1.Determination of Strychnine and Brucine in Strychnos Nux-vomica Linn by SPME-HPLC

Objective:To establish a SPME-HPLC method for the determination of strychnine and brucine in Stryehnos nux-vomiea Linn. Methods:A novel capillary microtube monolithic column coupled with HPLC was prepared for the selective solid-phase microextraction of strychnine and brucine in Stryehnos nux-vomiea Linn. A Hypersil ODS column(150 mm × 4. 6 mm, 5 μm)was used and the mobile phase was acetonitrile- 0. 01 mol·L -1 sodium heptanesulfonate mixed with the same quantity of 0. 02 mol · L -1 potassium dihydrogen phosphate(adjusting pH to 2. 8 with 10% phosphoric acid)(21 ∶79). The detection wavelength was 260 nm,the sample size was 20 μl,and the column temperature was 25℃ . Results:The linear range of strychnine was 1. 2-90. 0μg·ml -1(r = 0. 999 8)and that of brucine was 1. 0-75. 0μg·ml -1(r = 0. 999 6). The average recovery was 100. 05%(RSD = 0. 58% ,n = 6)for strychnine and 99. 98%(RSD = 0. 27% ,n = 6)for brucine. Conclusion:The method is accurate and highly efficient,which can be used for the determination of strychnine and brucine in Stryehnos nux-vomiea Linn.
2.Mechanism of long non-coding RNA MALAT1 in promoting migration of hepatocellular carcinoma cells

Objective To investigate whether the long non-coding RNA (lncRNA) metastasis associated lung adenocarcinoma transcript 1 (MALAT1) can act as a competitive endogenous RNA (ceRNA) to promote the migration of hepatocellular carcinoma (HCC) cells.Methods Transient transfection of small interfering RNA (siRNA) against MALAT1 was used to knockdown MALAT1 in HepG2 cells.Transwell assays were employed to assess the migration capabilities of HepG2 cells upon MALAT1 knockdown.RNA pull-down assays were performed to validate the direct binding between MALAT1 and miR-126*.Quantitative reverse transcription PCR (qRT-PCR) and Western blotting assays were used to detect the mRNA and protein levels of the miR-126* target genes.The dysregulation and prognostic significance of MALAT1 and miR-126* were analyzed in the public dataset of The Cancer Genome Atlas (TCGA).Results Compared with the control group, MALAT1 knockdown significantly inhibited the migration of HCC cells.MALAT1, with three miR-126* response elements, directly sponged miR-126* in a sequence-specific manner.The mRNA and protein levels of CXCL12, which was the miR-126* target gene, were significantly down-regulated upon MALAT1 knockdown.The TCGA database showed that MALAT1 was significantly up-regulated in HCC and high expression levels of MALAT1 were significantly associated with poor disease-free survival, whereas an opposite pattern of miR-126* was observed.Conclusion This study suggests that MALAT1 directly sponges miR-126* and upregulates the expression of CXCL12, which in turn promotes the migration of HCC cells.
3.Advances in AEG-1 mediated drug resistance

It has been demonstrated that astrocyte elevated gene 1 ( AEG-1 ) can promote tumor initia-tion and progression .Over expression of AEG -1 is correlated with tumor angiogenesis ,metastasis and chemother-apy resistance of tumor cells of different origins .The present article is a review on the mechanism of AEG -1 me-diated drug resistance .Studies have shown that AEG -1 participates in carcinogenesis through Ha -ras,myc,NF-κB and PI3K/Akt signalling pathways .AEG-1 can also promote autophagy through activating AMP Kinase . Other researchers demonstrate that AEG -1 promotes MDR1 protein translation by up-regulating MDR1 mRNA expression ,and thus increases polyribosome .It is testified that AEG-1 can influence drug susceptibility and ex-pression of MDR gene as a RNA binding protein .Multiple functions of AEG -1 in drug resistance in multiple cancers demonstrate that AEG -1 can be used as a novel target for antitumor drugs .

6.Changes of apoptosis and cell cycle of HSC in rat hepatic fibrosis by treatment with traditional Chinese medicine prescription (Dan-Shao-Hua-Xian )

AIM: To investigate the effect of a Chinese medicime, Dan-Shao-Hua-Xian capsule, on liver fibrosis induced by CCl_4 by observation of apoptosis and cell cycle variation in the liver cells. METHODS: Animal models were produced through eight-week treatment of the rats with CCl_4, alcohol and diet of high fat/low proteins, and then administration of Dan-Shao-Hua-Xian to the rats (1 g/kg) via stomach-tube-pouring for eight weeks was performed. Liver index, serum hyaluronic acid (HA) and glutamate pyruvate transaminase (ALT) were measured and hydroxyproline (Hyp) content in urine were determined. The extent of the liver fibrosis was observed under light microscope and apoptosis and cell cycle were also examined by cytometry between the two groups. RESULTS: Compared to the liver fibrosis group, the liver index, serum HA, ALT in the treatment group decreased, the development of liver fibrosis delayed, the urine Hyp and the number of apoptosed cells and the ratio of G_0/G_1 cells increased, as well as the S phase cells decreased, yet unable to return to normal. All those changes detected were statistically significant. CONCLUSIONS: The Dan-Shao-Hua-Xian is effective in treating the CCl_4-induced liver fibrosis in rats partly by virtue of inhibition of the growth of hepatic stellate cells and induction of apoptosis.

8.Study on the association between expression and clinicopathological significance of AEG-1 and MMP-9 in colorectal tissue

Objective To investigate the expression of astrocyte elevated gene -1( AEG-1) and matrix metallopeptidase 9(MMP-9)in colorectal tissues and evaluate its correlation with clinical pathologic factors of colorectal cancer.Methods AEG-1 and MMP-9 expression in normal colorectal mucous (n=45),low-grade adenoma(n=31),high-grade adenoma(n=15)and colorectal carcinoma(n=146)were examined by immuno-histochemistry .Statistical analyses were applied to test the significance of its expression .Results AEG-1 ex-pression levels were gradually elevated in normal tissues ,low-grade adenoma ,high-grade adenoma and colorec-tal carcinoma respectively .Furthermore ,there was a similar trend for MMP -9from normal mucous to adenoma and carcinoma .Statistical analysis revealed that AEG -1 expression was markedly correlated with the UICC stage,T classification,N classification,M classification,and histological differentiation in the colorectal cancer pa-tients,but not with age,gender,tumor location and tumor size.In addition,AEG-1 expression was positively cor-related with the MMP-9 expression in colorectal cancer .Besides,those patients with high AEG-1 and MMP-9 levels had shorter survival time ( P<0 .05 ) .Conclusion AEG-1 may be involved in carcinogenesis and pro-gression of colorectal carcinoma and can be a novel prognostic biomarker in patients with colorectal carcinoma .
9.Preliminary application of 3D printing to manufacture a customized guide plate for clinical percutaneous needling at the hip joint

Objective To investigate the safety and efficacy of a customized guide plate which is designed by digital medical technology and manufactured by 3D printing in clinical percutaneous needling at the hip joint.Methods Enrolled in this study were 9 patients with hip disorder who required hip joint needling during the period from April 2015 to August 2015.They were 7 males and 2 females,21 to 53 years of age (average,42 years).Laterality:5 left sides and 4 right sides.Conditions:4 cases of traumatic arthritis and 5 cases of osteoarthritis.All of them underwent lamellar CT scans whose data were imported into the software of Mimics 10.01 for 3D reconstruction.Then the software of 3-matic was used to design a customized guide plate on the reconstructed models.Finally,3D printing was used to manufacture the guide plate.The needle angle,direction and depth were adjusted in vitro and the operation was simulated before actual needling in clinic.After percutaneous hip joint needling was performed in the 9 patients separately,the accuracy of needling was checked using C-arm fluoroscopy.Results The percutaneous hip joint needling was performed successfully in the 9 patients in whom the customized guide plate was used.The results were consistent with what had been designed on the reconstructed models.The operation time in the 9 patients averaged 417.7 s (range,from 387 s to 590 s).Little intraoperative bleeding was observed at the site of needling,fluoroscopy was applied only once for check.No neurovascular injury or other operation-related complications occurred.Conclusion For patients who need repeated percutaneous needling at the hip joint,the customized guide plate designed by us can improve accuracy and ensure safety and efficacy of needling.
10.Relationship between High-risk HPV Infection and Cervical Lsesions in Cervical Cancer Opportunistic Screening

Objective:To analyze the infection rate of high-risk HPV(hr-HPV) and its relationship with cervical intraepithelial neoplasms in cervical cancer screening population.Methods:The positive rate of HPV 16,18 and other 12 kinds of HPV subtypes from 2015.Jun to 2016 Nov was calculated and the infection rate in CIN Ⅱ+ population were analysed.Results:In the cervical cancer screening population,the total HPV positive rate was 19.0%,the positive rate of HPV 16,HPV 18 and other 12 kinds of HPV were 5.0%,1.6% and 15.2% respectively The risk to develop CIN Ⅱ + in HPV 16,HPV 18 and 12 other hr-HPV positive TCT ASCUS/LSlL population was 55.2%,35.5% and 38.9% respectively The risk to develop CIN Ⅲ+ was 25.3%,7.7% and 6.9% in HPV 16,HPV 18 and other 12 kinds of hr-HPV positive TCT ASCUS/LSIL population respectively In the CIN Ⅱ + population,45.0% cases were 12 other hr-HPV positive while 46.3% were HPV 16 positive and the positive rate of HPV 18 was 8.7%.Conclusions:In the cervical cancer screening population,the rate of HPV 18 positive was low,the rate of other 12 kinds of HPV positive was high.It's necessary to establish exact classification for the positive rate of each subtype.hr-HPV genotyping plays an important role in cervical cancer screening.In the referral to colposcope population,the rate of HPV 18 positive was low,the TCT results showed ASCUS/LSIL risk of HPV 18 positive and other 12 kinds of HPV positive was low.Consider using shunt detection to improve screening specificity.
