1.COX-2 and its inhibitors in lung cancer drug resistance

COX-2 is the inducible form of cyclooxygenase,which is overexpressed in non-small cell lung cancer,especially in the adenocarcinoma,and is involved in the production of lung cancer drug resistance, reducing the sensitivities of tumor cells to chemotherapy drugs. COX-2 inhibitors have the effects of antitumor and prevention of tumor formation. Therefore,in order to reverse the drug resistance of tumor cells,improve the sur-vival rate and the prognosis of patients with tumor,the application of COX-2 inhibitors as adjuvant chemotherapy and combined with chemotherapy drugs has become a new direction in the treatment of lung cancer.
2.Effects of Muscle Relaxants on Motor Evoked Potentials Monitoring during Intracranial Surgery in Functional Area

Objective To investigate the effects of muscle relaxants on motor evoked potentials (MEPs) monitoring during intracranial surgery. Methods 62 patients with neurological tumor were divided into 2 groups: muscle relaxant group (n=21) and non-muscle relaxant group (n=41). The incidence of successful MEPs monitoring was investigated. Results The incidence of successful MEPs monitoring was 76.2% in the muscle relaxant group and 41.5% in the non-muscle relaxant group (P<0.05). Conclusion Muscle relaxants can affect the MEP monitoring, which would not be administered as possible during anesthesia for intracranial surgery in functional area.

4. Polyethylenimine-mediated BMP-7 gene transfection promotes fracture healing in elderly rats

Objective: To observe polyethylenimine-mediated BMP-7 gene transfection in promoting fracture healing in elderly rats. Methods: Male SD rats (20-month-old) were randomly divided into 3 groups: group A received BMP-7 gene therapy mediated by polyehtylenimine, group B was treated with normal saline and group C was treated with BMP-7 plasmid without polyethylenimine. Right femoral shaft fracture model was established in all rats. Animals in group A received transdermal injection of pcDNA3. 1-BMP-7/PET; those in group B and C received the same volume of normal saline and pcDNA3. 1-BMP-7, respectively. X-ray photography, histological observation (H-E staining), immunohistochemical staining of collagen type I, biomechanical and bone mineral density test were employed to assess the healing of fracture 2, 4, and 8 weeks after treatment. Results: The results of X ray and H-E staining showed no fracture healing in the 3 groups during the 8th week after fracture; however, the growth of coloboma in group A was better than that of the other 2 groups, with partial bone union between the fracture ends. Staining of collagen type I showed deeper, wider staining in group A compared with the other 2 group. Anti-bending intensity and bone mineral density tests showed that the parameters in group A were better than those of the other 2 groups (P<0.05 or 0.01). Conclusion: Polyethylenimine-mediated hBMP-7 gene transfer can effectively promote fracture healing in elderly rats.
5.Anatomical polysegmentectomy with reservation of segments Ⅰ and Ⅳ for the treatment of complicated hepatolithiasis

Anatomical hepatic segmentectomy is the treatment of choice for hepatolithiasis. However, in consideration of the volume of residual liver and the liver function, anatomical polysegmentectomy of the bilateral lobes for hepatolithiasis is restricted. Protection of the portal pedicles to the segments preserved and avoidance of ischemia/reperfusion injury to the residual liver parenchyma are critical steps during the operation.A female patient with hepatolithiasis and had a surgical history of choledocholithiasis removal and T-tube drainage received ana tomic polysegmentectomy with segments Ⅰ and Ⅳ preservation at the General Hospital of Kunming Medical College. During the operation, Portal pedicles to the segments Ⅰ , Ⅱ, right lobe,and segments Ⅱ and Ⅲ were isolated prior to liver parenchyma transection. Portal pedicles to segments Ⅰ and Ⅳ were protected under direct visualization. Hepatoduodenal ligament occlusion was not applied during liver parenchyma transaction. Segments Ⅱ- Ⅲ and Ⅴ-Ⅷ were anatomically resected, and segments Ⅰ ,Ⅳ were preserved with satisfactory vascularization. The patient recovered uneventfully and was discharged 14 days after the operation.
6.Changes of echocardiographic indices in patients receiving catheter ablation for atrial fibrillation with ;different outcomes

Objective To explore the changes of different echocardiographic indices in patients receiving catheter ablation for atrial fibrillation with different outcomes. Methods 75 patients received catheter ablation for atrial fibrillation during the period of January 2012 to February 2014. Before and 6 monthes after ablation , all the patients were examined left atrial volume index (LAVi), left atrium pressure (LAP), and left atrial ejection force (LAF) with echocardiography. Then after 6 months, according to the outcomes of the procedure, we divided the patients into two groups and compared the changes of these three parameters. Results In sinus group, LAVi decreased while LAF increased after treatment [LAVi: (29.3 ± 1.9) vs. (41.2 ± 9.3), LAF:(1.31 ± 0.61) (kg·cm)/s2 vs. (0.83 ± 0.22) (kg·cm)/s2, P < 0.05]; But LAP did not change [(55.7 ± 23.3)mmHg vs. (58.9 ± 18.6)mmHg, P > 0.05]; while in recurrent group, there were no changes in LAVi, LAP, and LAF after treatment (P > 0.05). Conclusions Radiofrequency catheter ablation is safe and effective in treatment of atrial fibrillation, resulting in significant improvements in left atrial volume and cardiac function.

8.Clinical value of serum soluble intercellular adhesion molecule-1 in patients with primary biliary cirrhosis and auto-immune hepatitis

Objective To investigate the clinical value of serum soluble intercellular adhesion molecule-1 (sICAM-1 )in patients with primary biliary cirrhosis (PBC)and autoimmune hepatitis (AIH).Methods Sixty-one PBC patients and 59 AIH patients,who were hos-pitalized or visited the outpatient department from June 2012 to September 2013,as well as 50 healthy controls,were included in the study. The PBC patients included 29 incipient cases,21 cases in remission,and 11 recurrent cases;the AIH patients included 26 incipient cases, 20 cases in remission,and 13 recurrent cases.Serum sICAM-1 level was measured by double-antibody sandwich enzyme-linked immu-nosorbent assay,and serum levels of alanine aminotransferase (ALT)and total bilirubin (TBil)were determined by biochemical enzyme as-say.Comparison between groups was made by analysis of variance;Pearson correlation analysis was performed.Results Among PBC pa-tients,the incipient group and recurrent group had significantly higher serum sICAM-1 levels than the remission group and control group (P=0.000 for all);there was no significant difference in serum sICAM-1 level between the incipient group and recurrent group (P=0.484);the remission group had a significantly higher serum sICAM-1 level than the control group (P=0.000).Among AIH patients, the incipient group and recurrent group had significantly higher serum sICAM-1 levels than the remission group and control group (P=0.000 for all);there was no significant difference in serum sICAM-1 level between the incipient group and recurrent group (P=0.802);no significant difference in serum sICAM-1 level was seen between the remission group and control group (P=0.281).For patients with PBC and AIH,serum sICAM-1 level was positively correlated with serum levels of ALT (r=0.664,P=0.000;r=0.784,P=0.000) and TBil (r=0.715,P=0.000;r=0.580,P=0.000).Conclusion Serum sICAM-1 may be involved in the immunologic injury in PBC and AIH.In patients with PBC and AIH,the elevation of serum sICAM-1 level is closely correlated with the severity of liver damage. Clinical monitoring of serum sICAM-1 level may play an important role in severity assessment,prognostic evaluation,and therapy guidance among patients with autoimmune liver diseases.
9.Expression of human BTC with biological activity

Objective To obtain abundant human betacellulin(BTC) with biological activity. Methods The whole mature protein coding sequence of BTC gene was amplified by polymerase chain reaction(PCR) method applied to human pancreatic ?-cell tumors cDNA.The fragment was cloned into prokaryotic expression vector pET32a(+) plasmid.The recombinant plasmid was transformed into E.coli BL21 and the fusion protein was expressed under isopropyl-beta-D-thiogalactopyranoside(IPTG).The fusion protein was purified by Ni2+ affinity chromatography.SDS-PAGE and Western blot were employed to determine the expression and purification of the expected protein.BTC was added to culture NIH3T3 cells for 5 days,and cell proliferation was detected by MTT. Results Lots of fusion protein were produced,and the purified protein can stimulate the proliferation of NIH3T3 cells. Conclusion The human BTC can be successfully obtained from the pET32a(+) system with the biological activity of stimulating the proliferation of NIH3T3 cells.
10.Construction and expression of human PDX-1 eukaryotic expression vector

Objective To construct eukaryotic expression vector of human pancreatic duodenal homeobox 1(PDX-1) gene,and to detect its expression in NIH3T3 cell lines. Methods The whole coding sequence of PDX-1 gene was amplified by polymerase chain reaction(PCR) from human pancreatic-cell tumors cDNA.The fragment was inserted into eukaryotic expression vector pcDNA3.1 plasmid.The recombinant plasmid was verified by double digestion and DNA sequencing.The expression of PDX-1 gene in NIH3T3 cells was assayed by Western blot. Results The length of specific fragment amplified by PCR was 852 bp,and the recombinant plasmid pcDNA3.1-PDX-1 showed two bands of 5.5 kb and 852 bp by digestion using respective restriction enzymes BamHⅠand EcoRⅠ.The sequence of PDX-1 gene was approved or confirmed by blasting to GenBank.It was suggested that PDX-1 gene had been cloned into pcDNA3.1 vector correctly.Western blot showed that PDX-1 gene was expressed,which was detected 24 h after pcDNA3.1-PDX-1 plasmid was transfected into NIH3T3 cells. Conclusion The recombinant eukaryotic expression vector pcDNA3.1-PDX-1 was successfully constructed and expressed in NIH3T3 cell lines.
