2.Mechanism of inhibitory effect of MN9202 on contraction and relaxation functions of cardiac myocyte

Objective To investigate the inhibitory effect on enhancement of contraction and relaxation functions of cardiomyocytes induced by of MN9202 TEA, Forskolin, and MB. Methods The twitch amplitude was measured with a video edge tracker method. The following indexes, including ph (peak height), peak height/baseline percent (ph/bl, %), maximal velocity of contraction (+dL/dt) and maximal velocity of diastolization (-dL/dt), were recorded by a computer. Results TEA, Forskolin and MB increased electrically-induced contraction as shown by the following indexes: ph, ph/bl%, +dL/dt and -dL/dt. Forskolin and MB markedly augmented function of cardiac myocyte contraction and diastolization. Comparing with the control group, Forskolin (10-8mol/L) increased ph from 0.12?0.03(um)to 0.24?0.06 (um), ph/bl % from 12?3% to 27?6%, +dL/dt from 1.8?0.5 (um/s) to 3.8?0.9 (um/s), -dL/dt from 1.8?0.22 (um/s) to 3.5?0.7 (um/s). These indexes increased by 91%, 123%, 110% and 89%, respectively. MB also increased significantly; ph, ph/bl %, +dL/dt and -dL/dt were 0.14?0.04um, 11?4%, 2.2?0.3um/s and 2.2?0.6um/s, respectively. MN9202 (3?10~ -6mol/L) decreased the indexes (+dL/dt and -dL/dt ) which were increased by Forskolin significantly. Conclusion TEA, Forskolin and MB increased function of cardiac myocyte contraction and diastolization. These drugs might elevate intracellular calcium content indirectly and directly. The effects of TEA, Forskolin and MB were attenuated by MN9202. MN9202 might not only block Dihydropyridine Receptor, but might also inhibit calcium influx. Further study is needed to elucidate exact mechanism of MN9202.
3.Protective effect of Rheum tanguticum polysaccharide against H_2O_2 induced IEC-6 cell apoptosis and its possible mechanism

Aim To investigate the protective effect of RTP1,one of the polysaccharide isolated from Rheum tanguticum,on H2O2 induced apoptosis in IEC-6 cells and its possible mechanism.Methods H2O2(100 mmol?L~-1)was used to induce IEC-6 cell apoptosis.Different doses(10,30,100 mg?L~-1)of RTP1 were administrated before H2O2 was added into IEC-6 cell culture.Cell viability was observed by MTT assay.Reactive oxygen species were measured with laser scanning confocal microscopy(LSCM).DNA content and percentage of apoptosis were assayed by DNA agarose gel electrophoresis,acridine orange staining and flow cytometry.The activation of Caspase-3 was detected with Western blot analysis.Results Following treatment with H2O2 for 2 h,H2O2 induced a significant decrease in cell viability,while DNA ladder was observed and apoptosis percentage was as high as 31.3%.Accumulation of intracellular ROS and increase in Caspase-3 activity were also detected.Pretreatment with RTP1 for 24 h exhibited cytoprotective effects in a dose-dependent manner.RTP1 obviously enhanced cell viability,reduced formation of DNA ladder and significantly reduced the number of cells labeled with Annexin V.The percentage of apoptosis/necrosis cells was markedly decreased to 24.4% and 21.5%,respectively.LSCM showed that RTP1 attenuated the accumulation of ROS.The significant decrease in Caspase-3 activity was detected.Conclusion RTP1 has cytoprotective capacity to antagonize H2O2-induced IEC-6 cell apoptosis and injury,and this effect may be related to decrease ROS and inhibit Caspase-3 activity
4.Alcoholic liver disease:gut microbiota and therapeutic perspectives

Alcoholic liver disease ( ALD ) , a chronic progres-sive disease, threatens human health seriously. An increasing number of studies have shown that gut flora dysbiosis plays an important role in the development of ALD. Intestinal microbiota maintains a steady state under normal conditions, regulating gut flora normal physiological function. However, chronic alcohol consumption produces intestinal bacteria overgrowth and dysbio-sis, including the alteration of the composition of intestinal mi-croflora, the increment of gut permeability and bacterial translo-cation. Subsequently, the host immune is activated, promoting the production of inflammatory cytokines in liver, which plays a central role in the development of ALD. Notably, the supple-ment of prebiotics or probiotics reverses the intestinal flora disor-der,ameliorating the clinical symptoms effectively in ALD pa-tients. The evidence impies that the modulation of dysbiosis may be effective in the prevention and treatment of ALD. This review summarizes the research progress on the mechanism of the devel-opment of dysbiosis-mediated ALD, to provide a theoretical basis for the research on intestinal flora and ALD.
5.Anti-inflammatory and Analgesic Actions of Essential Oil Extracted From Radix Angelica Sinensis by Ethanol

OBJECTIVE:To study the analgesic and anti-inflammatory actions of essential oil extracted from Radix Angelica Sinensis METHODS:The pharmacodynamic effects were obsersed in five experimental models of inflammation and pain,including:(1)mouse auricular edema induced by xylen;(2)increased vascular permeability induced by acetic acid in mice;(3)paw edema caused by albumen in rats;(4)granuloma caused by implanted cotton ball in rats;(5) writhe reaction induced by acetic acid in mice RESULTS:Essential oil from Angelica Sinensis could distinctively inhibit the inflammatory edema caused by various inflammatory agents and reduce the times of writhe induced by acetic acid in mice CONCLUSION:Essential oil from Angelica Sinensis has analgesic effect and could not only inhibit acute inflammation but also chronic ones in animals
6.Pharmaceutical Care Measures for Improving the Safety of Chemotherapy by Clinical Pharmacists

OBJECTIVE:To investigate the measures and roles of pharmaceutical care in improving the safety of chemotherapy by clinical pharmacists. METHODS:Chemotherapy drug-induced ADR medical records were collected from oncology department of our hospital during Jan.-Dec. 2013 (before intervention) and Apr. 2014-Mar. 2015 (after intervention),and then analyzed and compared in respects of chemotherapy plan,patient’age and gender,route of administration,involved organs and systems,severe ADR,etc. RESULTS:After the formulation and implementation of pharmaceutical care for improving safe use of chemotherapy drugs,the incidence of chemotherapy drugs-induced ADR decreased from 88.48%to 72.14%;the incidence of ADR of involved di-gestive system,skin and its appendants decreased from 67.28% and 7.37% to 42.29% and 1.99%,respectively;the incidence of severe nausea and vomiting decreased from 27.65%to 18.91%;the constituent ratio of single agent chemotherapy and oral adminis-tration route increased from 31.80%(69/217) and 14.29%(31/217) to 44.28%(89/201) and 22.39%(45/201),respectively (P<0.05). CONCLUSIONS:Pharmaceutical care intervention measures formulated by clinical pharmacists can significantly lower the incidence of ADR,promote safe use of chemotherapy drugs and guarantee the safety of drug use.
7.The diagnostic significance of anti-cyclic citrullinated peptide antibody and its predictive value for articular erosion in patients with rheumatoid arthritis

Objective To explore the diagnostic significance of anti-cyclic citrullinated peptide antibody(anti-CCP)and the role it plays in the articular erosion in patients with rheumatoid arthritis(RA).Methods The diagnostic significance of anti-CCP?AKA and RF were evaluated respectively.Seventy-five RA patients were devided into group1(limited radiographic damage group)and group2(severe radiographic damage group).The distribution of anti-CCP,AKA and RF in patients of the two groups were investigated.A univariate analysis was used to determine whether anti-CCP?AKA?RF?ANA?ESR?CRP or cutaneous nodules plays a role on articular erosion in RA.To determine which has the best predictive value for severe radiographic da-mage,all variables were entered into a logistic regression model.Results The sensitivity and specificity of an-ti-CCP?AKA and RF were49%,94%;50%,93%;79%,67%respectively.When any two markers were combined,the specificity would be raised.The positive rate of these three markers were much higher in group2than that in group1.Anti-CCP had the highest OR(6.71)for articular erosion in RA.Logistic regression analysis showed a strong correlation between anti-CCP,AKA,CRP or cutaneous nodules and less favourable disease outcomes.Cut.aneous nodules had the strongest correlation with severe radiographic damage.Conclusion Anti-CCP is a satisfactory marker for RA.Diagnostic accuracy appears to be raised when anti-CCP combined with AKA and RF.Anti-CCP has a strong correlation with severe radiographic damage.To investigate multiple risk factors of articular erosion will be helpful to pridict the outcome of RA.
8.Effects of essential oil from Radix Angelicae Sinensis on rat uterine contraction in vitro

AIM: To investigate the contraction effect of essential oil of Radix Angelicae Sinensis on isolated uterus in rats. METHODS: Isolated uterine strips of rats were used to observe the reactions in the presence of oxytocine and high k + depolarization solution. RESULTS: The isolated rat uterus contraction induced by oxytocin and Ca 2+ after high K + depolarization was dose dependently inhibited by the essential oil. The cumulative dose response curves of CaCl 2 shifted to right and the maximal responses were depressed. This showed that the essential oil was a non competitive antagonist. The extracellular and intracellular Ca 2+ dependent contraction evoked by oxytocin could also be inhibited by the essential oil. CONCLUSION: The essential oil of Radix Angelicae Sinensis could inhibit the contractility of the isolated uteri, and it might act as an Ca 2+ antagonist to relax the isolated rat uterus.
9.Protection of Mn~(2+) on Injury of human kidney tubular epithelial cell line induced by bentamicin

AIM: To evaluate the protection of Mn~(2+) on injury of human kidney tubular epithelial cell line induced by gentamicin, and investigate its principals. METHODS: Gentamicin was used to injury human kidney tubular epithelial cell line (HK-2) to produce model of kidney injury. MTT method was used to measure effect of Mn~(2+) on proliferation of cells after they were injured by gentamicin. The spectrophotometry method was used to observe the change of LDH,NAG and SOD effected by Mn~(2+). Ultrastructure was examined by electron microscopy. RESULTS: Although HK-2 cells were injured, Mn~(2+) promoted cells proliferation, decreased LDH activity and NAG content, increased SOD activity, and alleviated dilation of the mitochondria and crushing of lysosomes. CONCLUSION: Mn~(2+) can protect human kidney tubular epithelial cell line from injury induced by gentamicin. It may be in relation to inhibiting the oxidative injury, lightening dilation of the mitochondria, protecting the integrity of lysosomes and decrease the leakage of enzyme.
10.Effects of rheum tanguticum polysaccharides on ulcerative colitis induced by TNBS in mice

AIM: To explore the effects of rheum tanguticum polysaccharides(RTP) on TNBS-induced colitis in mice and its probable mechanisms.METHODS: Mice colitis model was induced by 2,4,6-trinitrobenzene sulfonic acid(TNBS).The mice were randomly divided into 5 groups,normal group,model group,RTP-100,200 and 400 group.The macroscopical and histological changes of the colon were evaluated and the cytokines IL-8,IL-10,TNF-?and IL-4 produced by splenocyts were analyzed with ELISA.RESULTS: Compared with the model group,both symptoms and the lesions of colonic mucosa of RTP-200 and 400 group were slighter on ulcerative colitis induced by TNBS in mice.
