1.Effects of high glucose and insulin on the mRNA expressions of TGF-β1 , and TIMP-1 of hepatic stellate cells in rat
Journal of Chinese Physician 2016;18(8):1194-1197
Objective To explore the mechanism of diabetic hepatic fibrosis by observing effects of high glucose and insulin on expressions of transforming growth factor beta 1 (TGF-β1) and tissue inhibitor of metalloproteinase (TIMP-1) mRNAs of hepatic stellate cell (HSC) in rat. Methods Hepatic stellate cell lines in vitro were administrated by different dose of glucose without insulin and glucose with insulin for 72 h,and mannitol was chosen as hyperosmosis control group.Real time fluorescent quantitation polymerase chain reaction (RT-FQ-PCR) was used to determine expressions of TGF-β1 and TIMP-1 mRNAs of HSC in each group. Results Expressions of TGF-β1 and TIMP-1 mRNAs of HSC in each group could be detected and showed no significant difference among groups (P > 0.05).However,in general, expression of TGF-β1 mRNA in glucose with insulin group decreased and TIMP-1 mRNA increased. Conclusions The mRNA expressions of TGF-β1 and TIMP-1 in HSC could not be induced by high glucose alone, high dose of insulin may increase expressions of HSC TIMP-1 and reduce TGF-β1 expression. The main mechanisms of diabetic hepatic fibrosis may not be TGF-β1 pathway but be closely related to TIMP-1 pathway.

Enterovirus 71 (EV71) is one of the primary causative agents of hand, foot and mouth disease in children and closely associated with severe neurological complications and even deaths.EV71 outbreaks have occurred throughout the Asia-Pacific region since 1990s, posing global public health threat;however, no specific therapeutic strategy exists for severe EV71 infection.Several inactivated vaccine products have entered or finished the clinical trial stage, and some novel vaccine candidates, including live attenuated, subunit, and virus-like particle, show great potential for further develop-ment.This review summarizes the present situation and progress in the development of EV71 vaccines.

Objective To investigate the clinical distribution characteristics of persisters isolated from the chronic infected pa-tients,so as to provide scientific basis for effective clinical measures to prevent, control and treat persister-associated chronic infection. Methods Clinical microbial samples cultured from Jan.2013 to Dec.2014 were analysed by WHONET5.6.Four bacteria species with the highest isolation rate were performed for screening of chronic infection.Concentration of bacteria were detected by viable plate count method and then the growth curve were drew of each sample under the presence of anti-biotics.Persisters were comfirmed according to the specific growth curve under the presence of antibiotics. Results Four highest isolated bacteria species of the clinical samples were Escherichia coli (1 3.2%),Klebsiella pneumoniae (9.6%), Pseudomonasaeruginosa (8%)and Staphylococcusaureus (6.6%).862 chronic infection samples were generated out of 14 216 microbial samples and 41 persisters (4 strains of Escherichia coli,23 strains of Klebsiella pneumoniae,8 strains of Pseudomonasaeruginosa and 6 strains of Staphylococcusaureus)were isolated finally. Conclusion With such a comprehen-sively retrospective analysis of the hospital clinical microbial samples, can tell that the ratio of persisters in chronic infection was not high. And chronic infections are mostly caused by gene-mutated drug-resistant bacteria. However, the isolation rate of Klebsiella pneumoniae was relatively high,of which more attention should be payed to the prevention and control.

Objective To study the changes of colonic motility in chronic stress-induced depression rat model, the association between interleukin 6 (IL-6) and depression and its effects on contraction of colonic smooth muscle in vitro.MethodsThere were 30 healthy adult Wistar rats,of which chronic stress-induced depression model was created in 15 rats and the other 15 rats were in control group. The ratio of sugar consumption to body weight and the percentage of sugar preference were tested in these two rats groups through sugar preference experiments. The behavioral changes of two rats groups were detected by open field test.Colonic motility was evaluated with simulated fecal discharge experiment, and the serum IL-6 level was measured with enzyme linked immunosorbent assay (ELISA).IL-6 in colon tissue at mRNA level was determined by RT-PCR.The contraction ability of rat colonic smooth muscle was observed. The colonic smooth muscle of control group was treated with different concentration of IL-6,the effects of IL-6 on normal colonic smooth muscle and its response to acetylcholine (Ach) were evaluated. Results In model group, the ratio of sugar consumption to body weight was 0.12±0.03,the percentage of sugar preference was 16.17% ±2.61 %,total walking distance was (741.54 ± 341.10) cm and total times of upright was 15.69 ± 8.00,which were all significantly lower than those of control group (0.18±0.02,25.54 % ± 2.32 %,1336.20 cm±698.80 cm and 24.87±7.90,respectively) (P=0.041,0.044,0.002 and 0.001).It took (109.78±48.00) min for defecating in model group, which was significantly longer than that of control group (28.00±11.10) min (P=0.002).The IL-6 levels were high both in the serum and colon tissue of model group rats. The response of colonic smooth muscle to Ach was weaker in model group than that of control group (P=0.035).In model group, there were no significant difference in the response of colonic smooth muscle to IL-6 at 200,500 and 1000 pg/ml (P =0.935、0.825、0.766 ).After Ach added, there was significant difference in the response to Ach between IL-6 at 500,1000 pg/ml (R=1.1480±0.1000 and 1.1361±0.1500) and control group only added with Ach (R=1.6100±0.4500) (P=0.000 and 0.004).ConclusionIL-6 level increase in chronic stress-induced depression rat model and causes smooth muscle contraction disorder through reducing the sensitivity of intestinal smooth muscle to Ach.
