1.Anti-inflammation Effects of Cacumen Biotae Water Decoction on Mice with Auricle and Abdominal In-flammation

OBJECTIVE:To study the anti-inflammation effects of cacumen biotae water decoction on mice with auricle and ab-dominal inflammation. METHODS:Mice were randomized into a normal control group,a model group,a positive control group (0.25 g/L aspirin)and cacumen biotae high-dose,medium-dose and low-dose [1,0.5 and 0.25 g(medicinal materials)/L] groups, respectively marked as groups A,B,C,D,E and F,with 10 mice in each group. The mice were given drugs ig for 5 consecutive days. The models of auricle and abdominal inflammation were established for all groups except the normal control group. The con-tents of tumor necrosis factor α(TNF-α)and interleukin 1(IL-1)in the serum of mice with auricle inflammation were determined, and the swelling degree and swelling rate were calculated. The contents of TNF-α,IL-1 and acid-sensing ion channels 1a(ASIC1a) in the liver tissues of mice with abdominal inflammation and the concentration of proteins in the peritoneal lavage fluid(ΔA)there-of were detected. RESULTS:For mouse models of auricle inflammation,the contents of TNF-α and IL-1 in serum in group B were higher than in group A;the swelling degree and swelling rate and the contents of TNF-α and IL-1 in groups C,D,E and F were lower than in group B,and those of group E and F were higher than in group C. There were statistical differences(P<0.05). For mouse models of abdominal inflammation,the contents of TNF-α,IL-1 and ASIC1a in liver tissues and ΔA in group B were higher than in group A;the contents of TNF-α,IL-1 and ASIC1a as well as ΔA in groups C,D,E and F were lower than in group B. There were statistical differences(P<0.05). CONCLUSIONS:Cacumen biotae water decoction has anti-effect on acute auricle and abdominal inflammation by a mechanism which may be related to decreasing TNF-α,IL-1 and ASIC1a.
2.Effects of exercise and low-fat diet on the TNF-alpha expression of insulin resistance in rats.

OBJECTIVETo investigate the effects of exercise (EX), low-fat diet (LFD) and their combination intervention on the tumor necrosis factor-alpha (TNF-alpha) expression of insulin resistance in rats.
METHODSOne hundred and thirty male rats randomly assigned to 2 groups: Control (CON)-10 rats consuming a low-fat diet; HFD-120 rats consuming a high-fat diet (HFD). The dietary regimen was sustained for 8 wk, at which point the 40 HFD group rats gaining the most weight were referred to as the obese rats. Glucose tolerance was assessed by an oral glucose tolerance test (OGTT). During the procedure of OGTT, the blood was drawn for insulin assay (insulin release test). The whole body insulin resistance was assessed by glucose-insulin index. The obese HFD group rats were randomized into one of four intervention groups: HFD-sedentary (HFD-SED), HFD-exercise(HFD-EX), low fat diet-SED (LFD-SED), LFD-EX. Ex rats performed 8-wk exercise training on a motorized treadmill. The CON group had access to low-fat diet for another 8 wk. After 8 wk of exercise and low-fat diet intervention, the OGTT and insulin release test were performed again. To use ELISA technique for detecting TNF-alpha in soleus muscle and adipose tissue.
RESULTSAfter being fed high-fat diet for 8 wk, glucose-insulin index in the HFD group were significantly greater than that in CON group (P < 0.01). After 8-week exercise and low-fat diet intervention, glucose-insulin index in HFD-SED group was significantly greater than that in CON group (P < 0.01). The index in three intervention groups was significantly less than that in HFD-SED group (P < 0.01) . TNF-alpha content in adipose tissue and the soleus muscle for HFD-SED group was significantly greater than that in CON group (P < 0.01). Three intervention groups were significantly less than HFD-SED group (P < 0.01).
CONCLUSIONExercise and low-fat diet interventions can decrease the TNF-alpha expression in insulin resistance rat.

3.Determination of Paeoniflorin in Qisheng Capsule

Objective To establish a content determination method for paeoniflorin in qisheng capsule. Methods The quantitative analysis of paeoniflorin in qisheng Capsule was carried out by high-performance liquid chromatography ( HPLC) . The chromatographic separation was achieved by using a Kromasil C18 chromatographic column (4. 6 mm×250 mm,5 μm) with a mobile phase consisting of methanol,water (1585) at a flow rate of 1. 0 mL·min-1 and 230 nm detection wavelength. Results The linear range was 2. 5-12. 5 μg·mL-1( r =0. 999 9). The average recovery and RSD of the method were 99. 97%and 0. 94%. Conclusion The method is accurate,specific,reproducible,which can effectively be used in quality control of paeoniflorin in qisheng capsule.
4.Application of BHP9504 fluorescent analytic instrument

Objective To introduce the clinical application of BHP9504 fluorescent analytic instrument.Methods The chemical immunofluorescent test was used in high-precision,high-stability photo-signal test by photon counting technology.Results Photo-signal test could examine strong photo-signals in dark background so that it was able to improve sensitivity of experiments.Conclusion This technique widens the range of application and reduces specimen amount.

6.Determination of Total Fatty Acid Esters of Chloropropanols in Edible Vegetable Oils by Gas Chromatography-Mass Spectrometry

A method was established for the simultaneous determination of the total fatty acid esters of chloropropanols in edible oils by gas chromatography-mass spectrometry combined with isotope dilution technology. The samples were hydrolyzed with sodium methylate-methanol, and then purified by diatomite cartridge. After being derivatized with heptafluorobutyrylimidazole ( HFBI ), the target analytes were determined by GC-MS with the deuteriumchloropropanols esters as the internal standards. An excellent linear correlation in the range of 0. 050-2. 000 mg / L was acquired for 3-monochloropropane-1,2-diol (3-MCPD) esters, 2-MCPD esters, dichloropropan-2-ol (1,3-DCP) esters and 2,3-dichloropropan-1-ol (2,3-DCP) esters, with all the correlation coefficients (r) higher than 0. 9995. The limits of detection (LODs) for 3-MCPD esters, 2-MCPD esters, 1,3-DCP esters and 2,3-DCP esters were 0. 015, 0. 015, 0. 030, and 0. 030 mg / kg, respectively, and the limits of quantitation (LOQ) were 0. 050, 0. 050, 0. 100, and 0. 100 mg / kg, respectively. The average spike recoveries of the four kinds of chloropropanols esters in blank extra virgin olive oil matrix were typically in a range of 87. 0% -110. 5% with the relative standard deviations (RSDs) less than 10. 1% . The detection rates of 3-MCPD esters, 2-MCPD esters, 1,3-DCP esters and 2,3-DCP esters in 74 edible oil samples were 94. 6% , 63. 5% , 5. 4% , and 0% , respectively. The contamination levels of 3-MCPD esters, 2-MCPD esters and 1,3-DCP esters were in the range of not detected (ND) to 10. 646 mg / kg, ND to 3. 617 mg / kg and ND to 0. 089 mg / kg, respectively. This method is accurate and rugged for the simultaneous determination of total fatty acid esters of chloropropanols in edible vegetable oils.
7.Polyketide Synthases Screening from Sponge-associated Culturable Microorganisms

PKS gene was screened by PCR from thirty strains of spone-associated bacteria including twenty-one actinomycetes isolated from Craniella anstrialiensis and nine bacillus isolated from Dysidea avara in the South China Sea.As a result,a 669 bp KS domain gene was successfully amplified from Bacillus C89.BLAST analysis showed that the KS domains were most closely related to the KS sequences of Bacillus subtilis subsp.subtilis str.168 with 96% similarity.Phylogenetic analysis demonstrated the KS domain belong to trans-AT KS domains.This study demonstrated the existence of PKS gene in bacteria associated with sponge Dysidea avara for the first time,and provided proof for the hypothesis that sponge-associated bacteria are perhaps the true producers of many novel bioactive compounds in sponge.Meanwhile,this study lays a basis for the microbial screening for polyketide compounds production.
8.Antimicrobial Resistance of Escherichia Coli from Different Clinical Samples

Objective:To investigate the antimicrobial resistance spectrum of Escherichia coli isolated from different clinical sam-ples to guide the nosocomial infection control and rational use of antibiotics. Methods:Escherichia coli from clinical samples were isola-ted and tested by microbiological assay system of BD Phoenix 100 and susceptibility test card in 2013. The drug resistance was analyzed and compared retrospectively. Results: There were 936 strains of Escherichia coli isolated from our hospital in 2013. Escherichia coli was resistant to common antibiotics at high level. Escherichia coli strains isolated from different samples varied in the drug susceptibili-ty. The resistance rate of the strains from sputum samples was higher than that from the other samples. Conclusion:The status of drug resistance of clinical isolated Escherichia coli to antibiotics is very serious. Escherichia coli isolated from different samples show different drug resistant features, and the antimicrobial therapy should be considered according to the antimicrobial susceptibility tests.
9.The relationship between apoptosis and neurological impairment in experimental intracerebral hemorrhage in rats

Objective To investigate the relationship between the apoptosis,expressions of Bcl-2 and Bax protein in perihematomal brain regions of rats and neurologic dysfunctions after intracerebral hemorrhage (ICH). Methods Seventy Sprague-Dawley rats were divided randomly into two groups:an experimental group and a control group.A model of ICH was established by injection of 0.5 U bacterial collagenaseⅦinto the caudate nucleus in the rats.Neurological impairment was evaluated at 6 h,12 h,24 h,48 h,72 h,7 d and 14 d after 1CH,respectively, before the rats were sacrificed.TUNEL method was used to detect apoptosis,and SP method to detect expressions of Bcl-2 and Bax protein in the perihematomal brain tissues.Results Neurological impairment occurred in all the rats after ICH,and peaked at 48 h after ICH.The apoptosis and expressions of Bcl-2 and Bax protein were peaked at 48 h,6 h and 48 h after ICH,respectively.Conclusion The degree of the neurological impairment after ICH is parallel to that of the apoptosis.Apoptosis may play an important role in neurological impairment after ICH.
10.Intervention effects of qingre jiangya capsule on brain hippocampus of spontaneously hypertensive rats based on metabonomic research.

Thirty SHRs were obtained randomly to hypertension, model group, captopril group and Qingre jiangya capsule group. Ten Wistar rats were used as control group. The hippocampus tissue was removed to explore the damage of spontaneously hypertensive rats (SHR) and the protective effect of Qingre jiangya capsule after continuously administered for 14 days. And then the data were processed by principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA). The research results revealed captopril group was significantly different from the other three groups. The classification of other three groups is also very clear after captopril group removed. This suggested that Qingre jiangya capsule could improve the overall metabolism compared with captopril. Four metabolites were identified: dimethylglycine, glycerophosphocholine, aldosterone and noradrenaline. Hypertension hippocampus damage may mainly be expressed in tyrosine metabolism, aldosterone-regulated sodium, vascular smooth muscle contraction reabsorption, and Qingre jiangya capsule could reverse the hippocampus tissue damage of SHR.
