1.Study on Bioequivalence of Cefdinir Capsules in Chinese Healthy Volunteers

Objective To evaluate postprandial pharmacokinetics and bioequivalence of two preparations of cefdinir capsules in Chinese healthy volunteers. Methods In a two-way cross-over study, 24 healthy male volunteers were divided into two groups randomly and a single dose of cefdinir capsules of test and reference preparation were administered orally, respectively.The concentration in plasma was determined by LC-MS/MS. Pharmacokinetic parameters and bioequivalence were calculated and evaluated by DAS. Results The main pharmacokinetic parameters of test and reference were as follows: AUCt (4.35±1.09) μg??h??mL-1 and (4.12±1.22) μg??h??mL-1, AUC0-∞(4.53±1.12) and (4.53±1.73) μg??h??mL-1, t1/2 (1.74±0.29) h and (2.13±1.65) h, tmax(4.44±0.86) h and (4.54 ±1.16) h, Cmax(900±250) ng??mL-1 and (876±269) ng??mL-1 . Conclusion The test and reference preparation of cefdinir capsules are bioequivalent.

3.Delayed protection of HO-1 in the exercise preconditioning from the myocardial relative ischemic reperfusion injury.

AIMTo explore the role of HO-1 in the exercise preconditioning protecting the rat myocardium from the relative myocardial ischemia/reperfusion injury (rI/R).
METHODS40 Wistar rats were divided into 5 groups randomly: control group (CN), relative ischemia/reperfusion group (IR), exercise preconditioning + relative ischemia-reperfusion group (EI), HO-1 revulsant group (HE) and exercise preconditioning + HO-1 inhibitor (EZ) group. We detected the cardiac function parameter--pressure-rate product (PRP), the levels of MDA in coronary effluent and the activity of HO-1.
RESULTSThe activities of HO-1 of EI group and HE group were higher significantly than the IR group. EZ group was lower than EI group. There was significant difference between EI group and HE group. The recovery rate of PRP in the 60 min point of reperfusion of EI group was higher significantly than IR group. EZ group was lower than EI group. There was significant difference between HE group and IR group in 30 min point of reperfusion. The levels of MDA in coronary effluent after rI/R of EI group, EZ group and HE group were lower significantly than IR group. There was significant difference between EI group and EZ group.
CONCLUSIONEP can activate the HO-1 which can ameliorate the rI/R injury occurred after 24 hours.

4.Nano-hydroxyapatite/collagen composite for bone repair.

OBJECTIVETo develop nano-hydroxyapatite/collagen (NHAC) composite and test its ability in bone repairing.
METHODSNHAC composite was developed by biomimetic method.
RESULTSThe composite showed some features of natural bone in both composition and microstructure. The minerals could contribute to 50% by weight of the composites in sheet form. The inorganic phase in the composite was carbonate-substituted hydroxyapatite (HA) with low crystallinity and nanometer size. HA precipitates were uniformly distributed on the type I collagen matrix without preferential orientation. The composite exhibited an isotropic mechanical behavior. However, the resistance of the composite to localized pressure could reach the lower limit of that of femur compacta. The tissue response to the NHAC composite implanted in marrow cavity was investigated. Knoop micro-hardness test was performed to compare the mechanical behavior of the composite and bone. At the interface of the implant and marrow tissue, solution-mediated dissolution and macrophage-mediated resorption led to the degradation of the composite, followed by interfacial bone formation by osteoblasts. The process of implant degradation and bone substitution was reminiscent of bone remodeling.
CONCLUSIONThe composite can be incorporated into bone metabolism instead of being a permanent implant.

5.The delayed protection of exercise preconditioning from the relative myocardial ischemia-reperfusion injury..

OBJECTIVETo explore the delayed protection of exercise preconditioning from the relative myocardial ischemia-reperfusion injury.
METHODSThe experiment included the vivo experiment and the vitro experiment, 32 Wistar rats in each experiment were divided into 4 groups randomly: control group (CN), relative ischemia reperfusion group (IR), exercise preconditioning group (EP) and Exercise preconditioning + relative ischemia-reperfusion group (EI). We detected the third loading exercise time, the levels of MDA in serum in vivo experiment and the Cardiac function parameter, the levels of MDA in coronary effluent in vitro experiment.
RESULTS(1) The vivo experiment: The third loading exercise time of EI group [(71.67 +/- 9.00) min] increased significantly compared with that of IR group [(58.67 +/- 4.13) min] (P < 0.05); The levels of MDA in serum of EP group (107.00 +/- 35.99) micromol/L and EI group [(152.23 +/- 29.94) micromol/L] decreased significantly contrasted to IR group (313.20 +/- 43.40 micromol/L) (P < 0.05). (2) The vitro experiment: The PRP (heart rate * left ventricular developed pressure) in reperfusion period of CN group and EP group were stable relatively, while it reached the peak after 30 minutes and almost recovered to the level before ischemia in EI group. The parameter of IR group recovered slightly but was lower significantly than that before ischemia. There was significant difference between the recovery rate of Cardiac function of EI group and that of IR group. The increase of MDA in coronary effluent after Ischemia-reperfusion of EP group (0.34 +/- 0.24 micromol/L) and EI group [(0.41 +/- 0.26) micromol/L] decreased significantly contrasted to that of IR group [(1.27 +/- 0.52) micromol/L] (P < 0.05).
CONCLUSIONEP has the obvious delayed protection from the relative myocardial ischemia-reperfusion injury.

6.Relationship between pesticide exposure and adverse pregnancy outcomes among famers: a meta-analysis.

OBJECTIVETo analyze the relationship between pesticide exposure and adverse pregnancy outcomes in famers.
METHODSA search was conducted to collect the articles about the relationship between pesticide exposure and adverse pregnancy outcomes published worldwide from 1990 to February 2012. Meta-analysis was performed on the collected articles using RevMan 4.2 software.
RESULTSTwelve articles were collected. Compared with the controls, the pesticide-exposed famers showed a combined odds ratio (OR) for spontaneous abortion of 1.52 (95%CI: 1.04 ∼ 2.21; P = 0.03), a combined OR for premature birth of 1.33 (95%CI: 1.09 ∼ 1.61; P = 0.005), a combined OR for dead fetus of 1.22 (95%CI: 1.16 ∼ 1.29; P < 0.01), a combined OR for stillbirth of 1.90 (95%CI: 0.58 ∼ 6.28; P = 0.29), a combined OR for birth defect of 2.02 (95%CI: 0.84 - 4.69; P = 0.12), a combined OR for low birth weight of 1.62 (95%CI: 0.60 ∼ 4.39; P = 0.34), a combined OR for neonatal death of 2.18 (95%CI: 0.54 ∼ 8.88; P = 0.28), and a combined OR for delayed conception of 1.43 (95%CI: 0.93 ∼ 2.18; P = 0.1). Pesticide exposure increased the risks for spontaneous abortion, premature birth, and dead fetus, but was not significantly associated with stillbirth, birth defect, low birth weight, neonatal death, and delayed conception.
CONCLUSIONPesticide exposure can cause adverse pregnancy outcomes in farmers, increasing the risks of spontaneous abortion, premature birth, and dead fetus.

7.Alterations of cardiac hemodynamics, sodium current and L-type calcium current in rats with L-thyroxine-induced cardiomyopathy.

The aim of the present study is to investigate the alterations of cardiac hemodynamics, sodium current (I(Na)) and L-type calcium current (I(Ca-L)) in the cardiomyopathic model of rats. The model of cardiomyopathy was established by intraperitoneal injection of L-thyroxine (0.5 mg/kg) for 10 d. The hemodynamics was measured with biological experimental system, and then I(Na) and I(Ca-L) were recorded by using whole cell patch clamp technique. The results showed that left ventricular systolic pressure (LVSP), left ventricular developed pressure (LVDP), +/-dp/dt(max) in cardiomyopathic group were significantly lower than those in the control group, while left ventricular end-diastolic pressure (LVEDP) in cardiomyopathic group was higher than that in the control group. Intraperitoneal injection of L-thyroxine significantly increased the current density of I(Na) [(-26.2+/-3.2) pA/pF vs (-21.1+/-6.3) pA/pF, P<0.01], shifted steady-state activation and inactivation curves negatively, and markedly prolonged the time constant of recovery from inactivation. On the other hand, the injection of L-thyroxine significantly increased the current density of I(Ca-L) [(-7.9+/-0.8) pA/pF vs (-5.4+/-0.6) pA/pF, P<0.01)], shifted steady-state activation and inactivation curves negatively, and obviously shortened the time constant of recovery from inactivation. In conclusion, the cardiac performance of cardiomyopathic rats is similar to that of rats with heart failure, in which the current density of I(Na) and especially the I(Ca-L) are enhanced, suggesting that calcium channel blockade and a decrease in Na(+) permeability of membrane may play an important role in the treatment of cardiomyopathy.

Objective To explore the protective effects of diazoxide on injury of human renal tubular cell(HK-2)induced by serum obtained from neonates with asphyxia.Methods HK-2 cells was used as the target cel1.The attacking concentration of serum obtained from neonates with asphyxia was 200 mL/L.The experiment was designed as 3 groups.HK-2 cells were divided into control group,asphyxia group,and diazoxide group.Control group:joined nutrient fluid including 100 mL/L embryo cow blood serum.Asphyxia group:joined nutrient fluid including the isometric 200 mL/L serum obtained from neonates with asphyxia.Diazoxide group:the diazoxide was joined nutrient including the isometric 200 mL/L serum obtained from neonates with asphyxia fluid.The diazoxide density finally was 100 ?mol/L.Then the change of morphology was observed and photographed under inverted microscope,and the cell viability was measured by methyl thiazolyl tetrazolium method,and the leakage rate oflactate dehydrogenase(LDH)was determined by biochemical methods.Results Under inverted microscopy,HK-2 cells in control group pastes the wall to be good,assumes the paving stone type,into flat polygon,fission many,the cell arrangement was close,connection large expanse,quantity were many.Compared with control group,the HK-2 cell to suffer injury obviously,the shape changed,become the anomalous circular or the ellipse by the model flat polygonal cell,the intercellular space crevice enlarged,the connection was loose,intercellular space obviously many cell fragmented.Living cell quantity reduced obviously,the cell vigor dropped,and the leakage rate of LDH increased significantly in asphyxia group(P
9.Application of Single Solid State Nanopore / Nanochannel Based on Polymer Membrane and Glass Nanopipette in Analytical Chemistry

Nanopore / nanochannel sensing technique drawing more attention in analytical chemistry due to its unique advantages and the fabricated electrochemical sensors and electrochemical responsive gates have been widely used for more target detection, including single molecule protein and DNA sequencing. Nanopore /nanochannel that used for fabricating electrochemical detection system is mainly divided into biological nanopore and solid state nanopore, and among them, solid state nanopore / nanochannel has a wide range of application due to its inherent properties, such as easy for modification, good mechanical property and stability. Resistive pulse sensing and current-voltage curves (ion current rectification) are two main methods of nanopore / nanochannel sensing technique used for target analysis, so in this review, we introduced the fundamentals and applications of nanopore sensing technique based on the above two methods. In addition, we concluded the application and development of single state nanopore / nanochannel in recent years, and also gave a brief look at the future challenges and prospects in the development of this field.
10.The risk factors and treatment of rituximab-induced interstitial pneumonia in patients with diffuse large B-cell lymphoma receiving R-CDOP regimen

Objective:To identify clinical features,possible risk factors and treatment related to rituximab-associated interstitial pneumonia(RTX-IP). Methods:The clinicopathological characteristics,immune phenotype and treatment of six patients with diffuse large B-cell lymphoma(DLBCL)develped after receiving R-CDOP treatment were retrospectively analyzed. Results:Six patients had agranulocytosis or granulocytopenia within 1 week before RTX-IP diagnosis,and the median interval time was 3 courses of treatment.All six patients had double-expressor lymphoma(DEL)or triple-expressor lymphoma(TEL).Of the six patients,four had germinal-center B-cell-like lymphoma(GCB),and two had non-GCB.The expression of Ki-67 was>70%,except for one patient with transformed lymphoma(TL).After treatment with methylprednisolone for about 1 week,all patients'chest CT showed inflammatory absorption.However,one patient developed pneumocystis carinii pneumonia during the process of hormone reduction,and recovered after 27 days of comprehensive treatment with hormones and anti-pneumocystosis therapy.All patients received CDOP regimen(a total of 8 courses)for the treatment of the primary disease,and the process was smooth. Conclusion:R-CDOP regimen may lead to a high incidence of RTX-IP in DLBCL patients(30.0%).The DLBCL patients with DEL or TEL,GCB subtype,TL and high Ki-67 expression were more liable to develop RTX-IP,and the recovery of agranulocytosis may be related to the pathogenesis of RTX-IP.High-resolution CT scan can provide valuable evidence for early diagnosis of RTX-IP.Metagenomic next-generation sequencing(mNGS)helps to distinguish IP from pathogen infections.High dose of glucocorticoids is effective treatment strategy.At the same time,it is necessary to strengthen the prevention and treatment of infection in the process of glucocorticoids application.
