1.Clinical significance and expression of Skp2, p27 and p21 in Ovarian Serous Cystadenocarcinomas

ObjectiveTo investigate the correlation of Skp2,p27kiP1 and p21WAF1 expression with the clinicopathological features of ovarian serous cystadenocarcinomas.Methods Expressions of Skp2 ,p27kiP1 and p21WAF1 were examined by immunohistochemical staining in 124 epithelial ovarian tumors (25 serous cystadenomas, 19 borderline serous cystadenomas, and 80 serous cystadenocarcinomas) Results(1) The expression of Skp2 in serous cystadenocarcinomas (47.5%)was significantly higher than that in borderline serous cystadenomas (0%)and serous cystadenomas (0%)(P < 0.001) .The p27kiP1 expression in serous cystadenocarcinomas (35.0%) was significantly lower than that in borderline serous cystadenomas(73.7%)and serous cystadenomas (80.0%) .The p21WAF1 staining frequency in serous cystadenocarcinomas (38.8%)was significantly lower than in borderline serous cystadenomas (73.7%)and serous cystadenomas (80.0%) .(2) The Skp2 protein expression in serous cystadenocarcinomas was positively correlated with clinicopathological stage,histological differentiation degree and lymph node metastasis of the tumors.The p27kiP1, p21WAF1 protein expression in serous cystadenocarcinomas was reversely correlated with clinicopathological stage and histological differentiation degree of the tumors(Ps < 0.05) .(3) The Skp2 protein expression in serous cystadenocarcinomas was reversely correlated with that of p27kiP1 , p21WAF1.Conclusion The Skp2 protein expression in serous cystadenocarcinomas was increased and positively correlated with the clinicopathological features of ovarian serous cystadenocarcinomas.Skp2 protein expression was reversely correlated with p27kip1 ,p21WAF1.Skp2 protein expression may play an important role in the development and progression of serous cystadenocarcinomas.
2.Study on cognitive of newborns′pain in medical staff of department of neonatal intensive care unit and influencing factors

Objective To learn the cognitive status for newborns medical pain in medical workers of neonatal intensive care unit of children′s hospitals and provide clinical basis for optimizing neonatal pain management. Methods Totally 320 medical staff in neonatal intensive care units of six children′s hospital in Wuhan, Shenzhen, Shanghai, Nanjing, Zhejiang, Chongqing were investigated using medical pain management survey questionnaires by means of convenience sampling method. Results The knowledge on neonatal pain were at quite high level, accuracy rate was (80.5 ± 18.1)%;but on pain feel, pain relief rate and anodyne-use, the accuracy rate was relatively low; chest drainage, lumbar puncture and PICC placement were generally considered to cause severe pain with scores of 8.03±2.20, 7.17±2.17, 7.09 ± 2.00, respectively. Medical staff with different degrees, having children or not, different titles and with different years of working at NICU had impact on neonatal pain assessment on heel prick and venipuncture among full-term newborns. The difference was statistically significant (χ2=6.208-23.314, P<0.05). Conclusions Neonatal pain management is gradually emphasized by clinical medical staff, but there exist a certain bias on the perception and assessment of pain. So, it needs to strengthen pain-related knowledge training on health care for newborns to prevent and reduce neonatal pain, improving neonatal pain management.
3.Effects of amantadine and biphenyl dimethyl dicarboxylate on hepatitis B virus in hepatitis B virus replication mice.

This study sought to investigate the in vivo antiviral effect of amantadine (AM) and biphenyl dimethyl dicarboxylate (DDB) on hepatitis B virus (HBV) in HBV replication mice. HBV replication-competent plasmid was transferred into male BALB/c mice by using hydrodynamics-based in vivo transfection procedure to develop HBV replication mouse model. The model mice were matched by body weigh, age and serum levels of hepatitis B e antigen (HBeAg) and were divided into four groups: AM group, DDB group, AM+DDB group and NS group, with the last one as control, and the mice of each group were administered corresponding agent orally twice a day, in a medication course lasting 3 d. On the third day, the mice were sacrificed 4-6 h after the last oral intake. HBV DNA replication intermediates in liver were analyzed by Southern blot hybridization. The serum hepatitis B surface antigen (HBsAg) and HBeAg were detected by enzyme linked immunosorbent assay (ELISA). Compared to the animals in the control group, HBV DNA replication intermediates in liver and HBsAg and HBeAg in serum from the AM and AM plus DDB group of mice decreased, and there was no difference between these two groups of mice. The levels of HBV DNA intermediate from liver and the serum HBsAg and HBeAg between the control and DDB group, however, were not obviously different. In conclusion, the inhibition effect of AM on HBV was detected, but treatment with DDB for 3 days did not influence the viral replication and expression of HBV in the HBV replication mice.

4.Academic level of papers on systematic reviews and meta-analysis published in Chinese Journal of Pediatrics and methodology they used

Objective To assess the academic level of papers on systematic reviews and meta-analysis published in Chinese Journal of Pediatricsand methodology they used.Methods Basic data were extracted from 13 papers on sys-tematic reviews and meta-analysis published in Chinese Journal of Pediatrics .The methodology they used was assessed according to the preferred reporting items for systematic reviews and meta-analysis (PRISMA) and ANSTAR Scale and analyzed using the RevMan5.0.Results The PRISMA score was 14-23.5 (mean 20.0±3.11) and the AMSTAR score was 3-7.5 (mean 6.04±1.38) for the methodology used in papers on systematic reviews and meta-analysis published in Chinese Journal of Pediatrics .Conclusion The methodology used in papers on systematic reviews and meta-analy-sis published in Chinese Journal of Pediatrics is not quite valid and should thus be improved .
5.Determination of Valproate Acid in Human Plasma by UFLC-MS/MS

To establish a UFLC-MS/MS method for the determination of valproate acid in human plasma. Methods:The sample was precipitated by methanol. The analysis of valproate acid and diclofenac sodium ( internal standard) was carried out on a Shim-pack XR-ODS II C18 column (2. 0 × 75 mm,2. 2 μm). Gradient elution was adopted with acetonitrile and water (containing 5 mmol·L-1 ammonium acetate) at a flow rate of 0. 3 ml·min-1. The detection was performed with multiple reactions monitoring (MRM) using nega-tive electrospray ionization (ESI) at m/z 142. 8→142. 8 for valproate acid and m/z 294. 0→249. 8 for diclofenac sodium. Results: The calibration curve of valproate acid was linear over the range of 8.4-168.0 μg·ml-1(r=0.997 5). Inter- and intra-day RSDs were less than 15%, and the analysis was proven to be stable. Totally 30 samples determined by EMIT were assayed by the method. And the results of the two methods analyzed by independent t-test showed no statistical significance. Conclusion:The method is rapid,sensitive and spe-cific,which can be applied in the determination of valproate acid in human plasma.
6.Application of service learning in practical teaching of traditional Chinese medicine nursing

Objective To Understand the learning effect of service learning mode in traditional Chinese medicine(TCM)nursing teaching. Methods By using convenience sampling method, 40 grade 2014 nursing students who volunteered to participate in TCM nursing learning and practice teaching were selected as research subjects from June 2016 to June 2017. Apply service learning mode to practice teaching in traditional TCM nursing teaching. Then, compared the examination results before and after participating practice teaching and investigated the satisfaction of the nursing students to practical courses. Results The examination results of the 40 nursing students after participating TCM nursing traditional practice teaching was (92.55±1.99), which was higher than those (90.28±2.75) before. The difference was statistically significant (t=4.246,P<0.01). 80.0% of the nursing undergraduates were satisfied with the content of TCM service practice teaching. The satisfaction with self-reflection was 47.5%. Conclusions Introducing the concept of service learning can enrich the TCM nursing practice teaching mode. Nursing students are willing to accept the course and it can promote their overall quality and stimulate their interest in learning. Teachers should pay attention to improving students' self-reflection and teamwork spirit in the future courses.
7.Effects of 3D printing technology combined with CBL teaching method in nursing teaching of adolescent idiopathic scoliosis

Objective:To explore the effect of three-dimensional (3D) printing technology combined with case-based learning (CBL) teaching method in clinical nursing teaching of adolescent idiopathic scoliosis.Methods:The convenience sampling was used to select 48 nursing students who had clinical practice in the Orthopedic Spine Ward of the Fourth Medical Center of Chinese People's Liberation Army General Hospital from January to November 2020 as the control group. A total of 50 nursing students in the same ward were selected as the experimental group from December 2020 to October 2021. The control group adopted the unified and standardized traditional teaching method of the Nursing Department, and the experimental group applied 3D printing technology combined with CBL teaching method on the basis of traditional teaching. After the internship, the two groups of intern nursing students were evaluated on their graduation examination results, teaching satisfaction and learning interest, and the knowledge assessment was carried out six months later.Results:The scores of the students in the experimental group in the academic theory, operation examination, teaching satisfaction and learning interest, as well as the scores of the related knowledge after half a year were higher than those in the control group, and the differences were statistically significant ( P<0.05) . Conclusions:The combination of 3D printing technology and CBL teaching method can help intern nursing students understand and master relevant teaching content, improve learning interest and teaching satisfaction, and help form long-term memory in the clinical nursing teaching practice of adolescent idiopathic scoliosis.

Objective:
To evaluate the outcomes and prognostic factors of patients with refractory and relapsed acute myeloid leukemia (AML) who received allogeneic hematopoietic stem cell transplantation (allo-HSCT) .
Methods:
The overall survival (OS) , disease free survival (DFS) , acute and chronic graft-versus-host disease (GVHD) , relapse rate (RR) , transplantation related mortality (TRM) and their related risk factors were analyzed retrospectively.
Results:
All the patients (median age 35 years, range 6 to 58) received myeloablative conditioning regimens. Objective:
To compare eficacy and safety of porcine antihuman lymphocyte immunoglobulin (pALG) and rabbit antithymocyte immunoglobulin (rATG) as a part of alternative donor allogeneic hematopoietic stem cell transplantation (AD allo-HSCT) for severe aplastic anemia (SAA).
Methods:
The clinical data of 46 SAA patients received AD allo-HSCT from January 2006 to November 2016 were retrospectively analyzed. The cohort of patients were divided into two groups based on rATG or pALG as a part of conditioning regimen to compare implantation rate, transplantation related complications and outcome.
Results:

Objective:
To explore the effectiveness of a novel GVHD prophylaxis regimen containing low-dose anti-T lymphocyte globulin (ATG) in patients undergoing peripheral blood stem cell transplantation (PBSCT) from HLA-matched sibling donors (MSD) given both the patients and donors were aged over forty years old.
Methods:
From March 2013 to April 2017, 98 patients with hematologic malignancies were enrolled in the study. Standard GVHD prophylaxis consisted of the administration of cyclosporine A/tacrolimus and a short course of methotrexate. In ATG group, 43 patients received low-dose rabbit ATG (Sanofi, 1.5 mg/kg per day for 3 consecutive days) before PBSCT. A retrospective matched-pair analysis was performed and 55 matched controls were available. The therapeutic process and clinical outcome were retrospectively analyzed.
