OBJECTIVE To understand the variance of antimicrobial resistance and the mutations of resistant genes of Mycobacterium tuberculosis isolates in Haiyan area and to be helpful for infection therapy. METHODS For the patients with the positive smears of sputum, the M. tuberculosis was isolated from by sputum. The antimicrobial resistance of the isolates was detected by high and low concentration of antimicrobial agents method. The resistant-genes mutations of rpoB ane katG genes were detected by real-time PCR. RESULTS Among the 131 strains of clinical M. tuberculosis isolates, the total antimicrobial-resistant rate was 14.5%. In the patients after 2 month therapy, the total antimicrobial-resistant rate was 37.8%. The mutations of rpoB ane katG genes were 14.5% and 8.4% in the initially isolated strains, but 39.2% and 25.7% in the strains after 2 month therapy. CONCLUSIONS The drug-resistance of M. tuberculosis isolates in this area existsd before therapy of anti-infection, and,it can increase therapy time. It is important to detect the resistance and the resistant-genes mutations of the isolates.

OBJECTIVE:To esbablish pre-dispensing management mode and computer system in outpatient dispensary to facilitate the practice of hospital pharmaceutical care.METHODS:The establisment and application of the pre-dispensing management mode and computer system in outpatient dispensary were analyzed from aspects of hardware preparation, design of system framework and functional modules of the system.RESULTS:The application of pre-dispensing management system brought down the dispensing error rate and reduced patients' waitting time. The stability of the computer system was the key to ensure smooth operation of the pre-dispensing management mode.CONCLUSION:The practice of the pre-dispensing management mode facilitates the formation of good pharmaceutical care system and is a good dispensing managment mode suitable for sound development of hospital pharmacy.

ObjectiveTo study the effects on growth characteristics of cells mutated by outer space. Methods5 tumor cell lines and 1 bio-pharmaceutical cell line were carried in the recoverable satellite No.18 and returned after 18 days flying in outer space. The cell living systems were utilized in passive carrying. Some survival cells were monocoloned, and the experimental methods such as cell morphological observation,MTT assay and FACS were used for analysis of cell growth characteristics. ResultsThe survival cell rates of tumor cell lines and bio-pharmaceutical cells were markedly different. Compared with the control group, mutated cells appeared to have multiple cell morphological changes and the cell number of G1 phase was increased markedly (P<0.05). Growth rate in mutated cells were varied without specific discipline. ConclusionEffects of outer space on growth characteristics were complicated and multiplex in mutated cells, which were not all heredity in further passage. The differences of the monoclonal cell lines provide the potential to obtain optimizing cell lines by screen.

ObjectiveTo develop the facility of mammalian cell culture in hypothermia for long-term, and to lay the base for carrying mammalian cells to outer space and establishing outer space biomedical research stage. MethodsMultiple cell lines were maintained in the spacial designed hypothermia cell culture facility and carried to space by Shenzhou 4 and No.18 recoverable satellite. Cell morphology, cell multiplication and the ability to be monocloned were observed.ResultsCells were successfully maintained in the hypothermia cell culture facility for up to 26 days without obvious changes of cell morphology. The cells could normally grow, multiple and be monocloned after inoculated in 37 ℃ for 8 h.ConclusionA hypothermia cell culture facility was successfully established, which ensure technically mammalian cells to be carried by space craft and further research on space radiation.

ObjectiveTo validate the feasibility of the outer space carrying system in culturing the CHO(dhfr-)cells in hypothermia, and to observe the effects of this carrying system on the growth characteristics of the CHO(dhfr-) cells.MethodsThe growth characteristics of the CHO(dhfr-) cells were observed and analyzed with cell morphological observation, MTT assay, FCM,3H incorporation and chromosome after the cells were cultured for 25 days in the carrying system. ResultsComparing with the control group, the CHO(dhfr-) cells appeared multiple cell morphological changes, the difference of cell cycle was not significant, the broken chromosome was not seen, the cell growth speed decreased markedly and big molecular biosynthesis increased obviously. ConclusionThe outer space carrying system has no outstanding effects on the survival and heredity of the CHO(dhfr-) cells, so that it can be used in cell carry.

Objective To explore the optimal dose-effect relationship of gene transfer according to the expression of pEGFP in tumor cells under different parameters mediated by microbubble contrast agent and ultrasound in vivo. Methods C57BL/6J mice were inoculated subcutaneously in the middle of the right flank with hepal-6 tumor cells. The mixture of SonoVue and enhanced green fluorescent protein(EGFP)plasmid was injected into the tail vein of each mouse, groups were randomly established according to different output intensity( 1 W/cm2, 2 W/cm2, 3 W/cm2 ), exposure time ( 1 min, 5 min, 10 min) and volume of SonoVue(30 μl,60 μl,90 μl), the expression of EGFP in tumor cells under different parameter was examined by flow cytometry and fluorescence microscopy. Results The expression of pEGFP in tumor cells was significantly higher with the increase of exposure time, output intensity and volume of SonoVue (1 min vs 5 min, P<0.05; 1 W/cm2 vs 2 W/cm2, P<0.05 ; 30/μl vs 60 μl, P<0.05).However, the transfection efficiency of EGFP can not be enhanced continuously with the increasing exposure time, output intensity and volume of SonoVue (5 min vs 10 min, P>0.05; 2 W/cm2 vs 3 W/cm2, P>0.05; 60 μl vs 90 μl, P>0.05).The highest transfection effieieney was achieved under intensities was 2 W/cm2 [(21.02±1.45)%]with 5 min[(23.22±1.91)%] exposed to ultrasound, and 60 /μl SonoVue [ (21.02±1.45) % ].Conclusions Gene transfeetion efficiency changded with the difference parameters.2 W/cm2,5 min and 60 μl SonoVue led to the optimum dose-effect relationship of gene transfer in vivo .

This article introduced professor WANG Shu-chen's experience in picture treating bronchiolitis obliterans syndrome (BOS) after hematopoietic stem cell transplantation with tougue picture as the key link. Patients with BOS showed deficiency of essence of tongue crack, which should be treated by warming kidney yang, replenishing essence and marrow; fester tongue for qi disorder, inflammation caused by fire, which should be treated by regulating qi, and clearing heat; exfoliative fur accumulation, which should be treated with blood stasis and toxin, removing blood stasis and toxin; thick and greasy fur, which should be treated by warming spleen and activating spleen.
