk of ADR.

Objective: To observe the clinical efficacy of heat-sensitive moxibustion for vascular dementia and explore its mechanism. Methods: A total of 70 patients with vascular dementia were randomized into an observation group and a control group, with 35 cases in each group. The control group was treated with piracetam, and the observation group was treated with heat-sensitive moxibustion on the basis of the treatment of the control group. The treatment lasted for 8 weeks. Before and after the treatment, the mini-mental state examination (MMSE), activity of daily living (ADL) and traditional Chinese medicine (TCM) symptom scores were assessed, and the levels of acetyl choline (Ach) and homocysteine (Hcy) were measured. The efficacy was evaluated after treatment. Results: The total effective rate of the observation group was significantly higher than that of the control group (P<0.05). After treatment, the MMSE and ADL scores in the observation group decreased significantly, and were lower than those in the control group (all P<0.05); the TCM symptom score of the observation group decreased significantly (P<0.05), while that of the control group had no significant change (P>0.05); the plasma Ach level in the observation group increased significantly (P<0.05), and the Hcy level decreased significantly (P<0.05), which were statistically different from those in the control group (both P<0.05). Conclusion: Heat-sensitive moxibustion plus piracetam is effective in treating vascular dementia. It can significantly improve dementia symptoms and ADL, which may be related to the correction of plasma Ach and Hcy levels.

Objective To investigate the relationship between diastolic function of the left ventricle and common carotid artery (CCA) elasticity in patients with hypertension. Methods Forty-one primary hypertension patients and 31 healthy volunteers underwent echocardiography to obtain the indices of LV diastolic function and also echo tracking techniqne (ET) to obtain the CCA elasticity indices. The LV diastolic function indices included Ve (early diastolic peak velocity), Va (late diastolic peak velocity ), Ve/Va, Em (early diastolic mitral annular motive velicity), Am (late diastolic mitral annular motive velicity) and Em/Am. The indices of CCA elasticity included Ep (pressure-strain elasticity modulus), β(stiffness parameter), AC (arterial compliance), AI (augmentation index) and PWVβ (pulse wave velocity). Results Va was higher and Em, Ve/Va and Em/Am were lower in patients than that in the healthy , P<0.05. Ep and PWVβ were higher in patients,P<0.05. Am and Va were correlated positively with PWV β and Ep , respectively in patients, P<0.05. Ve/Va was correlated positively with AC, and negatively with PWV β, P<0.05 in patients. Em/Am was correlated negatively with β and PWV β in patients. Conclusions The LV diastolic function and common carotid artery elasticity were decreased in patients with hypertension. The decreased common carotid artery elasticity was along with the LV diastolic function decreased.

Objective To study the value of CT angiography(CTA) and three dimensional contrast enhanced magnetic resonance angiography(3D CE-MRA) in the diagnosis of arteriosclerotic occlusive disease (ASO) of lower limb and evaluate arterial stenosis. Methods Thirty-eight patients clinically suspected ASO were studied with CTA and3D CE-MRA,twenty-two patients underwent DSA at the same time. The degree of seriousness of the stenosis shown by various imaging techniques were analyzed respectively. Results Using the DSA images as the standard reference, the sensitivity and specificity of CTA were 97.3% ,99. 1%, and MRA both were 100.0% in detecting the segments occluded. With regard the detection of segments that had more than moderate stenosis ,the sensitivity and specificity of CT were 99.3% ,97.6% ,and MRA were 97.9% ,96.4%. The degree of seriousness of the stenosis shown by MRA was overestimated than CTA on the segments that had more than moderate stenosis(x2 =5.80,P <0. 05). Conclusion CTA and 3D CE-MRA were reliable method in diagnosis of ASO.
