Objective To investigate the anti-migratory and anti-invasive effect of somatostatin receptor type 2(SSTR2) gene transfection mediated by adenovirus in human pancreatic carcinoma cell and the mechanisms involved in this effect. Methods The full length human SSTR2 cDNA was introduced into pancreatic cancer cell line BXPC-3 by adenovirus-mediated transfection, and stable expression of RNA and protein of SSTR2 were detected by RT-PCR and Westen-blot.The Matrigel coated Transwell was used to detect the migratory and invasive ability of SSTR2expressing cells,Adv-GFP control cells and mock control cells.Furthermore, the expressions of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase-2(TIMP-2) were detected by RT-PCR method in these cells. Results The stable expression of SSTR2 was detected in BXPC-3 cells transfected by Adv-GFP-SSTR2.A dramatic decrease of BXPC-3 expressing SSTR2 cell(migrated) through a Matrigel-coated filter was observed,as compared with Adv-GFP control cells and mock control cells(P

Objective To investigate the injury characteristics and prognostic risk factors of patients with craniocerebral injury so as to provide some suggestions for the improvement of medical prevention, intervention and treatment measures. Methods Trauma database system was applied to collect the medical records of 661 patients with traumatic brain injury admitted to the Department of Neurosurgery of 105th Hospital of Hefei in 2009.Their general conditions, injury characteristics and outcomes were analyzed and the risk factors affecting their outcomes were investigated by Logistic regression analysis. Results Of the patients with craniocerebral injury, the males accounted for 70% (463/661).The patients at 45 years old or so had the highest incidence of craniocerebral injury.Patient number in the second half of one year was 1.65 times more than that in the first half of one year (412/249 ).Most patients were injured from traffic accidents (87.7%,580/661 ) and 512 patients (77.5%) suffered from multiple injuries.ISS,injury causes, age and injury incidence quarters showed close correlation with the final outcome. Conclusions The patients with craniocerebral injury demonstrates distinctive characteristics in aspects of gender, age, incidence time, injury causes, and multiple injuries, but the ultimate outcomes are related to the injury severity, injury causes, age and other factors.

Objective To explore the value of Ligasure vessel sealing system(LVSS)during resection of retroperitoneal tumor. Methods Group 1 including 32 cases were performed resection of retroperitoneal tumor with LVSS and electrosurgical seapel from Jun.2004 to Oct.2008.group 2 including 26 cases were done with electrosurgieal seapel from Jan.2001 to Jun.2004.Operating blood loss, operating time, iatrogenic injury, postoperative blood loss and hospital stay were compared between the two groups. Results There was no significant difference in hospital stay[(11.7±0.7)d vs.(12.3±1.4)d)]and iatrogenic injury(9.38%vs.16.00%)between two groups statistically(P>0.05),but the intraoperative blood lose[(403.1±37.1)ml vs.(704.0±129.0)m1)s,postoperative blood loss[(131.5±18.4)ml vs.(214.8±29.2)ml)]and operating time[(166.5±8.9)min vs.(186.8±15.4)min]were less in group 1 than that in group 2(P<0.05).Conclusion Ligasure vessel sealing system has advantages of safe coagulation, shortening operation time in resection of retroperitoneal tumor.

Chinese Journal of Pathophysiology 1999;0(09):-
AIM: This study was designed to investigate the inhibition of tanshinone ⅡA on C6 glioma cell line and its mechanism. METHODS: MTT was used to measure the levels of the proliferation of C6 cultured with tanshinone ⅡA at different concentrations. The effects of tanshinone ⅡA on cell cycle of C6 were observed by FCM. The change of DNA was observed by Sepharose electrophoresis. The expression of proto-oncogenes c-myc was measured by RT-PCR. RESULTS: The proliferation of C6 was obviously inhibited by tanshinone ⅡA in a dose-dependent manner. The outcome of FCM showed that the apoptotic cell rate was 7.7%, when cultured with tanshinone ⅡA at 1.0 mg/L for 3 days. The apoptotic cell rate was 21.6%, when cultured with tanshinone ⅡA at 2.0 mg/L in 3 days. CONCLUSION: Tanshinone ⅡA inhibits the proliferation of C6 cells, induces apoptosis and inhibits the expression of proto-oncogene c-myc.

Objective To investigate the clinical characteristics and evaluate surgical experiences in Hashimoto's disease patients suffering from coexistent thyroid nodules. Method Clinical data in 299 cases of Hashimoto's disease coexistent with thyroid nodules treated surgically from Jun 1985 to Dec 2004 were analyzed. Results The coexistent rate of Hashimoto's disease (HD) with thyroid carcinoma (TC) . thyroid adenoma.nodular goiter.hypothyroidism and hyperthyroidism was 17. 4% , 34. 4% , 5. 7% , 3. 7% and 2. 3% respectively. The coexistent rate of HD with TC was 9. 2% before 1995, and 19. 7% after 1996. There were 35 cases of papillary carcinoma, 11 cases of follicular carcinoma, 4 cases of mixed type and one case of lymphoma. There were 17 cases of occult thyroid carcinoma, accounting for 32.7% of the malignancy. Operative procedures included biopsy only and thyroidectomy according to frozen pathology. Conclusions There is a high incidence of coexistent thyroid neoplasm in patients with Hashimoto's disease, the incidence of thyroid carcinoma especially occult carcinoma is rising recently.

Objective To explore the expression and clinical significance of EphA2 and E-cadherin in (pancreatic) cancer tissue and cells. Methods The expression of EphA2 and E-cadherin in 56 pancreatic(carcinomas) and 23 adjacent noncancer tissues were studied by immunohistochemistry, and their relationship to clinicopathological characteristics were analyzed.RT-PCR was performed to explore the expression of EphA2,BXPC-3,PC-3 and PANC-1 in pancreatic carcinoma cell lines. Results In pancreatic carcinomas tissuse showed increased EphA2 expression and reduced E-cad expression which compared with adjacent noncancer tissues. The expression level of EphA2 had a significant positive relationship with tumor differentiation degree, lymphatic metastasis and clinical stage. However, the expression level of E-cadherin had negative relationship with both the tumor clinical stage and lymphatic invasion.Furthermore,a significant negative relationship(between) the expression of EphA2 and E-cadherin was observed. The expressions of EphA2 were higher in high-invasive cell lines PXPC-3 and Panc-1 than in low-invasive cell line PC-3.Conclusions The(expression) and/or abnormal function of EphA2 and E-cadherin may together be involved in the development and progression of pancreatic cancer;the combined measurement of these two proteins may be useful for(determination) of metastatic potency of pancreatic carcinoma.
