2. Anesthesia selection in the establishment of the model of middle cerebral artery occlusion in rats

Objective: To study the effect of intraperitoneal injection anesthesia and endotracheal anesthesia during creating a rat model of middle cerebral artery occlusion (MCAO). Methods: Rats were anesthetized and maintained respectively with chloral hydrate by intraperitoneal injection and inbreathing 2% halothane and 30% O2 via tracheal intubation. MCAO model was created by method of Zea Longa. The time of induction, maintenance, recovery and physiological indexes were monitored, and the result of blood gas analyses, blood glucose, brain temperatures, rectal temperatures, neurological deficit, 48 hours' survival rate and the volume of brain infarction were also studied. Results: Intraperitoneal injection anesthesia inhibited the respiration significantly. Blood gas analysis indicated serious respiratory acidosis (pH 7.29 ± 0.03, PCO2 56.8 ± 4.2 mm Hg, PO2 119.1 ± 15.6 mm Hg). But it appeared approximately normal in rats with halothane anesthesia (pH 7.48 ± 0.06, PCO2 36.2 ± 0.3 mm Hg, PO2 219.1 ± 27.4 mm Hg). There was significant difference between two kinds of anesthesia in pH, PCO2, PO2 (all P < 0.01). Anesthesia with chloral hydrate by intraperitoneal injection can reduce brain and rectal temperature significantly (cerebral cortex in ischemia area 33.3 ± 0.8 °C, striatum 33.6 ± 0.3 °C, rectum 34.7 ± 0.5 °C), However, endotracheal anesthesia affected little on brain and rectal temperature (cerebral cortex in ischemia area 35.1 ± 0.39 °C, striatum 36.2 ± 0.3 °C, rectum 35.1 ± 0.3 °C). There were significant difference between two kinds of anesthesia in brain temperature (P < 0.01), but in rectal temperature they had not significant difference (P > 0.05). There was no significant difference of survival rate at 48 hours and brain infract volume between the two kinds of anesthesia (P > 0.05). Conclusion: This study suggestes that endotracheal anesthesia with 2% halothane and 30% O2 is better than intraperitoneal injection of chloral hydrate. It is effective and less influence on the brain temperature and other relative indexes during creating middle cerebral artery ocelusion rat model.
3.Study on sleep disorders in patients with Parkinson' s disease by polysomnogram

Objective To observe the polysomnographic manifestation of sleep disorder and the characteristics of sleep architecture in patients with PD.Methods 42 patients with PD and 40 normal controls underwent night polysomnography.The parameters of sleep architecture and progress in two groups and video-monitoring features were analyzed.Results According to PSG recordings,the incidence of difficulties in the initiation of sleep(73.8%),fragmented sleep(59.5%),excessive daytime sleepiness (46.1%)were respectively increased in PD patients group than that in controls(all P5)、 ESS were increased(P=0.022,0.000,0.007, 0.001,0.000,respectively).SOREMPs occurred in 6 patients(14.3%)in PD group,but didn't oecure in controls.In PD group REM without atonia(RWA)was demonstrated in 36 patients(85.7%),RBD in 19 patients;in control group,however,REM in 6 and RBD in 2 separately.The statistics analysis showed the incidence of RWA(85.7%)and RBD(45.2%)in PD group was significantly higher than that in control group(P
4.Absorbable coral hydroxyapatite as a scaffold for tissue engineered bone in vitro

?Objective:To investigate the behavior of absorbable coral hydroxyapatite (Interpore 500R) as bone tissue engineering scaffold. Methods: Marrow stromal stem cells harvested from legally aborted human fetus were cultured in DMEM containing 100 ml/L fetal boving serum and induced to differentiate towards osteoblasts.Then the cells were seeded onto absorbable coral hydroxyapatite , then the composite was implanted subcultaneously into the back of nude mice .Cell materia attachement was observed with scanning electron microscope,type I collegen secretion was studied with laser confocal microscope and new bone formation was studied histologicaley. Results: The cells could adhere to absorbable coral hydroxyapatite and secrete collagen I.Mature bone tissue was found in the implanted composite 40 day after transplantation. Conclusion: Absorbable coral hydroxyapatite Interpore 500R is a favorable bone tissue engineering scaffold.
5.The effect of peer assisted learning method in infectious disease surgical nursing students in evaluation.

Objective To improve the effectiveness of internship nursing students in infectious diseases by implementing peer teaching method. Methods The nursing students were divided into two groups according to the order of student number. The control group was given the teaching method of the general outline. On the basis of the outline teaching, the experimental group gave counseling and counseling to the students in their professional and psychological aspects. Results There were significant differences between the two groups in terms of the results of the three tests and the theoretical evaluation, and the results of the test group were better than those of the control group (P<0.05). Conclusion Peer- to- peer learning method can improve the practical effect of surgical nursing students in infectious diseases, and can be popularized in a certain extent.

8.Preparation and Quality Control of Dibing Gel

OBJECTIVE:To prepare Dibing gel and establish its quality control method.METHODS:The0.5%Carbopol940was taken as the base of gel.The content of dexamethasone acetate was determined by HPLC.The stability and the skin irritation test were studied,too.RESULTS:The calibration curve of dexamethasone acetate was linear in the range of20~120?g/ml.The average recovery of dexamethasone acetate was101.1%,RSD=0.92%.The preparation was stable and had no irritation to skin.CONCLUSION:The gel was resonable in formulation,convenient in preparation technique,and simple and accurate in method of quality control.
9.Nude mouse as in vivo bioreactor: The feasibility study of construction transplantable tissue engineered bone

ObjectiveTo investigate the feasibility of construction transplantable tissue engineered bone in nude mouse as in vivo bioreactor. MethodsSelected the 4th passage of rabbit bone mesenchymal stem cells, which were cultured, expanded and induced in vitro. Cells were seeded into porous natural coral. The complexes were implanted subcutaneous in nude mice. Regenerated bone was taken out 8 weeks after implantation and implanted into muscle tissue interspace in the back of rabbit itself.The rabbit-anti-mouse antibody titer was measured at 14,28,56 days and the level of IL-2 was detected the at 2, 7, 14, 28 and 56 days after implantation.Immunohistochemistry staining was carried on in the 8th week. ResultsDuring the whole experimental process, IL-2 activity couldn't be measured.The rabbit-anti-mouse antibody titer was positive.A specific antibody reaction was detected from 2ed week to 8th week after transplantation and touches the peak value four weeks later.Immunohistochemistry staining result was negative.ConclusionThe tissue engineered bone growth in nude mouse is seemed to be the reliable transplantable bone source.
