1.Research on the physical components stimulated by rotating grating using functional magnetic resonance imaging

AIM: Using functional magnetic resonance imaging, to inspect v sual cortex physical reactions stimulated by rotating grating, and to its components. METHODS: On 1.5T MR scanner, GRE-EPI imaging sequence was carried on 9 h discover ealthy volunteers, the visual cortex response data were processed after delinearation by SPM99.RESULTS: Different components of rotating grating excited different areas of the visual cortex. Dramatic response in the central part of the occipital lobe, which was related to white light stimuli, located at primary visual cortex. Response area in bilateral Broadaman 19 areas was related to vision-motion function. And weak response area in the central part of the occipital lobe was related to shape perception.CONCLUSION: Rotating grating conclude plenty of visual information, and it excites different areas of the optic center as a stimuli. fMRI is a valuable equipment to study the physiology of visual cortex.
2.Identification of senkyunolide I in Guanxinning Injection chromatographic fingerprint by GC-MS and HPLC-PDA-MS-MS

AIM: To identify an unknown peak in Guanxinning Injection(Radix et Rhizome Salviae Miltiorrhizae,Rhizoma Chuanxiong) chromatographic fingerprint. METHODS: The elution(44-46 min) separated and collected by HPLC was analyzed by GC-MS and HPLC-PDA-MS-MS. RESULTS: The unknown peak was corresponding to senkyunolidel I. CONCLUSION: It is quick method to identify the reported chemical in herbal medicine based on a small quantity of sample by GC-MS and HPLC-PDA-MS-MS.
3.Real-time three-dimensional echocardiographic assessment of left ventricular remodeling index and systolic function in patients with coronary artery disease

Objective To explore left ventricular remodeling index(LVRI),systolic function and their correlation by real-time three-dimensional echocardiography(RT-3DE)in patients with coronary artery disease(CAD).Methods According to coronary artery angiography,one main vessel stenosis no less than 50% was defined as positive standard,RT-3DE data of 24 patients with left circumflex coronary artery or/and right coronary artery stenosis(LCA/RCA group),21 patients with left anterior descending coronary artery stenosis(LAD group),27 patients with double or triplex coronary arteries stenosis which must included LAD(vessels group)and 22 normal controls(NC group)were acquired.Left ventricular end-diastolic volume(EDV),left ventricular end-diastolic epicardial volume(EDVepi),left ventricular ejection fraction(EF)were automatically measured by RT-3DE analysis software;left ventricular mass[1.05×(EDVepi-EDV)],LVRI(left ventricular mass/EDV)were calculated.The inter-group differences of LVRI,EF and intra-group correlations between LVRI and EF were analyzed.Results LVRI and EF significantly decreased in order(P<0.05 or P<0.01);and their correlation coefficents were r=0.10(P>0.05),0.86(P<0.01),0.83(P<0.01)and 0.77(P<0.01)in NC group,LCA/RCA group,LAD group,vessels group,respectively.Conclusions LVRI measured by RT-3DE can assess left ventricular remodeling,and reflect left ventricular systolic function in different CAD,which can be as a new approach for evaluating left ventricular remodeling in clinic.

5.Clinical and imaging characteristics of Percheron artery infarction with unilateral embryonic posterior cerebral artery

Objective Percheron artery is an uncommon anatomic variant. Percheron artery infarction with unilateral embryonic posterior cerebral artery ( PCA ) was rarely reported.The aim of this study was to characterize the clinical and imaging patterns of Percheron artery infarction with the unilateral embryonic PCA for early diagnosis and treatment.MethodsClinical and imaging data of 2 patients with Percheron artery infarction were reviewed retrospectively.ResultsTwo patients presented acute coma and one had paroxysmal blurred before coma.On neurological examinations,one patient had vertical gaze palsy besides two were unconsciousness. MRI showed symmetrical high signal intensity on diffusion weighed imaging (DWI)in bilateral paramedian thalami in two patients and a distinct pattern of V-shaped hyperintensity on DWI andFLAIR was present on the midbrain in one patient. Digital subtraction angiography and magnetic resonance angiography demonstrated unilateral embryonic PCA in two patients.ConclusionsThe classical clinical symptoms,symmetrical high signal in bilateral paramedian thalami and V-shaped sign in midbrain on DWI can improve recognition,evaluation and management of Percheron artery infarction.The unilateral embryonic PCA may be underlying risk factor of Percheron artery infarction.
6.DNA damage in peripheral blood lymphocytes of patients with autosomal dominant polycystic kidney disease

Objective To detect DNA damage of peripheral blood lymphocytes in patients and family members of autosomal dominant polycystic kidney disease (ADPKD),and to study the effect of irradiation on genomic stability of lymphocytes. Methods Before and after 0.5 Gy radiation dose,single-cell gel electrophoresis (SCGE) was employed to analyze DNA damage of lymphocytes in 10 ADPKD patients (group A),3 members without clinical symptoms of a ADPKD family (group B) and 20 healthy control people (group C).The damage was estimated based on the content of DNA in tail (TDNA%) with comet analysis software (CASP). Results Both before and after irradiation,the TDNA% (8.85%±0.14%,14.84%±0.77%) and the value-added (6.00%±0.77%) of TDNA% of group A were significantly higher than those of group C (7.50%±0.37%,12.46%±0.26%,4.96%±0.44%) respectively.There were no significant differences between group B and group C or group A and group B.While 1 person in group B had higher TDNA% as compared to group C both before and after irradiation. Conclusions The lymphocytes of ADPKD patients are more sensitive to ionizing radiation as compared to healthy people.The genomic instability in ADPKD patients or member of ADPKD family may trigger cystic formation in multi-organs when exposing to environmental agents. SCGE may provide a new approach to elucidate the pathogenesis and prognosis of ADPKD.
7.A method for isolated culture of bone microvascular endothelial cells of human femoral head.

OBJCETIVETo investigate the method of separation of culture of bone microvascular endothelial cells (BMECs) of human femoral head in vitro.
METHODSFrom October 2013 to January 2014,15 femoral heads without pathologic change from patients resected during hip replacement were selected involving 2 males and 13 females with a mean age of 71.2 years old ranging from 38 to 92. Cancellous bone in femoral head was bited into broken bone grain and transfered into medium in aseptic contidion. Cells were isolated by the methods of enzymic digestion and density gradient centrifugation,purified by differiential attachment. The characteristics of cells was observed by inverted microscope. vWF and CD31 immunofluorescence analysis was applied for identification of cells.
RESULTSThe number of cells was positively correlated with patients' age after 24 hours in primary culture. The older patients had the less cells numbered. After 4 to 5 days' culture, primary cells appeared short spindle,polygon shaped and cobblestone-like morphology. After 7 to 10 days' culture, primary cells proliferated densely, became fusion, arranged in swirl, and contact inhibition appeared significantly. Immunofluorescence staining revealed the cells were 100% positive for vWF and CD31, and it showed that the cultured cells were BMECs.
CONCLUSIONIt was a simple, steady, effective method with good reproducibility, by which highly purified human BMECs can be obtained.

Objective To analyse the high-dose dexamethasone suppression test(HDDST)-related differences in the clinical and biochemical features of the patients with Cushing's disease Methods Cases were drawn from 60 consecutive patients with Cushing's disease,who were then divided into two groups according to the response to the HDDST.The clinical and biochemical features between two groups were compared.Results(1) Of the 60 patients with Cushing's disease,23.3%(14/60)of patients(group A)did not yield results of suppression with the HDDST,and the others(group B)did.No difference was found in the age[(33.8?10.4 vs 36.2?11.2)years]and duration of illness[(2.1?1.6 vs 3.9?3.1)years]between two groups.(2)In clinical features,the patients in group A were more likely to have edema of lower limbs(64.3% vs 32.6%),hypokalemia (71.4% vs 28.3%),secondary diabetes(57.1% vs 26.1%)and purple striae(85.7% vs 54.3%,all P
9.Study on the extraction of the active component in Atractylode macrocephala with ultrasonic wave technology optimized by the response surface method.

OBJECTIVETo create Atractylode macrocephala inspissation decoction pieces. The effect of ultrasonic wave on extraction of the active components in A. macrocephala was studied in a water solution.
METHODThe factors including the ratio of material to liquid, ultrasonic power, ultrasonic time, soaking time, particle size etc, were studied. The best extraction method was found through the response surface method.
RESULTThe best extraction method was found as follows: the granularity of material 0.1 mm, the repetition times of ultrasonic process 3 times, the soaking time before the ultrasonic process 30 min, the ratio of liquid to material 10:1, the soaking time after the ultrasonic process 2.6 h, the time of the ultrasonic wave 15.5 min, the power of the ultrasonic wave 531 W, the rate of reservation of active components 88.5%, the rate of inspissation 1.6.
CONCLUSIONThe ultrasonic wave can used in the extraction of the active components in A. macrocephala and a model equation that can be used to predict the experiment was get through the response surface method.

10.Study of the gene expression pattern in the Kkay mouse with type 2 diabetes by genomic-scale gene expression analysis.

OBJECTIVETo study the genes differentially expressed in the liver of Kkay diabetic and normal mice by genomic-scale gene expression analysis.
METHODScDNA microarray chips containing 8,192 cDNAs were used to explore the gene expression pattern of Kkay mouse liver.
RESULTSOne hundred and fifty-four genes were screened out, including 68 complete cDNAs and expressed sequence tags, and among them 40 genes were up-regulated and 114 genes were down-regulated respectively.
CONCLUSIONMost of the gene expression analysis results were consistent with previous study, and the gene expression pattern of Kkay mouse based on cDNA microarray could be used for high-throughout screening out the genes associated with type 2 diabetes.
