1.Immunoloregulation effect of polysaccharide isolated from Angelica Sinensis on mouse

AIM: To investigate the immunoloregulation effect of Angelica polysaccharide extracted from Angelica Sinensis (Oliv.) Diels. METHODS: Index of immune organs were weighed and calculated. Phagocytosis of mononuclear macrophage (M) were determined with carbon particle clearance test. Spectrophotography was used to estimate levels of serum hemolysis IgG, IgM. The cell proliferation was measured by MTT assay. RESULTS: In certain doses (10-100 mg/kg), Angelica polysaccharide significantly increased phagocytic function and spleen index in immunosuppression mice caused by hydrocortisone, while showed no obviouse influnence on thymus index. It also remarkble decreased levels of serum hemolysis IgG, IgM. In vitro experiments, Angelica polysaccharide could increase the proliferation of mouse spleen cell and macrophage. CONCLUSION: These results suggest that Angelica polysaccharide has potent enhancement on non specific immunity, however, it could suppress humoral immunity.
2.Systemic lupus erythematosus patients with acute abdomen:a report of 20 cases and literature review

Objective To summarize the various acute abdomen manifestation of systemic lupus erythematosus (SLE) and their diagnosis and treatment.Methods Twenty SLE patients with AA were analysed and 35 year′s literature was reviewed.Results Most AA were of active SLE (70%),others were of non SLE related disease (30%).Cause of these cases was diversified,which often lead to misdiagnosis.Making correct diagnosis as early as possible was the key point to improve the patient′s survival rate.Conclusion SLE with AA indicates a critical condition.Investigating the cause of AA and working out appropriate treatment measures are most important.
3.The diagnostic significance of anti-cyclic citrullinated peptide antibody and its predictive value for articular erosion in patients with rheumatoid arthritis

Objective To explore the diagnostic significance of anti-cyclic citrullinated peptide antibody(anti-CCP)and the role it plays in the articular erosion in patients with rheumatoid arthritis(RA).Methods The diagnostic significance of anti-CCP?AKA and RF were evaluated respectively.Seventy-five RA patients were devided into group1(limited radiographic damage group)and group2(severe radiographic damage group).The distribution of anti-CCP,AKA and RF in patients of the two groups were investigated.A univariate analysis was used to determine whether anti-CCP?AKA?RF?ANA?ESR?CRP or cutaneous nodules plays a role on articular erosion in RA.To determine which has the best predictive value for severe radiographic da-mage,all variables were entered into a logistic regression model.Results The sensitivity and specificity of an-ti-CCP?AKA and RF were49%,94%;50%,93%;79%,67%respectively.When any two markers were combined,the specificity would be raised.The positive rate of these three markers were much higher in group2than that in group1.Anti-CCP had the highest OR(6.71)for articular erosion in RA.Logistic regression analysis showed a strong correlation between anti-CCP,AKA,CRP or cutaneous nodules and less favourable disease outcomes.Cut.aneous nodules had the strongest correlation with severe radiographic damage.Conclusion Anti-CCP is a satisfactory marker for RA.Diagnostic accuracy appears to be raised when anti-CCP combined with AKA and RF.Anti-CCP has a strong correlation with severe radiographic damage.To investigate multiple risk factors of articular erosion will be helpful to pridict the outcome of RA.

OBJECTIVE:To study the analgesic and anti-inflammatory actions of essential oil extracted from Radix Angelica Sinensis METHODS:The pharmacodynamic effects were obsersed in five experimental models of inflammation and pain,including:(1)mouse auricular edema induced by xylen;(2)increased vascular permeability induced by acetic acid in mice;(3)paw edema caused by albumen in rats;(4)granuloma caused by implanted cotton ball in rats;(5) writhe reaction induced by acetic acid in mice RESULTS:Essential oil from Angelica Sinensis could distinctively inhibit the inflammatory edema caused by various inflammatory agents and reduce the times of writhe induced by acetic acid in mice CONCLUSION:Essential oil from Angelica Sinensis has analgesic effect and could not only inhibit acute inflammation but also chronic ones in animals

6.Early detection of cardiac allograft vasculopathy and chronic rejection after heart transplantation-Report of one case

Objective To study clinically the feasibility of early diagnosis of cardiac allograft vascularopathy (CAV) and chronic rejection. Methods A 13 year old female patient with dilated cardiomyopathy received orthotopic heart transplantation for advanced heart failure, and subsequent immunosuppressive therapy including cyclosporine, prednisone and mofetil, and a monthly close follow up. Coronary angiography and left ventricular endomyocardial biopsy (EMB) was performed 9 months after the operation. Results The clinical and follow up data of the case showed that cardiac or systemic nonspecific symptoms such as exertional chest discomfort, palpitation, fatigue or fever of unknown reasons were the first and ignorable clinical symptoms, and found disappeared after dosage addition of cyclinsporine, which indicated a early clinical manifestations of rejection or vasculopathy. While persistent sinus tachycardia on electrocardiogram, decreased left ventricular ejection fraction (from 64?% ~68?% down to 47?%~50?%), enlarged right atrial (from 32~ 41?mm up to 44?mm in diameter), thickened intraventricular septal ( 13?mm ), repetitive tricuspid regurgitation on echocardiography, increased white blood cells without infection were observed respectively. Coronary angiography showed a typical (type B) vasculopathy with diffuse concentric stenosis of mid and distal left anterior descending artery and its small braches, the distal diagonal, circumflex, and right coronary artery were also involved to some extent. Furthermore, early mild chronic rejection of grade I A was proved by pathologic study. Conclusion Close clinical observation and follow up, serial echocardiography and electrocardiogram, reaction to enhanced anti rejection, and other related laboratory methods could be analyzed and colligated for early noninvasive diagnosis of cardiac allograft vasculopathy and chronic rejection, while EMB and coronary angiography are still the precise diagnostic ways.
7.A neonatal mouse model of AIDS induced by L6565 murine leukemia virus

Aim To establish a mouse model of AIDS and observe the effects of Combivir,an anti-AIDS drug.Methods The model was set up by infecting infant mice with L6565 murine leukemia virus.Quantitative RT-PCR method was used to measure viral load in plasma.The percentage of CD4~+ and CD8~+ lymphacytes was estimated by flowcytometric method.The number of WBC,RBC and marrow karyote was counted.indexes of immune organs were weighed and calculated.Results Compared with control,the viral load in plasma in the model was at high level.The percentage of CD4~+ lymphocyte,the ratio of CD4~+/CD8~+,the number of RBC and marrow karyote,and thymus index were significantly decreased in the model,while the number of WBC and spleen index was remarkably increased.Conclusion AIDS patient symptom is shown in mouse infected with L6565 murine leukemia virus.This suggests that this model is likely to be used as a mice model to evaluate the effect of anti-AIDS drugs.
8.Effects of Angelica polysaccharide and its sulfates on coagulation and platelet aggregation

Object To investigate the effects of Angelica polysaccharide (AP 0) and its sulfates (APS) on blood coagulation and platelet aggregation. Methods Infrared turbidimetric method was used to estimate platelet aggregation, prothrombin time (PT) and thrombin time (TT), actiated partial thromboplastin time (APTT); bleeding time (BT) was measured by cutting the mouse's tail and coagulate time (CT) was measured by glass method sheet; hemorheology was measured with heparin as anticoagulant.Results The AP 0 and APS prominently enhanced the platelet aggregation at 5 min (PAG ), while showed less effects on the maximum platelet aggregation (PAG ); they, especially APS 1, also markedly prolonged CT but shortened BT, both AP 0 and APS 1 significantly prolonged TT and APTT in all experimental dosage, while only APS 1 in high dosage (8.46 mg/kg) significantly prolonged PT; AP 0 increased the aggregation index (AI) of RBC and the whole blood viscosity (? b) at shear rate of 5 s -1 at a dose of 2.00 mg/kg, however, unlike AP 0, APS 1 decreased AI and ? b at shear rate of 5 s -1 and 230 s -1 . Conclusion These results suggest that AP 0 and APSs have potent anticoagulant and haemostasis effects. The haemostasis effect is related to promoted platelet aggregation. Ameliorative effect of Angelica polysaccharide sulfate on hemorheology is better than that of Angelica polysaccharide itself.

AIM:To explore the therapeutic effect of dexamethasone Angelica sinensis polysaccharide prodrug(DEX-AP) on trinitrobenzene sulfonic acid(TNBS) induced ulcerative colitis(UC) in rats and its side effects.METHODS: The experimental UC rats were induced by clusis of the solution of TNBS in 45% alcoho1(50(mg?ml~(-1))).The UC rats were orally administrated with(0.25)(?mol?kg~(-1)?d~(-1)) DEX and(0.05),(0.25),(1.25)(?mol?kg~(-1)?d~(-1)) DEX-AP(calculated by carried DEX in DEX-AP) for 7 days,respectively.The rats were killed after the amount of peripheral blood lymphocyte was counted,then the spleen,thymus and colon were separated and weighted.After the ulcerative area of colon was calculated,the colonic myeloperoxidase(MPO) activity was determined and parts of colon were paraffin sectioned and examined under light microscope by HE stain.RESULTS: After the UC rats were administrated with different doses of DEX-AP for 7 days,the ulcerative area,the weight and the MPO activity of colon reduced significantly.The reduction of MPO activity was correlated to the dose of DEX-AP and the MPO activity with DEX-AP at the doses of(0.25),(1.25)(?mol?kg~(-1)?d~(-1)) reduced more significantly than that with DEX at the the dose of(0.25)(?mol?kg~(-1)?d~(-1)).The number of peripheral blood lymphocyte,spleen weight and thymus weight of UC rats reduced significantly at the dose of(0.25)(?mol?kg~(-1)?d~(-1)) DEX(P
10.Colon-targeted delivery system of dexamethasone-angelica sinensis polysaccharides prodrug in rats

AIM: To explore the transport and delivery of active drug from dexamethasone-angelica sinensis polysaccharides prodrug in the gastrointestinal tract of rats. METHODS: Dexamethasone and the prodrug were orally administered to rats at the dose of 1.96 mg?kg~ -1 (calculated by carried dexamethasone). The drugs in the plasma and contents of different parts of the rats' gastrointestinal tract were determined by high performance liquid chromatography (HPLC). RESULTS: Dexamethasone carried by the prodrug was mainly released in the contents and mucosa of cecum and colon after oral administration of the prodrug. The absorption of released dexamethasone was reduced significantly. The peak time, peak concentration and AUC were 7.2 h , 42 ?g?L~ -1 and 334 ?g?h?L~ -1 , respectively. However, free dexamethasone was found mainly in the contents and mucosa of the stomach, proximal and distal small intestine after oral administration. The peak time, peak concentration and AUC were 2.2 h, 2 120 ?g?L~ -1 and 11 875 ?g?h?L~ -1 , respectively. CONCLUSION: Dexamethasone can be specifically delivered to the cecum and colon by using dexamethasone- angelica sinensis polysaccharides prodrug. The absorption of dexamethasone was reduced significantly and the drug concentration in colon was increased significantly. The prodrug has a potential in the treatment of colitis.
