1.Study on the Content Dynamic Changes of Caffeic Acid,Vicenin-2 and Isoschaftoside in Rabdosia lophanthoi-des

OBJECTIVE:To establish a method for simultaneous determination of caffeic acid,vicenin-2 and isoschaftoside in Rabdosia lophanthoides to assess its optimal harvest period. METHODS:HPLC was performed on the column of Diamonsil C18 with mobile phase of methanol-0.1% phosphoric acid solution (gradient elution) at a flow rate of 0.8 ml/min ,detection wave-length was 334 nm ,column temperature was 25 ℃ and volume injection was 15 μl. RESULTS:The linear range was 0.23-5.68 μg/ml for caffeic acid(r=0.999 9),0.31-7.76 μg/ml for vicenin-2(r=0.999 8)and 0.45-11.30 μg/ml for isoschaftoside(r=0.999 9);RSDs of precision,stability and reproducibility tests were lower than 2.0%,average recoveries were 95.1%-98.9%(RSD=1.8%, n=6),97.4%-101.3%(RSD=1.7%,n=6) and 95.5%-98.8%(RSD=1.4%,n=6),respectively. The contents of above-men-tioned 3 ingredients of R. lophanthoides were the highest harvested in Apr. to May and Jul. to Aug. in a year. CONCLUSIONS:The method is simple,accurate and reproducible,and can be used for the simultaneous determination of caffeic acid,vicenin-2 and isoschaftoside in R. lophanthoides. The optimal harvest period is Apr. to May and Jul. to Aug. in a year.

3.Right hemihepatectomy through the right retrohepatic tunnel

Objective To study the clinical value of anatomizing and ligating the short hepatic veins (SHVs) and right hepatic veins through the right Retrohepatic Tunnel (RT) in right hemibepatectomy of giant hepatic carcinoma.Methods We performed the right hemibepatectomy on 23 patients with huge tumors which diameters were lager than 8 cm.There were four main procedures during the operation.Firstly,sperated the interspace between right hepatic veins and middle hepatic veins through secundum porta hepatis.Secondly,seperated and ligated the right hepatic portal vein and hepatic artery in the Glisson's system.Thirdly,anatomized and ligated SHVs and right hepatic veins through the right RT conversly.Finally,set a blocking-belt at the left of the central liver and then cut the right hemi-hepar.Results The anatomy of right hepatic artery,Portal Vein,retrohepatic inferior vena cava (RHIVC),right hepatic veins was well done.The intraoperative blood loss of 7 patients was less than 400 mL,and 11 was 500-700 mL,2 was 800-1 000 mL,3 was 1 100-1 400 mL,and average was 640 mL.The hepatic function was changed in the third day afteroperation.The numerus of TBIL of 16 patients was 20-40 μmol/L,and 6 was 45-50 μmol/L,and 1 was 60 μmol/L.The numerus of ALT of 14 patients was 150-200 U/L,and 9 was 250-400 U/L.The numerus of AST of of 13 patients was 160-200 U/L,and 9 was 230-400 U/L,and 1 was 430 U/L.The numerus of GGT of 14 patients was 160-200 U/L,and 8 was 220-310 U/L,and 1 was 420 U/L.The hepaticfailure happened to 1 patient because of deep venous infection.Conclusions The right hemibepatectomy through the right retrohepatic tunnel is feasible in the tumor without invasion the RHIVC and diaphragm,or the volume was not too huge.The operation could reduce the blood loss and make for the recovery of hepatic funtions.
4.Bile leakage in transjugular intrahepatic portosystemic shunt: effect on growth and function of endothelium

Objective To evaluate the effect of bile on growth and function of endothelial cells in vitro. Methods In vitro, endothelial cells explanted from human umbilical vein were cultured. The experiment was designed as 6 groups with concentration of 5%, 10%, 15%, 20%, 25% bile and without bile respectively. Morphological changes of the cells was evaluated daily until to harvest after culture. MTT value of light absorption and total protein was assessed at 5 days. von Willebrand factor (vWF) was measured for identification of cells and estimation of function. Results The explanted cells were verified as endothelial cells by vWF test. Viability and morphology of endothelial cells in culture mediums added with 5%, 10%, 15% bile were similar to those cells without bile, while cells in mediums with 20% and 25% bile were markedly reduced and immature. 5.A correlation between benign prostate hyperplasia and atherosclerosis

ObjectiveTo investigate the correlation between benign prostate hyperplasia (BPH) and atherosclerosis (AS).Methods 95 cases were divided into two groups based on prostate volume (PV):24 cases with non-BPH (PV≤20 ml) and 71 cases with BPH (PV>20 ml).Carotid intinal medial thickness (IMT) was determined by high resolution color Doppler sonography.ResultsLevels of triglyeeride (TG),hypersensitive C reactive protein (hs-CRP),ln fasting insulin [ln (FINS)],ln insulin resistance index [ln (HO)MA-IR)] and IMT in BPH group were significantly higher than in non-BPHgroup.The morbidity rate of coronary heart disease and cerebral arteriosclerosis in the BPH group enhanced as compared with non-BPH group.PV were positively correlated with ln(FINS),ln(HOMA-IR),TG and high density lipoprotein (HDL-C).Conclusions BPH is closely correlated with AS and their co-pathogenesis might be insulin resistance,inflammatory reaction and endothelial dysfunction.
7.Expression of p16 and Ki67 in cervical intraepithelial neoplasia and cervical carcinoma

Two hundred and seventy four patients with suspicious cervical lesions underwent pelvic examination and biopsy. Tissue biopsy results revealed 153 cases of cervical intraepithelial neoplasia(CIN)and 10 cases of cervical carcinoma. p16 and Ki67 were detected by immunohistochemistry method in tissue specimens. The positive rates of p16 were 9.0%, 69. 9%, 93. 8%, 95. 5% and 10/10 in normal cervical mucosa, CIN Ⅰ , CIN Ⅱ , CIN Ⅲ and cervical carcinoma, respectively; while those of Ki67 were 76. 6%,89. 2%, 91.7%, 95.5% and 10/10, respectively. The expressions of p16 and Ki67 were significantly associated with cervical lesions ( P < 0. 01 ). The study suggests that p16 and Ki67 may be used as supplemental diagnostic markers for cervical lesions.

Kaposi sarcoma-associated herpesvirus (KSHV),also known as human herpesvirus 8 (HHV-8),is discovered in 1994 from Kaposi's sarcoma (KS) lesion of an acquired immunodeficiency syndrome (AIDS)patient.In addition to its association with KS,KSHV has also been implicated as the causative agent of two other AIDS-associated malignancies:primary effusion lymphoma (PEL) and multicentric Castleman's disease (MCD).KSHV is a complex DNA virus that not only has the ability to promote cellular growth and survival for tumor development,but also can provoke deregulated angiogenesis,inflammation,and modulate the patient's immune system in favor of tumor growth.As KSHV is a necessary but not sufficient etiological factor for KS,human immunodeficiency virus (HIV) is a very important cofactor.Here we review the basic information about the biology of KSHV,development of pathogenesis and interaction between KSHV and HIV.

Objective To evaluate the effect of bile on endothelial nitric oxide synthase(NOS) activity. Methods Endothelial cells explanted from human umbilical vein were cultured in vitro, then bile of different concentrations 5%,10%,15%,20%,25% were mixed with the standard culture medium. Viability and morphology of the endothelial cells were studied, and NO(nitric oxide) content in conditioned medium and NOS activity were assessed. Results Viability and morphology of endothelial cells in the culture mediums presented as 5%,10%,15% bile were similar to the cells without bile medium while endothelial cells in mediums presented with 20% and 25% bile were markedly reduced in number and immature. Endothelial cell NOS activities in mediums presented with various concentration of bile were statistically decreased comparing with the cells in medium without bile. The quantity of NO in conditioned culture mediums with or without bile did not reach a statistical significance. Conclusion Bile appears to inhibit endotholial cells growth as well as endothelial NOS activity.
10.Expression of Cytokine in Peripheral Blood of Newborns with Hypoxic-Ischemic Encephalopathy and Its Clinical Significance

Objective To observe the expression of CD_(25),CD_(25) mRNA,interleukin-1?(IL-1?) in peripheral blood of newborns with hypoxic-ischemic encephalopathy(HIE) and its clinical significance.Methods The peripheral blood mononuclear cells(PBMC) of newborns with HIE and normal controls were isolated by routine Ficoll-Hypaque,and the level of CD_(25) induced and non-induced by phytohernagglutinin(PHA) were detected by biotin-streptavidin(BSA),CD_(25) mRNA was detected by real-time PCR and IL-1? in serum was detected by enzyme-linked immunosorbent assay on the first,third and seventh day after birth.The final expressive level of CD_(25) mRNA was based on rate of CD_(25) cDNA and gluceraldehyde phosphase dehydrogenase(GAPDH).Results On the first day,the positive rates of CD_(25) induced and non-induced by PHA and IL-1? were(3.6?1.1)%,(20.9?4.8)%,(17.7?8.2) ng/L,respectively.There were significant differences in HIE group and normal controls(P0.05).Conclusions There is obvious inflammatory(rea)ction in babies with HIE.The main character of PBMC of newborns is all infantile and undifferentiated,which surface sign is infantile,and lower level of CD_(25) is expressed.However,the level of CD_(25) mRNA can be expressed consistently during the course of disease.The immunity and immune regulative response in newborns is gradually built up during the growth and can participate in recondition of brain injury caused by HIE.
