Objective To investigate the expression levels and clinical significance of programmed cell death 1 ligand (PD-L1) and programmed cell death factor-1 (PD-1) in peripheral T-cell lymphoma (PTCL). Methods Immunohistochem?istry was used to detect expression levels of PD-L1 and PD-1 in PTCL (test group, n=51) and benign proliferative lesions of lymph node tissues (control group, n=20). The correlations of PD-L1 and PD-1 expressions with clinical pathological param?eters and prognosis were analyzed between two groups. Results The expression level of PD-L1 was significantly higher in PTCL group than that in control group (74.51%vs 35.00%,χ2=9.662, P<0.05). The positive expression of PD-1 was signifi?cantly higher in PTCL group than that in control group (66.67%vs 25.00%,χ2=10.074, P<0.05). There were significant dif?ferences in PD-L1 and PD-1 expressions between different peripheral lactate dehydrogenase (LDH) levels of PTCL group (P<0.05). After two cycles of CHOP or ECHOP treatments, the response rate (RR) was higher in PD-L1 negative group than that in positive group (84.6%vs 47.4%,χ2=5.478, P<0.05), and RR was higher in PD-1 negative group than that in positive group(82.4%vs 44.1%,χ2=6.755, P<0.05). The median overall survival (OS) time was higher in PD-L1 negative group than that in positive group (29.8 months vs 17.6 months,χ2=4.413, P<0.05) and the median OS time was higher in PD-1 negative group than that in positive group (29.8 months vs 17.6 months,χ2=8.293, P<0.05). Conclusion There are high expression levels of PD-L1 and PD-1 in peripheral T-cell lymphoma, which is closely related with the elevated LDH in peripheral blood, poor response rate and shorter OS. Therefore, the expression levels of PD-L1 and PD-1 can be used as factors of worse effect of chemotherapy and poor prognosis.

Objective To study the relationship between the number of myeloid-derived suppressor cells (MDSCs) in peripheral blood and prognosis in patients with peripheral T-cell lymphoma (PTCL). Methods Twenty-three patients with PTCL were selected in this study and 14 healthy persons were used as the normal control. The proportion of MDSCs/mononu?clear in peripheral blood was detected by flow cytometry. The correlation between the number of MDSCs and the clinical character and prognosis in patients with PTCL was analyzed. Results The proportion of MDSCs in peripheral blood was sig?nificantly higher in patients with PTCL than that of healthy subjects [(4.59±1.41)%vs (1.36±0.59)%, P<0.01]. The number of MDSCs in peripheral blood was significantly decreased after chemotherapy compared with that before treatment [( 1.29 ± 2.95)%, P<0.05)]. There were significant differences in MDSCs expressions in patients between different extranodal involve?ment, bone marrow involvement, clinical stage and IPI score (P<0.05). Conclusion The higher expression of MDSCs may be related to the progression in patients with PTCL.

ObjectiveTo investigate the feasibility of construction transplantable tissue engineered bone in nude mouse as in vivo bioreactor. MethodsSelected the 4th passage of rabbit bone mesenchymal stem cells, which were cultured, expanded and induced in vitro. Cells were seeded into porous natural coral. The complexes were implanted subcutaneous in nude mice. Regenerated bone was taken out 8 weeks after implantation and implanted into muscle tissue interspace in the back of rabbit itself.The rabbit-anti-mouse antibody titer was measured at 14,28,56 days and the level of IL-2 was detected the at 2, 7, 14, 28 and 56 days after implantation.Immunohistochemistry staining was carried on in the 8th week. ResultsDuring the whole experimental process, IL-2 activity couldn't be measured.The rabbit-anti-mouse antibody titer was positive.A specific antibody reaction was detected from 2ed week to 8th week after transplantation and touches the peak value four weeks later.Immunohistochemistry staining result was negative.ConclusionThe tissue engineered bone growth in nude mouse is seemed to be the reliable transplantable bone source.

OBJECTIVETo explore the best treatment opportunity of acupuncture at Shiqizhui (Extra) for treating primary dysmenorrhea.

METHODSEighty cases with primary dysmenorrhea were randomly divided into an acupuncture at Shiqizhui (Extra) in the premenstrual period group (group A, n = 20), an acupuncture at Shiqizhui (Extra) when pain occurs group (group B, n = 20) and a blank group (group C, n = 40). Both of acupuncture groups were treated and followed up for 3 consecutive menstrual cycles, respectively. The therapeutic effects were compared by use of the Cox Menstrual Symptom Scale (CMSS).

RESULTSThere were no significant differences in CMSS between the two acupuncture groups during each course and the follow-up period (all P > 0.05), but the CMSS in the two acupuncture groups were all obviously lower than those in the blank group during the same period (P < 0.05, P < 0.01).

CONCLUSIONAcupuncture at Shiqizhui (Extra) has certain therapeutic effect on primary dysmenorrhea either in the premenstrual period or when pain occurs and there is no significant difference in acupuncture at different time.

Objective To investigate the clinical and pathological features of elderly patients with hepatocellular carcinoma (HCC)and the prognosis after radical resection.Methods From January 2013 to December 2014,98 elderly patients with primary liver cancer were enrolled in this study,with 120 non-elderly patients with primary liver cancer serving as the control group.Comparison was made concerning clinical and pathological characteristics,short term postoperative outcomes and long-term prognosis between the two groups.Results The average age of patients in the elderly group(68.4±3.7)was significantly higher than in the control group(53.6 ±5.3),and the difference was statistically significant(t=23.376,P＜0.001).The positive rate of HBsAg in the elderly group was 38.8 ％,higher than in the control group (70.0 ％),and the difference was statistically significant (x2 =21.341,P＜0.001).The incidence of liver failure in the elderly group was 4.1％,higher than in the control group (0.0 ％),and the difference was statistically significant (xe =4.990,P =0.026).There was no significant difference in survival rate at 6 months,1 year and 2 years between two groups (x2 1.427,2.127,and 0.510,each P＞0.05).There was no significant difference in recurrence rate between the two groups at 1 year and 2 years(x2 =0.205 and 0.706,each P＞0.05).Conclusions Elderly patients with hepatocellular carcinoma present favorable clinical and pathological features and show similar short and long-term outcomes,compared with non-elderly patients.Radical resection is valuable in the treatment of hepatocellular carcinoma in elderly patients.

Objective: To explore the effect of different culture conditions on protocorm-like bodies (PLBs) of Dendrobium candidum in bioreactor culture. Methods: Inoculation quantity, ventilation, sucrose concentration, and light intensity were taken to carry out the single factor experiment design. Biomass was determined by drying method, polysaccharide content by phenol-sulfuric acid method, and SPSS16.0 software was used for data analysis. Results: Under the bioreactor culture, the nutrient medium components for the PLBs proliferation of D. candidum and polysaccharide accumulation were 1/2 MS medium supplemented with 1.0 mg/L NAA, 5% coconutmilk, and 3% sucrose, pH value was 6.0. The culture conditions, such as inoculation with 40 g/L, 15 μm pore porous nozzle, aeration volumes by 1.0 and 0.5 L/min used alternately, and photon flux of 2000 lx could improve the proliferation coefficient and polysaccharide contents of PLBs. Conclusion: Different culture conditions have the significant effect on the growth of PLBs and the accumulation of polysaccharide. Suitable culture conditions have the very important practic significance to the biomass of PLBs and main medicinal ingredient production of PLBs.

Objective To investigate the protective effect of sevoflurane on the brain against focal ischemia-reperfusion(I/R)injury and its mechanism.Methods Twenty-four male SD rats weighing 250-300 g were randomly allocated into 3 groups(n=8 each):group Ⅰ sham operation;grouop Ⅱ I/R and group Ⅲ I/R + sevoflurane.The rats were anesthetized with intraperitoneal chloral hydrate 300 mg?kg~(-1).Middle cerebral artery occlusion(MCAO)was produced by insertion of a 4-0 mono-filament nylon thread with rounded tip at bifurcation of right common carotid artery into internal carotid artery.The nylon thread was advanced cranially until resistance was felt.The depth of insertion was 18-20 mm.After 3 h MCAO the thread was withdrawn to allow reperfusion.In group Ⅲ the animals inhaled 1.0 MAC sevoflurane for 30 min at 30 min before reperfusion.The rectal temperature of the animals was kept at 36.5-37.5℃.At the end of 24 h reperfusion the animals were weighed again.The animals'neurological deficit was evaluated using Zea Longa score(0=no defcit,4=unable to walk and unconscious).The animals were then killed.The neuronal apoptosis in striatum was assessed(TUNEL)and the PKC protein expression in striatum was determined by immunocyto-chemistry.Results The body weight of the animals in I/R group was significantly reduced after 24h reperfusion as compared to the body weight before ischemia (P＜0.01),while in control group and sevoflurane group there was no significant difference in the body weight before and after sham operation or I/R.The neurological deficit scores were significantly higher in I/R group than in sevoflurane group.The number of apoptotic neurons in striatum was significantly higher in I/R group than in sevoflurane group.The PKC expression in striatum was significantly higher in sevoflurane group than in I/R group (P＜0.01).Conclusion 1.0 MAC sevoflurane inhalation has protective effect on the brain against I/R injury. Upregulation of PKC expression in striatum decreased by I/R is involved in the mechanism.
