1.Effect of eoeulture with mouse dermis-derived mesenehymal stem cells on the secretion of collagen and expression of transforming growth faetor-betal by hnman dermal fibroblasts

Objective To explore the role of mouse dermis-derived mesenchymal stem cells (mdMSC) on skin repair. Methods mdMSC and human dermal fibroblasts were isolated and identified. Human dermal fibroblasts were cultured alone or eoeultured with mdMSC in Transwell chambers with the density ratio of human dermal fibroblasts to mdMSC being 2/5, 1/1, and 2/1. On day 4 and 8 of culture, the expression levels of hydroxyproline and transforming growth factor-beta (TGF-beta) 1 were measured in the supematant of monoculture and coculture by alkaline hydrolysis and ELISA respectively. Results The level of hydro-xyproline was significantly higher in the supematants of coculture system with a density ratio of 2/5 and 1/1 than that in monoculture supematants of human dermal fibroblasts on day 8 (both P < 0.05). Elevated level of TGF-betal was observed in all coculture supematants on day 8 (all P < 0.01) and in the supernatants of coculture system with a density ratio of 1/1 on day 4 (P < 0.05). There was no significant correlation between the expression level of TGF-betal and hydroxyproline in the coculture supernatants (r = 0.108, P > 0.05). Conclusion In vitro coculture with mdMSC can increase the production of hydroxyproline and TGF-betal by fibroblasts, which may be a mechanism underlying the facilitation of skin repair by mdMSC.
2.Effect of Erigeron breviscapus Hand-Mazz on learning and memory dysfunction in mice with cerebral ischemia/reperfusion injury

Objective To investigate the effects of Erigeron breviscapus Hand Mazz(EBH) on learning and memory dysfunction in mice with cerebral ischemia/reperfusion injury.Methods 40 mice were randomly divided into control group, sham operated group, model group and EBH group, with 10 mice each. Mice model of cerebral ischemia/reperfusion injury was established by bilateral common carotid artery blocking. 7 days before and after operation, the mice in EBH group were injected I.p.with EBH injection 0.15 ml/d, while those in other groups were injected I.p. with saline 0.15 ml/d. The learning and memory function was tested by Morris water maze.Results The latent period in the model group was longer than that in the normal group and the sham operated group (P<0.05). EBH could shorten the latent period significantly as compared with those in the model group (P<0.05).Conclusion EBH can improve learning and memory function in cerebral ischemia/reperfusion injured mice.

Aniracetam could shorten the latent period significantly as compared with those in the model group (P<0.05). 5.Influence about complications of gastrointestinal tract among ICU patients by three methods of nasal feeding pump

Objective To discuss the influence about three kinds of nasal feeding methods on ICU critical patients with gastrointestinal complications, thus providing guidance for ICU patients to choose reasonable way of nasal feeding pump injection. Methods 120 ICU patients were randomly assigned into three groups, which is the continuous 6-hour and intermittent 2-hour every three time pump group and continuous 24-hours pump group and intermittent 24-hour(continuous 2-hour and intermittent 1-hour every eight time units)pump group ,each group has 40 cases. To observe the the influence about three different way of nasogastric on ICU critical patients with gastrointestinal complications. Results The incidence of diarrhea and constipation among the three groups was not statistically significant (P>0.05) . The gastric retention rate of the continuous 24-hour pump group was significantly higher than that of the continuous 6-hour and intermittent 2-hour pump group and the intermittent 24-hour pump group, the differences were statistically significant (P < 0.05). There was no statistically significant difference between the continuous 6-hour and intermittent 2-hour pump group and the intermittent 24-hour pump group, (P > 0.05). Conclusions The continuous 24-hour pump group in gastric retention was slightly worse than the other two pump way. Compared with ICU class three a week and feeding times , the continuous 6-hour and intermittent 2-hour pump method is more suitable for ICU patients.
6.Distribution and genotypic subgroup of oral Saccharomyces albicans isolated from cancer patients receiving chemotherapy.

OBJECTIVEThe objective of this study is to study the distribution and molecular characteristics of the oral Saccharomyces albicans (S. albicans) in the cancer patients receiving chemotherapy.
METHODS390 cancer patients receiving chemotherapy were sampled by oral mucosal swab. The Candida species were identified by CHROMagar Candida differential medium. All the S. albicans were genotypic grouped by PCR using primers reported to span a transposable intron region in the 25S rDNA gene.
RESULTSThe frequency of oral Candida carriage of the cancer patients receiving chemotherapy was 53.85% (210/390). Most of them were Saccharomyces albicans, the frequency was 48.21% (188/390). The frequency of oral Candida glabrata carriage was 5.64% (22/390). Genotypic subgroup A, B, C of Saccharomyces albicans were determined, and genotypic group B was the predominant group 59.57% (112/188).
CONCLUSIONSaccharomyces albicans, especially genotypic subgroup B, rather than subgroup A, is the prevalence subpopulation in the oral Candida obtained from cancer patients receiving chemotherapy.

7.Different metabolites of leaves between Tripterygium wilfordii and Tripterygium hypoglaucum based on UPLC-Q-TOF-MS.

To analysis the differences between Tripterygium wilfordii and T. hypoglaucum, specimens of their leaves were collected from five production regions and analyzed by ultra performance liquid chromatography coupled with quadrupole time of flight mass spectrometry (UPLC-Q-TOF-MS). The data were analyzed by multivariate statistical method, such as hierarchical cluster analysis (HCA) principal component analysis (PCA) and orthogonal signal correction partial least square discrimination (OPLS-DA). Potential markers with VIP values above 5.0 and corresponding r values above 0.85, were selected and further tested by combining mann-Whitney nonparametric. Those with P < 0.001 and AUC = 1 were confirmed as metabolite markers to discriminate them from each other. Results revealed that the two species were obviously different in their leaf metabolites. Based on their mass spectra, 23 potential metabolite markers were identified to distinguish T. wilfordii from T. hypoglaucum.

8.A totally laparoscopic peritoneal free flap for reconstruction of hand.

Management of defects on the hand and foot with exposed tendons remains a major challenge for plastic surgeons. Here, we present a case of hand reconstruction with a totally laparoscopic peritoneal flap. The anterior rectus sheath was preserved in situ. The peritoneal free flap supplied by peritoneal branches of the deep inferior epigastric artery was retrieved by laparoscopy to cover the soft tissue defect of the hand. The defect of the dorsal hand was 17 cm ×12 cm. The peritoneal flap measuring 22 cm × 15 cm survived completely without any complications. A following split-thickness skin graft offered the suc- cessful wound closure. Motor and sensory function improved gradually within the first year follow-up. The totally laparoscopic peritoneal free flap is a good choice for reconstruction of the soft tissue de- fects accompanied by exposed tendons on the hand and foot.

9.Roles of drug efflux pump genes in the mechanism of Saccharomyces albicans biofilm drug tolerance.

OBJECTIVESaccharomyces albicons is the main opportunistic pathogen which is also the common member of oral microflora, and the biofilms they formed have spontaneous drug tolerance compared with planktonic ones. Saccharomyces biofilm population can produce subpopulation of cells which can tolerant high concentration of antifungal drugs. They are called persisters. Many researches indicated that drug efflux gene such as CDR or MDR gene plays the very important roles in yeast drug resistance. The objective of this study is to illuminate the mechanism of Saccharomyces albicons biofilm drug tolerance related with drug efflux gene, especially to the persisters formation.
METHODSTest the minimal inhibitory concentration (MIC) and SMIC80 (sessile minimal inhibitory concentration 80%) of 24 h biofim of totally 7 defective strains of drug efflux pump gene with Amphotercin B. And treat the 24 h biofilm by fluconazole, micronazole, clotrimazole combined with CDR1 inhibitor Enniatin B respectively and antifungal drugs alone as control, then scrapped the biofilm, cultured on the YPD agar. By CFU counting, the numbers of biofilm persisters were determined.
RESULTSAll the defective strains have the similar MIC and SMIC80 for 24 h biofilm with wide type strains. CDR1 inhibitor Enniatin B can help antifungal drugs especially micronazole and clotrimazole to eliminate the biofilm persisters.
CONCLUSIONSaccharomyces albicans drug efflux gene may minor associated with 24 h biofilm drug tolerance. Drug efflux gene CDR1 may play an role in persisiters related biofilm drug tolerance.
