Near-infrared (NIR) diffuse reflectance spectroscopy was used to analyze the impact of multi-class particle size of Chinese material medica (CMM) based on the spectral characteristics in overtone and combination region. Several types of CMM (60, 80, 100,120 mesh) were subjected to NIR spectra analysis. Spectral reproducibility was examined after sample repackage. The result showed that the effects of particle size on the NIR spectra were different according to different bands, in the combination region and first combination-overtone region. Spectroscopy intensity was proportional to the particle size and influence of particle size was greater as the wavelength increased. While in the second combination-overtone region, it was inversely proportional to particle size. To the sampling loading error, the result indicated that when the mesh number was larger than 60 mesh, the error was small. The appropriate particle size was clarified to guarantee the accuracy and reliability of NIR diffuse reflectance spectroscopy in CMM.

Objective To establish a key technological system for spider fibroin gene code tandem connection , vector construction , prokaryotic expression and purification using genetic engineering in order to achieve MaSp 1 heterologous ex-pression in Escherichia coli and its separation and purification .Methods Isocaudarner ligation method was used to connect synthetic spider fibroin gene monomer code in tandem , and a recombinant clone concatemer was obtained .The identified recombinant clones were connected with prokaryotic expression vector pET 28a(+), and then transformed into E.coli BL21 (DE3).After being induced by IPTG for 6 hours, the expression product was identified by SDS-PAGE and Western blot-ting.Engineering bacteria were fermented in high density , and the obtained protein was purified through ammonium sulfate fractionation.Results and Conclusion The expression plasmids of MaSp1concatemers were successfully constructed , and the induced expression genetic engineering MaSp 1 protein was of the expected relative molecular mass .In addition, the pu-rity of the purified protein was above 80%.This study has developed crucial technologies for mass production of genetic en-gineering spider silk proteins .

Near-infrared (NIR) was used as rapid analysis method to identify the sulfur fumigated Puerariae Lobatae Radix. NIR spectra of the cross-sectional and longitudinal selection of samples were acquired. Principal component analysis was conducted. The samples were randomly selected. The different pretreatment methods were compared. Discriminant models were established for every type of spectra to calculate the recognition rate. The orthogonal test and nonparametric test were used to test data normality. The result showed that absorbance values of different sections were different due to the different structure, and the raw spectra were analyzed by PCA method. The result founded that the cumulative contribution rate was arrived at 99.2% while the PC numbers were arrived at 3. The pretreatment method based on the MSC + 1D + Savitzky-Golay was the best to establish the model. For the 50 models constructed with cross-section and longitudinal spectra and total spectra, the recognition rate were (94.4 ± 0.66)%, (94.4 ± 0.66)%, (95.3 ± 0.65)%, respectively, and no difference was observed. The NIR method could be used to identify the sulfur fumigated Puerariae Lobatae Radix.

Objective To explore the clinical application of the Conners Parent Symptom Questionnaire (PSQ) and the Teacher Rating Scale (TRS) in 3~7 years old hearing-impaired children. Methods 30 hearing-impaired children were tested by PSQ and TRS. Results The detection rate of problem behavior is 53.33% in PSQ and 26.67% in TRS. There was no significant difference in the PSQ and TRS scores between different sex and age (P>0.05). There was no significant correlation between PSQ and TRS (P>0.05). Conclusion The Conners Rating scale is suitable to evaluate the behavior problem in 3~7 years old hearing-impaired children.

Experimental investigations were conducted on the process of combustion and explosion vent in a 200 mm (diameter) x 400 mm (length) vertical cylindrical vessel. When CH(4)-air mixture gases were used and the vent diameter was 55 mm, conditions of Phi (equivalent ratio)=0.8, Phi=1.0 and Phi=1.3 and two ignition positions (at the cylinder center and bottom) were selected. The venting processes and the correlated factors are discussed in this paper.

This study was aimed to investigate the effect of chemokine-like factor superfamily 8 (CKLFSF8) on proliferation and expression of epidermal growth factor receptor (EGFR) of HL-60 cells. Expression of CKLFSF8 mRNA on HL-60 cell line was assayed by RT-PCR; the target gene was transfected into the cells by lipid vector, cell proliferation was determined by MTT assay, while expression of EGFR in HL-60 was determined by immunocytochemical technique. The results indicated that expression of CKLFSF8 existed in HL-60 cells. After transfection, cell proliferation was inhibited (P < 0.05) and the expression of EGFR in HL-60 cells was also discovered to be inhibited (P < 0.05). It is concluded that the proliferation and expression of EGFR in HL-60 cells can be inhibited by transfection of CKLFSF8. The novel chemokine may provide a new approach in the treatment of leukemia.

Objective To observe the effects of remote ischaemic preconditioning on myocardium in heart operation under thoracoscope.Methods One hundred and twenty patients (72 males,48 females,aged 41-69 years,weighing 49-68 kg,falling into ASA physical status Ⅱ or Ⅲ) who received heart operation under thoracoscope were randomly divided into 2 groups (n =60 each):remote ischaemic preconditioning group (group RIPC) and control group (group C).Comparing the pH value of arterial blood around the RIPC;observing the variation of LVEF and CI at before the operation,before leaving room and 24 h after operation,and the changes of myocardial enzymes (including cTnI,CK-MB and LDH) at basic level and 6,24 and 48 h after RIPC;comparing the basic state of the two group during the perioperation.Results Compared with group C,the pH value of artery blood was lower in group RIPC after RIPC(P＜0.01).CI in RIPC group was higher than that in group C at 24 h after operation(P＜0.05);there was no statistic difference in LDH between the two groups.The value of cTnI in RIPC group was lower than that in group C at 6 and 24 h after operation (P＜0.05 or P＜0.01).The value of CK-MB in group RIPC was lower than that in group C at 6,24 and 48 h after operation (P＜0.05);there was no statistic difference in.LDH between the two groups.Basic conditions during and after the surgery in two groups had no statistic difference.Conclusion Remote ischaemic preconditioning can protect to myocardium during the heart operation under thoracoscope.

The aim of this study was to investigate whether exosomes derived from K562 cells and human monocyte-derived dendritic cells (DCs) transfected with total RNA of K562 cells are capable of inducing antigen-specific cytotoxic T lymphocytes (CTL) responses in vitro. DCs were generated from peripheral blood mononuclear cells (PBMNC) of healthy volunteers in the presence of GM-CSF and IL-4, and then were transfected with K562 RNA by using DOTAP lipofection. Exosomes was extracted from the supernatant of DCs and K562 cells. The T cell were activated to be tumor specific CTL after DCs and exosomes were co-cultured with autologous T cells derived from healthy volunteers' PBMNC. The effect of CTL on K562 cells was detected by MTT assay. The results showed that treatment of T cells with exosomes derived from K562 cells or DCs transfected with total RNA of K562 cells could significantly promote their killing ability on K562 cells as compared with untreated T cells (P < 0.05). The killing ability of T cells treated with exosomes on K562 cells was stronger than on HL-60 cells (P < 0.05). It is concluded that the specific CTL immune response to leukemia cells can be induced by exosomes derived from K562 cells.

OBJECTIVETo observe different effects of moxibustion on extracellular potassium ion in acupoint under physiological and pathological status and provide experimental evidence for exploring action mechanism of moxibustion on acupoint local.

METHODSForty female SD rats were randomly divided into a blank group, a blank-moxibustion group, a model group and a model-moxibustion group, 10 cases in each one. The complete Freund's adjuvant(CFA) was adopted to establish model of adjuvant arthritis (AA) in the model group and model-moxibustion group. No treatment was given in the blank group and model group while moxibustion was applied at "Zusan-li" (ST 36) for 30 min in the blank-moxibustion group and model-moxibustion group. The tissue fluid in "Zusanli" (ST 36) was collected with microdialysis and real-time analyzed by electrolytic analyzer. The change of concentration of potassium ion in "Zusanli" (ST 36) was observed.

RESULTS(1) Under physiological status, the concentration of extracellular potassium ion in the blank group was not changed within 150 min (P > 0.05); before the moxibustion, the concentration of extracellular potassium ion in the blank-moxibustion group was (1.21 +/- 0.31) mmol/L, and after treatment it was gradually increased and reached its peak at (2.38 +/- 0.42) mmol/L after 60 min (P < 0.05), then it was reduced. 150 min after the treatment, concentration of potassium ion was slightly higher than that before moxibustion as well as that in the blank group. The concentration in the blank-moxibustion group at 60 min was statistically significant compared with that in the blank group (P < 0.05). (2) Under pathological status, the concentration of extracellular potassium ion in the model group was not changed within 150 min, differences of which at each time point was not statistically significant (all P > 0.05). Before the moxibustion, the concentration of extracellular potassium ion was (1.09 +/- 0.12) mmol/L in the model-moxibustion group, and it was immediately increased to (1.96 +/- 0.18) mmol/L after moxibustion. 60 min and 90 min after the moxibustion, it still maintained a higher level, which was (1.87 +/- 0.29) mmol/L and (1.59 +/- 0.16) mmol/L respectively (both P < 0.05). The differences of each time point after moxibustion in the model-moxibustion group were statistically significant compared with those in the model group (all P < 0.05).

CONCLUSIONThe moxibustion could increase the concentration of potassium ion in rat's acupoint local under physiological status but time of effect is short; with moxibustion at "Zusanli" (ST 36) under pathological status, the concentration of local potassium ion is obviously increased and maintains for a long time.

Objective To observe the effects of remote ischemic preconditioning(RIPC) on Na+-K+-ATP enzyme and Ca2+-Mg2+-ATP enzyme in thoracoscopic heart operation.Methods One hundred and twenty patients with thoracoscopic heart operation were randomly divided into the thoracoscopic extracorporeal circulation control group (C)and RIPC plus thoracoscopic extracorporeal circulation group(RIPC).The acidity and alkalinity change of arterial blood before and after treatment was abserved in the RIPC group;the changes of myocardial enzymes spectrum,cTnI and oxidation indicators(SOD,MDA) were compared among different time periods,and preoperative and postoperative Na+-K+-ATP enzyme and Ca2+-Mg2+-ATP enzyme levels were also compared.Results Compared with the group C,the acidity and alkalinity of artery blood were lower after RIPC(P＜0.05);the levels of CK-MB and cTnI at postoperative 6,24 h in the RIPC group were lower than those in those in the group C(P＜0.05);the SOD activity was higher than that in the group C,while the MDA level was lower than that in the group C(P＜0.05);the postoperative Na+-K+-ATP enzyme and Ca2+-Mg2+-ATP enzyme levels were higher than those in the group C at different degrees.Conclusion RIPC can alleviate myocardial injury in the patients with thoracoscopic heart operation and this effect may be related with activation of corresponding ATP enzyme.
