A canine distemper virus strain was isolated from the lung of dog coming from Aksu in Xing Jiang using lung primary M cell during the CDV molecular epidemiological study. It was demonstrated to be a virulent strain of CDV by a series of systematic identification such as morphology , serology neutralization test, canine infection test, and molecular virology test.
2.Percutaneous vertebroplasty in treatment of compression fracture of thoracic and lumbar vertebrae

Objective To discuss the value and experience of the percutaneous vertebroplasty (PVP)in the treatment of vertebral body compression fracture(VCF)in aged osteoperosis.Methods PVP was performed in 44 cases with VCF including 28 with single vertebral compressed fracture,12 with double compressed fracture and four with triple compressed fracture,with 67 vertebrae,for clinical and radiologieal evaluation.Results The mean follow-up was 15 months(4-23 months).There could be seen immediate relief of pain in 40 cases,out-of-bed activities at operation day in 19 and out-of-bed activ- ities at second day after operation in 25.Postoperative X-ray showed uniformly distributed bone cement in the vertebral,without leakage.Conclusion PVP is a recommendable method for VCF,for it has ad- vantages of pain relief,vertebrae stabilization,minimal invasion and minor complications.
3.Hepatitis C virus F protein-mediated inhibition of hepatoma cell proliferation.

OBJECTIVETo investigate the biological function of the hepatitis C virus (HCV)-encoded F protein in hepatocytes.
METHODSThe full-length F gene was amplified by PCR from HCV genotype 1a and cloned into plasmid pSEB-3Flag by restriction enzyme digestion and ligation. Hepatoma cell lines, Huh7 and SMMC7721, were transfected with the resultant recombinant pSEB-3Flag-F or the original pSEB-3Flag (negative control) and screened with the selective antibiotic, blasticidin. Stable F gene and protein expression was verified by RT-PCR analysis. Analysis of cell growth and cell cycle was carried out by MTS assay, crystal violet staining and flow cytometry.
RESULTSHuh7 and SMMC7721 cells transfected with pSEB-3Flag-F plasmid (Huh7-F and SMMC7721-F, respectively) uniquely expressed the F gene and protein. The Huh7-F and SMMC7721-F cells showed significantly decreased proliferation rates, compared to the respective control groups. A similar HCV F-mediated growth-inhibiting activity was observed by the cell viability assay. Furthermore, cell cycle analysis revealed that the S-phase distribution was much lower in Huh7-F (47.12%) and SMMC7721-F (30.75%) cells than in the respective controls (55.35% and 33.23%, respectively) (P less than 0.05).
CONCLUSIONStable expression of the HCV F gene reduced the in vitro proliferation rate of hepatoma cell lines, indicating that the F protein may function as a growth inhibitor of infected cells.

4.Theory of local inflammatory infiltration and irritation in the pathogenesis of acquired cholesteatoma.

OBJECTIVETo explore the deep pathogenesis of acquired cholesteatoma.
METHODSThe temporal bone slides of 12 ears with retraction pocket were histopathologically studied under microscope, especially focusing on the location of retraction pocket and inflammatory pathology in the local middle ear cavity next to retraction pockets. The temporal bone slides of 11 ears with acquired cholesteatoma were histopathologically observed and 33 cases diagnosed as acquired cholesteatoma were clinically observed observed in the local middle ear cavity next to the part without retraction pocket of eardrum. The results of pathological observation of the temporal bone slides with acquired cholesteatoma and clinical observation during operation for acquired cholesteatoma show that cholesteatoma invade mainly and occupied the ossicular chain eara of the middle ear cleft.
CONCLUSIONIn the pathological process of otitis media, the intractable pathological changes in the ossicular chain area can inward adhere posterosuperior quadrant or pars flaccida of the eardrum to form retraction pocket and permanently infiltrate the external squamous epithelial layer of retraction pocket to excessively proliferate and keratinize, leading to formation of acquired cholesteatoma.

5.Detection of early organ dysfunction for the selection of treatment strategy on severe acute pancreatitis..

OBJECTIVETo investigate the severity related influencing factor and treatment strategy of severe acute pancreatitis with early organ dysfunction.
METHODSFrom July 2007 to December 2008, 167 patients with severe acute pancreatitis were treated in the Surgical Department of Ruijin Hospital. The relationships between the happening of early organ dysfunction and outcome of the patients were observed, with operative or nonoperative treatment strategy.
RESULTSAmong 167 patients, 68 patients have early organ dysfunction, in which 39 with single organ dysfunction and 29 with multiple organ dysfunction. The early organ dysfunction were involved in 47.1% in cardiovascular system, 35.3% in lung and 29.4% in kidney. Aging (P < 0.05) and higher APACHE II score (P < 0.05) predicted a poor prognosis, which were benefit from early operation.
CONCLUSIONSThe mortality of the patients with SAP is related to age, and the degree of organ dysfunction as well. In the first phase of the disease, the selection of operation depends on the trends and the degree of early organ dysfunction before infected necrosis happens, with the aid of SOFA score as a scale.

6.Construction and rescue of infectious cDNA clone of pigeon-origin Newcastle disease virus strain JS/07/04/Pi.

Based on the complete genome sequence of pigeon-origin Newcastle disease virus strain JS/07/04/ Pi(genotype VIb), nine overlapped fragments covering its full-length genome were amplified by RT-PCR. The fragments were connected sequentially and then inserted into the transcription vector TVT7/R resulting in the TVT/071204 which contained the full genome of strain JS/07/04/Pi. The TVT/071204 was co-transfected with three helper plasmids pCI-NP, pCI-P and pCI-L into the BSR cells, and the transfected cells and culture supernatant were inoculated into 9-day-old SPF embryonated eggs 60 h post-transfection. The HA and HI tests were conducted following the death of embryonated eggs. The results showed that the allantoic fluids obtained were HA positive and the HA could be inhibited by anti-NDV serum which indicated that the strain JS/07/04/Pi was rescued successfully. The rescued virus rNDV/071204 showed similar growth kinetics to its parental virus in CEF. The successful recovery of this strain would contribute to the understanding of the host-specificity of pigeon-origin NDV and to the development of the novel vaccines against the NDV infection in pigeons.

8.Primary clinical result of digital template as navigation to supper cervical pedicle instrumentation.

OBJECTIVETo observe the primary clinical result of digital template as navigation to the upper cervical pedicle instrumentation.
METHODSCT scan of upper cervical vertebrae was performed. 3-D model of upper cervical vertebrae was reconstructed by software Amira 3.1 and was preserved in STL format. Then 3-D model was run in software UG Imageware 12.0, the best pedicle channel was extracted according to the reverse engineering principle. A virtual navigational template was established according to he lamina anatomic trait, and the best pedicle channel. The virtual vertebrae and navigational template were manufactured using rapid prototyping. The navigational template was sterilized and used intra operative to assist with the placement of pedicle screw. The Accuracy of screw placement was confirmed with postoperative X-ray and CT scanning.
RESULTSThe digital navigational template had been established and used in the 3 cases, the good trajectory of cervical pedicle had been showed by the CT scan of post operation. There were not complications of related pedicle screw insertion.
CONCLUSIONSA novel method of upper cervical pedicle location using Reverse Engineering and rapid prototyping has been developed; the navigational template is found to be highly accuracy and has great expectation.

9.Effects of jing'an capsule on rat sperm quality.

OBJECTIVEThe effects of Jing'an capsule on the quality of rat sperm were studied to supply data for its safe clinical use.
METHODSEighty SD male rats were evenly and randomly divided into three test groups and one control group. Then the test groups were continuously given Jing'an capsule at different dosages: 557.1 mg/kg, 5,571 mg/kg or 11,420 mg/kg and the control group was given starch (20 g/L). Sixty days later, one of the epididymides, the sperm density was made and the sperm motility and morphology were investigated. The data obtained were analyzed by ANOVA, chi 2 test and Kruskal-Wallis test.
RESULTSThe sperm density, motility and morphology were variable at different dosages. Compared with the control, the sperm density increased significantly at the dosage of 557.1 mg/kg(P < 0.05), and a significant decrease was observed in the sperm density and motility (P < 0.05) at the dosage of 11,420 mg/kg. Although the rate of abnormal sperm morphology decreased at the dosage of 557.1 mg/kg and increased at the dosage of 5,571 mg/kg or 11,420 mg/kg compared with the control, there was no statistic significance(P < 0.05).
CONCLUSIONSA low dosage of Jing'an capsule might ameliorate the quality of sperm, while a high dosage could do damage to sperm.

10.Protective effect of essential oils from Blumea balsamifera on sunburn induced by UVB in mouse skin

AIM To study the protective effect of essential oils from Blumea balsamifera (L.) DC.(BBO) on UVB-induced sunburn in mouse skin and its mechanism of action.METHODS The model for sunburned mouse skin was established by acute UVB irradiation.Essential oils from B.balsamifera were applied to the surface of wound for external use.The pathological changes of sunburned skin tissue were observed by hematoxylin-eosin HE) staining.The activity of superoxide dismutase (SOD),and the contents of malondialdehyde (MDA) and glutathione (GSH) were measured.The levels of 8-hydroxy-desoxyguanosine (8-OHdG),interleukin-6 (IL-6) and nuclear factor kappa-B (NF-κB) in epidermis were detected by ELISA.Additionally,the expressions of tumor necrosis factor-alpha (TNF-α),P53 tumor suppressor protein and proliferation cell nuclear antigen (PCNA) were evaluated by immunohistochemistry (IHC).RESULTS Compared with the model group,treatment with essential oils from B.balsamifera significantly reduced the thickness of epidermis,and the activity of SOD and the contents of MDA,GSH in mouse skin were restored.In addition,the essential oils from B.balsamifera resulted in a significant decrease in levels of 8-OHdG,IL-6 and NF-κB,and an inhibition in expressions of P53 and PCNA.CONCLUSION The essential oils from B.balsamifera can alleviate UVB-induced sunburn.Its mechanism is related to enhanced antioxidant power,inhibited NF-κB signal passway,down-regulated release of IL-6 and reduced levels of 8-OHdG,PCNA.
