1.Changes of gene methylation profile in malignant transformation of immortalized human bronchial epithelial cell line induced by alpha-particle irradiation

Objective To identify the changes of DNA methylation profile in the process of malignant transformation of BEP2D cell induced by α particles.Methods The genomic DNAs were isolated from the malignant transformation BERP35T4 cells and immortalized human bronchial epithelial cell line BEP2D.Genomic DNAs were digested by MseI and ligated of PCR linkers.Methylated DNAs were digested by BstUI and amplified by PCR.The methylated DNA probes were prepared by labeling with Cy3 and Cy5 fluorescence dyes individually and hybridized to the methylation CpG-Island microarray.The hybridization results were scanned and analyzed.Intensity values were quality controlled and normalized.The normalized data were used to identify the differentially expressed genes based on a 1.5 fold difference of the expression level.Results There were 16 genes which showed changes of methylation level in malignant transformation BERP35T4 cells, 9 of them were hypermethylation and 7 were hypomethylation.These genes were including the SKIP gene, PPP3CC gene, MAP2K6 gene, KIR2DL1 gene, KIR2DL4 gene, KIR3DP1 gene, ZNF493 gene, ZNF100 gene, NKX2-5 gene, TFAP2D gene, DR1 gene, KCNJ16 gene, CCDC18 gene, FNBP1L gene, IRX4 gene, EPB41L3 gene, TCP10 gene and so on.Conclusions The DNA methylation might have effects on ionizing radiation drived tumorigenesis.

Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis is a group of systemic small vasculitis characterized by the detection of ANCA in serum. Bactericidal permeability enhancing protein (BPI) is one of the target antigens of ANCA. BPI-ANCA-associated vasculitis is not common clinically, and the combination of bronchiectasis is not accidental. The paper reported a case of BPI-ANCA-associated vasculitis with renal damage combined with bronchiectasis. We reviewed relevant literature to explore the characteristics of BPI-ANCA-associated vasculitis and the correlation between bronchiectasis and ANCA-associated vasculitis, so as to improve the clinician's understanding on this disease.
3.Operation of ehealth literacy in coronary disease patients

Objective To understand the ehealth literacy of coronary disease patients and to explore the problems encountered when searching for disease related health task by computer so as to provide a reference for constructing the ehealth literacy intervention for coronary disease patients. Methods From June 2018 to December 2018, this study selected 20 coronary disease inpatients at a class Ⅲ grade A hospital in Hangzhou by purposive sampling to carry out operational experiment. Subjects operated 6 experimental tasks related to coronary network health designed by researchers in computers. In the experiment, subjects were required to use the method of think-aloud. At the same time, screen video experts recorded the screen activity track, image and sound produced when subjects operated tasks to collected subjects' experimental data. Moreover, two researchers recorded the completion, expression and time of subjects when operating 6 tasks. Colaizzi 7-step analysis was used to summary and analyze the experimental data. Results Problems encountered when coronary disease patients searched for disease related health task by computer included four themes, operating computers and browsers, navigation and location on the Web, using search strategy as well as evaluating the information correlation and reliability. Conclusions Health professionals should pay attention to the basic computer skills of patients,health information search strategy and quality evaluation ability of network health information when providing ehealth literacy training for coronary disease patients to improve the ehealth literacy of coronary disease patients.
