1.Effect of hypothyroidism on oxidative stress status in developing rat brain

Objective To study the effect of hypothyroidism on oxidative stress status in developing rat brain and to further explore the mechanism of impaired brain development caused by hypothyroidism. Methods Perinatal hypothyroidism was induced by administering propylthiouracil(PTU) solution to the dams by gavage.The oxidative stress indexes were measured in brain homogenate of normal and hypothyroid pups which were sacrificed on the 21st d after birth. Results As compared to the control,the following indexes were found to be increased in the hypothyroid group: protein carbonyl contents,thiobarbital acid reactive substances,reduced glutathione,total antioxidative capacity,activities of superoxide dismutase and glutathione peroxidase(P0.05). Conclusion Hypothyroidism during rat brain development may cause oxidative stress,which may be related to the brain damage caused by hypothyroidism.
2.Significance of Detection of Level of Serum Sex Hormone in Healthy Children

Objective To build a normal value of serum sex hormone in the healthy children in favour of the diagnosis of children precocious puberty and the evaluation of the growth of the children.Method Serum of 462 healthy children(aged from 1 month to 14 years;205 females,257 males)was collected,and the level of testosterone(T) and 17-estradiol(E2).Results There were no significant deviation in the levels of E2 of deferent genders between 1-12 months,but the level of T in boys and girls were obviously different.The puberty of boys was la-ter than girls.There was no significant deviation in the level of E2 of the boys in different periods,but the levels of T in boys at different ages were obviously different.Conclusions With the improvement the detection methods,the normal values of the serum sex hormone in healthy children have been changing.It is helpful for diagnosis of children precocious puberty by rebuilding normal values of T and E2 .

5.CD133+cells in combination with human umbilical cord stem cells in mouse heart failure

BACKGROUND:Currently, conventional treatment methods for heart failure are al ineffective. OBJECTIVE:To explore the combined effects of human umbilical cord stem cel s and CD133+cel s in mice with heart failure, providing a new insight into the treatment of heart failure. METHODS:Ful-term newborn umbilical cord from vaginal delivery was col ected to isolate CD133+cel s and human umbilical cord stem cel s using lymphocyte separation medium method. Twenty Balb/C nude mice were randomly subjected to mononuclear cel injection (mononuclear cel group) or injection of CD133+cel s combined with human umbilical cord stem cel s (combined group) via the tail vein after establishing heart failure models. RESULTS AND CONCLUSION:Fourteen days after injection, the body weight and liver, heart and lung mass of mice were significantly larger in the combined group than the mononuclear cel group (P<0.05). After 30 days, myocardial cel s arranged regularly in the combined group, but disorderly in the mononuclear cel group;compared with the mononuclear cel group, the average area of myocardial col agen fibers was significantly decreased in the combined group (P<0.05), and the level of serum matrix metal oproteinase-9 was also significantly lower in the combined group (P<0.05). Masson staining showed that blue-stained col agen fibers in the combined group were less but arranged neatly;however, in the mononuclear cel group, the number of col agen fibers that arranged irregularly was increased to different extents. To conclude, the combined use of CD133+cel s and human umbilical cord stem cel s has desired outcomes in the treatment of heart failure in mice, indicating a higher clinical value.
6.Foodbome Viruses and its Detection Methods

Foodbome viruses are defined to be viruses that can lead to human diseases through food. In accordance with the different origin, foodborne viruses can be divided into two kinds: intestinal viruses and zoonotic viruses. The former include those viruses that can be transmitted to person via fecal-orally route. The latter include those zoonotic viruses that chiefly transmitted to person through livestock and poultry products. This paper expounds foodborne viruses categories, biology nature, epidemiology character, and study circumstance, and clarifies the molecular biological methods and problems on the base of the polymerase chain reactions, and presents the development direction and application perspective of the foodbome viruses study.
7.Biomechanical effects of osteoporosis vertebral augmentation on the adjacent intervertebral levels using a three-dimensional finite element analysis

Objective To investigate effects of volume,distribution and leakage of bone cement in ver- tebroplasty on the adjacent intervertebral bodies using an experimentally calibrated and anatomically accurate fi- nite-element model of elderly L4-L5 vertebral bodies.Methods Computed tomography(CT)scanning was done,at 1 mm intervals,on L4-L5 vertebral bodies of the lumbar spine of an old man cadaver that had no abnormal findings on rnentgenograms.The L4-L5 motion segment data were obtained from the CT scans to establish in computer a three-dimensional finite element model of L4-L5 functional spinal unit(FSU)with the software Ansys 7.0.Compressive load after virtual vertebroplasty on the damaged model,unipedicular and bipedieolar injections and leakage of cement into the intervertebral space,and the resulting endplate and disc stresses of the adjacent vertebral bodies were analyzed in various spatial distributions of the filling material and different loading conditions. An anatomically accurate finite-element model of elderly L4-L5 vertebral bodies was developed.Results The FSU study suggested that changes in stress and strain at adjacent levels were minimal.Furthermore,endplale and disc stresses of the adjacent vertebral bodies were not influenced by bone cement filling volumes or distributions except under bending,whereas asymmetric distribution and leakage of bone cement to the disk increased the stress of adjacent endplates.Conclusion Since asymmetric distribution and significant leakage of cement into the intervertebral space can increase the stresses of endplates of adjacent vertebral bodies and may lead to a fracture, symmetric placement of cement in operation should be achieved and leakage of cement avoided.

9.Effect of benazepril on expression of RAGE in renal tissues of diabetic rats

Objective To investigate the effects of benazepril on the expression of receptors for advanced glycation end-products(RAGE)in diabetic rat kidneys,and to explore its mechanisms of renal protection in diabetic rats. Methods Thirty SD rats were randomly divided into three groups(n=10 in each group): normal control group,diabetic control group and diabetic with benazepril group.Blood glucose,blood lipid,HbA1c,kidney to body weight ratio and 24 h urinary protein excretion were detected after 12 weeks.RAGE mRNA level was analyzed by quantitative RT-PCR. Results Compared with the normal control group,the blood glucose,HbA1c,triglyceride,total cholesterol and low density lipoprotein cholesterol in the diabetic control group and diabetic with benazepril group were significantly increased(P0.05).The kidney to body weight ratio,24 h urinary protein excretion and RAGE mRNA level in the diabetic with benazepril group were significantly higher than those in the normal control group(P
10.Effect of valsartan on expression of receptor for advanced glycation end-products in renal tissues of diabetic rats

Objective To investigate the effects of valsartan on the expression of receptors for advanced glycation end-products(RAGE) in kidneys of diabetic rats,and to explore its renoprotection mechanisms. Methods Thirty rats were divided into normal control group,diabetes control group and diabetes with valsartan group(n=10).Blood glucose,blood lipid,HbA1c,kidney to body weight ratio and 24 h urinary protein excretion were measured after 12 weeks.RAGE mRNA level was detected by real-time quantitative PCR. Results Compared with normal control group,kidney to body weight ratio,24 h urinary protein excretion and RAGE mRNA were significantly increased in diabetes control group.Compared with diabetes control group,kidney to body weight ratio,24 h urinary protein excretion and RAGE mRNA were significantly decreased in diabetes with valsartan group. Conclusion Valsartan can inhibit renal hypertrophy and decrease urinary protein excretion in diabetic rats.The renoprotective effects may be related to its inhibition on RAGE expression.
