Traditional Chinese Drug Research & Clinical Pharmacology 1993;0(01):-
Objective To develop a method for the determination of berberine hydrochloride, palmatine hydrochloride and jatrorrhizine hydrochloride in Yiqi Zhixue Granules. Methods The samples were extracted by ultrasonication with methanol for 30 min,and an ion pair RP-HPLC method was applied to determine three kinds of alkaloids. The column of C18 with temperature at 30 ℃ was used to separate the target components, the mobile phase consisted of the mixed solution of 0.01 mol/L sodium-heptanesulfonate solution and equal volume of 0.01 mol/L potassium dihydrogen phosphate solution(pH being adjusted at 3.1 with phosphoric acid) combined with acetonitrile(70 ∶ 30),the flow rate was 1.0 mL/min,and the detection wavelength was at 345 nm. Results Three kinds of alkaloids were separated perfectly, the average recoveries(n=6)were 100.95 %(RSD=2.10 % ) for berberine chloride,102.14 %(RSD=2.29 % ) for palmatine chloride, and 100.71 %(RSD=2.65 % ) for jatrorrhizine chloride. Conclusion The developed method is demonstrated to be simple, specific and accurate, which can be used to determine the contents of berberine chloride, palmatine chloride and jatrorrhizine chloride in Yiqi Zhixue Granula, and to control the quality of Folium Mahoniae in Yiqi Zhixue Granules.

OBJECTIVETo explore clinical outcomes of posterior malleolar fractures with medial-extension type through posterioromedial and posteriorlateral incision.
METHODSFrom January 2008 to January 2011,25 patients with posterior malleolar fractures with medial-extension type were treated by hollow lag screw. Among them, 15 patients were treated through posteromedial incision, including 9 males and 6 females, aged from 21 to 67 years old with an average of 48.1 +/- 1.3; there were 5 cases with type A, 6 cases with type B and 4 cases with type C,according to Denis-Weber classification. Ten patients were treated by through posterior-lateral incision, including 6 males and 4 females, aged from 23 to 64 years old with an average of 46.9 +/- 1.5; there were 3 cases with type A, 5 cases with type B and 2 cases with type C,according to Denis-Weber classification. Operation time, blood loss, length of incision, times of X-ray exposure and complications of two groups were recorded and compared, Baird-Jackson effective evaluation were applied for evaluate clinical outcomes.
RESULTSAll patients were followed up from 12 to 49 months with an average of 20.6 months. There were significant differences in operation time, blood loss, times of X-ray exposure and complications between two group (P < 0.05). While there was no obvious meaning in clinical outcomes between two groups (P > 0.05).
CONCLUSIONTreating posterior malleolar fractures with medial-extension type through posteromedial approach can expose and fix fracture under direct vision, has advantages of shorter operation time, less X-ray exposure and blood loss, is a good choice of surgical approach.

ObjectiveTo investigate the effects of Aniracetam on learning and memory dysfunction in mice with cerebral ischemia/reperfusion injury. Methods48 mice were randomly divided into control group, sham operated group, model group and Aniracetam group, with 12 mice each. Mice model of cerebral ischemia/reperfusion injury was established by bilateral common carotid artery blocking. The learning and memory function was tested by Morris water maze.ResultsThe latent period in the model group was longer than that in the normal group and the sham operated group (P<0.05). Aniracetam could shorten the latent period significantly as compared with those in the model group (P<0.05). ConclusionAniracetam has effects in improving learning and memory function in cerebral ischemia/reperfusion injured mice.

Objective To investigate the effects of Erigeron breviscapus Hand Mazz(EBH) on learning and memory dysfunction in mice with cerebral ischemia/reperfusion injury. Methods 40 mice were randomly divided into control group, sham operated group, model group and EBH group, with 10 mice each. Mice model of cerebral ischemia/reperfusion injury was established by bilateral common carotid artery blocking. 7 days before and after operation, the mice in EBH group were injected I.p.with EBH injection 0.15 ml/d, while those in other groups were injected I.p. with saline 0.15 ml/d. The learning and memory function was tested by Morris water maze. Results The latent period in the model group was longer than that in the normal group and the sham operated group (P<0.05). EBH could shorten the latent period significantly as compared with those in the model group (P<0.05).Conclusion EBH can improve learning and memory function in cerebral ischemia/reperfusion injured mice.

In this paper, metabolic networks of the Corynebacterium glutamicum GWY020 and the two de-rivatives carrying additional mutations HUI821and GUI089 were established and modified. The concentra-tions of extra-cellular metabolites were determined under sub-steady-state (50 h~52 h) of the batch culture. The metabolic flux distribution maps of the three strains were obtained, compared and analyzed. These re-sults indicate that the introduction of analog supersensitive marker or analog resistant marker skew the metabolic flux towards the formation of L-Arginine. This study revealed the usefulness of the metabolic flux analysis as a tool for verification of existing production strains. The analysis may play an important role in helping us to rationally re-design metabolism for further improvement of fermentation process.

Objective To analyze the effect of N-acetyl-L-cysteine(NAC)on endoplasmic reticulum stress mediated HepG2 cells apoptosis and evaluate the role of NAC in the treatment of liver injury. Methods HepG2 cells were treated with thapsigargin(TG)to establish the model of oxidative endoplasmic reticulum stress mediated apoptosis, and NAC was used to intervene in apoptosis. To evaluate the apoptosis, various methods such as MTT assay, flow cytometry,DNA ladder and Western blot were performed. Results After treated with 2 μmol/L TG for 0,24,36 and 48 hours, the vitality of HepG2 cells decreased. The ratio of apoptotic cells increased along with the prolonged treatment duration of TG(0.7%±0.5%,27.6%±6.3%,29.7%±3.3%,47.9%±3.5% respectively,P<0.05),and the production of reactive oxygen species(ROS)also increased in time-dependent manner(14.0%±0.5%,36.1%±3.0%,38.2%±6.0%,48.3%±12.4%,P<0.05).The HepG2 cells showed typical morphologic change of endoplasmic retieulum stress induced by 2 μmol/L TG after 36 h and 48 h.DNA ladder was observed at the same concentration and time point correspondingly.Endoplasmic reticulum stress mediated-apoptosis was confirmed by Western blot. Both 10 mmol/L and 20 mmol/L NAC could protect ceils from apoptosis. The ratio of apoptotic cells decreased to 14.0%±1.3% and 11.0%±0.3%,respectively.The production of ROS decreased to 34.7%±0.8% and 31.5%±2.9%,respectively.The effect was related to the concentration of NAC.Conclusions As a Ca2+-adenosine triphoshatase inhibitor,TG may disrupt intracellular calcium homeostasis, which can induce endoplasmie reticulum stress and apoptosis.NAC,the precursor of the synthesis of-SH,can directly inhibit the ROS reaction and alleviate liver damage, which may play a role in the treatment of liver failure.

A 2,4 -dichlorophenol degrading Pseudomonas strain GI241-1 was isolated from a soil sample. The dienelactone hydrolase gene, designated as dcpD which encodes dienelactone hydrolase involved in transforming cis-2-chloro-dienelactone into 2-chloromaleylacetic acid, was cloned from this bacterium strain. The gene cloning strategy was to construct genomic library after location of its neighbouring gene by Southem blot and to screen the aim transformant by dot blotting. Sequencing results showed that length of dcpD is 702bp. The sequence of dcpD and the deduced amino acid are different from the relative sequences registered in the GenBank.
