3.Essential role of thyroid hormones in brain developm ent
Chinese Journal of Endocrinology and Metabolism 2001;0(05):-
Thyroid hormones (TH) play an imp or tant role in the development and maturity of the mammalian brain. Maternal TH de ficiency, even subclinical hypothyroidism, leads to poor neurodevelopment of the fetus. Screening of pregnant women for hypothyroidism is recommended. Identific ation of the TH-responsive genes in the developing brain and investigation on t heir functions are helpful to provide insights into the molecular mechanisms by which TH influence brain development.

Objective To discuss effects of anti-inflammatory mechanism of amygdalin on rats with type II collagen-induced arthritis (CIA). Methods Wistar rats were randomized into normal group, model group, amygdalin group, and tripterygium group. Type II CIA rat models were established. From the 15th day after the modeling establishment, each administration group was given corresponding dose of medicine for continuous 28 days. Levels of TNF-αand sICAM-1 were detected by ELISA in serum of rats, and expression of TNF-α was detected by immuno-histochemical method. Results TNF-α positive expression in amygdalin group and tripterygium group was similar and significantly reduced compared with model group. Levels TNF-α and sICAM-1 in amygdalin group and tripterygium group significantly decreased compared with those in model group (P<0.05), without significant difference compared with normal group (P>0.05). Conclusion Amygdalin can inhibit the expression of TNF-α and levels of TNF-α and sICAM-1, in order to treat rheumatoid arthritis.

Objective To observe the effect of tongue-drawing at lateral position in nasogastric intubation for senile patients with conscious disorders. Methods One hundred senile patients with conscious disorders were randomly divided into observation group and control group with 50 cases in each group. The patients in the control group were given routine nasogastric intubation while those in the observation group were given nasogastric intubation by tongue drawing at the lateral position. The two groups were compared in terms of success rate of intubation, the rate of adverse reactions and intubation effects. Result Compared with the control group, the observation group showed significantly higher in one-time intubation success rate , comfort rate and lower rate of adverse reaction (all P<0.01). Conclusions The nasogastric intubation by tongue-drawing at lateral position is superior to the routine intubation. It can improve the success rate of one-time intubation, decrease the rate of adverse reactions and relieve the pains of patients.

Objective To study the effects of recombinant adenovirus vector containing vp3 gene (AdAFPvp3) on c57BL/6 murine hepatocellular carcinoma model. Methods The titers of adenovirus vectors AdAFPvp3 and AdCMV-eGFP were detected. c57BL/6 murine hepatocellular carcinoma models were reproduced by subcutaneous inoculation of murine Hepa1-6 cells, and tumors were monitored for their generation rates. Twenty-four mice were randomly divided into adenovirus vector AdAFPvp3 group, adenovirus vector AdCMV-eGFP control group and PBS control group (8 each) when the tumors grew into 5mm in diameter. AdAFPvp3 (5?108pfu/100?l), AdCMV-eGFP (5?108pfu/100?l) and PBS (100?l) was intratumorally injected, respectively, in the three groups every other day for 2 times. The volume of tumor and the presence of adverse effects were observed. Seven days after treatment, all mice were sacrificed for evaluation of antitumor effect. Tumors, livers and spleens were all harvested for routine pathological examination. At the same time, the way of tumor cell death induced by vp3 in vivo was identified by TUNEL. Results The titers of both adenovirus AdAFPvp3 and adenovirus AdCMV-eGFP were 5?109pfu/ml. The carcinogenesis rate of hepatocellular carcinoma was 100%. Compared with adenovirus AdCMV-eGFP control group and PBS control group, the volume of tumor was diminished evidently in AdAFPvp3 group after intratumoral injection (P

Journal of Third Military Medical University 2003;0(13):-
Objective To construct an recombinant adenovirus vector of vascular endothelial growth factor small interfering RNA(Ad5-VEGFsi) and to observe its inhibitory effect on the cell proliferation of human leukemia K562 cells. Methods The specific human VEGF siRNA was subcloned into the shuttle plasmid pSES-HUS and then cotransfected with plasmid pAdeasy-1 to produce pAd5-VEGFsi by homologous recombination. The identified recombinant plasmid pAd5-VEGFsi was packaged and amplified in 293 cells. At 72 h after the transfection of Ad5-VEGFsi into K562 cells, VEGF mRNA expression and the protein level of VEGF in the cell culture supernatant were determined by RT-PCR and ELISA, respectively. Cell proliferation was measured by MTT assay and cell apoptosis by flow cytometry. Results The recombinant adenovirus Ad5-VEGFsi was successfully constructed. Ad5-VEGFsi with the titer of 4.6?1011 pfu/ml was harvested by CsCl gradient purification. Compared with those in the control, EGF mRNA in cells decreased by 66.55% (P
