1.Joint Effects of Over Intake of Fluoride and Iodine-Deficiency on Thyroid Glands of Pregnant Mice and Their Offspring

Objective To discuss the effects of high-fluoride intake and iodine-deficiency on thyroid glands of pregnant mice and their offspring. Methods 200 mg/ L sodium fluoride solution was supplied ad.lib. for the adult female Kunming mice before pregnancy and during pregnancy with iodine-sufficiency and iodine-deficiency. The morphological changes and functions of thyroid glands and the body weights of the pregnant mice and their offspring were observed and measured. Results Under the condition of iodine-sufficiency, significantly higher levels of blood T4 in pregnant mice exposed to high-dose fluoride and their offspring were observed compared with those of control group. Asymmetric dimethylarginine(ADMA)is an endogenous inhibitor of nitric oxide synthase(NOS).it reduces nitric oxide production. ADMA is correlated with the risk factors of atherosclerosis, such as hypercholesterolemia, hypertension and hyperglycaemia. Accumulating evidence suggests that a derangement of the NOS pathway plays a critical role in atherogenesis and ADMA may participate in the process.

5.Breeding of Arachidonic Acid Producting Strain with Mortierella Sabellina by Ultraviolet Mutation

In order to gain the strain which has high growth vigor and whose biomass concentration satisfy further fermentation to produce arachidonic acid, we took the yield of microbes olein and arachidonic acid as evaluative index, adopted twice ultraviolet mutation, determined the time of ultraviolet irradiation by single factor expriment, and then the content of the arachidonic acid was measured by GC. The results of this experiment showed that the power of viltalight lamp was 20 W, exposure distance was 30 cm and exposure time was 80 s, lethality of spores of Mortierella isabellina was 76.4%. After twice ultraviolet mutation and repeatedly screen, a mutation of Mortierella isabellina Z80s2-109 whose arachidonic acid concentration in biomass were 3.77 times of the control strains was obtained and genetic stability.

Objective To investigate the influence of all-round intervention on the quality of life and the emotion of cancer patients who have accepted the gamma knife therapy. Method Using health education, psychotherapy and the behavior treatment for 260 patients with malignant tumor who had accepted the gamma knife therapy. Evaluated the quality of life and the emotion of these patients before and after the intervention respectively by using questionnaire of QOL, SCL-90, SDS and SAS. Results The values of indicated the QOL, SCL-90, SDS and SAS before the all-round intervention were 23.12?4.68, 68.12?33.26, 40.12?10.28 and 42.48?12.16 respectively. Nuclear factor-E2 related factor 2 (Nrf2)can protect the normal cells from the damage of oxidants and electrophones,preventing tumorigenesis and metastasis.However,the abnormal activation of Nrf2 in tumor cells can lead to tumor development.Chemicals acting on Nrf2 pathway include activators and inhibi-tors,Nrf2 inhibitors can increase the chemosensitivity of the tumor,providing a new idea for the drug design of high-efficiency and low-toxicity.
8.Development and application of reference materials containing mixed degradation products of amoxicillin and ampicillin.

Reference materials containing mixed degradation products of amoxicillin and ampicillin were developed after optimization of preparation processes. The target impurities were obtained by controlled stress testing, and each major component was identified with HPLC-MS and compared with single traceable reference standard each. The developed reference materials were applied to system suitability test for verifying HPLC system performed in accordance with set forth in China Pharmacopeia and identification of major impurities in samples based on retention and spectra information, which have advantages over the methods put forth in foreign pharmacopoeias. The development and application of the reference materials offer an effective way for rapid identification of impurities in chromatograms, and provide references for analyzing source of impurities and evaluation of drug quality.

Objective To evaluated the effect of hyperbaric oxygenation therapy(HBO)on the RhoA ex- pression and nerve function after transient focal cerebral ischemia in a rat model of middle cerebral artery occlusion. Methods One hundred and twenty-six healthy Sprague-Dawley rats were used and randomly divided into a sham op- eration group(shame group,n=42),a treatment group(n=42),and a control group(n=42).The animal model of middle cerebral artery occlusion(MCAO)was established by using the Zea-Longa method with the animals in the treatment and the control groups,and sham operation was performed with those in the sham group.HBO was applied to the animals in the treatment group.The RhoA protein expression was observed by using immunohistochemistry technique,and the neurological function was evaluated by Bederson's scale at different time points after MCAO.Re- sults(1)Weakly positive expression of RhoA could be located in bilateral cortex and the basal ganglia in the sham group.The expression of RhoA in the treatment group and control group was increased as early as 6 hours after MCAO when compared with that of the sham group,and peaked at 48 h after MCAO and decreased after then,but was still higher than that of the shame group at 7th day to 14th day after MCAO.It was also found that the expression of RhoA of the treatment group was significantly lower than that of the control group(P
10.Influence of cationic cyclopeptide on microstructure and permeability of Caco-2 cell membrane.

The microstructure of cationic cyclopeptide (TD-34) treated Caco-2 cell membrane was observed, and we discussed the relationship between membrane structure and insulin transmembrane permeability. Atomic force microscope (AFM) was used to observe living cell membrane in air condition and tapping mode. Results showed that the surface of Caco-2 cell membrane treated with TD-34 lost its smoothness and nearly doubled its roughness. Apparent permeability coefficients (P(app)) of insulin in Caco-2 cell monolayers increased 2.5 times. In conclusion, AFM can be used to observe microstructure of cationic cyclopeptide treated cell membrane and cationic cyclopeptide enhanced insulin delivery across Caco-2 cell membrane by increasing membrane fluidity.
