1.Effects of Dahuang Zhechong Pills on CD44,VEGF and the metastasis suppressor in mice with Lewis Lung carcinoma

Objective:By observing effects of Dahuang Zhechong Pills on CD44,VEGF and the metastasis suppressor in mice with Lewis Lung carcinoma,to discuss its mechanism of anti-tumor metastasis preliminarily.Methods:The mice were divided randomly into five groups the day after modeling and each group included ten mice.According to the pre-test,high-dose,medium-dose and low-dose were quantitated with 0.18g/ml,0.09 g/ml,0.045g/ml.The model group was intragastric administrated with NS 0.2ml for ten days;the chemotherapy group was Intraperitoneal administrated 50mg/kg CTX on the first day.Observing and numerating the pulmonary metastases with nodules by pulmonary pressure slide film on the 12th day.Based on immunohistochemical method to detect the expression of VEGF and the activity detection by flow cytometry CD44,Its anti-tumor mechanism and the role of target were preliminarily studied.Results:The inhibition rates of lung spontaneous metastasis in experimental groups of high(H),medium(M),low(L)dosages and the chemotherapy group were respectively 57.44 percent, 55.32 percent,38.72 percent,60.00 percent.CD44,respectively,values were 0.59?0.86,0.21?0.28,0.33?0.31,1.36?0.52; the experimental group(M)compared with model group,VEGF expression was significantly reduced,there was a significant difference(P0.05).Conclusion:Dahuang Zhechong Pills reduced the lung metastases incidence of C57BL mice with Lewis lung carcinoma and prolonged their lives.These results were obvious in the medium-dose experimental group.Medium-dose of Dahuang Zhechong Pills had effects on C57BL mice with Lewis lung carcinoma,whose CD44 activity were inhibited and the expression of VEGF was reduced.
2.Application effect of systematic rehabilitation intervention in postradiotherapy dysphagia of patients with nasopharyngeal carcinoma

Objective To analyze the application effect of systematic rehabilitation intervention in postradiotherapy dysphagia of patients with nasopharyngeal carcinoma.Methods Totally 60 patients with nasopharyngeal carcinoma and postradiotherapy dysphagia admitted in our hospital from June 1st,2013 to Feburary 28th,2016 were selected and divided into the treatment group and the control group according to random number table,with 30 cases in each group. Patients in the control group were treated with conventional rehabilitation exercises,combined with routine care and conventional drug therapy,while those in the treatment group were treated with systematic rehabilitation intervention model,including rehabilitation exercises and swallowing apparatus. Improvement of swallowing function and satisfaction toward the nursing were compared between two groups.Results There were 14 effective cases and 16 ineffective cases in the control group. Two cases in the treatment group were cured. There were 23 effective cases and five ineffective cases in the treatment group. The total efficiency of the treatment group (83.33%) was significantly higher than that of the control group (46.67%) (χ2=2.159,P<0.05). The experimental results of drinking water and improvement of limitation of mouth opening in the treatment group were obviously better than that in the control group (Z=5.012,7.018;P<0.05). Satisfaction in the treatment group (86.67%) was obviously higher than that in the control group (43.33%) (χ2=6.321,P<0.05).Conclusions Systematic rehabilitation intervention in postradiotherapy dysphagia of patients with nasopharyngeal carcinoma can significantly improve patients' swallowing difficulties.
3.Mechanism of Danggui Niantongtang on Adjuvant Arthritis Rats with Wind-dampness-heat Arthralgia Based on Quantitative Proteomics

ObjectiveTo explore the mechanism of Danggui Niantongtang (DGNT) against adjuvant arthritis (AA) rats with wind-dampness-heat arthralgia by quantitative proteomics. MethodSixty SD rats were randomly divided into normal group, model group, angelica came pain soup low, medium and high dose group and methotrexate (MTX) group, each group of 10, only the rat tail root subcutaneously inactivated mycobacterium tuberculosis (Mtb) of adjuvant to build model of AA, artificial climate box intervention 16 d rheumatic fever bi syndrome model is set up, building the day began to drug intervention, The intervention lasted for 28 days. The proteins of synovial tissues in experimental rats were extracted. The differential proteins in the medium-dose DGNT group and the model group were detected and analyzed by 4D label-free quantification (4D-LFQ) proteomics. The differentially expressed proteins associated with mitochondrial pathway apoptosis were verified by immunohistochemistry and Western blot. ResultA total of 4 756 proteins were identified from rat synovial tissues, of which 4 234 proteins contained quantitative information. There were 814 differential proteins between the model group and the DGNT group. As revealed by Gene Ontology (GO) and Kyoto Encyclopedia of Gene and Genome (KEGG) enrichment analyses, DGNT had an effect on the synovial proteome of AA rats with wind-dampness-heat arthralgia, and the differential proteins were enriched in the regulation of the immune system, response to acute inflammation, and apoptosis regulation. As demonstrated by the results of immunohistochemistry and Western blot, compared with the model group, the DGNT groups and the MTX group showed increased protein expression of B-cell lymphoma 2 (Bcl-2)-associated X protein (Bax) and cytochrome C (Cyt C)(P<0.05, P<0.01), reduced Bcl-2 level (P<0.05, P<0.01), elevated level of cleaved cysteinyl aspartate-specific protease 9 (Caspase-9)/Caspase-9 (P<0.01), and decreased level of phosphorylated protein kinase B (p-Akt)/Akt(P<0.05, P<0.01). ConclusionDGNT involved multiple targets in the treatment of AA with wind-dampness-heat arthralgia and it may exert its effect in the prevention and treatment by regulating the Akt/Bax/Bcl-2 pathway and promoting the cell apoptosis in the mitochondrial pathway.
