OBJECTIVE To evaluate the value of HPV-DNA test in the screening of cerical lesions.METHODS Patients with cervical lesions were detected by HPV-DNA test Papanicolaou(Pap) smear and pathological inspection, and the pathological results were regarded as gold standard. From the 178 positive cases,we compared the sensitivity of HPV-DNA test and Pap smear, and the HPV-DNA detection rate from patients with different histocytological classification.RESULTS All of 178 cases were positive in pathological inspection, the HPV-DNA test sensitivity was 85.4%,whereas Pap smear was 50.6%,the difference between HPV-DNA test and Pap smear was significant(P

Chinese Journal of Tissue Engineering Research 2013;(49):8474-8480
BACKGROUND:According to the previous studies, it is known that bone marrow mesenchymal stem cells live in the physiological environment of lower oxygen concentration compared with the common culture concentration (20%-21%). Hypoxia can promote bone marrow mesenchymal stem cells proliferation and maintain the characteristics of differentiation potential.
OBJECTIVE:To understand the biological characteristics of bone marrow mesenchymal stem cells after hypoxic preconditioning, and search for in vitro basis for bone marrow mesenchymal stem cells therapy in vivo with hypoxic preconditioning strategy.
METHODS:After passage and inoculation, bone marrow mesenchymal stem cells were cultured in the hypoxic incubator (1%O2, 5%CO2). Bone marrow mesenchymal stem cells cultured in the normoxic incubator (20%O2, 5%CO2) served as controls.
RESULTS AND CONCLUSION:Compared with bone marrow mesenchymal stem cells cultured under normoxia (20%O2), bone marrow mesenchymal stem cells cultured under hypoxia (1%O2) began to display exponential growth phase after a longer incubation period. Hypoxia (1%O2) was not the lethal condition for bone marrow mesenchymal stem cells. cellsurvival rates of both groups reduced over time, while those of bone marrow mesenchymal stem cells cultured under normoxia reduced faster (P<0.05). Surface markers expression of bone marrow mesenchymal stem cells, including CD29(+), CD90(+), CD31(-), CD45(-), after hypoxic preconditioning did not change greatly when compared with the results before hypoxic preconditioning. The hypoxia preconditioned bone marrow mesenchymal stem cells did not differentiate into vascular endothelial cells. Hypoxia-inducible factor-1αprotein expression was promoted after 24 hours to 48 hours hypoxic preconditioning, and there was significant difference between the two time points (P<0.05). Vascular endothelial growth factor expression increased at both transcriptional and protein levels after 48 hours hypoxic preconditioning (P<0.05). Culture under hypoxia (1%O2) does not produce a lethal effect on bone marrow mesenchymal stem cells and can be used for hypoxic preconditioning. Surface markers of bone marrow mesenchymal stem cells after 48 hours hypoxic preconditioning exhibit no great changes and the hypoxia preconditioned bone marrow mesenchymal stem cells maintain the characteristics of stemness. Hypoxic preconditioning can promote the ability of bone marrow mesenchymal stem cells to survive under hypoxia/ischemic condition. The activation of the hypoxic response signal transduction pathway through hypoxia-inducible factor-1αmay explain this.

Objective To study the hypoxia-inducible factor-1α (HIF-1α) expression in the col-lagen induced arthritis(CIA) model and its role in rheumatoid arthritis (RA) pathogenesis. Methods We established CIA model, and taked CIA model of the ankle joint to have them HE staining and HIF-1α immu-nohistochemistry staining, then the HIF-1α expression in the tissues of RA was observed. Results The ex-pression of CIA synovial membrane of rat and the lower synodal both was expressed in HIF-1α. The positive expression of HIF-1α reached the highest volume in the first 21 days. Then, with the disease progression, the expression gradually decreased, which was in significantly positive relation with the synovial pathology score, synovial hyperplasia and angingenesis rating score, however, the expression was no positively correla-ted with the inflammatory infiltration. Conclusion The expression of HIF-1α in RA tissue is positively re-lated with inflammation severity, It indicates that HIF-1α is closely related to the occurrence and develop-ment of RA.

Objective To investigate the effects of exercise on the expression of synapotophysin around hemotomas after intracerebral hemorrhage (ICH). Methods Ninety-five male Sprague-Dawley rats weighing 270 to 300 g were divided into 3 groups: a trial group (ICH-induction and exercise, n = 20), a control group ( ICH-in-duction only, n =20) and a sham-operation group (no ICH and no exercise, n = 20). The rats' brains were re-moved at 7, 14, 21, and 28 days after the operation. The other 35 rats were divided into 7 groups (6 h, 12 h, 24 h, 48 h, 72 h after ICH, no ICH and normal ). The activation of synapotophysin was measured by immunohis-tochemical techniques. The rats in the trial group begin cage-running exercise at 72 h after the operation. The oth-ers lived in standard cages. Results Synapotophysin-positive cells were seen in the tissues around the hematomas and in the cortex. There was no synapotophysin expression in the centres of the hematomas. The number of synapo-tophysin-positive cells in ICH group was significantly less than in the sham-operation and normal groups from 6 h to 24 h after the operation. There was then a gradual increase again from 48 h onwards until the 28th day after the op-eration. The trial group had the largest increases in expression. There was a significant difference compared with the control group. Compared with the sham operated group, the trial and control groups showed very significant differences. Conclusion The results suggest that synapotophysin is involved in neuron plasticity after ICH, but exercise training (cage-running) can accelerate the expression of synapotophysin, thereby improving functional re-covery.

Aim To investigate the effect of simvastatin ( Sim ) on endogenous antioxidant system after acute myocardial infarction ( AMI ) and its potential mecha-nisms. Methods The acute myocardial infarction ( AMI ) rat models were made by ligation left anterior descending of coronary artery. Then the successful models were randomly divided into myocardial infarc-tion group ( MI group) and simvastatin group ( Sim,20 mg·kg-1·d-1), another group without ligation left anterior descending of coronary artery served as sham group(Sham group). The Sim group was administered simvastatin by gavage for 7 days. MI group and Sham group received saline. Hemodynamic parameters, lipid levels, troponinI ( c-TnI ) and lactate dehydrogenase ( LDH) concentrations were examined after 7days, and the levels of superoxide dismutase ( SOD) and glutathi-one peroxidase ( GP) of myocardial antioxidant system were detected by ELISA. The expression of cardiac p-Akt and p-eNOS protein were detected by Western blot. Results Acute myocardial infarction significant-ly lowered cardiac hemodynamic parameters, increased serum c-TnI and LDH levels, lowered levels of SOD and GP, and lowered the expression of p-Akt and p-eNOS protein. However, Sim could effectively prevent the deterioration of cardiac function, reduce serum c-TnI and LDH levels, increase levels of SOD and GP, and increase p-Akt and p-eNOS protein expression. Conclusion Early using Sim can effectively improve heart function after acute myocardial infarction, acti-vate myocardial antioxidant system, and reduce myocar-dial necrosis, which may be related to increasing the expression of p-Akt and p-eNOS.
