Objective:To investigate the antioxidative effect of rosiglitazone(RSG),a new insulin sensitizer,on kidney of rats with unilateral ureteral obstruction(UUO).Methods: Male adult Sprague-Dawley rats were randomly divided into 3 groups: Sham-operated group(n=6),UUO model group(UUO model was established by ligating the left ureter,n=18),and UUO+RSG group(UUO model rats were treated with RSG,n=18).The latter 2 groups were divided into subgroups according to the times(3 d,7 d and 14 d) after intervention(n=6).Malondialdehyde(MDA),catalase(CAT) and superoxide dismutase(SOD) contents in the homogenized lysate of obstructive renal cortex were determined.Results: Compared with those in Sham-operated group,MDA contents in UUO group increased by 8.9(P=0.000),3.5(P=0.000) and 0.7 times on d 3,d 7 and d 14,respectively,while the contents of CAT and SOD decreased dramatically,with CAT contents reduced by 66.0%,(72.6%) and 83.8%,respectively(P=0.000) and SOD contents reduced by 42.0%,50.2% and 67.7%,respectively(P

4.A spleno-pancreas preserving lymph node clearance in the radical resection of gastric carcinoma

ObjectiveTo evaluate the results of a novel operative procedure of spleen and pancreas preserving clearance of lymph nodes on spleen hillus and along the splenic artery in gastric resection for gastric carcinoma.Methods The postoperative complications and survival rates were observed in 61 patients of gastric carcinoma undergoing gastrectomy with preservation of the spleen and pancreas (PSP) and clearance of lymph nodes on spleen hillus and along splenic artery.Results were compared with those undergoing traditional operations. Results The postoperative complications and mortality were lower and survival rates higher in patients with preservation of spleen and pancreas than those with spleen and a portion of the pancreas resected.Conclusions Spleno-pancreas preserving lymph node clearance on the splenic hillus and along pancreatic artery is safe and effective procedure in radical gastrectomy for patients with gastric carcinoma.
5.Investigation of Quality of 31 Batches of Compound Glycyrrhiza Oral Solutions

OBJECTIVE:To investigate the internal quality of 31 batches of Compound Glycyrrhiza oral solutions from different manufacturers.METHODS:The contents of Morphine and Guaifenesin in Compound Glycyrrhiza oral solutions were determined by HPLC,and the pH value of the solution was detected as well.RESULTS:Of the total 31 batches of samples,13 was unqualified in Morphine content,1 was unqualified in Guaifenesin content,and 1 was unqualified in pH value.CONCLUSION:The manufacturers should strictly obey the standards recorded in China Pharmacopoeia in respect of quality control to ensure the safety and efficacy of clinical medication.

7. Protective effect of peroxisome proliferator-activated receptor γ on TNFα-induced injury of inner medullary collecting duct cells

Objective: To observe the protective effect of peroxisome proliferator-activated receptor γ (PPARγ) on TNFα-induced injury of the inner medullary collecting duct (IMCD) cells. Methods: Cultured IMC D cells were randomly divided into blank control, TNFα stimulation, PPARγ transfection (pPPARγ-control) and PPARγ transfection + TNFα stimulation groups. Stimulation was induced with 50 ng/ml TNFα for 24 h and mouse wild-type PPARγ plasmid was used to transfect IMCD cells. Cell supernatant MCP-1 or TGFβ1 was detected by ELISA method. Results: IMCD cells transfected with wild-type PPARγ pla smid had high expression of PPARγ mRNA and protein. The contents of MCP-1 and TGFβ1 in the supernatant were similar in blank control group and pPPARγ-control group. Compared with blank control group, TNFα stimulation group had decreased contents of MCP-1 and TGFβ1 in the supernatant (P<0.005), but the contents of TNFα stimulation group were significantly higher than those of PPARγ transfection + TNFα stimulation group (P<0.05 and P<0.005, respectively). Conclusion: IMCD cells over-expressing PPARγ have anti-inflammatory and anti-fibrosis effects.

Conclusion: Rosiglit azone can suppress oxidative stress in rats with UUO by reducing lipid peroxidation production and improving antioxidative enzyme contents.
9. Influence of rosiglitazone on peritoneal fibrosis induced by peritoneal dialysate in rats

Objective: To investigate the effect of rosiglitazone (RGZ) in prevention of peritoneal fibrosis induced by peritoneal dialysis in rats. Methods: Fifty SD rats were randomly divided into six groups: control group (n = 5), normal saline (NS) group(n = 9), model group (n=9), dimethyl sulfoxide(DMSO) group(n = 9), low-dose RGZ group (n = 9), and high-dose RGZ group (n = 9). Peritoneal catheters were implanted in the last five groups, and peritoneal fibrosis models were induced in the last four groups by high-glucose peritoneal dialysate and erythromycin with rats. Animals in the low-dose RGZ and high-dose RGZ groups were treated with 1.5 mg/kg RGZ and 15 mg/kg RGZ, respectively. The one hour peritoneal equilibration test was performed and the plasma glucose and serum lipids were estimated five weeks after dialysis. The ultrafiltration volume (UF), dialysate-to-plasma urea ratio (D/Purea), and glucose reabsorption (D1/D0) were calculated. The visceral peritoneum tissues of rats were stained with hematoxylin-eosin(H-E) and Masson trichrome staining to observe the changes of peritoneal morphology. Blood vessels and leukocytes of peritoneum were quantified as n/mm2 within the histological sections with H-E staining. The expression of TGF-β1 and α-SMA in the parietal peritoneum was detected by immunohistochemistry assay. Results: Compared with the control group, the numbers of peritoneal vessels, leukocytes, peritoneal thickness, and the expressions of TGF-β1 and α-SMA were significantly higher in the model group and DMSO group (P<0.05). Administration of RGZ improved the above changes and significantly decreased the expression of TGF-β1 and α-SMA in the model and DMSO groups (P<0.05). Compared with the control group, the UF and D1/D0 were significantly lower and D/Purea was significantly higher in the rest five groups (P<0.05), and there were no significant differences between the low-dose RGZ group and the high-dose RGZ group(P>0.05). Conclusion: Administration of rosiglitazone can effectively protect the ultrafiltration function of peritoneum during peritoneal dialysis and delay the progression of peritoneal fibrosis.
10.Effects of dexamethasone on acute lung injury in rats induced by lipopolysacharide

Objective: To study the difference between the preventive and therapeutic effects of dexamethasone on acute lung injury models induced by lipopolysacharide(LPS) in different phases.Methods: Forty adult male Wistar rats were randomly divided into four groups:(1)control group to receive intraperitoneal NS injection(2 mL/kg).(2)LPS group to receive intraperitoneal LPS injection(5(mg/kg).)(3)one-hour group to receive intraperitoneal dexamethasone injection(2 mg/kg) one hour after LPS injection.(4)three-hour group to receive intraperitoneal dexamethasone injection(2 mg/kg) three hours after LPS injection.Then histopathology,arterial blood gases,lung permeability,wet-to-dry weight ratio and immunohistochemistry AQP1 were performed 24 hours later.Results: Dexamethasone could improve biological indexes.Lung permeability,wet-to-dry weight ratio and immunohistochemistry AQP1 were(5.73?1.37),(4.92?0.23),(19.92?6.47) in LPS group,(2.4?0.51),(4.89?0.21),(33.47?9.41) in one-hour group and(2.15?0.63),(4.57?0.14),(40.69?9.18) in three-hour group,respectively.Dates in three-hour group were prior to those of one-hour group,and there was slight but no significant difference between the two groups.Conclusion: Dexamethasone can improve lung permeability and reduce lung edema.There is no need to be treated with glucocorticoids in advance.
