1.Isolation and Identification of Endophytic Bacteria H-6 with High Antifungal Activity from Huperzia serrata

In this study,nearly 200 endophytic bacteria were isolated from different part of Huperzia serrata, over 60 bacterium with clear antifungal activity were selected from those cultures.Among them,strain H-6 exhibited the highest antifungal activity which was strongly inhibits the growth of many plant pathogenic fungi such as Sclerotinia scleroliorum,Fusarium graminearumt,Sclerotinia libertiana,Phytophthora capsici Leonia and Sesame fusarium wilt.According to the characteristics of morphology,physiology and biochem- istry tests and the comparison of 16S rDNA sequence,the strain H-6 was similar to the Burkholderia.So strain H-6 was identified as Burkholderia sp.H-6.The results also showed that Burkholderia sp.H-6 was markedly different from Burkholderia cepacia that was applied widely in agriculture as antagonistic bacteria. The medium and culture conditions of the strain all were optimized by single factor and orthogonal experiments.In the investigation of the culture condition,growth was carried out in a basal medium(potato juice)and gradually supplemented with the various ingredients to be investigated.The major ingredients be- ing investigated included carbon sources and nitrogen sources.The optimal antifungal activity production condition is growth in a medium(potato juice with 2.5%mannitol and 0.1%NaNO3),initial pH 4.0 at 28℃.
2.Study on Flavonoids Producing and Kinetics in Cell Suspension Culture of Eucommia ulmoides Oliv.

The type of basic media and the contents of plant growth substances were investigated by orthogonal design experiment,and also the effects of different culture conditions on the growth of suspension cells and the accumulation of total flavonoids in Eucommia ulmoides were studied.The results showed that B5 medium supplemented with 0.5mg/L NAA,0.6mg/L 6-BA and 30g/L sucrose,at initial pH 5.0~5.5,20g(FW)/L inoculation quantity and 110 r/min of rotation speed was a preferable culture conditions for E.ulmoides suspension cells growth and flavonoids synthesis.The results of metabolic kinetics analysis for E.ulmoides cell suspension culture showed that the logistic and Luedeking-Piret equations can be used for describing the kinetics of cell growth,sucrose consumption and flavonoids production during the process.The maximum specific growth rate(?m),the actual growth yield based on sucrose(YG) and maintenance coefficient(m) were 0.417/d,0.619g/g and 0.0206g/(g?d-1) respectively.All these outcomes could give a basis for establishing the suspension cell culture of E.ulmoides and production of the natural active components in large-scale.
3.Effect of adult-onset hypothyroidism on neurogranin expression in hippocampus of adult male mice

Adult male Kunming mice were divided into normal control group,propylthiouracil(PTU) treated group and PTU+T_4 treated group.Neurogranin(Ng)protein expression in the hippocampus after 8 weeks was assayed by Western blotting and immunohistochemical method.Results showed that Ng protein was decreased in the hippocampus of male mice with adult-onset hypothyroidism,suggesting that decreased Ng,which is correctable by T_4,is involved in cognitive dysfunction in adult-onset hypothyroid male mice.

Objective To explore the investigate of X-chromosome-linked inhibitor of apoptosis pro- tein(XIAP)and its effect on chemotherapeutic sensitivity in bladder carcinoma.Methods Using immu- nohistochemistry methods,the expression of XIAP was evaluated in 47 bladder carcinomas and 6 normal bladder tissues.The XIAP gene was transfected into bladder cancer cell line T24 by liposome and the positive clone was screened by G418.Cellular XIAP mRNA level was detected by RT-PCR.The apoptosis of T24 cells was induced by low-dose of mitocycin C(0.005 mg/ml and 0.05 mg/ml,respectively).The in vitro cellular growth activities were assayed by MTT color imetry;and the apoptosis rate was assayed by TUNEL methods. Results The expression rate of XIAP was 78.7%(37/47)in bladder carcinoma samples,with no corre- lation with carcinoma stages and grades(P>0.05).XIAP mRNA level in transfected T24 ceils was signifi- cantly increased by 3.8 times.Treated with 0.005 mg/ml and 0.05 mg/ml of mitomycin C,the growth rates of XIAP transfected T24 cells were increased [(11.60?0.25)% and(16.51?0.87)% ,respectively,P<0.05];and the apoptosis rates were decreased [(10.1?0.2)% and( 11.9?0.2)% ,respectively,P<0.05]compared with those in control cells.Conclusions XIAP is highly expressed in humun bladder car- cinoma samples.Overexpression of XIAP in T24 cells results in decrease in bladder carcinoma cell apoptosis induced by MMC,which may decrease the chemotherapeutic sensitivity of T24 cells.
5.Reconstruction of mandible defect in osteoradionecrosis patients with free fibula osteomyocutaneous flap

Objective To explore the clinical applicating and efficacy of free fibula osteomyocutane- ous flap in mandible defect reconstruction in osteoradionecrosis patients.Methods The mandible defects were reconstructed by free fibula flaps with or without muscle cuff.The soft tissue defects were repaired by skin paddles.Status of osteotomy in fibula and flap survival was recorded.The complication in recipient site and donor site,as well as mouth opening and occlusion were reviewed.Facial contour and chewing function after reconstruction were evaluated.Results Patients were followed up 3-16 months.4 free fibula flaps with muscle cuff and 5 without muscle cuff survived well.The size of mandible defects covered from 6cm to 17cm. And the harvested fibula flaps with length of 8.6-17cm were cut into 3 segments in 2 cases,and 2 segments in 5 cases.Fibula flap was divided into 2 segments and overlapped in 2 cases.No serious complication was oh- served in recipient site and donor site.Satisfying esthetic result and normal occlusiong of heath mandible were obtained in all cases.The degree of mouth opening was 2.5-3.3cm.Fair chewing function was revealed in re- constructive region after prosthesia repaired.Conclusion Free fibula osteomyocutaneous flap is relatively ideal reconstruction material of mandible defect in osteoradionecrosis patients for its high survival rate and well esthetic results.
6.Protective effect of garlic oil given at different time against acute liver injury induced by CCl4.

OBJECTIVETo observe and compare the protective effect of garlic oil against carbon tetrachloride (CCL)-induced acute liver injury.
METHODSThe experiments include 4 preventive groups and 2 therapeutic groups. In every preventive and therapeutic group, the mice were randomized into 6 groups with 15 each, including one negative control group, one solvent control group, one CCl4 model group and 3 garlic oil groups (25, 50, and 100 mg/kg body weight). Before given a single gavage of CCl4 (80 mg/kg), the mice were pretreated with garlic oil by gavage in preventive group 1 (30 days, once daily), preventive group 2 (5 days, once daily), preventive group 3 (ahead of 2 h, once), preventive group 4 (immediately, once) or the vehicle (corn oil, 10 ml/kg) in solvent control group. In therapeutic groups, the mice were gavaged garlic oil 2 h (once, in therapeutic 1) or for 5 days (once daily, in therapeutic 2) after administration CCl. After 24 h of the last administration, blood was collected and centrifuged at 2500 r/min at 4 degrees C for 10 min, and serum was removed to measure ALT and AST activities. The liver was dissected, weighed to calculate the liver coefficient (relative liver weight). At the same time, the liver samples were studied by histological examinations.
RESULTSCompared with negative group, the liver coefficient and the activities of ALT and AST in serum of model group were increased remarkably (P < 0.01). Compared with CCl model group, the liver coefficient and the activities of ALT and AST in serum were decreased significantly (P < 0.01) by garlic oil dose-dependently in each preventive group. Simultaneously, histological assessment showed that garlic oil effectively alleviated hepatocyte injuries induced by CCl4. Comparing the preventive effects of garlic oil in every group, it was better in preventive group 3 than others. However, all indexes and histological examinations in therapeutic group 1 did not show the difference with those of CCl4 model group. In therapeutic group 2, all indexes recovered after 5 d of CCl4 administration.
CONCLUSIONSGarlic oil can prevent acute liver injury induced by CCl4 and the effect is better in ahead of 2 h group than others.

7.Advances of studies on acupuncture reinforcing-reducing of twirling-rotating the needle.

The theories and clinical and experimental studies about reinforcing-reducing manipulation of twirling rotating the needle in recent 15 years are reviewed and a brief account of manipulation methods, mechanisms and effects of reinforcing-reducing manipulation of twirling-rotating the needle is given, and it is indicated that the following several aspects in the studies at present need to be completed. Standardization and establishment of national standard of acupuncture manipulation must be enforced; quantitative parameters of acupuncture manipulation and mechanisms of acupuncture reinforcing-reducing of twirling-rotating the needle need to be further studied.

8.Analysis on indications and dominant diseases of acupotomology.

To analyze the indications and dominant diseases of acupotomology by literature research. The findings reveal that the 64.4% of all literature focused on eight categories of diseases, including the third lumbar vertebrae transverse process syndrome, tendinitis stenosans, cervical spondylosis, heel pain, scapulohumeral periarthritis, external humeral epicondylitis, lumbar disc herniation and osteoarthritis. The 87.5% of all literature focus on diseases of chronic strain of movement system, cervical spondylosis, cervicogenic disease, lumbar disc herniation and osteoarthritis. The indications of acupotomology are various, but not evenly distributed; the dominant diseases are comparatively concentrated. The acupotomology has great potential to treat the indications and dominant diseases. Therefore, acupotomology should be promoted scientifically in the future.

9.Ursolic acid inhibits T-cell activation through modulating nuclear factor-κ B signaling.

OBJECTIVETo investigate the effects of ursolic acid (UA) on T-cell proliferation and activation, as well as to examine its effect on nuclear factor-κB (NF-κB) signaling pathway in T cells.
METHODST-cells isolated from BALB/c mice were incubated with UA at concentrations ranging from 5-30 μmol/L in the presence of phorbol 12-myristate 13-acetate (PMA) or PMA plus ionomycin. The proliferation of T cells was measured by the MTT assay. The expressions of CD69, CD25, and CD71 on T-cell surface were analyzed using flow cytometry. The level of interleukin-2 (IL-2) in the culture supernatant of activated T cells was quantified by enzyme-linked immunosorbent assay (ELISA). The level of phosphorylated IκB-α (p-IκB-α) in total protein and p65, a subunit of NF-κB, nuclear translocation were measured by Western blot analysis.
RESULTSUA in a dose-dependent manner significantly decreased the proliferation and inhibited the surface expressions of CD69, CD25, and CD71 in murine T lymphocytes upon in vitro activation (P<0.01). Significant reduction of IL-2 production was found in activated T cells treated with UA (P<0.01). The PMA-induced increase in p-IκB-α protein was inhibited, and nuclear translocation of p65 from the cytoplasm was blocked by UA.
CONCLUSIONUA is a potent inhibitor for T cell activation and proliferation; these effects are associated with the inhibition of NF-κB signaling pathway.

10.Detection of cytokeratin20 mRNA in peripheral blood from colonic cancer patients by fluorescence quantitative polymerase chain reaction.

OBJECTIVETo investigate the expression level of cytokeratin20 mRNA (CK20 mRNA) of cancer cells in peripheral blood from colorectal cancer patients, detected by fluorescence quantitative polymerase chain reaction (FQ-PCR), and to evaluate its clinical value.
METHODSTo systemically study the reproducibility, quantitative range and amplification efficiency of CK20 mRNA detection by FQ-PCR, analyze and compare the result consistency with conventional RT-PCR and immunohistochemistry, separately. The expression level of CK20 mRNA of cancer cells in peripheral blood was examined in 136 colorectal cancer cases with or without hepatic metastasis.
RESULTSThe within-run and between-run CV of FQ-PCR to assay CK20 mRNA were 3.6% and 5.3%, respectively, quantitative range was from 10(3) copies/ml to 10(8) copies/ml and amplification efficiency was 87.4%. Comparing with traditional RT-PCR and immunohistochemistry, the Kappa value was 0.87 and 0.83, respectively. The expression level of CK20 mRNA of cancer cells in peripheral blood from colorectal cancer patients was (3.52 +/- 1.47) x 10(4) copies/ml, and the expression positive rate was 48.5%. None was found among the 75 cases in the control group. The positive rate of CK20 mRNA of cancer cells in peripheral blood was 9.5%, 25.0%, 48.8% and 87.5% in the patients at Dukes stage A, B, C and D, respectively (P < 0.05). The positive rate of CK20 mRNA was 87.5% in patients with hepatic metastasis and 32.3% in patients without hepatic metastasis (P < 0.05). CK20 mRNA showed a tendency to decline in 35 cases of colorectal cancer within the 1st, 3rd and 5th week after operation. There was no difference among the data of pre-operation cases and on the 1st, 3rd week (P > 0.05), but a significant difference between pre-operation and the 5th week (P < 0.05).
CONCLUSIONFQ-PCR is a rapid and sensitive method for quantitating CK20 mRNA. The expression of CK20 mRNA of cancer cells in peripheral blood from colorectal cancer patients has a correlation with recurrence and metastasis of the tumors. Detection of CK20 mRNA is helpful to monitor hematogenous dissemination of colorectal cancers.
