Objective To simultaneously determine the quantify liquiritin, glycyrrhetinic acid and glycyrrhizic acid inXianlong-Jiedu Yin by high performance capillary electrophoresis (HPCE).Methods HPCE method was performed on a fused silica capillary coates 75μm×85 cm (effective column length 78.5 cm) using 50 mmol/L borax solution-acetonitrile (95:5), with adjusted pH=8.50 as running buffer, lytical 22 kV, and electrokinetic injection 10 s, detected wave length of 254 nm, and temprature maintaining 25℃.Results The liquiritin, glycyrrhetinic acid and glycyrrhizic acid were completely separated within 30 minutes, showing good linear relationship in the range of 12-120, 15-150, 8-160μg/ml (r>0.999 3). The average recoveries were 100.70%, 98.51%, 98.40% withRSD 2.49%, 2.79%, 0.40%, respectively.Conclusions The HPCE was suitable for the determination of liquiritin, glycyrrhetinic acid and glycyrrhizic acid inXianLong-JieDu Yin, which can be used for the quality control.

Objective To study the feasibility,efficacy,and safety of MRI-guided percutaneous cryoablation for the treatment of hepatic carcinoma. Methods From January to September 2008,27 cases of hepatic carcinoma were treated with MRI-guided percutaneous cryoablation with Cryo-Hit system via two cycles of freezing-thawing. During the procedure,MRI showed the expansion of ice globe wrapping the tumor. Ten days after the freezing treatment,TACE was carried out in 5 cases. Results MRI-guided percutaneous cryotherapy were completed successfully in all of the cases. In 26 patients (96.3%),expanded ice globe wrapping the tumor was detected by MRI,showing a dark zone with clear margin. Only in one patient,the tumor was wrapped. The patients were followed up for 1 to 12 months (mean,3.5 months),during which the level of AFP decreased to a normal value in 14 cases (51.9%),continued to increase in 1 cases,and increased after an reduction in 12 cases (44.4%). The patients received more than one times of re-examination by imaging,which showed completed necrotic tumor tissues or no confirmed living tumors in 14 cases (51.9%); and incomplete tumor necrosis or living tumor lesions in 13 (48.1%). No severe complications occurred in this series.Conclusions MRI-guided percutaneous cryoablation is feasible,effecitve and safety for hepatic carcinoma.

AIM:To investigate the roles of maspin in the biological behaviors of non-small-cell lung cancer A549 cells.METHODS:Full-length coding region of maspin was amplified by PCR from human normal breast tissue and cloned into MSCV vector .Virus supernatants was produced by Phoenix A packaging system , and target cell line was infec-ted with the virus supernatants .The transfected cells were screened with puromycin .The cell line with maspin over-expres-sion was identified by real-time PCR and Western blot.Cell growth, migration and invasion were investigated by xCelli-gence system .RESULTS:Maspin over-expression vector was successfully constructed .The cell line with maspin over-ex-pression was established .No difference of the growth between A 549-control cells and A549-maspin cells was observed .The migration and invasion were quite different between A 549-control cells and A549-maspin cells.The expression level of inte-grin β1 in A549-maspin cells was decreased compared with A 549-control cells .CONCLUSION:Maspin has no effect on the growth of A549 cells.Maspin suppresses the migration and invasion of A 549 cells, in which integrin β1 is involved.

OBJECTIVEThis work lays the foundation for establishing a digital model database with normal occlusion. A digital dental cast is acquired through grating projection, and model features are measured through reverse engineering.

METHODSThe grating projection system controlled by a computer was projected onto the surface of a normal dental model. Three-dimensional contour data were obtained through multi-angle shooting. A three-dimensional model was constructed, and the model features were analyzed by using reverse engineering. The digital model was compared with the plaster model to determine the accuracy of the measurement system.

RESULTSThe structure of three-dimensional reconstruction model was clear. The digital models of two measurements exhibited no significant difference (P > 0.05). When digital and plaster models were measured, we found that the crown length and arch width were not statistically different (P > 0.05), whereas the difference between the crown width and arch length was statistically significant (P < 0.05).

CONCLUSIONThe reconstruction of a digital model by using the grating projection technique and reverse engineering can be used for dental model measurement in clinic al and scientific research and can provide a scientific method for establishing a digital model database with normal occlusion.

Objective:To optimize the extraction technology for Sanren Tongbian mixture. Methods:The water amount,decoction time and decoction frequency as the evaluation factors,and the content of amygdalin and the paste-forming rate as the evaluation indi-ces,the extraction process of Sanren Tongbian mixture was optimized by L9 (34 ) orthogonal design. Results: The optimal extraction conditions were as follows:adding 8-fold amount of water, decocting twice with 1. 5h for each time. Conclusion: The study provides scientific basis for the research of the extraction technology of Sanren Tongbian mixture, and the further experiments proved that the op-timized process is stable and feasible.

Objective To explore the histocompatibility and plasticitity of hydroxyapatite/high-density polyethylene composite (HMPE). Design Experimental study. Participants Forty-five rabbits. Methods Rabbits were randomly devided into HA/PE, HA and PE surgical groups, every group included 15 rabbits. Osteoeetomy of orbital rim and implantation (HA/PE, HA and PE materials were used) were performed on 3 groups of the rabbits. At 1st week, 4th week, 8th week, 12th week and 24th week of postoperation, we took out the samples from the rabbits separately in each group and did histopathologic and electron microscope examinations. Main Outcome Measures Physical signs of the implant combination with the bone. The bone cell appeared in the implants. Results After 12 weeks, the calcium salts deposit like bone plate with some osteoblast inside the HA/PE implants was observed. After 8 weeks, the calci- um salts deposit inside the HA plants was observed. In the PE implants, nothing could be seen but some fibrous connective tissue. Conclusion HMPE is an ideal repair material especially for orbital bone.

AIM: To study the therapeutic effect of Angeng Recipe(AGR) on the female climacteric rats. METHODS: Female SD rats were collected and randomly divided into climacterium,AGR(low dose,medium dose and high dose),nilestriol and young control groups.The level of hormone in serum and hypothalamus was analyzed after using AGR,the alteration of organ coefficient of uterus,adrenal,appendix cerebri and thymus were detected,the apoptosis of ovary granulosa cells was detected by fluorescence microscope and flow cytometer. RESULTS: E_2 contents in high dose group were higher than those in the climacterical group.FSH contents in high dose group were lower than those in the climacterical group.Uterus and adrenal gland viscera coefficient of high dose group,medium dose and low dose group were higher than that of climacterical group,The apoptosis of cells in medium dose group showed lower than that in the climacterical group. CONCLUSION: AGR could adjust the function of endocrine secretion by the effect on ovaries and appendices suprasphenoidalis directly or indirectly,and the mechanism of action was multitarget and the regulation was entirety.AGR could relieve the degeneration of sexual gland axis,and improve symptomatic in climacteric.

Objective To evaluate the effect and significance of apoptosis inducing factor (AIF) on promoting apoptosis of hypertrophic cardiomyocytes induced by hypoxia/reoxygenation. Methods Cardiomyocytes of neonatal Kunming strain rats were isolated and cultured. Cardiomyocyte hypertrophy was induced by angiotensinⅡ (0.1?mol/L for 12h), and the cells were cultured under the condition of hypoxia and reoxygenation. The cultured cardiomyocytes were then divided into seven groups: hypertrophic cardiomyocyte control (HC) group, hypoxia for 8h (H8h) group, hypoxia for 12h (H12h) group, hypoxia for 8h and reperfusion for 4h (H8h/R) group, hypoxia for 12h and reperfusion for 4h (H12h/R) group, hypoxia for 12h+siRNA transfection (H12h+siRNA) group, and hypoxia for 12h and reperfusion for 4h+siRNA transfection (H12h/R+siRNA) group. Cardiomyocytes were transfected with AIF siRNA. Apoptosis was detected with Hoechst 33258 staining. The AIF mRNA and protein expressions were detected by RT-PCR and Western blotting. Results The levels of AIF mRNA and protein expression were significantly higher in H8h group and H12h group (mRNA: 0.52?0.04, 0.85?0.10, respectively; protein: 2.07?0.15, 3.12?0.19, respectively) compared with that in HC group (mRNA: 0.29?0.08; protein: 1.00?0.04, P0.05). Objective To detect the variation of vascular endothelial growth factor (VEGF) in the splanchnic vessels under portal hypertension (PHT) and explore its mechanism and influence on the process of hyperdynamic circulation.Methods In humans,the level of VEGF pathway related proteins were detected in the splenic artery of PHT patients in clinical trials.In animal experiments,the following parameters were observed for rats in the control group (group N,n =7) and the CCl4 induced portal hypertension group (group PHT,n=7):portal vein diameter,portal vein blood flow velocity (PBV),portal vein blood flow (PBF),portal vein pressure (PP),norepinephrine (NE) reactivity in the isolated mesenteric artery microcirculation,contractile reactivity and degree of endothelial ni tric oxide synthase (eNOS) activation in the mesenteric artery by selectively inhibiting the VEGF signal pathway with SU5416.In cell experiments,primary culturing of arterial endothelial cells in vitro were used to verify the effects of VEGF on eNOS activation.Results The results showed that VEGF expression levels in the splenic artery of PHT patients significantly increased.In animal experiments,there was not a significant difference in portal vein diameter between group N and group PHT.How ever,the PBV and PBF of group PHT were lower than those of group N,and SU5416 had no clear effect on PBV and PBF in group PHT.PP of group PHT was much higher than that of group N,and SU5416 had little influence on reducing PP in group PHT.The contractile response of mesenteric artery microcirculation to NE in group PHT decreased significantly,EC50 increased a lot,and SU5416 improved this hypoergia partially.The protein levels of VEGF,VEGFR-2,eNOS,and p-eNOS in the mesentery artery of group PHT raised quite a lot compared to group N,and SU5416 decreased the protein level of VEGFR-2 and activation of eNOS significantly.Experiments in vitro confirmed that VEGF could promote the activation of eNOS.Conclusion The excessive VEGF produced in visceral arteries under PHT may participate in the process of hyperdynamic circulation partially through promoting the synthesis and activation of eNOS and then reducing visceral arteries' response to NE.
