1.Effect of Mild Hypothermia on Anti-brain Antibody and Cerebral Haemodynamics of Patients with Severe Traumatic Brain Injury

Objective To study the protective mechanism of mild hypothermia from severe traumatic brain injury (sTBI). Methods 160 cases with sTBI were treated under normothermia (NT group, n=80) and mild hypothermia (HT group, n=80) respectively. Their serum level of anti-brain antibody (ABAb) was measured with ELISA, cerebral haemodynamics with transcranial Doppler, and intracranial pressure (ICP) with ICP monitor 1 d, 3 d, 5 d, 7 d and 14 d after injury, while they were assessed with Glasgow coma scale (GCS) and Glasgow outcome scale (GOS). Results The level of ABAb was lower in HT group than in NT group (P<0.05) at every point. The scores of GCS increased (P<0.05), and the volume of systole, mean and pulse index improved more in HT group 5 d after injury (P<0.01). The ICP was lower in HT group 3 d after injury (P<0.01). The score of GOS was better in HT group than in NT group when discharged (P<0.05). Conclusion Mild hypothermia can reduce the level of serum anti-brain antibody, improve cerebral haemodynamics, decrease ICP and improve the prognosis in patients with sTBl.
2.Establishment of fingerprint of processing Gentianae scraba Bge and its mutual mode by HPLC

AIM:To establish the HPLC fingerprint of methanol-resoluble component of processing Gentiana scraba Bge collected from different habits to provide evidence for quality control and quality evaluation of Gentiana scraba Bge.METHODS:Ten batches of processing Gentiana scraba Bge were measured by RP-HPLC,and the mutual mode was made by the similarity degree evaluating system 2004 of Chromatographic Fingerprint of Traditional Chinese Medicine promulgated by Chromatographic condition,consisted of Agilent-Eclipse-XDB-C_ 18-column and gradient elution with methanol-water,the detection wavelength was at 254 nm,flow rate was at 1.0 mL/min and the column temperature was at 20 ℃.RESULTS:The mutual mode of processing Gentiana scraba Bge was set up and the similarity of fingerprints of processing Gentiana scraba Bge from different habits was gained.CONCLUSION:According to the analysis of statistics,this method is steady,accurate with good repeatability and can be used as standardization implantation and quality control of Gentiana scraba Bge.
3.A Meta-analysis on effect of antibody blood screening in preventing transmission of cytomegalovirus by transfusion

Objective To determine the effect of using cytomegalovirus-seronegative blood components in preventing transfusion-acquired cytomegalovirus infection,which laid foundation for the application of blood antibody screening of cytomegalovirus.Methods The documents of studies about the comparison in transfusion-acquired cytomegalovirus ratio between using cytomegalovirus-seronegative blood components with using cytomegalovirus-unscreened /non-WBC-reduced blood were retrieved from the databases of PubMed,MEDLINE,Ovid,ProQuest,EBSCO,Cochrane Library,EMbase,CNKI,VIP,CBM and WanFang Library,and the reference in studies were retrieved by hands at the same time.The documents were screened,extracted and evaluated according to inclusion and exclusion criteria,and then given a Meta-analysis by using Rev Man 5.1 software.Results There were totally 7 controlled studies(430 patients) included.The results of Meta-analysis showed that compared with using cytomegalovirus-unscreened/non-WBC-reduced blood,the effect of using cytomegalovirus-seronegative blood components in preventing transfusion-acquired cytomegalovirus infection had a statistical difference(OR=0.07,95%CI:0.03-0.18,P<0.01).Conclusion Application of blood antibody screening of cytomegalovirus is effective in preventing transfusion-acquired cytomegalovirus infection,especially organ transplantation and neonate patients.
4.Assessment of journals according to their weighted citations

A method to assess library-holding journals according to their weighted citations was proposed in light of the actual needs of users for core journals, the use of library-holding journals and references by authors in different orders of precedence.The Western journal of rheumatology, highly cited by authors of Second Military Medical U-niversity, were assessed using this method, showing that this method is better than the citation analysis-based tra-ditional method in assessment of journals.
5.Effect of naloxone in combination with morphine on the growth of subcutaneous tumor of human gastric cancer MGC-803 cells in nude mice

Objective To observe the effects of naloxone in combination with morphine on the growth of subcutaneous tumor of human gastric cancer MGC-803 cells in nude mice.Methods The model of subcutaneous tumor of human gastric cancer MGC-803 cells in nude mice was established.Fifty nude mice were randomly divided into 5 groups: control group (group C),normal saline group (group S),20 mg/kg morphine group (group M),and 1 mg/kg naloxone group (group N),and 1 mg/kg naloxone+20 mg/kg morphine group (group NM).The mice in group C received no treatment,while the mice in group S,group M,group N and group NM were injected with 1.5 ml/kg saline,20 mg/kg morphine,1 mg/kg naloxone,and 1 mg/kg naloxone+20 mg/kg morphine per day,respectively.The caliper was used to measure the tumor sizes every the other day.The mice in each group received intraperitoneal injection of the drugs for 2 week.Then the relative volume (RTV) of tumor was calculated.The expression of Cyclin D1,VEGF,MMP-9 mRNA and proteins were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR),immunochemistry staining and Western blot.Results RTV in group M (2.21±0.62)% was significantly lower than that in group C (3.16±0.68)%,group S (2.98±0.61)%,group N (3.16±0.35)% and group NM (2.64±0.37)% (P<0.05).RTV in group NM was significantly lower than that in group C,group S and group N (P<0.05).Compared with group C,the expression of Cyclin D1,VEGF,and MMP-9 in group M were significantly decreased (P<0.05).The organizational structure of the subcutaneous tumor in groups C,S,N and NM was almost normal.Cytoplasm vacuolization,disruption of nuclear membrane and chromatin margination were occured in group M.While the level of Cyclin D1,VEGF,and MMP-9 in group NM was increased compared to group M (P<0.05).Conclusion Morphine could inhibit the growth of subcutaneous tumor of human gastric cancer MGC-803 cells in nude mice by downregulating the expression of Cyclin D1,VEGF,and MMP-9.Naloxone could antagonize the anti-growth effects of morphine.
8.Structural difference of gut microbiota in obese patients with or without acanthosis nigricans

Objective To investigate the distribution of gut microbiota in obese patients with or without acanthosis nigricans .Methods Totally 131 obese patients and 25 healthy participants were divided into three groups:the obesity with acanthosis nigricans (AN) group (n=59), the simple obesity (OB) group (n=79), and the control (CON) group (n=25).The fresh stool samples were collected , and the clinical and biochemistry markers were measured .Pyrosequencing technology was performed based on the 16s rRNA of fecal samples to identify and analyze the distribution pattern of gut microbiota in each group .Results The AN group had signifi-cantly higher body mass index [ (37.45 ±5.12) kg/m2 vs.(33.34 ±2.54) kg/m2 vs. (20.35 ±1.68) kg/m2, P=0.045, P<0.001], insulin [32.77 (25.18) mU/L vs.20.73 (9.30) mU/L vs.8.70 (6.18) mU/L, P<0.001, P<0.001], insulin resistance [7.78 (6.87) vs.4.71 (2.88) vs.1.81 (1.40), P<0.001, P<0.001], and interleukin (IL) -6 [ (3.64 ±2.23) ng/L vs.(2.71 ±0.78) ng/L vs.(2.17 ±0.86) ng/L, P=0.040, P=0.009] levels than OB and CON groups compared with OB and CON groups , AN group had sig-nificantly decreased diversity of bacterial flora ( P=0.015 , P=0.001 ) , while no significant difference was observed in the abundance of bacterial flora .At the phylum level , the composition of flora among these three groups was similar, mainly including bacteroidetes , firmicutes, proteobacteria, and actinomycetes.Although the proportions of main bacteria flora were different , the difference was not statistically significant .At the genus level, the bacteria flora in AN and OB groups were primarily composed of Bacteroides, Megamonas, Faecalibac-terium and Escherichia-Shigella.In addition, compared to OB and CON groups , AN group had significantly lower proportion of Ruminococcus ( P=0.023 , P=0.043 , respectively ) and higher proportion of Veillonella (P=0.048, P=0.043, respectively).Furthermore, the proportion of Weissella was higher in AN and OB groups than in CON group ( P=0.045 , P=0.025 ) .Conclusion Obese patients with AN have more severe in-sulin resistance and inflammation status than those with simple obesity , and the distribution feature of gut micro-biota also differ between these two patient populations .
9.Molecular subtypes of Uygur and Han patients with breast cancer in Kashgar, Xinjiang

Objective:This study aimed to compare the differences in molecular subtypes between Uygur and Han patients with breast cancer. This study was also conducted to provide clinical recommendations. Methods:The new typing standard of St. Gallen International Expert Consensus on the Primary Therapy of Early Breast Cancer 2011 was used to classify the molecular subtypes of 369 breast cancer cases from the First People's Hospital of Kashgar Prefecture, Xinjiang Uygur Autonomous Region, China. Four immunohistochemical markers (ER, PR, HER-2, and Ki-67) were used to divide the patients into four intrinsic subtypes:Luminal A;Luminal B (divided into HER-2 negative subtype and HER-2 positive subtype);HER-2 enriched;and triple negative breast cancer (TNBC) subtype. Statistical analysis was then conducted to evaluate the differences in molecular subtype characteristics of Uygur and Han patients with breast cancer. Results:The proportion of Luminal A, Luminal B with HER-2 negative subtype, Luminal B with HER-2 positive subtype, HER-2 enriched, and TNBC subtype were 12.67%(36/284), 34.51%(98/284), 20.07%(57/284), 14.79%(42/284), and 17.96%(51/284) for 284 Uygur patients with breast cancer;the corresponding proportions were 16.47%(14/85), 37.65%(32/85), 10.59%(9/85), 10.59%(9/85), and 24.71%(21/85) for 85 Han patients with breast cancer, respectively. The HER-2 positive rates of Uygur and Han patients with breast cancer were 34.86%(99/284) and 21.18%(18/85), respectively. The proportions of Luminal B with HER-2 positive subtype and HER-2 positive rate were significantly higher in Uygur patients than in Han patients (P=0.045 and P=0.030, respectively).Conclusion:A larger proportion of Luminal B with HER-2 positive subtype and a higher HER-2 positive rate were observed in Uygur patients with breast cancer than in Han patients with the same disease. HER-2-targeted therapy could be more effective for Uygur patients with breast cancer than for Han patients.
10.Design and Construction of an Embedded Telemetry System for Potentiometric Sensors

A new embedded telemetry system for potentiometric sensors was developed. The system consisted of a transmitter unit, a receiver unit and a personal computer ( PC) . The transmitter unit included a current amplifier, a∑-Δanalog-to-digital converter ( ADC) , a microcontroller unit ( MCU) and a radio module. The receiver unit was composed of a radio module, a microcontroller unit and a serial-to-USB converter module. The receiver unit was connected to an upper computer via a universal serial bus ( USB ) . The embedded software written in C language controlled the signal acquisition and transmission. The computer software written in LabVIEW language was used for data storage and display. The range of the acquisition voltages was from-1. 17 V to +1. 17 V. In order to verify the accuracy and reliability of this system, a control experiment had been done with this system and a digital multimeter. Moreover, a response test of acidity changes had been done with the self-made H+ selective electrode. The results showed that the accuracy of this system could be up to 0. 1 mV and it had strong anti-noise ability. The square of the linear correlation coefficient of the response of the changes in pH values was 0 . 998 . The curve of the results measured by this system was consistent with that measured by the commercial electrochemical analyzer. The system was built with standard hardware components and the size of the transmitter unit was only 29 mm×14 mm×11 mm. It can be easily used for remote and real-time detection for the potentiometric sensor.
