1.Protection for regional systolic function of left ventricle by ischemic postconditioning in rabbits with ischemic reperfusion detected by echocardiography

Objective To detect cardiac function in rabbits models with isehemic reperfusion and to evaluate the short-term protective effects of ischemic postconditioning on regional systolic function of left ventricle by echocardiography. Methods Twenty-eight rabbits were divided into 2 groups:control group and ischemic postconditioning(IP) group. Echocardiography were performed before tests and 2 weeks after tests. The conventional echocardiography indices included left ventricular end-diastolic diameter(LVDd) ,left ventricular anterior wall end-diastolic thickness (AW) and left ventricular ejection fraction (LVEF). Quantitative tissue velocity imaging and strain rate imaging indices included the peak velocity in systole(Vs) and radial peak strain rate in systole(SRs). Results Before tests, there were no difference in conventional echocardiography indices between the two groups; compared with before test, the two groups had significantly larger LVDd,thiner AW and lower LVEF(P <0.05) ; compared with the controls, the group IP had significantly smaller LVDd, thicker AW and higher LVEF 2 weeks after tests(P<0.05) ; compared with controls, the number of segments with abnormal wall motion in group IP significantly decreased. Before tests,there were no differences in Vs and radial SRs between the two groups; compared with before test,the two groups had significantly lower Vs and radial SRs(P<0.05) ; compared with the controls,Vs and radial SRs in group IP significantly increased 2 weeks after tests (P<0.05). Conclusions Ischemic postconditioning can protect regional systolic function of rabbits models with ischemic reperfusionin in short-term,which can be effectively evaluated by strain rate imaging.
2.Study of estradiol on treatment of preeclampsia in rat model

Objective To evaluate whether estradiol can inhibit and cure the inflammation of experimental preeclampsia in rats. Methods Experimental preeclampsia was induced in 14-day-pregnant rats by infusion of endotoxin (1.0 ?g/kg). Rats with normal pregnancy were infused with sodium chloride solution.A group of preeclampsia rats was injected with 17?-estradiol (17?-E_2, 1 mg?kg -1 ?d -1 ). Blood pressure, albuminuria,inflammation associated adhesion molecule CD_ 49d and tumor necrosis factor-?(TNF-?) were assessed. Results On pregnant day 19, for normal pregnancy group(group C) the blood pressure was (120.4?2.0)mm Hg (1 mm Hg=0.133 kPa),urinary protein (0.47?0.06)mg/24 hours;for experimental preeclampsia group(group A) blood pressure was (134.2?2.4) mm Hg,urinary protein(0.79?0.10)mg/24 hours; for experimental preeclampsia with 17?-E_2 treatment group (group B) blood pressure was(123.3?1.7)mm Hg,urinary protein (0.51?0.08)mg/24 hours. A significant increase of blood pressure and urinary albumin was observed in group A. CD_ 49d expression and TNF-? concentration were also increased. 17?-E_2 reduced the expression of CD_ 49d , concentration of TNF-?,blood pressure and albuminuria of experimental preeclampsia. However, the weight of fetuses in 17?-E_2 treatment group were less than that in other groups. Conclusion 17?-E_2 can improve the symptoms of experimental preeclampsia,but its effects on fetus need to be further studied.
3.A calibration phantom system for QCT bone mineral density determination.

This paper describes a calibration phantom system for QCT bone mineral density determination, which consists of 4-standard-solid-sample calibration phantom, a quality assurance (QA) phantom and the bone mineral density analysis software. The system adds to the new applications of CT systems, and provides a new method with a good accuracy and reliability for the examination, diagnosis, prevention, treatment of osteoporosis diseases and the observation of curative effect of drugs.

4.Effect on M1 macrophages of shenlian extracts.

This study discusses the effects of Shenlian extracts (SL) on M1 macrophages in atherosclerosis. The MTT assay was used to detect the growth inhibition rates of RAW264.7 cells. RAW264.7 cells were stimulated with murine interferon-gamma (IFN-gamma) plus lipopolysaccharide (LPS) to induce M1 macrophages. The different concentrations of SL extracts (high-dose 50 mg x L(-1), moderate-dose 25 mg x L(-1), low-dose 12.5 mg x L(-1)) were added. The CD86 of M1 macrophages in cell membrane was measured by flow cytometry. The mRNA expression of iNOS and TNF-alpha gene was detected by reverse transcription PCR (RT-PCR). And the supernatants were collected, the content of IL-6 and TNF-alpha were detected with ELISA kits. The results of this experiment show that the expression of the cell membrane molecule CD86, iNOS and TNF-alpha gene, the content of IL-6 and TNF-alpha was obviously increased in M1 macrophages by IFN-gamma and LPS. The different doses of SL extract could reduce the expression of the above indicators. The above experimental results demonstrate that IFN-gamma combined LPS can induce RAW264.7 cell to type into M1 macrophages, and SL extracts can inhibit M1 macrophages.

5.Synthesis and LAR inhibition of 7-alkoxy analogues of illudalic acid.

To obtain higher potency and specificity, a series of 7-alkoxy analogues of illudalic acid was synthesized on the base of structure-activity relationship (SAR). All of these compounds exhibited submicromolar inhibition of the enzyme when tested against human leukocyte common antigen-related phosphatase (LAR) (for example, for 15e, IC50 = 180 nmol x L(-1)). They represent the most potent small-molecule inhibitors of LAR so far. These analogues also display excellent selectivity for LAR over other protein tyrosine phosphatases (PTPs) except for the highly homologous PTPsigma. The compound 15f is of 120-fold selectivity for LAR versus PTP-1B inhibition. The development of potent enzyme-specific inhibitors is so important that they may serve both as tools to study the role of LAR and as therapeutic agents for treatment of type II diabetes.
6.Treatment of femoral intertrochanteric instable fractures with cemented artificial bipolar arthroplasty for senile patients

Objective To discuss the feasibility and effects of cemented artificial bipolar arthroplasty for senile patients with femoral intertrochanteric instable fractures. Methods From January 2004 to June 2008,46 aged cases with femoral intertrochanteric instable fractures were cured with cemented artificial bipolar arthroplasty. According to improving-classfication of Evans,there were 19 cases with type Ⅲfractures and 27 cases with type Ⅳ fractures. Results All cases were successfully operated. The surgery time was 50-90 (68.00 ±20.43) minutes. Bleeding volume was 160-600 (210.00 ±40.26) ml. The time in hospital was 16-26 (19.30 ± 4.56) days. All cases were retrospectively reviewed for 6-39 (17.10 ± 4.33)months. Three cases died of other diseases, 43 cases were graded by the system of Harris. The Harris scale was 70-94 (86.70 ±5.16) scores,and the choiceness rate was 90.7%(39/43). No dislocation,loosening,subsidence and late infection occurred in all the implants. Conclusions Cemented artificial bipolar arthroplasty has good advantages of reduced laying up period and few complications. The short-term outcome is satisfactory.
7.Progress of cytogenetic detection in myelodysplastic syndromes.

In recent years, significant progresses have been got in study on pathogenesis, treatment and prognosis of myelodysplastic syndromes (MDS), especially on use of new technology, that has great importance for cytogenetics of MDS. Recently, the progress of cytogenetic detection in MDS is very remarkable. Based on the metaphase cytogenetics (MC) method, prognostic significance of cytogenetics in MDS was clarified gradually. For example, people have known the prognostic significance of 12 p-, 11 q-, +21, t(11(q23)), although these genetic abnormalities are rare in the MDS. In addition, chromosome mutation emerged in the process of MDS may indicate the poor prognosis. On the other hand, with the use of SNP-A and aCGH in the study of genetics, MDS cytogenetic abnormality detection rate has been further improved and can reach to 78%. At the same time, some of MDS patients with the "normal karyotype" detected by MC have new hidden aberrations through the SNP or CGH detection, and these patients have a poorer prognosis. In this review, the advances of study on cytogenetic detection for MDS based on MC and SNP-A or aCGH methods are summarized.

8.Study on No.7 lymph nodes micrometastasis in patients with node- negative gastric carcinoma by routine examination.

OBJECTIVETo investigate the micrometastatic characteristics in the gastric cancer patients with negative lymph node invasion by routine pathological examination and evaluate the relationship between micrometastasis and clinicopathological features.
METHODSOne hundred and thirty-eight lymph nodes from No. 7 group in 46 patients with node-negative gastric cancer by routine examination were examined by consecutive sections. Telomerase activity was determined by RT-PCR and ELISA. Clinicopathological data were analyzed by statistical method.
RESULTSMicrometastasis was found in 32 lymph nodes (23.2%) from 13 cases (28.3%) by consecutive sections. Telomerase activity was positive in 49 lymph nodes (35.5%) from 20 cases (43.5%). The sensitivity, specificity, positive and negative predictive value and diagnostic accuracy of TRAP-ELISA was 100%, 84%, 65%, 100% and 88% respectively. The micrometastasis in lymph node was correlated with age, sex, primary tumor location, histological classification and metastatic lymph node type (P> 0.05), but correlated with the gross type of the primary tumor, size and serosa invasion (P< 0.05).
CONCLUSIONSIt is necessary to discern the micrometastasis in gastric cancer with negative lymph node by routine examination in order to objectively evaluate the clinicopathological classification and prognosis. TRAP-ELISA can be a complementary method to traditional histological examination.

9.Effects of different transplanting conditions on survival rate and growing status of Anoictochilus roxburghii plantlets.

The growing status of Anoectochilus roxburghii seedling was observed and the survival rate of seedlings, height, stem diameter and plant fresh weight under the conditions of different transplanting substrate compositions, planting density, shading rate were measured. The results showed that the effects of different transplanting substrates, planting densities, shading rates and nutrient solutions on the growing status of A. roxburghii plantlets varied greatly. A. roxburghii plantlets demonstrated a high survival rate and better growing status under the Following conditions: the ratio of peat and river sand as 2: 1, the planting density as 3 cm x 3 cm, the shading rate as 70%, and the nutrient solution as 1/4MS. The findings of the study provide a solid technical solution for the artificial cultivation of A. roxburghii plantlets.

10.Effect of Dietary Fatty Acids on Colon Tumorigenesis Induced by Methyl Nitrosourea in Rats

To study the effect of dietary fatty acid on the colon tumorigenesis induced by methyl nitrosourea in rats, male SD rats were fed five semi-synthetic diets composed of different proportions of beef tallow, soybean oil, alkana oil, corn oil and fish oil for 180 days. The experimental groups were injected with a solution of methyl nitrosourea in phosphate buffer intraperitoneally once a week for six weeks. The control groups were injected with phosphate buffer solution only. The incidence of colon cancer, the avergage volume of the tumors, proliferation cell nuclear antigen, cell kinetics, membrane lipid fluidity, alkaline phosphatase activities and the content of prostaglandin E2 in colon mucosa and the fatty acid of testis pad fat were measured at the end of the experiment. The results showed that the incidence of colon cancer and the average volume of tumors in animals fed with diets composed mainly of beef tallow, soybean oil or alkana oil were significantly higher than those that were fed fish oil. The most effective anticancer diet in our study contained saturated fatty acid, monounsaturated fatty acid and polyunsaturated fatty acid of fish oil in the proportion of 13.9%, 16.4% and 68.8% respectively. Inhibition of colon tumorigenesis appeared to be related to the regulation of membrane lipid fluidity, and a decrease in the proliferation of cell nuclear antigen in colon cells. In addition, a decrease was noted in the number of cells in S phase and alkaline phosphatase activity, along with inhibition of arachidonic acid products and a corresponding decrease in the amount of prostaglandin E2.
