1.Clinical observation of Esculin and Digitalisglycosides eye drops and acupoint massage on patients with video display terminal asthenopia

? AlM: To evaluate the efficacy of Esculin and Digitalisglycosides eye drops and acupoint massage alone or combination of both on video display terminal ( VDT ) asthenopia.
?METHODS: Totally, 224 patients with VDT asthenopia were randomly divided into eye drops group ( n = 56 ) received the intervention by Esculin and Digitalisglycosides eye drops, acupoint massage group (n=56) received the intervention by acupoint massage around the eyes, and combination group ( n=56 ) received the intervention by combination of both and control group received the intervention by ocular hygiene instruction. The effect was evaluated after 4wk. The symptoms curative effect index, tear film break-up time ( BUT) , Schirmer l test ( Slt) and corneal fluorescein staining score were as the indexes of evaluation.
? RESULTS: The symptoms curative effect index of combination group (78. 96±5. 29)% was higher than that of eye drops group (69. 55±6. 23)% and acupoint massage group (71. 15±6. 41)%, which of after both groups was higher than that of control group ( 33. 01±4. 26 )% ( P<0. 01). The main effects of Esculin and Digitalisglycosides eye drops and acupoint massage and its interations for the curative effect index were statistically significant ( P<0. 01). BUT and Slt of after intervention in eye drops group were significantly higher than that of before intervention, acupoint massage group and combination group (P<0. 05); After intervention, BUT of combination group was higher than that of eye drops group, acupoint massage group and control group; Slt of control group was lower than that of eye drops group, acupoint massage group and combination group.
? CONCLUSlON: Esculin and Digitalisglycosides eye drops and acupoint massage could improve symptoms and ocular physiology in patient with VDT asthenopia, and the effects of the combinative use of both are synergistic.
2.Investigation of radiation exposure protection of cardiovascular intervention

Objective To investigate the radiation doses in different areas around the operation table during intervention procedure and to study the effect of X-ray protection equipments.Methods A total of 37 procedures(12 diagnostic cardiac angiographies and 25 percutaneous coronary intervention therapy) were investigated.The detection of X-ray was carried out in 2 separate groups.The first group included the operators,assistants and nurses.There were 6 detecting points on each person resulting in a total of 18 detecting points.The second group consisted of 10 detecting points on different areas of the patients and the protecting lead shields around.The measurement of X-ray doses was performed by the specialist from the radiation protection agency.Results The doses of X-ray was directly proportional to the duration of exposure.The X-ray doses varied among the 28 detecting points.The highest amount of radiation were detected on the left arm of the operator and on the left axilla and the back of the patients.Above 90% of radiation could be protected by lead shields and lead clothes.Conclusion Lead protective products available at precent are efficient in protecting radiation exposure during cardiovascular intervention procedures.More attention still need to be paid to certain highly exposed areas.
3.Construction and identification of a lentiviral vector harboring RNAi sequence targeting the human high mobility group A1 gene

Objective: To construct and identify a lentiviral vector harboring RNAi sequence targeting the human high mobility group A1(HMGA1) gene.Methods: The effective sequence of siRNA targeting the HMGA1 gene confirmed in our previous study,the complementary DNA containing both sense and antisense Oligo DNA of the targeting sequence was designed,synthesized and cloned into the pGCL-GFP vector diced by the restriction enzyme of HpaⅠ and XhoⅠ,which contained the U6 promoter and green fluorescent protein(GFP).The resulting lentiviral vector containing HMGA1 shRNA was named LV-sh HMGA1 and confirmed by PCR and DNA sequencing.A total of 293T cells were cotransfected with LV-sh HMGA1,pHelper 1.0 and pHelper 2.0.All the virus stocks were produced by Lipofectamine2000-mediated transfection.The titer of the virus was tested according to the expression level of GFP.Results: PCR analysis and DNA sequencing demonstrated that the RNAi sequence targeting the human HMGA1 gene was successfully inserted into the lentiviral vector.The titer of the recombinant lentiviral vector was 5?107 TU/ml.Conclusion: The successful construction of the lentiviral vector of HMGA1 has prepared the ground for further studies on the functions of the HMGA1 gene with the RNAi technique.
4.Inhibition of colorectal cancer cell adhesion by cimetidine

Objective To investigate the effect of Cimetidine on the adhesion between colorectal cancer LOVO cells and endothelial ECV304 cells;and study whether Cimetidine can inhibit the expression of E-selectin in ECV304 cells. Methods Cellular uptake of rose Bengal stain was used to measure the adhesion between LOVO cells and ECV304 cells. Enzyme-linked immunosorbent assay(ELISA)and flow cytometry (FCM, indirect fluorescence staining and labeling)were used to detect the expression of E-selectin. Results ECV304 cells were exposed to different concentrations of Cimetidine. Both the adhesion between LOVO cells and ECV304 cells and the levels of E-selectin significantly decreased with increasing concentration of Cimetidine(P =0. 001 and 0. 001 respectively). Conclusion Cimetidine can inhibit the adhesion of human colorectal cancer LOVO cells on endothelial ECV304 cells by blocking E-selectin expression. Our observations indicated a potential of anti-adhesion therapy using Cimetidine in cancer treatment.

Wearable technology, widely applied in health and medical field, has become an important part of in-telligent healthcare and medicare. However, the data security and privacy protection of wearable health and medi-cal devices are greatly concerned and need to be solved. The characteristics of data security and privacy protection of wearable health and medical devices were thus analyzed and compared with those of traditional health and medi-cal devices. The related studies and laws on domestic and foreign wearable health and medical devices were summa-rized, and the advances, experiences and problems in domestic and foreign wearable health and medical devices were analyzed and compared in order to provide reference for establishing the mechanisms underlying their data se-curity and privacy protection.
7.Clinical characteristics of hypertrophic cardiomyopathy in the elderly

Objective To investigate the clinical characteristics of hypertrophic cardiomyopathy (HCM) in the elderly. Methods The clinical characters of HCM in 70 elderly patients were retrospectively analysed. Results Among the 196 identified HCM patients, 70 were elders. Out of them, 7 patients(10.0%) were suspected as HCM according to the clinical symptoms, 29 patients (41.4%)were suspected as other cardiac diseases, 34 (48.3%) were diagnosed HCM due to other reasons. Among the 70 patients, 12 patients(17.1%) had history of cerebrovascular diseases, 54 (77.1%) manifested symptoms after 45 years of age and 4 (5.7%) showed no obvious symptoms. Among them, 18 patients were examined by UCG for two times and HCM was diagnosed at the second time. Conclusions HCM in the elderly is not an uncommon disease. The onset of cardiac symptoms is relatively late in the elderly HCM and frequently misdiagnosed; cerebrovascular diseases are commonly seen in elderly HCM patients.
8.Expression of MMP-1, MMP-3 and iNOS in cartilage of experimental rabbit osteoarthritis induced by anterior cruciate ligament transection

Objective To observe the expression of matrix metalloproteinases(MMP)-1,MMP-3 and iNOS in cartilage of experimental rabbit osteoarthritis at different time intervals after anterior cruciate ligament transection(ACLT)operation.The aim of this study is to provide the theoritical evidence for using ACLT rabbit model in Osteoarthritis(OA)research.Methods Unilateral ACLT was performed on 27 randomly selected while rabbits and underwent unilateral arthrotomy was performed on the other 9 white rabbits as the control group.Nine randomly selected white rabbits in experimental group were killed and 3 white rabbits in the control group at 4th,8th and 12th week respectively.Cartilage degradation of femoral condyles was evaluated macr-oscopically,mRNA expression level and protein expression level of MMP-1,MMP-3 and iNOS was measured by reverse transcription-polymerase chain reaction(RT-PCR)and immunohistochemistry respectively.Results Forepart OA cartilage degradation was observed at the 4th week and became more severe at the 8th week after ACLF.Afterpart cartilage degradation was evident at the 12th week after ACLT while cartilage still remained normal in the control group,mRNA expression level and protein expression level of MMP-1.MMP-3 and iNOS were increased at the 4th week and became higher gradually at the 8th,12th week after ACLT compared with the control group.Expression distribution of MMP-1,MMP-3 and iNOS bad different patterns respectively.Conclusion It is suggested that the process of OA cartilage degradation can be simulated by ACLT model and MMP-1,MMP-3 and iNOS may be good markers in therapeutical research of OA.
9.Breast tumor size assessment:comparison of conventional ultrasound and real-time ultrasonic elastography

Objective Tumor size discrepancy in measurement between real-time ultrasonic elastography and conventional uItrasound(US) was found in some breast lesions.The study was designed to investigate the value of the feature in the diagnosis of breast cancer,and its relationship with the margin features on gray-scale US.Methods Both US and real-time elastogram using a Hitachi EUB-8500 US system were performed in 308 consecutive Datients.The margin features on US and the size measured on both methods were documented.A lesion was defined as a significant size discrepancy when a larger size measured at elastogram.Sensitivity,specificity,and overall accuracy of feature to diagnosis malignancy were determined by surgical pathology as gold standard.Results There were total 166 benign,163 malignant and 3 borderline lesions.of 99 lesions with larger measurement on elastography,there were 91 malignant lesions and 8 benign lesions.By the feature of size discrepancY,the sensitivity,specificity and accuracy was achieved 55.8%,96.9%,75.7%,respectively.The size measured on elastogram was statistically larger than that on gray scale US(t=-11.0,P<0.05).The ratio of indistinct,angular and spiculated margin characteristics were significantly higher in breast cancers with larger size measurement than those with unchanged or decreased size measurement(P<0.05).Conclusions The increased size-measurement at elastography is helpful to diagnosis breast cancers.And it is more likely to be present in breast cancers with indistinct,angular or spiculated margin.
10.Gloning and Sequence Analysis of Dienelactone Hydrolase Gene

A 2,4 -dichlorophenol degrading Pseudomonas strain GI241-1 was isolated from a soil sample. The dienelactone hydrolase gene, designated as dcpD which encodes dienelactone hydrolase involved in transforming cis-2-chloro-dienelactone into 2-chloromaleylacetic acid, was cloned from this bacterium strain. The gene cloning strategy was to construct genomic library after location of its neighbouring gene by Southem blot and to screen the aim transformant by dot blotting. Sequencing results showed that length of dcpD is 702bp. The sequence of dcpD and the deduced amino acid are different from the relative sequences registered in the GenBank.
