Protein phosphatase 2A(PP2A)is a major Ser/Thr phosphatase involved in sevelral cellular signal transduction pathways.The reversible phosphorylation of proteins is accomplished by opposing activities of kinases and phosphatases.PP2A controls the specific dephosphorylation of thousands of phosphoprotein substrates and thus regulates physiological and biochemical processes of organism.The changes of the physiological activities of PP2A closely relate to apoptosis and regeneration of injured intestinal epithelial cell,and PP2A may be a regulatory factor in balancing the apoptosis and regeneration of intestinal epithehal cell.

Objective To explore the value of the multi-phasic spiral CT scan in the diagnosis of gallbladder carcinoma.Methods The characteristic CT findings were reviewed in 42 cases with gallbladder carcinoma proven pathologically.Results The gall bladder carcinoma had certain enhancing characteristics: high density in hepatic artery period,same or high density in porch vein period,and no clear fadeaway in delaying period.In other words,the gall bladder carcinoma was higher enhancement than normal gallbladder wall and liver,and had long-term enhancement.Conclusion The multi-phasic spiral CT scan plays an important role in diagnosis and differential diagnosis of gallbladder carcinoma.

Background Gene transfection is an effective therapeutic avenue to target many kinds of eye diseases.Non-viral vectors with high transfection efficiency,long-term expression,low toxicity and high expression levels are pivotal in gene therapy of corneal disease.Objective This study was to evaluate and compare the safety and efficiency between EntransterTM and liposome vectors for transfer of CD25 siRNA in rat cornea.Methods Eighty male SPF SD rats were randomly divided into EntransterTM-CD25 siRNA group,liposome-CD25 siRNA group,simple CD25 siRNA group and normal saline solution (NSS) group with the right eye as experimental eyes.Corneal epithelia of the rats were completely removed after ocular surficial anesthesia,and 50 μl EntransterTM-CD25 siRNA,liposome-CD25 siRNA,CD25 siRNA solution and NSS were topical administered in the eyes respectively.Ocular response and green fluorescence number on the corneas were examined under the slit lamp assisted microscope 12 hours,24 hours,3 days and 7 days after use of the drugs.The rats were sacrificed and the corneas were obtained,and corneal histopathological examination was performed by using hematoxylin eosin stain.The gene transferred efficiency in the corneas was evaluated by fluorescence technology,and the safety of EntransterTM and liposome carriers was assessed using TUNEL stain.The expression and location of CD11b in the corneas were detected by immunofluorescence technology.The use and care of the experimental animals complied with Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Committee.Results The quantity and intensity of fluorescence staining in the corneas were significantly increased in the EntransterTMCD25 siRNA group in comparison with the liposome-CD25 siRNA group,and the corneal fluorescence appeared earlier in the simple CD25 siRNA group,but it disappeared in 24 hours after transfection.Corneal histopathological examination revealed that the corneal edema and inflammatory cell infiltration in corneal epithelium after gene transfection were more dominant in the liposome-CD25 siRNA group than those in the EntransterTM-CD25 siRNA group,simple CD25 siRNA group and NSS group,and no abnormality was seen in the stroma and endothelium.The number of inflammatory cells was more in the liposome-CD25 siRNA group than that in the EntransterTM-CD25 siRNA group,simple CD25 siRNA group and NSS group (all at P =0.00).The number of apoptosis cells was significantly more in the liposome-CD25 siRNA group than that in the EntransterTM-CD25 siRNA group,simple CD25 siRNA group and NSS group in 12 hours and 3 days after transfection (all at P =0.00).Immunofluorescence assay showed the expression of CD11b primarily located in the corneal epithelial and stromal layers.The expression of CD11b was gradually enhanced over time in the liposome-CD25 siRNA group and peaked in 24 hours after transfection.However,the expression was absent in the EntransterTM-CD25 siRNA group,simple CD25 siRNA group and NSS group.Conclusions EntransterTM nanometer material-mediated transfection of CD25 siRNA in corneas of normal SD rats appears to have high transfection efficiency,low toxicity and slight irritating response to corneas,and EntransterTM vector is currently available for the gene therapy of corneal disease.

Objective To investigate the calcification of thyroid nodules detected by ultrasound and its relation with thyroid carcinoma.Methods Four thousand one hundred and eighty-six cases of thyroid disease were surgically treated during a period of five years.Retrospective analysis was made to compare preoperative color Doppler ultrasound and postoperative pathology.Results The incidence of calcification,non-microcalcification,and microcalcification were significantly higher in thyroid carcinoma than that in benign diseases(P＜0.05).For predicting thyroid cancer,microcalcification was more significant than non-microcalcification(P＜0.01).There was a significant difierence in the microcalcification between the different age groups(2.4%for patients of 45 and older,16.8%for patients younger than 45;P＜0.05):There was no difference in the calcification between male and female groups(P＞0.05);There was a significant difierence in malignant calcified nodules between difierent nodule groups(70.7%for patients of solitat'y.49.1%for patients of muhiple:P＜0.05).Conclusion Microcalcifications in the thyroid is related to thyroid cancer.especially to micro-papillary carcinoma.

BACKGROUND: Preparing the tracheal prosthesis by biomaterials is crucial for studying the implant of long tracheal defect.The biocompatibility and biodegradability of collagen/hydroxyapatite (Col/HAp) and poly(lactic-co-glycolic acid) (PLGA) in vivo, as a artificial material for tracheal prosthesis, need to be observed.OBJECTIVE: To explore in vivo biodegradability of Col/HAp and PLGA of tracheal prosthesis in rats.DESIGN: Randomized controlling observation.SETTING: Department of Thoracic and Cardiovascular Surgery, Clinical College of Yangzhou University; Department of Thoracic and Cardiovascular Surgery, Changzheng Hospital of the Second Military Medical University of Chinese PLA.MATERIALS: The experiment was carried out in the National Experimental Animal Center of the Second Military Medical University of Chinese PLA from March 2003 to December 2004. Sixteen SD rats of either gender were offered by this center, aged three months and weighed 150-170 g. Col/HAp and PLGA (the copolymer of polylactic acid and polyglycolic acid according to the percent of 90:10) were self-made.METHODS: ①Empirical process: Rats were fed adaptively for 1 week and anesthetized. Then undermining dissection was performed along with musculi dorsal surface toward spine bilaterally, so as to form two capsular gaps, which were implanted with Col/HAp sponge and PLGA fiber mesh cloth respectively, in a size of 10 mm ×10 mm. ②Empirical evaluation: The postoperative activity, incision healing and rejection of rats were observed; 2, 4, 6, 8 weeks postoperatively, the Col/HAp sponge and PLGA mat that were embedded subcutaneously as well as surrounding tissues were determined using scanning electron microscope or transmission electron microscope.MAIN OUTCOME MEASURES: In vivo biodegradability of two materials implanted in rats at different time points.RESULTS: ①All the animals could carry out normal activity, respiration and diet; no incision infection, fluidity, necrosis or sinus tract appeared; there was no edema in the skins of implanted area, neither hypersensitiveness nor toxicity was found; the implant materials had no rejection. ②The features of the implanted materials varied throughout the implantation. During the follow-up, the infiltration of inflammatory cells was found in the interface between the implants and neighboring host tissues in the early stage, phagocytes and fibroblastic cells were also observed later. As the process went on, the materials were biodegraded gradually, encapsulated by phagocytes, and replaced by newly generated fibrous tissues. No remarkable harmful influences of the composite materials on the neighboring host tissues such as apomorphosis, necrosis, hyperplasia and foreign body reaction were observed grossly and microscopically.CONCLUSION: All the implants show a good biocompatibility and biodegradability. Both Col/HAp and PLGA go through a gradual process of biodegradability, biological absorption and replacement by host tissues ultimately in vivo, which suggest that these two kinds of composite biomaterials will be used safely in developing tracheal prosthesis.
