1. Induction of cytotoxic T lymphocytes against cervical cancer cell line CaSki using therapeutic dendritic cell vaccine

Objective: To observe the inductive effect of cytotoxic T lymphocytes (CTLs) against human cervical cancer cell line CaSki using therapeutic dendritic cells (DCs) vaccine in vitro . Methods:. Immature mouse DCs were isolated and cultured. The expressions of cell-surface CD40, CD86, major histocompatibility complex (MHC)-. and CD11c in immature DCs were detected by flow cytometry (FCM). Then the immature mouse DCs were infected with recombinant adenoviral vector carrying human papillomavirus (HPV )16 E 6/E 7 (pAd-E6/E7), and the CaSki cell lysate-loaded autologous DCs vaccine was prepared. The expression of green fluorescent protein in pAd-E6/E7-infected immature mouse DCs was observed under a laser scanning confocal microscope, and the expression of E6 protein was detected by Western blotting. DCs vaccine was used to induce specific CTLs, were subsequently co-cultured with CaSki cells. The killing effect of CTLs against CaSki cells was determined using cell counting kit8(CCK8) assay. Results: HPV16 E6/E7-specific DCs vaccine was successfully prepared. CTLs which induced by DCs vaccine exerted a killing effect on CaSki cells. This killing effect was higher in pAd-E6/E7-infected group than those in CaSki cell lysate-loaded group and the untreated control group (P<0.05). Conclusion: Genetically modified DC vaccine can successfully be prepared by infection with pAd-E6/E7, and it has a significant effect on triggering of specific CTLs against CaSki cells.

3.Modified hemispherectomy for intractable epilepsy in patients with infantile hemiplegia.

OBJECTIVETo explore the effectiveness of modified hemispherectomy for intractable epilepsy in patients with infantile hemiplegia.
METHODSEighteen cases of patients were treated with modified hemispherectomy and the effectiveness was studied and followed up.
RESULTSThe seizures in all 18 cases of patients were controlled effectively and stopped completely in 16 cases of them, without nervous disfunction worsened. The patients' cerebral peduncles on healthy side were much thicker than those on sick side (t = 58.32, P < 0.001) and healthy peoples' (t = 14.63, P < 0.001) and the patients' cerebral peduncles on sick side were much thinner than those of healthy peoples' (t = 51.27, P < 0.001).
CONCLUSIONThe modified hemispherectomy can effectively control the seizures of patients with infantile hemiplegia without superficial cerebral hemosiderosis happened.

4.Clinical diagnosis and treatment of superior tibial fracture complicating with crotch injury of distal popliteal artery.

OBJECTIVETo investigate the clinical effects of treatment for superior tibial fracture complicating with crotch injury of distal popliteal artery by an alternative micro-surgical operation.
METHODSDuring Feb. 2002 to Oct. 2007, there were 19 patients with superior tibial fracture complicating with crotch injury of distal popliteal artery included 15 males and 4 females aged from 21 to 48 years (means 35 years). There were 6 cases complicating with fracture of tibial plateau, 3 cases complicating with nerve injury. The tibial fracture were fixed with external fixator and the anterior tibial artery and vein and posterior tibial artery and vein were treated by transplantable great saphenous vein (Y-shape) combined with windowing for interosseous membrane by the posterior and anterolateral incision. Evaluations of clinical effect were performed according to Rasmussen functional score system.
RESULTSAll patients survived after operation. All fracture achieved bony union, the union time was from 3 to 14 months (means 5.5 months). All patients were followed-up for from 8 to 23 months (means 13 months). The mean Rasmussen functional score was (27.0 + 2.9). The results were excellent in 11 cases, good in 7, and fair in 1.
CONCLUSIONThe surgical exploration should be done as soon as possible when diagnosis of injuries of large arteries is definite or highly suspected. Simultaneous reconstruction of both posterior tibial artery and anterior tibial artery associated with vein can reduce the rate of disability and recover function of limb.

The herbs used as the material for traditional Chinese medicine are mostly planted at mountain areas where there is appropriate weather condition. Due to the complex terrain condition in mountain area, the planting at such places is always associated with a scattered pattern, which inevitably bring difficulty to the work of accurate measurement for the planting area. Advanced research about monitoring the planting area for medicine-herb under complex environmental condition is so critical and meaningful for developing the medicine-herb planting to a large scale and an industry. Therefore, this research, taking the planting of Herpetospermum pedunculosum in Luding county as an example, utilizes RS&GIS: using the image data from TM and ETM datum as the remote sensing information source, integrating GPS data from quadrat survey on the spots, and analyzing on a GIS platform, to extract the planting area of H. pedunculosum in Luding county. The results indicate that: (1) the method and technology used to estimate the planting area of H. pedunculosum are feasible and can be applied to monitoring the planting area of medicine-herb at mountain area under complex environmental conditions: (2) the planting area of H. pedunculosum in Luding county is estimated to be 857. 246 acres by the methods utilized in this research, which is closed to the value from local statistical data with an error range less than 5%; (3) in consideration of those inevitable errorrs generating from the procedures such as remote sensing data acquisition, data preprocessing, data interpretation, and so on, and the fact that the classification of remote sensing data, which must be implemented by those above procedures, was used directly to extract the herb type, the accuracy of area estimation for the herb is significantly influenced by the accuracy of classification for herb type, and the estimated area is inevitably associated with some error.

6.Effect of full-responsibility nursing model in the rehabilitation exercise on patients with chronic heart failure

Objective To observe the effect of full-responsibility nursing model in rehabilitation exercise on patients with chronic heart failure (HF).Methods Eighty-six patients with HF were randomly divided into observation group and control group,with 43 cases in each group.sixteen nurses were randomly divided into A and B group ; A group carried out full - responsibility nursing model and was in charge of the rehabilitation exercise of observation group,B group carried out routine holistic nursing model and was in charge of the rehabilitation exercise of control group.After 4 weeks,the effects including EF,6MWT,the knowing of rehabilitation exercise,satisfaction to the hospital were evaluated.Results Before rehabilitation exercise,there were no significant differences in EF and 6MWT( P > 0.05 ).After 4 weeks,EF was( 54.13 ± 9.12 ) and 6MWT was (402.63 ± 74.81 ) in the observation group,while EF was (43.26 ± 8.57 ) and 6MWT was ( 336.16 ±75.82) in the control group.There were significant differences between the two groups ( t =2.093,2.145 ; P <0.05).there were significant differences before and after rehabilitation exercise in two groups ( P < D.05 ).After 4 weeks,EF and 6MWT of the two groups were both increased,but the effect of the observation group was better (P < 0.05 ). After 4 weeks,there were significant differences in the knowing of rehabilitation exercise and the satisfaction to the hospital in the two groups ( P < 0.05 ).Conclusions Full-responsibility nursing model in rehabilitation exercise can make an effective effect.
7.Genotype and phenotype polymorphisms of NAT2 and CYP2E1 in the Han Chinese pediatric population.

OBJECTIVEN-acetyltransferase 2 (NAT2) and cytochrome P450 2EI (CYP2E1) play a crucial role in the drug metabolic process. The aim of this study was to understand the genotype and phenotype polymorphisms of NAT2 and CYP2E1 in the Han Chinese pediatric population in order to provide a theoretical basis for individualized drug treatment.
METHODSA total of 341 (211 males and 130 females) randomly sampled Han Chinese children, aged from 2 months to 14 years, were enrolled in this study. Genotyping was carried out by PCR method, and metabolic phenotypes were identified.
RESULTSIn this study population, wild genotype was found as a major genotype in seven SNPs of NAT2, rs1801279, rs1041983, rs1801280, rs1799929, rs1799930, rs1208 and rs1799931. The frequency of NAT2 fast metabolism was highest (61.3%), followed by middle to slow metabolism (34.1%). Wild genotype also predominated in the four SNPs of CYP2E1 (rs2031920, rs3813867, rs6413432 and rs72559720) named as CYP2E1*5, *6 and *2, with a frequency of 61.3%, 60.1% and 99.4% respectively. As the relationship between CYP2E1 genotype and phenotype was unknown, phenotyping of CYP2E1 was not done.
CONCLUSIONSThe important SNPs of NAT2 and CYP2E1 are predominantly wild genotype in the Han Chinese pediatric population. Fast metabolic phenotype predominates in important SNPs of NAT2.

8.Effect of hyperbaric oxygen on cytochrome C, Bcl-2 and Bax expression after experimental traumatic brain injury in rats.

OBJECTIVETo explore the effects of hyperbaric oxygen (HBO) treatment on the neuronal apoptosis at an earlier stage and the expressions of Cytochrome C (Cyt C), Bcl-2 (B-cell lymphoma-2 family) and Bax (Bcl-2 associated X protein) in rat brain tissues after traumatic brain injury (TBI).
METHODSForty adult rats were divided into two groups, i.e., Group A (the rats with untreated TBI) and Group B (rats with HBO treatment after TBI). Sections of brain tissues of these two groups were then detected at 3, 6, 12, 24, 72 hours after TBI by immunohistochemistry and electronmicroscope, respectively.
RESULTSHBO treatment could up-regulate the expression of Bcl-2 within 72 hours, reduce the release of Cyt C from mitochondria, attenuate the formation of dimeric Bax and alleviate the mitochondrial edema within 24 hours after TBI.
CONCLUSIONSHBO treatment can alleviate neuronal apoptosis after TBI by reducing the release of Cyt C and the dimers of Bax and up-regulating the expression of Bcl-2.

9.A comparative study on therapeutic method of traumatic epidural hematoma.

OBJECTIVETo explore the therapeutic methods, surgical indications and clinical practice of minimally invasive surgery on traumatic epidural hematoma (EDH).
METHODSRetrospective study was made on 135 patients with traumatic EDH admitted into our hospital from June 2002 to August 2005. Sixty-five patients were treated with mini-invasive negative pressure drainage (treatment group), 70 patients with comparable condition used traditional craniotomy (control group). The mean time of operation, average days in hospital, expenditure and prognosis of two groups were recorded and analyzed.
RESULTSThere was no significant difference in therapeutic efficacy between two groups. Patients in treatment group had a shorter hospital stay and less expenditure than those in control group.
CONCLUSIONMini-invasive negative pressure drainage is simple, effective, economical and applicable to some traumatic EDH patients.

10.Influencing factors for posttraumatic hydrocephalus in patients suffering from severe traumatic brain injuries.

OBJECTIVETo detect the influencing factors for posttraumatic hydrocephalus in patients with severe traumatic brain injuries and provide theoretical reference for clinical treatment.
METHODSRetrospective study was made on 139 patients with severe traumatic brain injuries in our hospital. The patients were divided into two groups: hydrocephalus group and non-hydrocephalus group. Single factor analysis and multiple factor analysis were used to determine the related factors and hydrocephalus. Multiple factor analysis was conducted with logistic regression.
RESULTSPosttraumatic hydrocephalus was found in 19.42% of patients. Age(OR equal to 1.050, 95%CI: 1.012-1.090), decompressive craniectomy (OR equal to 4.312, 95%CI: 1.127-16.503), subarachnoid hemorrhage(OR equal to 43.421, 95%CI: 7.835-240.652) and continuous lumbar drainage of cerebrospinal fluid (OR equal to 0.045, 95%CI: 0.011-0.175) were screened out from nine factors as the influencing factors for posttraumatic hydrocephalus.
CONCLUSIONSRisk factors for PTH are as follows: age, decompressive craniectomy and subarachnoid hemorrhage (SAH). Continuous lumbar drainage of cerebrospinal fluid can greatly reduce posttraumatic hydrocephalus.
