1.Evaluation on the performance of MicroScan WalkAway in detecting carbapenem-resistant Enterobacteriaceae

Objective To investigate the performance of MicroScan WalkAway 96 Plus (MSW) system in detection of carbapenem-resistant Enterobacteriaceae (CRE).Methods A total of 81 stock CRE strains were used in this study. Bacterial identification and antimicrobial susceptibility test were performed by MSW system. Beta-lactamases genes blaKPC,blaIMP,blaVIM, blaOXA-48 and blaNDM were amplified by PCR and subjected to sequencing analysis. Disk diffusion method and PCR were used as gold standard to evaluate the performance and reliability of MSW system in identifying carbapenem-resistant and carbapenemase-producing Enterobacteriaceae.Results Overall, 69.1 % (56/81) of the Enterobacteriaceae strains were identified as CRE by the MSW system. The results of PCR showed that 48 strains were carbapenemase-producing Enterobacteriaceae. When carbapenemase-producing Enterobacteriaceae strains were identified by the instrument using an advanced expert system, the sensitivity was 93.8 % and specificity was 42.4 %. The positive predictive value was 70.3 %, the negative predictive value was 82.4 % and the predictive accuracy value was 72.8 %.Conclusions The MicroScan WalkAway 96 Plus system has shown good performance in detection of CRE.
2.A longitudinal study on the influence of students relationships with teachers, peers students and school attitude on externalizing behaviors among migrant children

Objective:
To explore the impact of teacher student relationship and peer relationship on externalizing behaviors among migrant children, and to investigate the mediating role of school attitude.
Methods:
A longitudinal follow up study was designed among 378 migrant children from Hubei, Guizhou and Chongqing city with the teacher student relationship, peer relationship questionnaire, school attitude and the externalizing behavior collected through questionnaire survey.
Results:
After controlling covariates and baseline (T1) externalized problem behaviors, baseline (T1) teacher student relationship and peer relationship negatively predicted externalizing behavior at T3 ( β teacher student =-0.12, P =0.08; β classmate =-0.20, P <0.01), The mediating effect of school attitude at T2 on the association between externalizing behaviors with teacherstudent relationship (-0.11) and peer relationship(-0.05).
Conclusion
Teacher student relationship and peer relationship affect externalizing behaviors among migrant children through school attitude. High quality teacher student relationship and peer relationship might help to reduce externalizing behaviors among migrant children.

Vine tea has been used as an herbal tea by several ethnic minorities for hundreds of years in China.Flavonoids,a kind of indispensable component in a variety of nutraceutical,pharmaceutical and cosmetic applications,are identified to be the major metabolites and bioactive ingredients in vine tea.Interest-ingly,vine tea exhibits a wide range of significant bioactivities including anti-oxidant,anti-inflammatory,anti-tumor,antidiabetic,neuroprotective and other activities,but no toxicity.These bioactivities,to some extent,enrich the understanding about the role of vine tea in disease prevention and therapy.The health benefits of vine tea,particularly dihydromyricetin and myricetin,are widely investigated.However,there is currently no comprehensive review available on vine tea.Therefore,this report summarizes the most recent studies investigating bioactive constituents,pharmacological effects and possible mechanisms of vine tea,which will provide a better understanding about the health benefits and preclinical assessment of novel application of vine tea.
4.Research progress of nitroxide radical derivatives and their biological activities

Nitroxide radicals are a kind of stable organic free radicals. Due to the presence of N-O· and unpaired electrons in its structure, it has many characteristics, and thus can be used as a spin marker to explore the mechanism of biological reactions; with its magnetic properties, it can be used for the development of multifunctional magnetic molecular materials and used as a polymerization inhibitor and catalyst in organic reactions. More importantly, it has a variety of biological activities such as anti-oxidation and anti-tumor, and so has attracted much attention in the research and development of new drugs. For example, the spin labeling of nitroxide radicals on anticancer drug podophyllotoxin can enhance the efficacy and reduce the toxicity, and can be easily to be absorbed by the body, thus obtaining a new anti-cancer drug 4-[4″-(2″,2″,6″,6″-tetramethyl-1″-piperidinyloxy nitroxide radical) amino]-4′-demethyl epipodophyllotoxin(GP-7). It is an effective way to seek new drugs by introducing pharmacophore to modify nitroxide radicals or it can be spin-labeled on active natural products to obtain new compounds with high efficiency and low toxicity. The research progress of derivatives and its biological activitives of nitroxide radicals are summarized, aiming to provide theoretical basis for the developing and utilizing of nitroxide radicals and searching for new drugs.
5.Identification of genetic characteristics of B19 virus in a human parvovirus outbreak

Objective To analyze the genetic characteristics of B19 parvovirus in a human parvovirus outbreak in Hubei Province in 2019, and to provide scientific basis for the prevention and control of parvovirus outbreaks. Methods Among of the 17 serum specimens and 20 throat swab specimens from a parvovirus B19 outbreak were collected in Hubei Province in 2019. The nucleic acid-positive specimens were identified by fluorescence quantitative RT-PCR. The specific sequences of NS1-VP1u (1157 bp) of B19 positive specimens were amplified by nested PCR to determine B19 genotype. Results The genotype of B19 virus in 17 patients were subgenotype 1a, and the nucleotide homology of subgenotype 1a was 99.90% - 100%. The B19 subgenotype 1a was located in the same small branch as the Chinese Beijing strain KR819772, the Chinese Beijing strain KR819773 and the Chinese Hangzhou strain KT310174, and the distance was the closest. Conclusion The B19 virus in a human parvovirus outbreak in Hubei Province in 2019 belongs to subgenotype 1a.
6.Effect and mechanism of tournefolic acid B on the improvement of glucose metabolism and renal function in diabetes nephropathy model mice

OBJECTIVE To explore the role and underlying mechanism of tournefolic acid B (TAB) on the improvement of glucose metabolism and renal function in diabetic nephropathy (DN) model mice. METHODS DN model mice were established by high-fat diet combined with streptozotocin, and then randomly divided into model group, positive control group (vitamin E, 20 mg/kg), TAB low-dose, medium-dose and high-dose groups (1, 2, 4 mg/kg), with 12 mice in each group; normal control group was given regular diet. Each group was given relevant medicine or normal saline intragastrically, once a day, for 4 consecutive weeks. The glucose metabolic function was estimated by fasting blood glucose, glucose tolerance test, insulin tolerance test and serum insulin concentration. The renal coefficients and biochemical indicators related to renal function [serum uric acid, blood urea nitrogen, creatinine levels, and ratio of urine microalbumin to creatinine] were detected in mice; the contents of biochemical indicators related to oxidative stress [superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), malondialdehyde (MDA), 8-hydroxydeoxyguanosine (8-OHdG)] were determined in renal tissue of mice; the pathological morphology of renal tissue was observed; the expressions of extracellular matrix (ECM) deposition related factors [transforming growth factor β1 (TGF- β1), fibronectin (Fn), type Ⅳ collagen (Col Ⅳ)] and protein kinase B (Akt)/nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) pathway related proteins were determined in renal tissue of mice. RESULTS Compared with normal control group, fasting blood glucose, area under glucose tolerance curve, area under insulin tolerance curve, serum insulin content, the levels of uric acid, urea nitrogen and creatinine @qq.com and ratio of urinary microalbumin to creatinine in serum, the contents of MDA and 8-OHdG and the protein expressions of TGF-β1, Fn and Col Ⅳ were increased significantly in model group (P<0.05), while the contents of SOD, GSH-Px and the protein expressions of p-Akt, Nrf2, HO-1 in renal tissue were decreased significantly (P<0.05); the significant thickening of the basement membrane, accumulation of mesangial matrix, glomerulosclerosis and interstitial fibrosis of the renal tubules were all found. Compared with model group, above indexes of mice were all reversed significantly in TAB groups (P<0.05), and pathological changes were alleviated in a dose-dependent manner. CONCLUSIONS TAB can improve blood glucose metabolism and kidney function and alleviate renal tubulointerstitial fibrosis in DN model mice, the mechanism of which may be associated with activating the Akt/Nrf2/HO-1 signaling pathway and suppressing ECM deposition.
7.Effects of mild moxibustion on intestinal microbiome and NLRP3 inflammasome in rats with 5-fluorouracil-induced intestinal mucositis.

OBJECTIVE:
The present study investigated how mild moxibustion treatment affects the intestinal microbiome and expression of NLRP3-related immune factors in a rat model of intestinal mucositis (IM) induced with 5-fluorouracil (5-Fu).
METHODS:
Forty male Sprague-Dawley rats were randomly divided into control, chemotherapy, moxibustion and probiotics groups. The IM rat model was established by intraperitoneal injection of 5-Fu. Mild moxibustion treatment and intragastric probiotic administration were provided once daily for 15 days. Tissue morphology, serum levels of inflammatory factors and the expression levels of tight junction proteins, caspase-1, gasdermin D and NLRP3 were evaluated in colon tissue, through hematoxylin and eosin staining, electron microscopy, enzyme-linked immunosorbent assay, Western blotting, quantitative real-time reverse transcription polymerase chain reaction and immunofluorescence. Gut microbiome profiling was conducted through 16S rRNA amplicon sequencing.
RESULTS:
Moxibustion and probiotic treatments significantly increased the expression levels of tight junction proteins, reduced cell apoptosis and the expression levels of caspase-1, gasdermin D and NLRP3; they also decreased the serum levels of tumor necrosis factor-α, interleukin (IL)-6, IL-1β and IL-18, while increasing serum levels of IL-10. Moxibustion and probiotic treatments also corrected the reduction in α-diversity and β-diversity in IM rats, greatly increased the proportion of the dominant bacterial genus Lactobacillus and reduced the abundance of the genera Roseburia and Escherichia in chemotherapy-treated rats to levels observed in healthy animals. We also found that these dominant genera were firmly correlated with the regulation of pyroptosis-associated proteins and inflammatory factors. Finally, moxibustion and probiotic treatments elicited similar effects in regulating intestinal host-microbial homeostasis and the expression of NLRP3 inflammasome-related factors.
CONCLUSION
Moxibustion exerts its therapeutic effect on IM by ameliorating mucosal damage and reducing inflammation. Moreover, moxibustion modulates the gut microbiota, likely via decreasing the expression levels of the NLRP3 inflammasome.
