1.Effects and mechanisms of exogenous nesfatin-1 injection in the central nucleus of the amygdale on exploratory behavior in rats

Objective To explore the effects and mechanisms of nesfatin-1 injection in the central nucleus of the amygdale(CeA) on exploratory behavior in rats.Methods Fifty rats were divided into 5 groups randomly : nesfatin-1 low dose group, nesfatin-1 high dose group, SHU9119 group, SHU9119+nesfatin-1 low dose group and control group.Drugs were administrated via CeA to examine behavioral changes of rats by elevated plus maze model test and nesfatin-1 mRNA expression in CeA of anxiety rats (10 rats).Results Compared with the control group,anxiety behaviors of rats in nesfatin-1 low dose group and nesfatin-1 high dose group were improved remarkably,showing that the open arm entries were significantly reduced (OE, t=4.16-12.87, P<0.01), open arm entry proportion was decreased remarkably (OE%, t=2.39-4.39, P<0.01-0.05), and time proportion in open arm was decreased significantly (OT%, t=5.43-20.55, P<0.01),which presented dose dependent.Compared with the control group, anxiety degree of rats in nesfatin-1 low dose group and nesfatin-1 high dose group significantly enhanced, showing that the rats head dipping reduced obviously (HD, t=6.97-16.73, P<0.01) ,while rearing was increased significantly (RE, t=6.60-13.25, P<0.01) ,which was presented dose dependent.Melanocortin receptor antagonist SHU9119 can partly eliminate nesfatin-1 induce-anxiety (t =2.11-3.08, P< 0.01-0.05).Real-time PCR results showed that nesfatin-1 mRNA expression of anxiety model rats was significantly increased (t =3.40-5.58, P<0.05).Conclusion Amygdala nesfatin-1 can increase anxiety and fear of rats,which may be associated with melanocortin systems.
2.Nursing of digestive tract fistula after orthotopic liver transplantation

Objective To summarize nursing experience of digestive tract fistula after orthotopic liver transplantation (OLT). Methods From January 2000 to December 2010,1173 patients received liver transplantation,among whom 61 recipients got digestive tract fistula during early stage after operation.The clinical data and major nursing measures of 61 patients were studied retrospectively. Results The incidence rate of digestive tract fistula after OLT was 5.20%.The incidence rate of bile leakage,gastric fistula,duodenal fistula,jejunal fistula,ileal fistula and transverse colon fistula were 3.90%,0.42%,0.08%,0.34%,0.08% and 0.34%,respectively.The onset time of digestive tract fistula was from 3 to 24 days post transplantation,and atypical symptoms such as fever,abdominal pain,abdominal distention,weakened enterocinesia and even septic shock were presented.And the rising or descending of white blood cells counts,rising of level of transaminase and total bilirubin in laboratory examination were simultaneously presented.Four patients died of bile leakage,and other seven patients died of intestinal fistula throughout conservative or operative treatment.The rest of 50 were discharged healthily. Conclusions The morbidity of digestive tract fistula after OLT is low,but its mortality rate is high.It is necessary to closely observe patients' condition and confirm diagnosis in early stage,in order to promote the healing of digestive tract fistula after OLT.At the same time,psychological,basic and dietary nursing should be given in order to enhance the survival rate and quality of life of patients.
3.Intervention of NGAL and HO-1 in valve replacement surgery-induced acute kidney injury.

OBJECTIVE:
To determine the pathological mechanism and prevent heart-renal syndrome after heart valve replacement surgery.
METHODS:
A total of 46 patients were admitted for selective valve replacement, and divide into 3 groups randomly: a control group (Con, n=16), a remote ischemic perconditioning (RIPerC) group (n=15) and a remote ischemic postconditioning (RIPostC) group (n=15). The serum creatinine (SCr), blood urea nitrogen (BUN), serum heme oxygennase-1 (HO-1), serum iron and urinary neutrophil gelatinase associated lipocalin (NGAL) level in the 3 groups were compared preoperatively and 6, 12, 24, 48 h after aortic cross-release.
RESULTS:
Compared with the preoperative level, the SCr, BUN, urinary NGAL, serum iron (6 and 12 h) and serum HO-1 values were significantly increased after the heart valve replacement surgery in the control patients, RIPreC and RIPostC groups (P<0.05). Compared with the control group, the serum HO-1 was significantly increased at 6, 12, 24, 48 h after the heart valve replacement surgery in both the RIPerC and RIPostC groups (P<0.05); the SCr, BUN, urinary NGAL and serum iron values were decreased at 6, 12, 24, 48 h after the heart valve replacement surgery in both the RIPerC and RIPostC groups (P>0.05).
CONCLUSION
Abnormal change in urinary NGAL, serum iron and HO-1 can be used as early warning indicators of acute kidney injury when cardio-renal syndrome occurrs among patients under heart valve replacement surgery. Remote ischemic conditioning plays a preventive role in the occurrence of cardio-renal syndrome and renal protection.

4.Effect of Compound Shi-Chang-Pu Granula on Behavior, Structure of Hippocampus in Epileptic Mice Induced by Pentylenetetrazol

This study was aimed to observe the effect of Compound Shi-Chang-Pu (CSCP) granula on behavior, structure changes of hippocampus with epileptic seizure of mice induced by pentyenetetrazole (PTZ). Sixty Kunming mice, which were half male and half female, were randomly divided into 6 groups, which were the blank control group, animal model, phenytoin (PHT) group, high-dose CSCP (4 000 mg·kg-1) group, middle-dose CSCP (2 000 mg·kg-1) group and low-dose CSCP (1 000 mg·kg-1) groups. The blank group and model group were given intragas-tric administration the same volume of saline, the others animals treated with continuous oral administration of drugs for 7 days. Intraperitoneal injection of PTZ (80 mg·kg-1) was given 1 h after the last medication to establish the a-cute epilepsy model (expect the blank control group). Observation was made on the influence of CSCP on incubation period, behavior degree and times of epileptic seizure among PTZ induced epilepsy mice, the structure changes of mice were detected by the brain tissue HE staining. The results showed that CSCP can improve the degree of seizure attack, prolong the incubation period of mild attack, and decrease Ⅳ and Ⅴ degree epileptic seizure, the PTZ in-duced epilepsy mice could damage the structure of hippocampus, reduce the number of cells, PHT and CSCP could ameliorate these changes. It was concluded that CSCP had certain inhibition on epileptic seizure mice induce by PTZ and ameliorate the damage of hippocampus. The antiepileptic mechanism still requires futher study.
5.Observation and nursing for patients complicated with pancreatic leakage after upper abdominal multiple organ transplantation

Objective To investigate the nursing points of pancreatic leakage after upper abdominal multiple organ transplantation. Methods A retrospective study was conducted on the nursing experience of two patients with end-stage liver disease and type 2 diabetes mellitus who were complicated with pancreatic leakage after upper abdominal multiple organ transplantation from March 2009 to July 2015. Results The blood glucose of these patients returned to normal level within 1 week after operation and insulin was discontinued. Pancreatic leakage was occurred in the two patients at 14 and 21 days after operation, respectively. They were both successfully discharged after active treatments and nursing cares including completely drainage, the application of drugs that inhibited the secretion of pancreatic enzymes and digestive tract glands, strengthening infection control, nutritional support and other conservative treatments. Conclusions It is the key to improve the recovery of pancreatic leakage after upper abdominal multiple organ transplantation with careful observation of abdominal signs and abdominal drainage tube, accurate use of somatostatin, nutritional support, maintenance of water and electrolyte balance, and psychological intervention.
6.The mechanism of interleukin-22 induced the secretion of vascular endothelial growth factor A in gastric cancer cell line AGS

Objective To investigate the mechanism of interleukin-22(IL-22) induced the secretion of vas-cular endothelial growth factor A(VEGF-A) in gastric cancer cell line AGS .Methods Gastric cancer cell line AGS were cultured in vitro ,and recombination cytokine IL-22 were added ,or signal pathway inhibitor were pre-incubated with AGS for 1 hour and then IL-22 were added ,the level of VEGF-A were detected by enzyme-linked immunosorbent assay .Results Compared with the unstimulated group ,the secretion of VEGF-A in IL-22-stimulated group was significantly increased ,the difference was statistically significant (P<0 .05) ,which was in a dose and time dependent manner .In addition ,IL-22-stimulated the secretion of VEGF-A by AGS was significantly decreased while pre-incubated by the signal transducers and activators of transcription 3(STAT3) inhibitor ,the difference was statistically significant (P<0 .05) ,but such effect was not observed while AGS were pre-incubated with the nuclear factor kappa B inhibitor ,c-Jun N-terminal kinase inhibitor ,mitogen-acti-vated protein kinase kinase 1/2 inhibitor and p38 mitogen-activated protein kinase inhibitor ,there was no sta-tistical significance(P>0 .05) .Conclusion IL-22 could induce the secretion of VEGF-A in gastric cancer cell line AGS via STAT3 signal pathway ,which may contribute to tumor progression .
7.Effects of the PEMFs of different intensity on BMD and biomechanical properties of rabbits' femur.

The effects of the pulsed electromagnetic fields (PEMFs) of different intensity on bone mineral density (BMD) and biomechanical properties of rabbits' femur had been studied. Twenty-seven female white big ear rabbits were randomly divided into three groups. The magnetic groups were fed in 15 Hz PEMFs, which pulse duration was set to be 5 ms (6 h x d(-1)), the magnetic intensity was 10 x 10(-4) T and the other was 20 x 10(-4) T. Control group were just fed in coils, and the instrument of PEMFs was powered off. After six weeks, by examine BMD and biomechanical properties of the rabbits' femur, the effects of these PEMFs were studied. Compared with control group, the values of BMD, maximum load and structural rigidity of magnetic group were significantly increased (P < 0.05). In addition, there was significant increase in values of BMD and structural rigidity in group 10 x 10(-4) T in comparison with group 20 x 10(-4) T (P < 0.05). PEMFs is effective in improving BMD and biomechanical properties. The experiment indicated that there was evident "window-effect" during the treatment by PEMFs. It is favorable to the treatment and prevention of osteoporosis.

8.Raman spectral characteristics of oral squamous cell carcinoma, epithelial dysplasia and normal mucosa

Objective To investigate the Raman spectral characteristics of oral squamous cell carcinoma,high-grade epithelial dysplasia and normal mucosa.Methods Fifty-six fresh samples of oral carcinoma,50 of high-grade epithelial dysplasia and 32 of normal mucosa were collected.The i-Raman spectrometer with an optical fiber tube was applied to acquire Raman spectrum.The diagnostic model established by principle component analysis(PCA) and discriminant function analysis(DFA) was used to analyze and classify the spectra of different samples.Results There were significant differences among the Raman spectra of these samples.Compared with the spectra of normal mucosa,the spectra of oral carcinoma and dysplasia showed strong peaks which were contributed to nucleic acids,proteins and lipids.The diagnostic models established by PCA-DFA could successfully classify these Raman spectra of different samples with a high accuracy of 96.4% (133/138).The model was evaluated by 'Leave one out' crossvalidation and reached a high accuracy of 92.8%(128/138).Conclusions The proliferation and metabolism of oral squamous cell carcinoma and epithelial high-grade dysplasia are more active than normal mucosa.The diagnostic models established by PCA-DFA can classify these Raman spectra of different samples with a high accuracy.
9.Efficacy and safety of domestic disposable pressure sensor:a prospective,self-parallel controlled double-center clinical study

Objective To evaluate the effectiveness and safety of domestically manufactured disposable pressure monitoring sensor in clinical application.Methods The self-parallel control was adopted.A total of 128 patients undergoing surgery for cardiovascular related disease requiring both invasive arterial blood pres-sure and central venous pressure simultaneously monitoring in the two collaborating hospitals of Chongqing Kanghua Zhonglian Cardiovascular Disease Hospital and the Second Affiliated Hospital of Army Medical Uni-versity from January to March 2021 were selected as the study subjects.The two kinds of disposable invasive pressure sensors were simultaneously utilized for continuous monitoring and recording invasive arterial blood pressure and central venous pressure prior to anesthesia induction(T1),at 1 min after the last intravenous an-esthetic drug injection(T2)and at tracheal intubation completion(T3).Additionally,the performance,safety indicators of the product,and success rate of arterial blood collection were recorded.Results At different time points T1,T2 and T3,the the consistency rates of systolic blood pressure measured by two sensors were both 100.0%.The consistency rate of diastolic blood pressure at T1 measured by the two kinds of sensors was 99.2%,which at T2 and T3 was 100.0%.The consistency rates of central venous pressure at 3 time points by the two kinds of sensors were both 100.0%.The correlation coefficient showed that the two kinds of sensors kept the high consistency in measuring systolic blood pressure,diastolic blood pressure and central venous pressure.The concordance rates of arterial blood gas analysis indexes pH,PO2 and PCO2 reached 100.0%,94.5%and 99.2%respectively.Conclusion The effectiveness,accuracy and safety of domestically produced dis-posable invasive blood pressure sensor meet clinical needs.
10.Investigation of present situation about pulmonary rehabilitation among COPD patients in community and the analysis of influence factors

Objective To study the present situation and influence factors of pulmonary rehabilitation among COPD patients in community.Methods The present situation of pulmonary rehabilitation was investigated using questionnaire in 201 COPD patients,including one community in Guangzhou and three communities in Qingyuan,and the influencing factors were analyzed by Logistic regression.Results Total of 201 COPD patients,20.4% COPD patients could insist on oxygen therapy,just one patient's oxygen time more than 15 hours;40.8% COPD patients would increase the oxygen flow if exacerbation;17.9% COPD patients could insist on respiratory exercising,and 75.5% COPD patients took exercise in wrong way.50.7% COPD patients took exercise in their life.3-4 times in hospital with a high rate of home oxygen therapy.Farmers and the course more than ten years with a high rate of exercising.Conclusions The cognition of COPD patients about pulmonary rehabilitation is poor,and also the compliance of pulmonary rehabilitation exercise is bad.The medical staff should strengthen the education of pulmonary rehabilitation of COPI patients in community,and pay attention to patients' hospitalization,occupation,and course of disease.
