Objective: To investigate the effect of sodium butytate (different concentrations) on the growth and proliferation of rat liver oval cell line WBF344, and to discuss the conditions and tules for sodium butyrate-inducd WB-F344 cells differentiation into biliary lineage in vitro. Methods: WB-F344 cells were treated with sodium butyrate (0.70, 2.25, 3.75, 4.5 mmol/L) and the cell growth and morphological changes were observed; routinely cultured WB-F344 cells were taken as control. The changes of CK19 protein expression were examined immunohistochemically after WB-F244 cells were treated with 3.75% sodium butyrate; and the expression of phenotypic markers, such as γ-glutamyltransferase (GGT), β4-integrin, CK19, AFP and ALB at toRNA level were determined by RT-PCR. Untreated WB-F344 cells were used as blank control. Results: We found that sodium butyrate inhibited the growth of WB-F344 cells. The optical densities were significantly decreased in 3.75 and 4.5 mmol/L groups compared with that in control group(P<0.01); but no significant difference was found between 0.75, 2.25 mmol/L groups with control group. WB-F344 cells treated with 3.75, 4.5 mmol/L sodium butyrate became larger and round, with increased nuclei and decreased nucleus to cytoplasm ratio; those treated with 0.75, 2.25 mmol/L had no obvious changes. Immunohistochemical results showed that sodium butyrate significantly increased CK19 expression compared with control group ([92.3±1.1]% vs [1.3±0.2]%, P<0.01). RT-PCR showed increased expression of β4-integrin in sodium butyrate treated groups, but not in control group; the expression of GGT and CK19 was higher than that of control group. Alpha-fetoprotein (AFP) expression was observed in blank control group, but not in sodium butyrate treated cells. Albumin expression was not detected in the 2 groups. Conclusion: Sodium butyrate at 3.75 mmol/L is suitable for inducing WB-F344 cells differentiate into the biliary lineage in vitro.

Objective: To evaluate the effect of preoperative transcatheter arterial chemoembolization (TACE) on liver function and survival of patients after resection of large hepatocellular carcinoma (HCC) by a randomized controlled approach. Methods: From July 2001 to December 2003, a total of 108 patients with resectable large HCC(≥ 5 cm) ,who met the inclusion criteria, were prospectively randomized into surgical resection group (OP group, n=56) or preoperative TACE group (TACE + OP group, n=52). Operative outcomes, resection rate, 1-,3-,and 5-year tumor-free survival rates and overall survival rate were compared between the two groups. Results: The preoperative baseline conditions were equivalent between the two groups. The γ-globulin level in TACE + OP group was significantly higher than that in the OP group(P = 0. 046) after chemoembolization. The prealbumin level was significantly lower than that of the OP group seven days after operation(P = 0. 031). Compared with TACE + OP group, OP group had a significantly higher resection rate (100% vs 89. 4% ,P=O. 017) ,a less average operative time (P=O. 042) ,and less metastases (2 vs 9,P=O. 018). There were no significant differences between the two groups in intraoperative blood loss, warm ischemic time, 1-,3-,and 5-year tumor-free survival rates,or overall survival rate. Conclusion: The preoperative TACE can not improve post-operative tumor-free and overall survival rates, and it may result in tumor metastasis or hepatic function damages.

Objective To observe the therapeutic effect of acupuncture plus oral taking Chinese medicine compared with that of oral taking Chinese medicine only for the treatment of insomnia.Methods Totally 64 patients were randomized into treatment group(33 cases)and control group(31 cases).The treatment group was treated by acupuncture with filiform needle[Baihui(GV 20),Sishen- cong(EX-HN1),Shenting(GV 24),Neiguan(PC 6),Shenmai(BL 62)and Zhaohai(KI 6)being the main points] plus oral taking Chinese medicine.The control group was treated by oral taking Chinese medicine only.All 64 cases were examined by SPIEGEL scale. OBJECTIVE:To compare the relative bioavailability of 2 kinds of domestic oxaprozin enteric tablets.METHODS:20 healthy volunteers were administered with single oral dose of trial tablet 400g and reference tablet 400g by crossover design,whose plasma oxaprozin level was determined by HPLC.The pharmacokinetic parameters and bioavailability of oxaprozin enterosoluble tablets were calculated by 3p97 software.RESULTS:The pharmacokinetic parameters of tested enteric tablets vs.reference tablets were as follows,t 1/2?(73.468?24.354),(73.556?24.406)h,t max(13.275?8.012),(13.200?15.154)h,C max(44.283?7.535)、(45.429?15.107)?g/ml,AUC 0～Tn(4471.792?1387.724),(4234.328?1741.380)(?g?h)/ml,AUC 0～inf(5040.407?2092.744),(4858.292?2423.656)(?g?h)/ml;No significant differences were noted between 2 tablets.The relative bioavailability of tested tablet was(112.8?38.5)%.CONCLUSION:2 kinds of oxaprozin enterosoluble tablets were bioequivalent.

OBJECTIVE: To propose a HPLC method for quantitative assessment of cilostazol in human plasma. METHODS: Chromatogram column was C18(150mm?4.6mm, 5?m), mobile phases were acetonitrile-water(40∶60,V/V), the flow rate was 1.4ml/min,determined wavelength was 254nm. RESULTS: Cilostazol concentration was in the linear range of 25～2000ng/ml(r=0.9 999)with the lowest determinable concentration of 12.5ng/ml,recoveries of cilostazol from plasma were 99.99%～101.44%,the relative standard variation of intra-day and inter-day were 0.20%～2.90% and 0.19%～2.13%(n=5) respectively. CONCLUSIONS: This method is s simple, convenient, accurate, and applicable for study of pharmacokinetics of cilostazol in clinic.

AIM: To study intra-ocular distribution and pharmacokinetics of musk eye drops in the rabbit eyes.METHODS: After administration,twenty seven rabbits were euthanatized and taken eye in 0.083 h,0.167 h,0.5 h,1 h,2.0 h,4.0 h,8.0 h and 12 h,and then isolated aqueous humor,vitreous body,further cornea and ciliary margin of iris from rabbits’eyes.The intraocular concentrations of muscone were measured by GC after topical instillation with 10% musk solution in rabbits.The pharmacokinetic parameters were calculated with 3p97 program.RESULTS: Results showed that the peak concentrations(max) of muscone in various ocular tissues were (107.52 ?67.07),(2.15 ?1.49),(0.034 ?0.007 6),(2.87 ?1.50) and(0.013 ?0.004 5)?g/g or ?g/mLin lacrimal fluid,cornea,aqueous humor,iris and vitreous body;Its half-Life(T1/2) was(8.08 ? 3.08),(2.87? 2.24),(3.37 ? 0.68),(4.69 ? 1.32) and(8.37 ? 2.70) h,respectively.AUC(0→T) of various tissues was (114.57 ?37.41),(11.57 ?7.16),(0.18 ?0.056),(2.86 ?0.42) and(0.079 ?0.017)?g/(h.g) or(?g/h.mL),respectively.CONCLUSION: The results indicate that musk in eye drop has a good permeability and high concentration in various intraocular tissues of rabbit after ocular instillation.

It is vital for reducing the occurrence of complications, such as liver cirrhosis and hepatic carcinoma, to early diagnose liver fibrosis in chronic hepatitis B and intervene in time with antiviral therapy. Although liver biopsy is the "golden standard" for the diagnosis of fibrosis, it has certain disadvantages, such as complicated operation, invasion, and occurrence of complications. Therefore, non-invasive diagnosis models have gained more and more attention clinically. In this review, we summarizes the non-invasive diagnosis models for chronic hepatitis B that have been established throughout the world. It is noticed that there are a number of models being studied and each model has a diagnostic value, to some degree, but a lack of consensus exists. The development of more efficient models that can replace liver biopsy still needs further research to assess liver fibrosis in chronic hepatitis B and guide antiviral therapy.

OBJECTIVETo explore the mechanisms of Qianjing Decoction in the treatment of oligoasthenospermia (OAS).

METHODSWe randomly divided 100 SPF male rats into five groups of equal number: normal, model, Huangjingzanyu, levocarnitine, and Qiangjing. OAS models were established in the animals followed by intragastrical administration of normal saline, ornidazole, Huangjingzanyu Capsules (200 mg per kg body weight per day), levocarnitine (100 mg per kg body weight per day), and Qianjing Decoction (10 g per kg body weight per day), respectively, qd, for 4 successive weeks. Then, we detected the concentration and motility of the epididymal sperm, obtained the contents of superoxide dismutase (SOD), malonaldehyde (MDA), glutathione peroxidase (GSH-Px), lactate dehydrogenase (LDH), α-glucosidase, and fructose in the epididymis, and determined the mRNA expressions of nuclear factor erythroid 2-related factor 2 (Nrf2) and succinate dehydrogenase (SDH) in the epididymal tissue of the rats by real-time PCR.

RESULTSThe concentration and motility of the epididymal sperm in the model, Huangjingzanyu, levocarnitine, and Qianging groups were (35.34 ± 4.22) x 10(6)/ml and (40.04 ± 7.05)%, (48.12 ± 5.56) x 10(6)/ml and (62.46 ± 7.12)%, (47.14 ± 4.87) x 10(6)/ml and (63.23 ± 6.34)%, and (50.25 ± 5.08) x 10(6)/ml and (66.34 ± 7.58)%, respectively, all significantly lower than in the normal group ([53.05 ± 4.55] x 10(6)/ml and [70.20 ± 8.54]%) (P < 0.05), but remarkably higher in the Huangjingzanyu, levocarnitine, and Qiangjing groups than in the model rats (P < 0.05). Compared with the thinned epididymal lumen walls, decreased sperm count, and disorderly and loose arrangement of the lumens in the OAS models, the rats in the Huangjingzanyu, levocarnitine, and Qiangjing groups showed evidently thicker epididymal lumen walls, with the lumens full of sperm cells and arranged regularly and compactly, similar to those of the normal rats. The levels of SOD and GSH-Px were significantly lower but that of MDA markedly higher in the model rats ([84.12 ± 23.25], [10.56 ± 3.02], and [14.04 ± 2.06] nmol/mg) than in the normal group ([110.04 ± 19.56], [17.25 ± 3.56], and [8.87 ± 1.35] nmol/mg) (P < 0.05), while the former two indexes remarkably higher and the latter one significantly lower in the animals treated with Qiangjing Decoction ([120.56 ± 23.68], [16.34 ± 3.12], and [8.45 ± 1.56] nmol/mg), Huangjingzanyu Capsules ([115.34 ± 21.35], [15.23 ± 3.67], and [8.33 ± 1.54] nmol/mg), and levocarnitine ([116.67 ± 22.67], [15.35 ± 3.45], and [8.05 ± 1.78] nmol/mg) than in the models (P < 0.05). The levels of fructose, LDH and α-glucosidase were decreased markedly in the OAS models ([100.22 ± 12.12] mg/[ ml x g], [322 ± 46.13] U/[ ml x g], and [10.48 ± 2.33] U/[ml x g]) as compared with the normal rats ([128.12 ± 13.45] mg/[ml x g], [428 ± 35.12] U/[ml x g], and [15.34 ± 3.12] U/[ ml x g]) (P < 0.05), remarkably higher in the rats treated with Qiangjing ([130.23 ± 13.67] mg/[ml x g] [455 ± 51.50] U/[ml x g], and [18.56 ± 4.67] U/[ml x g]), Huangjingzanyu ([124.16 ± 14.02] mg/[ml x g], [ 419 ± 43.14] U/[ml x g], and [17.64 ± 4.08] U/[ml x g]), and levocarnitine ([123.34 ± 14.02] mg/[ml x g], [430 ± 31.80] U/ [ml x g], and [16.85 ± 5.55] U/[ml x g]) than in the models (P < 0.05). The Nrf2 mRNA expression was significantly reduced in the models as compared with the normal rats (P < 0.05) but remarkably increased in the Huangingzanyu, Qiangjing and levocarnitine groups as compared with the model and normal animals (P < 0.05). The SDH mRNA expression was significantly lower in the model than in the normal rats (P < 0.05) but markedly elevated in the Huangjingzanyu, Qiangjing and levocarnitine groups as compared with the model and normal animals (P < 0.05), remarkably higher in the Qiangjing than in the Huangjingzanyu group (P < 0.05).

CONCLUSIONOrnidazole induces OAS in rats, which is closely associated with excessive oxidation and energy metabolism dysfunction. Qiangjing Decoction can improve and even reverse ornidazole-induced OAS in rats as well as improve the ultrastructure of their testicular and epididymal tissues. Antioxidation and improvement of energy metabolism are probably the action mechanisms of Qiangjing Decoction in the treatment of OAS.
